| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 3 | Semen parameters in men recovered from COVID-19显示文摘The novel coronavirus disease(COVID-19)pandemic is emerging as a global health threat and shows a higher risk for men than women.Thus far,the studies on andrological consequences of COVID-19 are limited.To ascertain the consequences of COVID-19 on sperm parameters after recovery,we recruited 41 reproductive-aged male patients who had recovered from COVID-19,and analyzed their semen parameters and serum sex hormones at a median time of 56 days after hospital discharge.For longitudinal analysis,a second sampling was obtained from 22 of the 41 patients after a median time interval of 29 days from first sampling.Compared with controls who had not suffered from COVID-19,the total sperm count,sperm concentration,and percentages of motile and progressively motile spermatozoa in the patients were significantly lower at first sampling,while sperm vitality and morphology were not affected.The total sperm count,sperm concentration,and number of motile spermatozoa per ejaculate were significantly increased and the percentage of morphologically abnormal sperm was reduced at the second sampling compared with those at first in the 22 patients examined.Though there were higher prolactin and lower progesterone levels in patients at first sampling than those in controls,no significant alterations were detected for any sex hormones examined over time following COVID-19 recovery in the 22 patients.Although it should be interpreted carefully,these findings indicate an adverse but potentially reversible consequence of COVID-19 on sperm quality. | Tong-Hang Guo Mei-Ying Sang Shun Bai Hui Ma Yang-Yang Wan Xiao-Hua Jiang Yuan-Wei Zhang Bo Xu Hong Chen Xue-Ying Zheng Si-Hui Luo Xue-Feng Xie Chen-Jia Gong Jian-Ping Weng Qing-Hua Shi | 2021 | Asian Journal of Andrology2021,23,5: | 2 |
| 4 | Transport of intensity diffraction tomography with non-interferometric synthetic aperture for three-dimensional label-free microscopy显示文摘We present a new label-free three-dimensional(3D)microscopy technique,termed transport of intensity diffraction tomography with non-interferometric synthetic aperture(TIDT-NSA).Without resorting to interferometric detection,TIDT-NSA retrieves the 3D refractive index(RI)distribution of biological specimens from 3D intensity-only measurements at various illumination angles,allowing incoherent-diffraction-limited quantitative 3D phase-contrast imaging.The unique combination of z-scanning the sample with illumination angle diversity in TIDT-NSA provides strong defocus phase contrast and better optical sectioning capabilities suitable for high-resolution tomography of thick biological samples.Based on an off-the-shelf bright-field microscope with a programmable light-emitting-diode(LED)illumination source,TIDT-NSA achieves an imaging resolution of 206 nm laterally and 520 nm axially with a high-NA oil immersion objective.We validate the 3D RI tomographic imaging performance on various unlabeled fixed and live samples,including human breast cancer cell lines MCF-7,human hepatocyte carcinoma cell lines HepG2,mouse macrophage cell lines RAW 264.7,Caenorhabditis elegans(C.elegans),and live Henrietta Lacks(HeLa)cells.These results establish TIDT-NSA as a new non-interferometric approach to optical diffraction tomography and 3D label-free microscopy,permitting quantitative characterization of cell morphology and time-dependent subcellular changes for widespread biological and medical applications. | Jiaji Li Ning Zhou Jiasong Sun Shun Zhou Zhidong Bai Linpeng Lu Qian Chen Chao Zuo | 2022 | Light(Science & Applications)2022,11,10: | 2 |
| 5 | Association of sexually transmitted infection with semen quality in men from couples with primary and secondary infertility显示文摘This study aims to compare the prevalence of sexually transmitted infections(STIs)with semen quality in men from couples with primary and secondary infertility.Semen samples were collected from 133 men who requested fertility evaluation.Seminal tract infection with Ureaplasma spp.(UU),Mycoplasma hominis(MH),Mycoplasma genitalium(MG),Chlamydia trachomatis(CT),Neisseria gonorrhoeae(NG),and herpes simplex virus-2(HSV-2)was assessed by PCR-based diagnostic assays.Among all patients,the prevalence of STIs was higher in men from couples with primary infertility than that in men from couples with secondary infertility(39.7%vs 21.7%,P=0.03).The prevalence of UU was 28.8%and 13.3%in men from couples with primary and secondary infertility,respectively.Men from couples with primary infertility were more likely to be positive for UU than men from couples with secondary infertility(P=0.04).Regarding the UU subtype,the prevalence of Ureaplasma urealyticum(Uuu)and Ureaplasma parvum(Uup;including Uup1,Uup3,Uup6,and Uup14)did not differ between the two groups.No associations between the prevalence rates of MH,MG,and CT were found in men from either infertility group.A lower sperm concentration was associated with STI pathogen positivity in men with primary infertility according to the crude model(P=0.04).The crude and adjusted models showed that semen volume(both P=0.03)and semen leukocyte count(both P=0.02)were independently associated with secondary infertility.These findings suggest the importance of classifying the type of infertility during routine diagnosis of seminal tract infections. | Shun Bai Yuan Li Mei-Hong Hu Li Wu Li-Jun Shui Xiao-Han Wang Yi-Xun Liu Qiu-Ling Yue Li-Na Yu Kai-Qiang Fu Xian-Hong Tong Xue-Chun Hu Bo Xu | 2022 | Asian Journal of Andrology2022,24,3: | 2 |

| 17 | Tungsten combustion in impact initiated W-Al composite based on W(Al) super-saturated solid solution显示文摘Element W can effectively improve the density of energetic structural materials. However, W is an inert element and does not combust in air. To change the reaction characteristics of W, 60 at.% Al was introduced into W through mechanical alloying. XRD analysis shows that after 50 h of ball milling, the diffraction peak of Al completely disappears and W(Al60) super-saturated solid solution powder is obtained. Further observation by HAADF and HRTEM reveals that the W(Al60) super-saturated solid solution powder is a mixture of solid solution and amorphous phase. Based on the good thermal stability of W(Al60) alloy powder below 1000℃, W(Al60)-Al composite was synthesized by hot pressing process.Impact initiation experiments suggest that the W(Al60)-Al composite has excellent reaction characteristics, and multiple types of tungsten oxides are detected in the reaction products, showing that the modified W is combustible in air.
