| 1 | Cyclooxygenase-2 polymorphisms and the risk of esophageal adeno-or squamous cell carcinoma显示文摘AIM:To determine whether-1195 A→G and/or-765 G→C polymorphisms in Cyclooxygenase-2(COX-2 ) may have a risk modifying effect on the development of esophageal carcinoma in a Dutch Caucasian population.METHODS:Two study groups were recruited, 252 patients with esophageal carcinoma and 240 healthy controls, matched for race, age, gender and recruiting area.DNA was isolated from whole blood and used for genotyping.PCR products were digested with restriction enzymes and products were analyzed by agarose gel electrophoresis.Odds ratios(OR) and 95% confldence intervals(CI) were estimated.RESULTS:The distribution of the-1195 A→G polymorphism was signif icantly different in esophageal cancer patients compared to controls.The-1195 GG genotype resulted in a higher risk of developing esophageal adenocarcinoma(OR = 3.85, 95% CI:1.45-10.3) compared with the-1195 AA genotype as a reference.The-765 G→C genotype distribution was not different between the two groups.The GG/ GG haplotype was present more often in esophageal adenocarcinoma patients than in controls(OR = 3.45, 95% CI:1.24-9.58;with AG/AG as a reference).The same trends were observed in patients with squamous cell carcinomas, however, the results did not reach statistical signif icance.CONCLUSION:Presence of the COX-2-1195 GG genotype and of the GG/GG haplotype may result in a higher risk of developing esophageal carcinoma. | Jón O Kristinsson Paul van Westerveld Rene HM te Morsche Hennie MJ Roelofs T Wobbes Ben JM Witteman Adriaan CITL Tan Martijn GH van Oijen Jan BMJ Jansen Wilbert HM Peters | 2009 | World Journal of Gastroenterology2009,15,28: | 11 |
| 2 | Transcriptomic and metabolomic analyses of cucumber fruit peels reveal a developmental increase in terpenoid glycosides associated with age-related resistance to Phytophthora capsici显示文摘The oomycete,Phytophthora capsici,infects cucumber(Cucumis sativus L.)fruit.An age-related resistance(ARR)to this pathogen was previously observed in fruit of cultivar‘Vlaspik’and shown to be associated with the peel.Young fruits are highly susceptible,but develop resistance at~10–12 days post pollination(dpp).Peels from resistant(16 dpp)versus susceptible(8 dpp)age fruit are enriched with genes associated with defense,and methanolic extracts from resistant age peels inhibit pathogen growth.Here we compared developing fruits from‘Vlaspik’with those of‘Gy14’,a line that does not exhibit ARR.Transcriptomic analysis of peels of the two lines at 8 and 16 dpp identified 80 genes that were developmentally upregulated in resistant‘Vlaspik’16 dpp versus 8 dpp,but not in susceptible‘Gy14’at 16 dpp.A large number of these genes are annotated to be associated with defense and/or specialized metabolism,including four putative resistance(R)genes,and numerous genes involved in flavonoid and terpenoid synthesis and decoration.Untargeted metabolomic analysis was performed on extracts from 8 and 16 dpp‘Vlaspik’and‘Gy14’fruit peels using Ultra-Performance Liquid Chromatography and Quadrupole Time-of-Flight Mass Spectrometry.Multivariate analysis of the metabolomes identified 113 ions uniquely abundant in resistant‘Vlaspik’16 dpp peel extracts.The most abundant compounds in this group had relative mass defects consistent with terpenoid glycosides.Two of the three most abundant ions were annotated as glycosylated nor-terpenoid esters.Together,these analyses reveal potential mechanisms by which ARR to P.capsici may be conferred. | Ben N Mansfeld Marivi Colle Yunyan Kang A Daniel Jones Rebecca Grumet | 2017 | Horticulture Research2017,4,1: | 3 |
| 3 | Relevance of proteolysis and proteasome activation in fatty liver graft preservation: An Institut Georges Lopez-1 vs University of Wisconsin appraisal显示文摘AIM To compare liver proteolysis and proteasome activation in steatotic liver grafts conserved in University of Wisconsin(UW) and Institut Georges Lopez-1(IGL-1)solutions.METHODS Fatty liver grafts from male obese Zücker rats were conserved in UW and IGL-1 solutions for 24 h at 4 ℃and subjected to 'ex vivo ' normo-thermic perfusion(2 h; 37 ℃). Liver proteolysis in tissue specimens and perfusate was measured by reverse-phase high performance liquid chromatography. Total free amino acid release was correlated with the activation of the ubiquitin proteasome system(UPS: measured as chymotryptic-like activity and 20 S and 19 S proteasome), the prevention of liver injury(transaminases), mitochondrial injury(confocal microscopy) and inflammation markers(TNF 1 alpha, high mobility group box-1(HGMB-1) and PPAR gamma), and liver apoptosis(TUNEL assay, cytochrome c and caspase 3).RESULTS Profiles of free AA(alanine, proline, leucine, isoleucine, methionine, lysine, ornithine, and threonine, among others) were similar for tissue and reperfusion effluent. In all cases, the IGL-1 solution showed a significantly higher prevention of proteolysis than UW(P < 0.05) after cold ischemia reperfusion. Livers conserved in IGL-1 presented more effective prevention of ATP-breakdown and more inhibition of UPS activity(measured as chymotryptic-like activity). In addition, the prevention of liver proteolysis and UPS activation correlated with the prevention of liver injury(AST/ALT) and mitochondrial damage(revealed by confocal microscopy findings) as well as with the prevention of inflammatory markers(TNF1alpha and HMGB) after reperfusion. In addition, the liver grafts preserved in IGL-1 showed a significant decrease in liver apoptosis, as shown by TUNEL assay and the reduction of cytochrome c, caspase 3 and P62 levels.
