1MICROSTRUCTURAL MODEL OF GATORIZED WASPALOY IN THE ISOTHERMAL FORGING PROCESS显示文摘A microstructural model, which was to predict the evolution of microstructure of Gatorized Waspeloy in the isothermal forcing process, was developed in terms of dynamic recrystallization and grain growth. Three steps of experiment were conducted during developing the model: (1) .Specimens were compressed in M FS testing machine; (2) Dynamic recrystallization and grain growth were discussed; (3) Dynamic recrys- tallization model and grain growth mode were set up. 3Invasive mechanism and control strategy of Ageratina adenophora (Sprengel)显示文摘In order to ascertain the invasive mechanism and control strategy of the invasive Crofton weed, Ageratina adenophora, its ecological adaptability and population differentiation,the formation of single dominant population, displacement of native plants and sustainable management strategies were investigated. 4Telomere erosion is independent of microsatellite instability but related to loss of heterozygosity in gastric cancer显示文摘AIM To correlate the length of the telomere to microsatellite instability (MSI) and loss of heterozygosity (LOH) of APC, MCC and DCC genes in gastric carcinomas.METHODS Telomeric restriction fragment (TRF) length of gastric cancer was measured with Southern blot. LOH of APC, MCC and DCC genes, microsatellite instability (MSI) and frameshift mutation of hMSH6, TGF-βR Ⅱ and BAX genes were analyzed by PCR-based methods.RESULTS Sixty-eight cases of sporadic gastric carcinoma were studied for MSI using five microsatellite markers. MSI in at least one locus was detected in 17 (25%) of 68 tumors analyzed. Frameshift mutations of hMSH6, TGF-βR Ⅱ and BAX were detected in 2,6 and 3 of gastric carcinomas respectively showing high MSI (≥ 2 loci, n = 8), but none was found in those showing Iow MSI (only one locus, n = 9) or MSS (tumor lacking MSI or stable, n = 51). Thirty-five cases, including all high MSI and Iow MSl, were studied for TRF. The mean TRF length was not correlated with clinicopathological parameters.No association was observed between TRF length and MSI or frameshift mutation. On the contrary, LOH at the DCC locus was related to telomere shortening (P< 0.01). This tendency was also observed in APC and MCC genes,although there was no statistical significance.CONCLUSION The development of gastric cancer can arise through two different genetic pathways. In high MSI gastric cancers, defective mismatch repair allows mutations to accumulate and generate the high MSI phenotype. 5Inhibitive effect of cordyceps sinensis on experimental hepatic fibrosis and its possible mechanism显示文摘AIM: To investigate the inhibitive effect and its possible mechanism of Cordyceps Sinensis (CS) on CCl4-plus ethanolinduced hepatic fibrogenesis in experimental rats.METHODS: Rats were randomly allocated into a normal control group, a model control group and a CS group. The latter two groups were administered with CCl4 and ethanol solution at the beginning of the experiment to induce hepatic fibrosis. The CS group was also treated with CS 10 days after the beginning of CCl4 and ethanol administration. All control groups were given corresponding placebo at the same time. At the end of the 9th week, rats in each group were humanely sacrificed. Blood and tissue specimens were taken.Biochemical, radioimmunological, immunohistochemical and molecular biological examinations were used to determine the level change of ALT, AST, HA, LN content in serum and TGFβ1, PDGF, collagen Ⅰ and Ⅲ expression in tissue at either protein or mRNA level or both of them.RESULTS: As compared with the model control group,serum ALr, AST, HA, and LN content levels were markedly dropped in CS group (86.0±34.4 vs224.3±178.9, 146.7±60.2vs272.6±130.1, 202.0±79.3 vs316.5±94.1 and 50.4±3.0vs 59.7±9.8, respectively, P<0.05). Tissue expression of TGFβ1 and its mRNA, collagen I mRNA were also markedly decreased (0.2±0.14 vs1.73±1.40, 1.68±0.47 vs3.17±1.17,1.10±0.84 vs 2.64±1.40, respectively, P<0.05). More dramatical drop could be seen in PDGF expression (0.87±0.43vs1.91±0.74, P<0.01). Although there was no statistical significance, it was still strongly suggested that collagen Ⅲ mRNA expression was also decreased in CS group as compared with model control group (0.36±0.27 vs0.95±0.65,P=0.0615). In this experiment, no significant change could be found in PDGF mRNA expression between two groups (0.35±0.34 vs 0.70±0.46, P>0.05).CONCLUSION: Cordyceps sinensis could inhibit hepatic fibrogenesis derived from chronic liver injury, retard the development of cirrhosis, and notably ameliorate the liver function. 6A study of interaction between surface water and groundwater using environmental isotope in Huaisha River basin显示文摘The surface water and groundwater are important components of water cycle, and the interaction between surface water and groundwater is the important part in water cycle research. As the effective tracers in water cycle research, environmental isotope and hydrochemistry can reveal the interrelationships between surface water and groundwater effectively. The study area is the Huaisha River basin, which is located in Huairou district, Beijing. The field surveying and sampling for spring, river and well water were finished in 2002 and 2003. The hydrogen and oxygen isotopes and water quality were measured at the laboratory. The spatial characteristics in isotope and evolution of water quality along river lines at the different area were analyzed. The altitude effect of oxygen isotope in springs was revealed, and then using this equation, theory foundation for deducing recharge source of spring was estimated. By applying the mass balance method, the annual mean groundwater recharge rate at the catchment was estimated. Based on the groundwater recharge analysis, combining the hydrogeological condition analysis, and comparing the rainfall-runoff coefficients from the 1960s to 1990s in the Huaisha River basin and those in the Chaobai River basin, part of the runoff in the Huaisha River basin is recharged outside of this basin, in other words, this basin is an un-enclosed basin. 8Mutation analysis of APC gene in gastric cancer with microsatellite instability显示文摘AIM: To evaluate the role of APC mutation in gastric carcinogenesis and to correlate APC mutation with microsatellite instability (MSI) in gastric carcinomas. METHODS: APC mutation was measured with multiplex PCR, denaturing gradient gel electrophoresis and DNA sequencing; and MSI was analyzed by PCR-based methods.RESULTS: Sixty-eight cases of sporadic gastric carcinoma were studied for APC mutation at exon 15 and MSI. APC mutaions were detected in 15(22.1%) gastric cancers. Frequence of APC mutation (33.3 %) in intestinal type of gastric cancer was significantly higher than that in diffuse type (13.1 %, P<0.05).On the contrary, no association was observed betweerAPC mutation and tumor size,differentiation, depth of invasion, metastasis or clinical stages.Using five microsatellite markers, MSI in at least one locus was detected in 17 of 68 (25 %) of the tumors analyzed.APC mutations were all detected in MSI-L (only one locus,n=9) or MSS(tumor lacking MSI or stable, n=51), but no mutation was found in MSI-H (≥2 loci, n=8).CONCLUSION: APC mutation is involved in carcinogenesis of intestinal type of gastric cancer and is independent of MSI phenotype but related to the LOH pathway in gastric cancer.Dian-Chun Fang Yuan-Hui Luo Shi-Ming Yang Xiao-An Li Xian-Long Ling Li Fang Department of Gastroenterology,Southwest Hospital,Third Military Medical University,Chongqing 400038,China 2002World Journal of Gastroenterology2002,8,5:34

10Endoscopic patterns of gastric mucosa and its clinicopathological significance显示文摘AIM: To explore the correlation of magnifying endoscopic patterns and histopathology, Helicobacter pylori (H pylori) infection of the gastric mucosa.METHODS: Gastric mucosal patterns in 140 patients with chronic gastritis were studied using Olympus GIF-Q240Z magnifying endoscope. Histopathological examination, rapid urease test and Warrthin-Starry staining were taken with biopsy samples from the magnified sites of stomach. The magnifying endoscopic patterns were compared with histopathological results and H pylori detection.RESULTS: The pit patterns of gastric mucosa were classified as types A (round spot), B (short rod), C (branched), D (reticular) and E (villus). The detection rate of chronic atrophic gastritis (CAG) by magnifying endoscopy was 94.3 % (33/35),which was significantly higher than that by routine endoscopy (22.9 %, 8/35) (P<0.01). The pit patterns of 31 cases of intestinal metaplasia (IM) appeared as type E in 18 cases (58.1%), type D in 8 cases (25.8 %) and type C in 5 cases (16.1%). Fourteen out of 18 patients (77.8 %) with complete type (type Ⅰ) of IM appeared as type E of pit patterns, whereas only 4 of 13 (30.8 %) patients with incomplete type (types Ⅱ and Ⅲ) of IM appeared as type E (P<0.05). Collecting venules in the anterior of lower part of gastric corpus were subgrouped into types R (regular), I (irregular) and D (disappeared).H pyloriinfection was found in 12.2 %(9/74),60 %(9/15) and 84.3 %(43/51) cases in these types respectively. 11Hepatitis C virus infection of human hepatoma cell line 7721 in vitro显示文摘AIM To establish a cell culture system with long-term replication of hepatitis C virus in vitro.``METHODS Human hepatoma cell line 7721 was tested for its susceptibility to HCV by incubating with a serum from a patient with chronic hepatitis C. Cells and supernatant were harvested at various phases during the culturing periods The presence of HCV RNA, the expression of HCV antigens in cells and/or supernatant were examined by RT-PCR, in situ hybridization and immunohistochemistry respectively.``RESULTS The intracellular HCV RNA was first detected on d 2 after infection and then could be intermittently detected in both cells and supernatant over a period of at least three months. The expression of HCV NS3, CP10antigens could be observed in cells. 12Pancreatic encephalopathy in 24 patients with severe acute pancreatitis显示文摘BACKGROUND: Pancreatic encephalopathy (PE), an un- familiar complication of severe acute pancreatitis ( SAP) , is difficult to diagnose and treat, and it has a high mortality. The aim of this study was to investigate the manifestation, classification, mechanism and therapy of PE. METHODS: Of 132 patients with SAP treated at our hospi- tal from March 1994 to March 2004, 24 patients complicated by PE were analyzed retrospectively. RESULTS: The causes of SAP were mostly biliary and alco- holic. Twenty-four patients (18.2%) were complicated by PE within 3 hours-38 days ( average 6. 6 days) [ 21 (87. 5%) within 2 weeks, and 3(12.5%) after 2 weeks]. Eleven pa- tients were male and 13 female, with an average of 47 years (range 25-72 years). Excitement or restrain was the main manifestation. Nine patients (37.5%) received surgery and 15 (62.5%) conservative treatment, with a mortality of 11.1 % (1/9) and 66.7% (10/15), respectively. CONCLUSIONS: PE occurs 2 weeks after SAP and is part of multiple organ failure (MOF). Some patients have PE in the late stage of SAP because of lack of VitB1 and nutri- tion. 13The integration of methanogenesis with denitrification and anaerobic ammonium oxidation in an expanded granular sludge bed reactor显示文摘The integration of methanogenesis with denitrification and anaerobic ammonium oxidation(ANAMMOX) was studied in an expanded granular sludge bed(EGSB) reactor in this work. Experimental results from the continuous treatment of wastewater with nitrite and ammonium, which lasted for 107 days, demonstrated that wastewater with high nitrite and ammonium could be anaerobically treated in an expanded granular sludge bed reactor. More than 91% to 97% of COD were removed at up to about 3.9 g COD/(L·d) of COD volumetric loading rate. More than 97% to 100% of nitrite was denitrified at up to about 0.8 g NO -2-N/(L·d), which is 16 times higher than that in a conventional activated sludge system with nitrification/denitrification(0.05 gN/(L·d)). No dissimilatory reduction of nitrite to ammonium occurred in the process. However, maximum of about 40% ammonium was found to be lost. Batch tests of 15 days with sludge from the reactor showed that 100% of nitrite was denitrified completely, and about 3% of ammonium was removed when only ammonium (34.3 mg/L) and nitrite(34.3 mg/L) were added into the sludge suspension medium. Furthermore, about 15% of ammonium amounts were lost with organic COD addition. It suggested that the methanogenesis in the system could enhance ANAMMOX because of intermediate hydrogen produced during methanogenesis.ZHANG Dai-jun 1,2 (1. 16Investigation of the effect of berberines alkaloids in Coptis chinensis Franch on Bacillus shigae growthby microcalorimetry显示文摘The inhibitory effects of five berberines alkaloids (BAs) from rhizoma of Coptis chinensis Franch, a traditional Chinese medicinal (TCM) herb, on Bacillus shigae (B. shigae) growth were investigated by microcalorimetry. The power-time curves of B. shigae with and without BAs were acquired; meanwhile, the extent and duration of inhibitory effects on the metabolism were evaluated by growth rate constants (k1, k2), half inhibitory ratio (IC50), maximum heat output (Pmax), and peak time (tp). The values of k1 and k2 of B. shigae in the presence of the five BAs decreased with the increasing concentrations of BAs. Moreover, Pmax was reduced, and the value of tp increased with increasing concentrations of the five drugs. The inhibitory activity varied with different drugs. IC50 of the five BAs was respectively 75 μg/mL for berberine, 90 μg/mL for coptisine, 115 μg/mL for palmatine, 220 μg/mL for epiberberine, and 400 μg/mL for jatrorrhizine. The sequence of antimicrobial activity of the five BAs: berberine > coptisine > palmatine > epiberberine > jatrorrhizine. The functional groups methylenedioxy at C2 and C3 on phenyl ring improve antimicrobial activity more strongly than methoxyl at C2 and C3 on phenyl ring. 17Correlation between gene expression of CCK-A receptor and emptying dysfunction of the gallbladder in patients with gallstones and diabetes mellitus显示文摘BACKGROUND: The incidence of gallstone is higher in patients with diabetes mellitus than in general population because of hypomotility and lowered emptying function. The aim of this study was to investigate the gene expression of cholecystokinin-A ( CCK-A) receptor in patients with gallstones and diabetes mellitus and its correlation with the hypomotility of the gallbladder. METHODS: Smooth muscle was taken from the gallbladder after cholecystectomy and total RNA was extracted from it by Trizol reagent to compose cDNA. CCK-A receptor was enlarged by reverse transcription-polymerase chain reaction (RT-PCR) and identified by electrophoresis. Based on the analysis of images, the relative expression of the CCK-A re- ceptor was obtained after comparison of the CCK-A recep- tor and β-action. The emptying function of the gallbladder was measured with B ultrasound before operation. RESULTS: Gallbladder ejetion fraction ( GBEF2 ) (%) (24.9±12.7) decreased more significantly in patients with gallstones and diabetes mellitus than in those with simple gallstone (61.5±8.5) (P <0.01). In the two groups, a frac- tion of 540bp of CCK-A receptor could be testified respec- tively, but the relative expression was different. Compared to the expression of β-action, it was 0. 400 ±0.068 in pa- tients with gallstones and diabetes mellitus, but 0. 622 ± 0.070 in those with gallstone only (F =7.169, P <0.01). The result was consistent with the decreased contractility. CONCLUSIONS: The gene expression of CCK-A receptor on the smooth muscle of the gallbladder may decrease more significantly in patients with gallstone and diabetes mellitus than in those with gallstone only; this result is con- sistent with the decreased motih’ty. 18A study of recombinant protective H.Pylori antigens显示文摘AIM: To construct a recombinant vector which can express Mr 26 000 outer membrane protein (OMP) from Helicobacter pylori (Hp), and to obtain the vaccine protecting against Hp infection and a diagnostic reagent kit quickly detecting Hp infection.METHODS: The gene encoding the structural Mr 26000 outermembrane protein of Hp was amplified from Hpchromosomal DNA by PCR, and inserted in the prokaryoticexpression vector pET32a ( + ), which was transformed intothe Topl0 E. coli strain. Recombinant vector was selected,identified and transformed into BL-21(DE3) E. coli strain.The recombinant fusion proteins were expressed. Theantigenicity of recombinant protein was studied by ELISA orimmunoblotting and immunized Balb/c mice.RESULTS: The gene of Mr 26 000 OMP was amplified to be594 base pairs, 1.1% of the cloned genes was mutated and1.51% of amino acid residues was changed, but there washomogeneity between them. The recombinant fusion proteinencoded objective polypeptides of 198 amino acid residues,corresponding to calculated molecular masses of Mr 26000.The level of soluble expression products was about 38.96 %of the total cell protein. After purification by Ni-NTA agaroseresin columniation, the purity of objective protein becameabout 90 %. The EESA results showed that recombinantfusion protein could be recognized by patient serum infectedwith Hp and rabbit serum immunized with the recombinantprotein. 19Mutation and methylation of hMLH1 in gastric carcinomas with microsatellite instability显示文摘AIM: To appraise the correlation of mutation and methylation of hMSH1 with microsatellite instability (MSI) in gastric cancers.METHODS: Mutation of hMLH1 was detected by Twodimensional electrophoresis (Two-D) and DNA sequencing;Methylation of hMLH1 promoter was measured with methylation-specific PCR; MST was analyzed by PCR-basecl methods.RESULTS: Sixty-eight cases of sporadic gastric carcinoma were studied for mutation and methylation of hMLH1 promoter and MSI. Three mutations were found, two of them were caused by a single bp substitution and one was caused by a 2 bp substitution, which displayed similar Two-D band pattern.Methylation of hMLH1 promoter was detected in 11(16.2%)gastric cancer. By using five MSI markers, MSI in at least one locus was detected in 17/68(25 %) of the tumors analyzed.Three hMLH1 mutations were all detected in MSI-H (≥2 loci,n=8), but no mutation was found in MSI-L (only one locus,n=9) or MSS (tumor lacking MSI or stable, n=51). 20Plasma endothelin and nitric oxide levels in patients with acute pancreatitis显示文摘Objective: To explore the changes of plasma endothelin(ET) and nitric oxide (NO) levels in patients with a-cute pancreatitis.Methods: The level of plasma ET was measured by ra-dioactive-immunoassay, and NO by spectrophotometry.Results: The levels of ET, NO and the ET/NO ratioin patients with severe acute pancreatitis(SAP) within24 hours in hospital were all significantly higher thanthose in other groups of patients [(176±8)pg/ml,(97±11) μmol/L, and 1.83±0.12, P<0.01]. Com-pared to healthy controls(N), the levels of ET and NOin patients without pancreatitis acute abdomen (NAP)and patients with mild acute pancreatitis (MAP) in-creased significantly (P<0.01). After appropriate treat-ment, the levels of ET and NO in the MAP groupwere lower (P<0.01).
