| 1 | Pathogenesis, prevention, diagnosis and treatment of breast cancer显示文摘Breast cancer is the most common cancer affecting women worldwide. Prediction models stratify a woman's risk for developing cancer and can guide screening recommendations based on the presence of known and quantifiable hormonal, environmental, personal, or genetic risk factors. Mammography remains the mainstay breast cancer screening and detection but magnetic resonance imaging and ultrasound have become useful diagnostic adjuncts in select patient populations. The management of breast cancer has seen much refinement with increased specialization and collaboration with multidisciplinary teams that include surgeons, oncologists, radiation oncologists, nurses, geneticist, reconstructive surgeons and patients. Evidence supports a less invasive surgical approach to the staging and management of the axilla in select patients. | 2 | Quantitative analysis of ruminal methanogenic microbial populations in beef cattle divergent in phenotypic residual feed intake(RFI)offered contrasting diets显示文摘Background:Methane(CH_4) emissions in cattle are an undesirable end product of rumen methanogenic fermentative activity as they are associated not only with negative environmental impacts but also with reduced host feed efficiency.The aim of this study was to quantify total and specific rumen microbial methanogenic populations in beef cattle divergently selected for residual feed intake(RFI) while offered(i) a low energy high forage(HF) diet followed by(ii) a high energy low forage(LF) diet.Ruminal fluid was collected from 14 high(H)and 14 low(L) RFI animals across both dietary periods.Quantitative real time PCR(qRT-PCR) analysis was conducted to quantify the abundance of total and specific rumen methanogenic microbes.Spearman correlation analysis was used to investigate the association between the relative abundance of methanogens and animal performance,rumen fermentation variables and diet digestibility.Results:Abundance of methanogens,did not differ between RFI phenotypes.However,relative abundance of total and specific methanogen species was affected(P< 0.05) by diet type,with greater abundance observed while animals were offered the LF compared to the HF diet.Conclusions:These findings suggest that differences in abundance of specific rumen methanogen species may not contribute to variation in CH_4 emissions between efficient and inefficient animals,however dietary manipulation can influence the abundance of total and specific methanogen species. | Ciara A Carberry David A Kenny Alan K Kelly Sinead M Waters | 2014 | Journal of Animal Science and Biotechnology2014,5,4: | 6 |
| 3 | Incorporation of Biochar Carbon into Stable Soil Aggregates:The Role of Clay Mineralogy and Other Soil Characteristics显示文摘Aggregation and structure play key roles in water-holding capacity and stability of soils.In this study,the incorporation of carbon(C) from switchgrass biochar into stable aggregate size fractions was assessed in an Aridisol(from Colorado,USA) dominated by 2:1 clays and an Alfisol(from Virginia,USA) containing weathered mixed 1:1 and 2:1 mineralogy,to evaluate the effect of biochar addition on soil characteristics.The biochar was applied at 4 levels,0,25,50,and 100 g kg^(-1),to the soils grown with wheat in a growth chamber experiment.The changes in soil strength and water-holding capacity using water release curves were measured.In the Colorado soil,the proportion of soil occurring in large aggregates decreased,with concomitant increases in small size fractions.No changes in aggregate size fractions occurred in the Virginia soil.In the Colorado soil,C content increased from 3.3 to 16.8 g kg^(-1),whereas in the<53 μm fraction C content increased from 5.7 to 22.6 g kg^(-1) with 100 g kg^(-1)biochar addition.In the Virginia soil,C content within aggregate size fractions increased for each size fraction,except the>2 000 μm fraction.The greatest increase(from 6.2 to 22.0 g kg^(-1)) occurred in the 53–250 μm fraction.The results indicated that C was incorporated into larger aggregates in the Virginia soil,but remained largely unassociated to soil particles in the Colorado soil.Biochar addition had no significant effect on water-holding capacity or strength measurements.Adding biochar to more weathered soils with high native soil organic content may result in greater stabilization of incorporated C and result in less loss because of erosion and transport,compared with the soils dominated by 2:1 clays and low native soil organic content. | Charlene N. | 4 | Overview of recent advances in metastatic triple negative breast cancer显示文摘Metastatic triple negative breast cancer(TNBC)has an aggressive phenotype with a predilection for visceral organs and brain.Best responses to chemotherapy are predominately in the first line.Recent studies have demonstrated improved progression free survival with the combination of atezolizumab/pembrolizumab and chemotherapy in programmed death-ligand 1 positive metastatic TNBC.However,a recent trial in a similar population showed no benefit for atezolizumab and paclitaxel which led to a Food and Drug Administration alert.Two phase III trials(OLYMPIAD and BROCADE3)demonstrated a benefit in progression free survival(PFS)but not overall survival in patients with BRCAassociated metastatic TNBC treated with Olaparib or Talazoparib respectively.For those treated with Talazoparib,the time to deterioration in health related-quality of life was also longer compared to chemotherapy.The BROCADE3 trial demonstrated that the combination of a platinum and veliparib increased PFS in first-line metastatic TNBC but at the cost of increased toxicity.There are no headto-head comparisons of a poly(adenosine diphosphate-ribose)polymerase inhibitors(PARPi)and platinums.There are unanswered questions regarding the role of PARPi maintenance after platinum therapy as is standard of care in BRCAassociated ovarian cancer.Other areas of therapeutic interest include targeting aberrations in the phosphoinositide 3-kinase pathway,protein kinase B,mammalian target of rapamycin or utilising antibody drug conjugates.This review focusses on recent and emerging therapeutic options in metastatic TNBC.We searched PubMed,clinicaltrials.gov and recent international meetings from American Society of Clinical Oncology,San Antonio Breast Cancer Conference and the European Society of Medical Oncology. | David O'Reilly Maha Al Sendi Catherine M Kelly | 2021 | World Journal of Clinical Oncology2021,12,3: | 5 |

| 8 | Inhibiting the kynurenine pathway in spinal cord injury: multiple therapeutic potentials?显示文摘Chronic induction of the kynurenine pathway(KP) contributes to neuroinflammation by producing the excitotoxin quinolinic acid(QUIN). This has led to significant interest in the development of inhibitors of this pathway, particularly in the context of neurodegenerative disease. However, acute spinal cord injury(SCI) also results in deleterious increases in QUIN, as secondary inflammatory processes mediated largely by infiltrating macrophages, become predominant. QUIN mediates significant neurotoxicity primarily by excitotoxic stimulation of the N-methyl-D-aspartate receptor, but other mechanisms of QUIN toxicity are known. More recent focus has assessed the contribution that neuroinflammation and modulations in the KP make in mood and psychiatric disorders with recent studies linking inflammation and modulations in the KP, to impaired cognitive performance and depressed mood in SCI patients. We hypothesize that these findings suggest that in SCI, inhibition of QUIN production and other metabolites, may have multiple therapeutic modalities and further studies investigating this are warranted.
