2Study of transmembrane La^(3+) movement in rat ventricular myocytes by the patch-clamp technique显示文摘We have studied transmembrane La3+ movement in rat ventricular myocytes for the first time by using the whole-cell patch-clamp recording mode. La3+ (0.01-5.0 mmol/L) could not bring out inward currents through the L-type calcium channel in rat ventricular myocytes, while it could enter the cells by the same way carried by 1μmo1/L ionomycin. When the outward Na+ concentration gradient is formed, La3+ can enter the cells via Na-Ca exchange, and the exchange currents increase with the increase of external La3+ concentrations. But compared with Na-Ca exchange currents in the same concentration, the former is only 14%-38% of the latter. 3Effects of lithium chloride on outward potassium currents in acutely isolated hippocam-pal CA1 pyramidal neurons显示文摘Although lithium possesses neuropro- tective functions, the molecular mechanism underly- ing its actions has not been fully elucidated. In the present paper, the effects of lithium chloride on volt- age-dependent potassium currents in the CA1 py- ramidal neurons acutely isolated from rat hippocam- pus were studied using the whole-cell patch-clamp technique. Depolarizing test pulses activated two components of outward potassium currents: a rapidly activating and inactivating component, IA and a de- layed component, IK. Results showed that lithium chloride increased the amplitude of IA in a concentra- tion-dependent manner. Half enhancement concen- tration (EC50) was 22.80±5.45 μmol?L?1. Lithium chloride of 25 μmol?L?1 shifted the steady-state acti- vation curve and inactivation curve of IA to more negative potentials, but mainly affected the activation kinetics. The amplitude and the activation processes of IK were not affected by lithium chloride. 4Morphology, mitochondrial development and adipogenic-related genes expression during adipocytes differentiation in grass carp(Ctenopharyngodon idellus)显示文摘To investigate the differentiation mechanism of grass carp preadipocytes, a primary adipocytes culture system was established. Confluent preadipocytes were induced to differentiation, and the morphology and gene expression were evaluated at different stages. It was shown that preadipocytes were gradually filled with droplets and the cellular lipid content increased during the differentiation. Ultrastructure observation indicated that the number of mitochondria increased with adipocytes differentiation.Consistently, the mitochondrial protein content was elevated in the differentiating adipocytes. q RT-PCR showed that the expression level of lipogenesis-related genes such as peroxisome proliferator activator receptor c(PPAR c),lipoprotein lipase(LPL), fatty acid synthase(FAS) and stearoyl-Co A desaturase(SCD) increased during adipocytes differentiation. The mitochondrial relevant gene also elevated when adipocyte differentiation, such as PPAR coactivator-1(PGC-1 a), PGC-1 b and nuclear respiratory factor(NRF-1). However, the expression of carnitine palmitoyltransferase I a(CPT-1 a) gene decreased at the initial stage, but increased at the last stage of cell differentiation. These results indicated that the differentiation process of grass carp preadipocytes is similar to that of landanimals, but the molecular mechanisms are not exactly the same. 5Biomass-derived hard carbon microtubes with tunable apertures for high-performance sodium-ion batteries显示文摘Sodium-ion batteries(SIBs)are considered the most up-and-coming complements for large-scale energy storage devices due to the abundance and cheap sodium.However,due to the bigger radius,it is still a great challenge to develop anode materials with suitable space for the intercalation of sodium ions.Herein,we present hard carbon microtubes(HCTs)with tunable apertures derived from low-cost natural kapok fibers via a carbonization process for SIBs.The resulted HCTs feature with smaller surface area and shorter Na+diffusion path benefitting from their unique micro-nano structure.Most importantly,the wall thickness of HCTs could be regulated and controlled by the carbonization temperature.At a high temperature of 1,600℃,the carbonized HCTs possess the smallest wall thickness,which reduces the diffusion barrier of Na+and enhances the reversibility Na+storage.As a result,the 1600HCTs deliver a high initial Coulombic efficiency of 90%,good cycling stability(89.4%of capacity retention over 100 cycles at 100 mA·g^(−1)),and excellent rate capacity.This work not only charts a new path for preparing hard carbon materials with adequate ion channels and novel tubular micro-nano structures but also unravels the mechanism of hard carbon materials for sodium storage.Pin Song Shiqiang Wei Jun Di Jun Du Wenjie Xu Daobin Liu Changda Wang Sicong Qiao Yuyang Cao Qilong Cui Pengjun Zhang Liaobo Ma Jiewu Cui Yan Wang Yujie Xiong 2023Nano Research2023,16,4:1

9An acetylcholinesterase inhibitor, 3-benzidino-5-methyl-6-phenylpyridazine, blocking outward potassium currents in acutely isolated rat hippocampal pyramidal neurons显示文摘3-benzidino-5-methyl-6-phenylpyridazine (BMP) inhibited electric eel acetylcholinesterase (AChE), with IC50 being 0.58 μmol·L-1. As an AChE inhibitor, the effects of BMP on delayed rectifier potassium current (IK(DR)) and transient outward potassium current (IK(A)) in acutely isolated rat hippocampal pyramidal neurons were studied using the whole cell patch-clamp technique. BMP (0.1―50 μmol·L-1) inhibited IK(DR) and IK(A) in a concentration-dependent and voltage-independent manner. The IC50 value for the blocking action of BMP on IK(DR) and IK(A) was calculated to be 2.92 and 2.11 μmol·L-1, respectively. At the concentration of 10 μmol·L-1, BMP shifted the activation curve of IK(DR) to negative potential by 8.85 mV. Meanwhile, at the concentration of 10 μmol·L-1, BMP also shifted the activation and the steady-state inactivation curve of IK(A) to negative potential by 5.82 mV and 10.02 mV, respectively. 10STUDIES ON UNIFORM VELOCITY DISTRIBUTION IN CYLINDRICAL VESSELS WITH ANNULAR PACKED-BED显示文摘The uniform distribution of radial velocities of flow is of great importance for a cylindrical vessel with annular packed-bed (CVAPB). In this paper, a theoretical analysis for producing a uniform radial velocity distribution within a vessel is presented and a design method is established for a specially designed conical chock (SDCC). A differential equation for determining the contour size of SDCC is derived. Experimental verification is performed in a test model of CVAPB. 12Ciprofloxacin stress changes key enzymes and intracellular metabolites of Lactobacillus plantarum DNZ-4显示文摘Ciprofloxacin(CIP)is an antibiotic used to treat infections caused by bacteria.In this experiment,key enzymes and intracellular metabolites of Lactobacillus plantarum DNZ-4 was researched under CIP stress.The results showed that the activities of hexokinase,pyruvate kinase,β-galactosidase and Na^(+),K^(+)-ATPase after 1/2 minimum bacteriostatic concentration(MIC)CIP treatment were significantly decreased(P<0.01).Gas chromatography-mass spectrometry was used to analysis the changes of main metabolites in the cells and principal component analysis and partial least square model were constructed.The results indicated that CIP could cause changes in intracellular fatty acids,carbohydrates and amino acids,and the mechanism of amino acid metabolism under CIP stress was significantly inhibited.L.plantarum DNZ-4 made stress response to CIP by regulating the ratio of saturated fatty acids and unsaturated fats.This experiment revealed the changes of growth and metabolism mechanism of L.plantarum DNZ-4 under CIP stress,which help to provide technical means for the development of effective probiotics preparation products.Pin Chen Xiaoqian Chen Wei Yu Bo Zhou Lihua Liu Yuzhuo Yang Peng Du Libo Liu Chun Li 2022Food Science and Human Wellness2022,11,2:0
13Steroid Receptor Coactivator 3 Regulates Synaptic Plasticity and Hippocampus-dependent Memory显示文摘Steroid hormones play important roles in brain development and function.The signaling of steroid hormones depends on the interaction between steroid receptors and their coactivators.Although the function of steroid receptor coactivators has been extensively studied in other tissues,their functions in the central nervous system are less well investigated.In this study,we addressed the function of steroid receptor coactivator 3(SRC3)–a member of the p160 SRC protein family that is expressed predominantly in the hippocampus.While hippocampal development was not altered in Src3^(+/-) mice,hippocampus-dependent functions such as short-term memory and spatial memory were impaired.We further demonstrated that the deficient learning and memory in Src3^(+/-) mice was strongly associated with the impairment of long-term potentiation(LTP)at Schaffer Collateral-CA1 synapses.Mechanistic studies indicated that Src3^(+/-) mutation altered the composition of N-methyl-D-aspartate receptor subunits in the postsynaptic densities of hippocampal neurons.Finally,we showed that SRC3 regulated synaptic plasticity and learning mainly dependent on its lysine acetyltransferase activity.Taken together,these results reveal previously unknown functions of SRC3 in the hippocampus and thus may provide insight into how steroid hormones regulate brain function.Hai-Long Zhang Bing Zhao Pin Yang Yin-Quan Du Wei Han Jianming Xu Dong-Min Yin 2021Neuroscience Bulletin2021,37,12:0
14XIAP expression and its predictive significance of prognosis in stage IIB osteosarcomas显示文摘Objective: To evaluate X-linked inhibitor of apoptosis protein (XIAP) expression in biopsy specimens from the patients with stage IIB osteosarcomas before chemotherapy to study XIAP expression and its predictive significance of prognosis in stage IIB osteosarcomas. Methods: The expression of XIAP was retrospectively detected by SP immunohistochemistry in 31 cases biopsy specimens from stage IIB osteosarcomas before chemotherapy, and the relationship between XIAP expression and clinicopathologic features of the patients with stage IIB osteosarcomas was analyzed. Results: The osteo-sarcoma specimens showed strong cell nucleus immunoreaction of XIAP while normal bone specimens showed weak. High XIAP positive expression independently predicted the patient’s poor outcome and had no relationship with clinicopathologic variables such as age, gender, location, tumor size, rate of tumor necrosis to preoperative chemotherapy, local recurrence (P > 0.05). Significant correlations were found between high XIAP expression and metastasis (P = 0.022), and also between high XIAP expression and final survival (P = 0.011).
