| 1 | Metal alginates for polyphenol delivery systems: Studies on crosslinking ions and easy-to-use patches for release of protective flavonoids in skin显示文摘Incorporation of bioactive natural compounds like polyphenols is an attractive approach for enhanced functionalities of biomaterials.In particular flavonoids have important pharmacological activities,and controlled release systems may be instrumental to realize the full potential of these phytochemicals.Alginate presents interesting attributes for dermal and other biomaterial applications,and studies were carried here to support the development of polyphenol-loaded alginate systems.Studies of capillary viscosity indicated that ionic medium is an effective strategy to modulate the polyelectrolyte effect and viscosity properties of alginates.On gelation,considerable differences were observed between alginate gels produced with Ca^2+,Ba^2+,Cu^2+,Fe^2+,Fe^3+and Zn^2+as crosslinkers,especially concerning shrinkage and morphological regularity.Stability assays with different polyphenols in the presence of alginate-gelling cations pointed to the choice of calcium,barium and zinc as safer crosslinkers.Alginate-based films loaded with epicatechin were prepared and the kinetics of release of the flavonoid investigated.The results with calcium,barium and zinc alginate matrices indicated that the release dynamics is dependent on film thicknesses,but also on the crosslinking metal used.On these grounds,an alginate-based system of convenient use was devised,so that flavonoids can be easily loaded at simple point-of-care conditions before dermal application.This epicatechin-loaded patch was tested on an ex-vivo skin model and demonstrated capacity to deliver therapeutically relevant concentrations on skin surface.Moreover,the flavonoid released was not modified and retained full antioxidant bioactivity.The alginate-based system proposed offers a multifunctional approach for flavonoid controllable delivery and protection of skin injured or under risk. | João Silva Pavlo Vanat Dorinda Marques-da-Silva Joaquim Rui Rodrigues Ricardo Lagoa | 2020 | Bioactive Materials2020,5,3: | 2 |
| 2 | Impaired T-cell differentiation in diabetic foot ulceration显示文摘Foot ulceration is one of the most debilitating complications associated with diabetes,but its cause remains poorly understood.Several studies have been undertaken to understand healing kinetics or find possible therapies to enhance healing.However,few studies have been directed at understanding the immunological alterations that could influence wound healing in diabetes.In this study,we analysed the T-cell receptor(TCR)repertoire diversity in TCR-αβ^(+)T cells.We also analysed the distribution and phenotype of T cells obtained from the peripheral blood of healthy controls and diabetic individuals with or without foot ulcers.Our results showed that diabetic individuals,especially those with foot ulcers,have a significantly lower naive T-cell number and a poorer TCR-Vβrepertoire diversity.We also showed that the reduced TCR-Vβrepertoire diversity in diabetic individuals was mainly owing to the accumulation of effector T cells,the major source of tumour necrosis factor-αproduction,which was even more pronounced in patients with acute foot ulceration.Moreover,the expression of several inflammatory chemokine receptors was significantly reduced in diabetic patients.In conclusion,effector T-cell accumulation and TCR repertoire diversity reduction appear to precede the development of foot ulcers.This finding may open new immunological therapeutic possibilities and provide a new prognostic tool in diabetic wound care. | João Moura João Rodrigues Marta Gonçalves Cláudia Amaral Margarida Lima Eugénia Carvalho | 2017 | Cellular & Molecular Immunology2017,14,9: | 6 |
| 3 | Dexamethasone intravitreal implant(Ozurdex) in diabetic macular edema: real-world data versus clinical trials outcomes显示文摘AIM: To investigate the safety and efficacy of intravitreal dexamethasone implants(Ozurdex?/DEX) in patients with diabetic macular edema(DME) either na?ve or nonna?ve to anti-VEGF therapies who switched to DEX implant independent of response to anti-vascular endothelial growth factors(anti-VEGFs).METHODS: This was an audit retrospective review of medical records of patients with DME who switched to the DEX intravitreal implant. Patients were divided into 2 groups: patients na?ve to antiangiogenic therapy and patients who were previously treated with anti-VEGFs. Data regarding demographics, changes in mean best-corrected visual acuity(BCVA), central macular thickness(CMT), and intraocular pressure(IOP) was collected over 6 mo. The demographic data mean changes in BCVA, CMT, and IOP were compared. Six-month follow-up data of 47 patients(57 eyes), who either switched to DEX implant irrespective of response to previous treatments or were treatment na?ve before receiving DEX implant, was collected.RESULTS: Improvement in mean BCVA was observed from 1-4 mo after injection with a decreased effect at month 6 as expected, with better outcomes in na?ve compared to non-na?ve patients. A statistically relevant decrease in mean CMT was observed during the follow-up period. An increase in mean IOP was observed in the first 2 mo after DEX therapy. The mean number of injections of the overall population during the 6 mo was 1.3. A subgroup analysis showed no relevant difference between phakic versus pseudophakic patients relative to measured outcomes. There was no cataract progression during the follow-up period and no adverse events reported.CONCLUSION: This real-life setting study shows that intravitreal DEX implant is effective and safe. | 6 | Understanding how environmental heterogeneity and elevation drives the distribution of woody communities across vegetation types within the campo rupestre in South America显示文摘The campo rupestre sensu lato is a vegetation type that occurs in South American mountains,supports a distinctive flora characterized by high rates of endemism,high herbaceous species richness and often-neglected but also species-rich of the arboreal stratum.We aimed to investigate how environmental factors and elevation are associated with the distribution and diversity of woody species in different rupestrian vegetation types across South America.Using a database of 2,049 woody species from 185 sites across four vegetation types within the campo rupestre,we assessed how the vegetation types were grouped according to their floristic composition and number of shared indicator species,as well as by using different beta diversity indices.The most important variables from a set of 27 variables(e.g.altitude,geo-edaphic and climatic)explaining species distribution were identified using redundancy analysis(RDA)and variation partitioning methods.The distribution of vegetation types was related to both environmental and spatial fractions,with a set of 17 variables retained(e.g.rockiness,grass cover and temperature seasonality as the most important variables).There was an association between the floristic composition of each vegetation type and the elevation range.Although the identified vegetation types are floristically related,they are distinguished by exclusive and habitat-specialist woody species.This uniqueness of vegetation types should be considered in terms of complementarity for the conservation of campos rupestres. | Marcelo Leandro BUENO Vanessa Leite REZENDE Luiza Fonseca A.DE PAULA João Augusto Alves MEIRA-NETO JoséRoberto Rodrigues PINTO Andreza Viana NERI Vanessa PONTARA | 2021 | Journal of Mountain Science2021,18,5: | 0 |
| 7 | Prevalence of Kudoa in Fish Fillets Caught in Para State显示文摘Kudoa is a myxozoan that causes myoliquefaction in marine fishes.Most of species only affect fish,but a K.septempunctata outbreak was reported in 358 people.Although many species of Kudoa are known,none was described in Brachyplatystoma filamentosum,Brachyplatystoma rousseauxii,Mugil curema,Plagioscion squamosissimus or Oxydoras niger until now.Due to the economic cost of eliminating seafood presenting myxozoan lesions,this study aimed to describe lesions found at necropsy and histopathology,as well as to detect this myxozoan by molecular techniques.For this purpose,were sampled 85 fish of the following species:Brachyplatystoma filamentosum,Brachyplatystoma rousseauxii,Mugil curema,Plagioscion squamosissimus,and Oxydoras niger from Colares and Vigia,Pará,Brazil.Necropsies were carried out to describe lesions and molecular techniques(PCR and sequencing)were applied for identification.Although muscle lesions were not observed at necropsy,histopathology revealed bacterial colonies,coagulative necrosis,dystrophic calcification,eosinophils,hemorrhage,parasitic pseudocysts,protozoan,and vacuolization.After sequencing,K.shiomitsui(GENBANK:LC128646)was identified as the most similar causative agent of fishes infection,but due to phylogenetic results and identity we suggest that the myxozoan found could be a new specie.Also,high parasitism of this myxozoan was observed in fishes sampled,i.e.,90%in Colares and 100%in Vigia. | Marianna Vaz RODRIGUES Patrícia Tidori MIURA Jéssica Fernandes de OLIVEIRA Maria das Dores Correia Palha João Pessoa Araújo JÚNIOR | 2020 | Journal of Zoological Research2020,2,3: | 0 |

| 12 | Can gadoxetic acid–enhanced magnetic resonance imaging be used to avoid liver biopsy in patients with nonalcoholic fatty liver disease?显示文摘BACKGROUND Nonalcoholic fatty liver disease(NAFLD)is a major cause of liver disease worldwide.The diagnosis of nonalcoholic steatohepatitis(NASH),the most severe form of NAFLD,is crucial and has prognostic and therapeutic implications.However,currently this diagnosis is based on liver biopsy and has several limitations.AIM To evaluate the performance of gadoxetic acid–enhanced magnetic resonance imaging(GA-MRI)in differentiating isolated steatosis from NASH in patients with NAFLD.METHODS In this prospective study,56 patients with NAFLD(18 with isolated steatosis and 38 with NASH)underwent GA-MRI.The contrast enhancement index(CEI)was calculated as the rate of increase of the liver-to-muscle signal intensity ratio from before and 20 min after intravenous GA administration.Between-group differences in mean CEI were examined using Student's t test.The area under the receiver operator characteristic curve and the diagnostic performance of gadoxetic acid–enhanced magnetic resonance imaging were evaluated.RESULTS The mean CEI for all subjects was 1.82±0.19.The mean CEI was significantly lower in patients with NASH than in those with isolated steatosis(P=0.008).Two CEI cut-off points were used:<1.66(94%specificity)to characterize NASH and>2.00(89%sensitivity)to characterize isolated steatosis.CEI values between 1.66 and 2.00 indicated liver biopsy,and the procedure could be avoided in 40%of patients with NAFLD.CONCLUSION GA-MRI is an effective noninvasive method that may be useful for the differentiation of NASH from isolated steatosis,and could help to avoid liver biopsy in patients with NAFLD. | Viviane Brandão Amorim Daniella Braz Parente Fernando Fernandes Paiva Jaime Araújo Oliveira Neto Amanda Almeida Miranda Cláudia Cravo Moreira Flávia Ferreira Fernandes Carlos Frederico FerreiraCampos Nathalie Carvalho Leite Renata de Mello Perez Rosana Souza Rodrigues | 2020 | World Journal of Hepatology2020,12,9: | 0 |
