1RNAi-Mediated Silencing of ITPK Gene Reduces Phytic Acid Content,Alters Transcripts of Phytic Acid Biosynthetic Genes,and Modulates Mineral Distribution in Rice Seeds显示文摘Phytic acid is the principal storage form of phosphorus in plant seeds and an essential signalling molecule in several regulatory processes of plant development.However,it is known as an anti-nutrient compound owing to its potent chelating property.Thus,reducing the phytic acid content in crops is desirable.Studies involving regulation of MIPS and IPK1 genes to generate low phytate rice have been reported earlier.However,the functional significance of OsITPK and the effect of its down-regulation on phytic acid content and the associated pleiotropic effects on rice have not yet been investigated.In this study,tissue specific RNA interference(RNAi)-mediated down-regulation of a major ITPK homolog(OsITP5/6K-1)resulted in 46.2%decrease in phytic acid content of T2 transgenic seeds with a subsequent 3-fold enhancement in the inorganic phosphorus content.Silencing of OsITP5/6K-1 altered the transcript levels of essential phytic acid pathway genes,without significantly affecting the transcript levels of other OsITPK homologs.Furthermore,the mapping of elements through X-ray microfluorescence analysis revealed significant changes in the spatial distribution pattern and translocation of elements in low phytate seeds.Additionally,low phytate polished seeds exhibited 1.3-fold and 1.6-fold enhancement in iron and zinc content in the grain endosperm,respectively.Silencing of OsITP5/6K-1 also altered the amino acid and myo-inositol content of the transgenic seeds.Our results successfully established that RNAi-mediated silencing of OsITP5/6K-1 gene significantly reduced the phytate levels in seeds without hampering the germination potential of seeds and plant growth.The present study provided an insight into the mechanism of phytic acid biosynthesis pathway.Aritra KARMAKAR Sananda BHATTACHARYA Shinjini SENGUPTA Nusrat ALI Sailendra Nath SARKAR Karabi DATTA Swapan K.DATTA 2020Rice science2020,27,4:7
2Core Shell Au Ag Nanoparticles in Dielectric Nanocomposites with Plasmon-Enhanced Fluorescence: A New Paradigm in Antimony Glasses显示文摘The nano era demands the synthesis of new nanostructured materials,if possible by simplified techniques,with remarkable properties and versatile applications.Here,we demonstrate a new single-step reproducible melt-quench methodology to fabricate core-shell bimetallic(Au0 Ag0)nanoparticles(2889 nm)embedded glasses(dielectrics)by the use of a new reducing glass matrix,K_(2)O B_(2)O_(3) Sb_(2)O_(3)(KBS)without applying any external reducing agent or multiple processing steps.The surface plasmon resonance(SPR)band of these nanocomposites embedded in KBS glass is tunable in the range 554-681 nm.More remarkably,taking advantage of the selective reduction capability of Sb_(2)O_(3),this single-step methodology is used to fabricate inter-metallic:rare-earth ions co-embedded(AuAg:Sm^(3+))dielectric(glass)-based-dnanocomposites and study the effect of enhanced local fi eld on the red upconversion fl uorescence of Sm^(3+)ions at 636 nm.The enhancement is found to be about 2 folds.This single-step in-situ selective reduction approach can be used to fabricate a variety of hybrid-nanocomposite devices for laser based applications(see supplementary information).Tirtha Som Basudeb Karmakar 2009Nano Research2009,2,8:7
3Novel virulence factor dupA of Helicobacter pylori as an important risk determinant for disease manifestation:An overview显示文摘Helicobacter pylori(H.pylori)is a microaerophilic,Gram-negative,human gastric pathogen found usually in the mucous lining of stomach.It infects more than 50%of the world’s population and leads to gastroduodenal diseases.The outcome of disease depends on mainly three factors:Host genetics,environment and bacterial factors.Among these,bacterial virulence factors such as cagA,vacA are well known for their role in disease outcomes.However,based on the global epidemiological results,none of the bacterial virulence(gene)factors was found to be associated with particular diseases like duodenal ulcer(DU)in all populations.Hence,substantial importance has been provided for research in strain-specific genes outside the cag pathogenicity island,especially genes located within the plasticity regions.dupA found within the plasticity regions was first demonstrated in 2005 and was proposed for duodenal ulcer development and reduced risk of gastric cancer in certain geographical regions.Due to the discrepancies in report from different parts of the world in DU development related to H.pylori virulence factor,dupA became an interesting area of research in elucidating the role of this gene in the disease progression.In this review,we shed light on the detailed information available on the polymorphisms in dupA and their clinical relevance.We have critically appraised several pertinent studies on dupA and discussed their merits and shortcomings.This review also highlights dupA gene as an important biomarker for DU in certain populations.Jawed Alam Avijit Sarkar Bipul Chandra Karmakar Mou Ganguly Sangita Paul Asish K Mukhopadhyay 2020World Journal of Gastroenterology2020,26,32:5
4A review of stress concentration factors in tubular and non-tubular joints for design of offshore installations显示文摘Tubular structures are widely used in offshore installations,trusses,high rise buildings,towers for wind turbines,ski-lift installations,lightning,road pole signals etc.,owing to their excellent structural performance and attractive appearance.Stress concentration,especially in the welded joints of these structures,is an important design consideration particularly for fatigue design.In the context of tubular and non-tubular joints,this paper provides a review of the experimental and numerical studies that have been carried out so far to determine the stress concentration factor(SCF).Emphasis is also placed on the complexity of capturing different types of stresses in tubular/non-tubular joints for estimation of SCF.Present code provisions for evaluation of SCF are also discussed.Further,a few issues,which require significant research effort to advance our understanding and to improve the current design guidelines,have been identified.©2016 Shanghai Jiaotong University.Published by Elsevier B.V.Dikshant Singh Saini Debasis Karmakar Samit Ray-Chaudhuri 2016Journal of Ocean Engineering and Science2016,1,3:4
5玫瑰茄花萼乙醇提取物的抗痛觉敏感、抗炎及止泻作用的实验研究(英文)显示文摘目的:研究玫瑰茄(Hibiscus sabdariffa)花萼的乙醇提取物的抗痛觉敏感、抗炎及止泻作用。方法:使用95%乙醇提取玫瑰茄花萼干品用于测定其功效。用小鼠扭体实验检测其抗痛觉敏感作用,二甲苯致耳水肿模型小鼠检测其抗炎症作用,蓖麻油致腹泻模型小鼠检测其止泻作用。结果:在乙酸致小鼠扭体实验中,玫瑰茄花萼的乙醇提取物对小鼠扭体的抑制与空白对照组相比差异有统计学意义(P<0.01),对二甲苯致耳水肿模型小鼠的耳水肿的抑制与空白对照组相比差异有统计学意义(P<0.01),且显著减少了蓖麻油致腹泻小鼠的排便次数并增加了排便间隔时间(P<0.01)。结论:本研究的结果证实了玫瑰茄花萼的乙醇提取物具有显著的抗痛觉敏感、抗炎及止泻作用,验证了其在传统医学中的应用。Md.Khadem Ali Ayesha Ashraf Nripendra Nath Biswas Utpal Kumar Karmakar Shamima Afroz 2011中西医结合学报2011,9,6:4
6Development of a set of SSR markers for genetic polymorphism detection and interspecific hybrid jute breeding显示文摘Corchorus capsularis(white jute) and C. olitorius(dark jute) are the two principal cultivated spedes of jute that produce natural bast fiber of commercial importance. We have identified 4509 simple sequence repeat(SSR) loci from 34,163 unigene sequences of C. capsularis to develop a non-redundant set of 2079 flanking primer pairs. Among the SSRs, trinudeotide repeats were most frequent(60%) followed by dinucleotide repeats(37.6%). Annotation of the SSR-containing unigenes revealed their putative functions in various biological and molecular processes, including responses to biotic and abiotic signals. Eighteen expressed gene-derived SSR(eSSR) markers were successfully mapped to the existing single-nucleotide polymorphism(SNP) linkage map of jute,providing additional anchor points. Amplification of 72% of the 74 randomly selected primer pairs was successful in a panel of 24 jute accessions, comprising five and twelve accessions of C.capsularis and C. olitorius, respectively, and seven wild jute spedes. Forty-three primer pairs produced an average of 2.7 alleles and 58.1% polymorphism in a panel of 24 jute accessions. The mean PIC value was 0.34 but some markers showed PIC values higher than 0.5, suggesting that these markers can efficiently measure genetic diversity and serve for mapping of quantitative trait loci(QTLs) in jute. A primer polymorphism survey with parents of a wide-hybridized population between a cultivated jute and its wild relative revealed their efficacy for interspecific hybrid identificatioii. 7Medicinal potential of Passiflora foetida L. plant extracts: biological and pharmacological activities显示文摘OBJECTIVE: To investigate analgesic, antidiarrhoeal and cytotoxic activities of the ethanol extract of Passifl ora foetida L.(Passifl oraceae) by three experimental methods. METHODS: Analgesic activity of the ethanol extract of Passifl ora foetida L.(EEPF) was carried out using acetic acid-induced writhing inhibition in mice. The method of castor oil-induced diarrhoea in mice was utilized to evaluate antidiarrhoeal activity. The cytotoxic activity of EEPF was explored with a brine shrimp lethality bioassay.RESULTS: The extract showed 68.75% and 30.00% inhibition of writhe at the doses of 500 and 250 mg/kg body weight, respectively. The extract increased the mean latent period prior to diarrhoeal onset to about 1.55 h and 1.17 h, and decreased the mean number of stools to 4.4 and 5.6 at the doses of 500 and 250 mg/kg body weight. The extract also demonstrated cytotoxic activity in the brine shrimp lethality assay, and the median lethal concentration for brine shrimp nauplii was 80 μg/mL. CONCLUSION: The results suggest that the plant extract has analgesic and antidiarrhoeal activities, supporting its uses in traditional medicine.
