| 3 | Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs. | Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang | 2019 | Genes & Diseases2019，6，3： | 11 |
| 4 | Free-breathing radial volumetric interpolated breathhold examination vs breath-hold cartesian volumetric interpolated breath-hold examination magnetic resonance imaging of the liver at 1.5T显示文摘AIM: To compare breath-hold cartesian volumetric interpolated breath-hold examination(cVIBE) and freebreathing radial VIBE(rVIBE) and determine whether rVIBE could replace cVIBE in routine liver magnetic resonance imaging(MRI).METHODS: In this prospective study, 15 consecutive patients scheduled for routine MRI of the abdomen underwent pre- and post-contrast breath-hold cVIBE imaging(19 s acquisition time) and free-breathing rVIBE imaging(111 s acquisition time) on a 1.5T Siemens scanner. Three radiologists with 2, 4, and 8 years post-fellowship experience in abdominal imaging evaluated all images. The radiologists were blinded to the sequence types, which were presented in a random order for each patient. For each sequence, the radiologists scored the cVIBE and rVIBE images for liver edge sharpness, hepatic vessel clarity, presence of artifacts, lesion conspicuity, fat saturation, and overall image quality using a five-point scale. RESULTS: Compared to rVIBE, cVIBE yielded significantly(P < 0.001) higher scores for liver edge sharpness(mean score, 3.87 vs 3.37), hepatic-vessel clarity(3.71 vs 3.18), artifacts(3.74 vs 3.06), lesion conspicuity(3.81 vs 3.2), and overall image quality(3.91 vs 3.24). cVIBE and rVIBE did not significantly differ in quality of fat saturation(4.12 vs 4.03, P = 0.17). The inter-observer variability with respect to differences between rVIBE and cVIBE scores was close to zero compared to random error and inter-patient variation. Quality of rVIBE images was rated as acceptable for all parameters. CONCLUSION: rVIBE cannot replace cVIBE in routine liver MRI. | 8 | Biosynthesis of Unsaturated Fatty Acids via Mortierella Isabellina Cultivated in a Medium Containing Butanol显示文摘Mortierella isabellina was found to accumulate large amounts of unsaturated fatty acids when it was grown in a medium containing butanol(5 g/L) and yeast extract(10 g/L) and cultivated at 25 ℃ for 5 d during which additional feeding butanol of 2 g/L was fed after being cultivated for 48 h . The resultant mycelial lipids accounted for 40 1% of the dry mycelia, while about 34% of butanol in the medium was converted. The mycelial lipids contained four kinds of unsaturated fatty acids, i.e ., palmitoleic(4 9%), oleic(54 1%), linoleic(10 4%) and linolenic(5 4%) acids. Those accounted for 74 8% of the total fatty acids. | 13 | Docosahexaenoic acid suppresses arachidonic acid-induced proliferation of LS-174T human colon carcinoma cells显示文摘AIM:To investigate the impact of arachidonic acid (AA) and docosahexaenoic acid (DHA) and their combination on colon cancer cell growth.METHODS:The LS-174T colon cancer cell line was used to study the role of the prostaglandin precursor AA and the omega-3 polyunsaturated fatty acid DHA on cell growth. Cell viability was assessed in XTT assays. For analysis of cell cycle and cell death,flow cytometry and DAPI staining were applied. Expression of cyclooxygenase-2 (COX-2),p21 and bcl-2 in cells incubated with AA or DHA was examined by real-time RT-PCR. Prostaglandin E2 (PGE2) generation in the presence of AA and DHA was measured using a PGE2-ELISA.RESULTS:AA increased cell growth,whereas DHAreduced viability of LS 174T cells in a time-and dose-dependent manner. Furthermore,DHA down-regulated mRNA of bcl-2 and up-regulated p21. Interestingly,DHA was able to suppress AA-induced cell proliferation and significantly lowered AA-derived PGE2 formation. DHA also down-regulated COX-2 expression. In addition to the effect on PGE2 formation,DHA directly reduced PGE2-induced cell proliferation in a dose-dependent manner. | 15 | 血根碱对肉鸡生长性能、相对器官重量、盲肠菌群、血清胆固醇水平以及肉品质的影响显示文摘研究旨在探索日粮添加血根碱(SGV)对肉鸡生长性能、相对器官重量、盲肠菌群、血清胆固醇水平和腿肌丙二醛含量的影响。选用840日龄雄性肉鸡(罗斯品种)饲养于28个稻草垫上,随机分为4个处理:玉米-豆粕型基础对照日粮、基础日粮中添加卑霉素10 ppm作为生长促进剂(AGP)、基础日粮中添加SGV 20 ppm(SGV20)和50 ppm(SGV50)。与饲喂基础日粮的肉鸡相比,基础日粮中添加AGP组、SGV20组和SGV50组的肉鸡最终体重(BW)、日增重、饲料转化率(FCR)显著改善(P<0.05)。与对照组相比,SGV20喂养组的肉鸡相对空肠重量显著降低(P<0.05),所有添加SGV组肉鸡相对空肠或回肠长度显著升高(P<0.05)。SGV20组较对照组肉鸡肠道菌群有所改变,尤其是盲肠乳酸菌数增加,但AGP组无改变。SGV20组或SGV50组肉鸡的总胆固醇水平较对照组显著降低(P<0.05),控制饮食物显著降低(P<0.05)。添加SGV后,作为脂质过氧化反应指示剂的腿肌丙二醛含量显著降低(P<0.05)。研究表明,在肉鸡日粮中补充SGV 20或50 ppm可以提高肉鸡生长性能,改变多种生物和生理参数,如相对器官重量、血清胆固醇水平、肠道菌群和肉品质。 | Lee K W Kim J S Oh S T Kang C W An B K | 2015 | 饲料博览2015，0，3： | 2 |
