| 1 | Experimental models of non-alcoholic fatty liver disease in rats显示文摘Non-alcoholic fatty liver disease(NAFLD)is the most common chronic liver disease in the Western world,and it persists at a high prevalence.NAFLD is characterised by the accumulation of triglycerides in the liver and includes a spectrum of histopathological findings,ranging from simple fatty liver through non-alcoholic steatohepatitis(NASH)to fibrosis and ultimately cirrhosis,which may progress to hepatocellular carcinoma.The pathogenesis of NAFLD is closely related to the metabolic syndrome and insulin resistance.Understanding the pathophysiology and treatment of NAFLD in humans has currently been limited by the lack of satisfactory animal models.The ideal animal model for NAFLD should reflect all aspects of the intricate etiopathogenesis of human NAFLD and the typical histological findings of its different stages.Within the past several years,great emphasis has been placed on the development of an appropriate model for human NASH.This paper reviews the widely used experimental models of NAFLD in rats.We discuss nutritional,genetic and combined models of NAFLD and their pros and cons.The choice of a suitable animal model for this disease while respecting its limitations may help to improve the understanding of its complex pathogenesis and to discover appropriate therapeutic strategies.Considering the legislative,ethical,economical and health factors of NAFLD,animal models are essential tools for the research of this disease. | Otto Kucera Zuzana Cervinkova | 2014 | World Journal of Gastroenterology2014，20，26： | 16 |
| 2 | 供体细胞和曲古抑菌素(TSA)对奶牛克隆胚胎发育的影响显示文摘本研究探讨供核细胞处理策略(新鲜消化、-196℃冻存组(复苏后)和曲古抑菌素(TSA)处理组)和TSA-CR1aa胚胎培养液处理时间对克隆胚发育的影响。利用体细胞核移植技术生产奶牛克隆胚胎,将部分克隆胚移植给自然发情同步的受体,检验克隆胚的体内发育能力。结果显示:TSA处理体细胞组的克隆胚卵裂率和囊胚率与新鲜消化和-196℃冻存组相比差异显著(P<0.05);-196℃冻存组与新鲜消化组差异不显著;用TSA-CR1aa分别处理新鲜消化细胞构建的克隆胚0、24、48和60h,其中,60h处理组的囊胚率最高(36.11±1.78%)与其他组对比差异显著(P<0.05);用TSA-CR1aa分别处理TSA处理组构建的重构胚24、48和60h,结果发现,60hTSA-CR1aa处理组囊胚率(37.39±1.78%)最高,显著高于24h(25.48±1.34%)TSA-CR1aa处理组,且差异显著(P<0.05)。将所获得的克隆胚胎分别移植给40头自然发情的受体母牛,并观察移植25,45,65和90d后的返情情况,结果显示,各组受孕率无统计学差异,但经TSA处理细胞和胚胎组移植的受体中,获得一头存活的体细胞克隆奶牛。说明优化的处理方法处理胚胎可以完成体内发育。 | 章美玲 张运海 陶勇 刘亚 曹鸿国 李运生 Oto Hanus Josef Kucera 章孝荣 | 2011 | 农业生物技术学报2011，19，4： | 3 |
| 3 | Epithelial markers of colorectal carcinogenesis in ulcerative colitis and primary sclerosing cholangitis显示文摘AIM:To evaluate the expression of epithelial markers of colorectal carcinogenesis in patients with long-term ulcerative colitis(UC) and primary sclerosing cholangitis(PSC) before and after transplantation.METHODS:Eight patients with UC and PSC prior to liver transplantation(PSC-UC),22 patients with UC after liver transplantation for PSC(OLT),9 patients with active ulcerative colitis without PSC(UCA),7 patients withUC in remission(UCR) and 10 controls(N) underwent colonoscopy with multiple biopsies.Specimens were analysed histologically and semi-quantitatively immunohistochemically for p53,Bcl-2 and cyclooxygenase-2(COX-2) markers.Statistical analysis was performed by Kruskal-Wallis and Fisher's exact tests.RESULTS:PSC-UC had a statistically significantly higher expression of p53 in the nondysplastic mucosa as compared to OLT,UCA,UCR and N(P < 0.05).We also found a statistically significant positive correlation between the incidence of PSC and the expression of p53(P < 0.001).UCA had a higher p53 expression as compared to UCR.OLT had a significantly lower expression of p53 as compared with PSC-UC(P < 0.001).Bcl-2 had a significant higher bcl-2 expression as compared with controls.No difference in COX-2 expression between PSC-UC,UCR and UCA was found.UCA had higher COX-2 expression as compared to UCR.We also found a statistically significant positive correlation between the expression of COX-2 and p53.Patients after liver transplantation for PSC had a statistically significantly lower expression of the p53 compared with PSCUC(P < 0.001).PSC-UC had the same inflammatory endoscopic activity as OLT and UCR when evaluated with the Mayo score.CONCLUSION:Our study shows that the nondysplatic mucosa of UC patients with PSC is characterised by a higher expression of the tumour suppressor gene p53,suggesting a higher susceptibility of cancer.This p53 overexpression correlates with the presence of PSC whilst it is not present in patients with UC after liver transplantation for PSC. | Pavel Wohl Tomas Hucl Pavel Drastich David Kamenar Julius Spicak Eva Honsova Eva Sticova Alena Lodererova Jan Matous Martin Hill Petr Wohl Milos Kucera | 2013 | World Journal of Gastroenterology2013，19，14： | 2 |
