1p53 gene therapy in combination with transcatheter arterial chemoembolization for HCC:One-year follow-up显示文摘AIM:To evaluate the efficacy and safety of combination therapy with recombinant adenovirus p53 injection (rAdp53) and transcatheter hepatic arterial chemoembolization (TACE) for advanced hepatocellular carcinoma (HCC).METHODS:A total of 82 patients with advanced HCC treated only with TACE served as control group.Another 68 patients with HCC treated with TACE in combination with recombinant adenovirus-p53 injection served as p53 treatment group.Patients were followed up for 12 mo.Safety and therapeutic effects were evaluated according to the improvement in clinical symptoms,leukocyte count,Karnofsky and RECIST criteria.Survival rate was calculated with Kaplan-Meier method.RESULTS:The total effective rate was 58.3% for p53 treatment group,and 26.5% for control group (P < 0.05).The incidence of gastrointestinal symptoms was lower in p53 treatment group than in control group (P < 0.05).The 3-,6-and 12-mo survival rates were significantly higher for p53 treatment group than for control group (P < 0.01).The combination treatment was well tolerated with such adverse events as fever (51.5%,P=0.006) and pain of muscles and joints (13.2%,P=0.003),which were significantly higher than the chemotherapy.Except for these minor adverse effects,no severe vector-related complications were identified.With respect to the efficacy,patients in p53 treatment group had less gastrointerestinal symptoms (P=0.062),better improvement in tumor-related pain (P=0.003),less downgrade of leukocyte counts (P=0.003) and more upgrade of Karnofsky performance score (P=0.029) than those in control group.The total effective rate (CR + PR) for p53 treatment group and control group was 58.3% and 26.5%,respectively,with distributions of different effect in two groups (P=0.042).The survival rates were 89.71%,76.13%,and 43.30% for p53 treatment group,and 68.15%,36.98%,and 24.02% for control group,respectively,3,6 and 12 mo after treatment,suggesting that the survival rates are significantly higher for p53 treatment group than for control group (P=0.0002).CONCLUSION:The rAd-p53 gene therapy in combination with TACE is a safe and effective treatment modality for advanced HCC.Yong-Song Guan Yuan Liu Qing He Xiao Li Lin Yang Ying Hu Zi La 2011World Journal of Gastroenterology2011,17,16:21
2Adenovirus-mediated wild-type p53 gene transfer in combination with bronchial arterial infusion for treatment of advanced non-small-cell lung cancer,one year follow-up显示文摘Objective:In the present study,we have examined the safety and efficacy of recombinant adenovirus encoding human p53 tumor suppressor gene(rAd-p53) injection in patients with advanced non-small-cell lung cancer(NSCLC) in the combination with the therapy of bronchial arterial infusion(BAI).Methods:A total of 58 patients with advanced NSCLC were enrolled in a non-randomized,two-armed clinical trial.Of which,19 received a combination treatment of BAI and rAd-p53(the combo group),while the remaining 39 were treated with only BAI(the control group).Patients were followed up for 12 months,with safety and local response evaluated by the National Cancer Institute's Common Toxicity Criteria and response evaluation criteria in solid tumor(RECIST),respectively.Time to progression(TTP) and survival rates were also analyzed by Kaplan-Meier method.Results:In the combo group,19 patients received a total of 49 injections of rAd-p53 and 46 times of BAI,respectively,while 39 patients in the control group received a total of 113 times of BAI.The combination treatment was found to have less adverse events such as anorexia,nausea and emesis,pain,and leucopenia(P<0.05) but more arthralgia,fever,influenza-like symptom,and myalgia(P<0.05),compared with the control group.The overall response rates(complete response(CR)+partial response(PR)) were 47.3% and 38.4% for the combo group and the control group,respectively(P>0.05).Patients in the combo group had a longer TTP than those in the control group(a median 7.75 vs 5.5 months,P=0.018).However,the combination treatment did not lead to better survival,with survival rates at 3,6,and 12 months in the combo group being 94.74%,89.47%,and 52.63%,respectively,compared with 92.31%,69.23%,and 38.83% in the control group(P=0.224).Conclusion:Our results show that the combination of rAd-p53 and BAI was well tolerated in patients with NSCLC and may have improved the quality of life and delayed the disease progression.A further study to better determine the efficacy of this combination therapy is warranted.Yong-song GUAN Yuan LIU Qing ZOU Qing HE Zi LA Lin YANG Ying HU 2009Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2009,10,5:20
3Aqueous Supercapacitors on Conductive Cotton显示文摘Wearable electronics offer the combined advantages of both electronics and fabrics.In this article,we report the fabrication of wearable supercapacitors using cotton fabric as an essential component.Carbon nanotubes are conformally coated onto the cotton fibers,leading to a highly electrically conductive interconnecting network.The porous carbon nanotube coating functions as both active material and current collector in the supercapacitor.Aqueous lithium sulfate is used as the electrolyte in the devices,because it presents no safety concerns for human use.The supercapacitor shows high specific capacitance(~70-80 F·g^(-1) at 0.1 A·g^(-1))and cycling stability(negligible decay after 35,000 cycles).The extremely simple design and fabrication process make it applicable for providing power in practical electronic devices.Mauro Pasta Fabio La Mantia Liangbing Hu Heather Dawn Deshazer Yi Cui 2010Nano Research2010,3,6:9
4Study of temperature and precipitation change in upstream mountain area of the Hexi inland river basin since 1960s显示文摘All rivers in the Hexi inland region of Gansu Province, China, originate from the northern slope of the Qilian Mountains. They are located in the southern portion of the region and respectively belong to the three large river systems from east to west, the Shiyang, Heihe and Shule river basins. These rivers are supplied by precipitation, snowmelt and ice-melt runoff from the Qilian Mountain area. Therefore, changes of precipitation and temperature in the upstream watersheds of these rivers have an important effect on changes of mountainous runoff and reasonable utilization of water resources in this region. For this reason, the Qilian Mountain area, upstream watersheds and runoff forming areas of these rivers are chosen as the study area. The change characteristics and variation trend of temperature and precipitation in this area under the backdrop of global warming are analyzed based on observational data of relational weather and hydrologic stations in the area. Results show that temperatures in the upriver mountain areas of these three large river basins have been increasing, although the increasing degree is differentially affected by global warming. The rising extent of annual and seasonal temperatures in the upstream mountain area of the Shule river basin located in the western Qilian Mountains, were all largest over the past 50 years. Precipitation in the upstream mountain areas of Hexi region' three river basins located respectively in the western, middle and eastern Qilian Mountains have been presenting an increasing trend to varying degrees as a whole for more than 50 years. 5The Tibetan Plateau Surface–Atmosphere Coupling System and Its Weather and Climate Effects: The Third Tibetan Plateau Atmospheric Science Experiment显示文摘The Tibetan Plateau(TP) is a key area affecting forecasts of weather and climate in China and occurrences of extreme weather and climate events over the world. The China Meteorological Administration, the National Natural Science Foundation of China, and the Chinese Academy of Sciences jointly initiated the Third Tibetan Plateau Atmospheric Science Experiment(TIPEX-Ⅲ) in 2013, with an 8–10-yr implementation plan. Since its preliminary field measurements conducted in 2013, routine automatic sounding systems have been deployed at Shiquanhe, Gaize, and Shenzha stations in western TP, where no routine sounding observations were available previously. The observational networks for soil temperature and soil moisture in the central and western TP have also been established. Meanwhile, the plateau-scale and regional-scale boundary layer observations, cloud–precipitation microphysical observations with multiple radars and aircraft campaigns, and tropospheric–stratospheric air composition observations at multiple sites, were performed. The results so far show that the turbulent heat exchange coefficient and sensible heat flux are remarkably lower than the earlier estimations at grassland, meadow, and bare soil surfaces of the central and western TP. Climatologically, cumulus clouds over the main body of the TP might develop locally instead of originating from the cumulus clouds that propagate northward from South Asia. The TIPEX-Ⅲ observations up to now also reveal diurnal variations, macro-and microphysical characteristics, and water-phase transition mechanisms, of cumulus clouds at Naqu station. Moreover, TIPEX-Ⅲ related studies have proposed a maintenance mechanism responsible for the Asian 'atmospheric water tower' and demonstrated the effects of the TP heating anomalies on African, Asian, and North American climates. Additionally, numerical modeling studies show that the Γ distribution of raindrop size is more suitable for depicting the TP raindrop characteristics compared to the M–P distribution, the overestimation of sensible heat flux can be reduced via modifying the heat transfer parameterization over the TP, and considering climatic signals in some key areas of the TP can improve the skill for rainfall forecast in the central and eastern parts of China.
