| 1 | Over-expression of an Arabi-dopsisd δ-OAT gene enhances salt and drought tolerance in transgenic rice显示文摘d-OAT, ornithine--aminotransferase, is the key enzyme involved in proline biosynthesis. In this study the Arabidopsis d -OAT gene was transferred into rice (Oryza sativa L. ssp japonica cv. Zhongzuo 321), whose successful integration was demonstrated by PCR and Southern blot analysis. The over-expression of the gene in transgenic rice was also confirmed. Biochemical analysis showed that, under salt or drought stress conditions, proline contents in the leaves and roots in transgenic rice plants were 5- to 15-fold of those in non-transgenic controls. Under stress conditions, germinating rate of transgenic lines is higher than that of controls. Although the growth of rice plants tested were more and more retarded with the increasing of NaCl concentration, the transgenic plants grow faster compared to the controls under the same stress condition. Meanwhile, the resistance to KCl and MgSO4 stresses was also found enhanced in trans-genic rice. Furthermore, the over-expression of d -OAT also improved the yield of transgenic plants under stress condi-tions. The average yield per plant of transgenic lines in-creases about 12%—41% more than that of control lines under 0.1 mol/L NaCl stress. | 2 | EXAFS study on arsenic species and transformation in arsenic hyperaccumulator显示文摘Synchrotron radiation extended X-ray absorption fine structure (SR EXAFS) was employed to study the transformation of coordination environment and the redox speciation of arsenic in a newly discovered arsenic hyperaccumulator, Cretan brake (Pteris cretica L. var ner-vosa Thunb). It showed that the arsenic in the plant mainly coordinated with oxygen, except that some arsenic coordinated with S as As-GSH in root. The complexation of arsenic with GSH might not be the predominant detoxification mechanism in Cretan brake. Although some arsenic in root presented as As(V) in Na2HAsO4 treatments, most of arsenic in plant presented as As(III)-O in both treatments, indicating that As(V) tended to be reduced to As(III) after it was taken up into the root, and arsenic was kept as As(III) when it was transported to the above-ground tissues. | 3 | The input/removal rates of size-fractionated ^210Po in the Jiulongjiang estuary and its implications显示文摘The geochemical behaviors of dissolved (<0.4 mm), low molecular weight (<10 kDa) and colloidal (10 kDa- 0.4 mm) 210Po in surface waters from the Jiulongjiang estuary were examined and the input/removal rates of different 210Po species were estimated. Both dissolved and low molecular weight 210Po were considerably removed during estuarine mixing. However, colloidal 210Po was removed only at low salinity regions (S <15) but had input behavior at high salin-ity surface waters (S >15). The input/removal rates of three species indicate that 210Po was removed rapidly in the Jiu-longjiang estuary. The discrepancy of input/removal rates between 210Po species suggests different geochemical be-havior and a dynamic transformation between 210Po species. | 5 | Different patterns of agonist-induced modulation of α_(1B)-adrenoceptor density at different initial expression levels显示文摘We compared the norepinephrine (NE) induced α1B-adrenoceptor (α1B-AR) expression modulation between two transfected human embryonic kidney (HEK) 293 cell lines in which α1B-AR densities were (6 336 ±913) and (773±164) fmol · mg -1, respectively. Treatment of cells with NE (10 μmol · L1) for 48 h decreased high-level expressed α1B-AR density, but increased low-level expressed α1B-AR density. The protein kinase C inhibitor Calphostin C or Ro-31-8220 reversed, and its activator PMA mimicked the NE-induced down-regulation of high-level expressed α1B-AR. Moreover, PMA induced a down-regulation of low-level expressed α1B-AR. The endoplasmic reticulum Ca2+-ATPase inhibitor cyclopiazonic acid (CPA) and the calcium chelator BAPTA/AM did not affect the down-regulation of high-level expressed α1B-AR, but inhibited the up-regulation of low-level expression α1B-AR induced by NE. | 6 | Construction of cytopathic PK-15 cell model of classical swine fever virus显示文摘No cytopathic effect (CPE) can be observed on classical swine fever virus (CSFV) infected cell culture in vitro. This brings an obstacle to the researches on reciprocity between CSFV and host cells. Based on the construction of full-length genomic infectious Cdna clone of Chinese CSFV standard virulent Shimen strain, partial deletion is intro- duced into genomic Cdna to obtain a 7.5 kb subgenomic Cdna. A new subgenomic CSFV is derived from transfection with the subgenomic Cdna on PK-15 cells pre-infected by CSFV Shimen virus. Typical CPE induced by this subgenomic virus is observed on PK-15 cells. Coexistence of wild- type and subgenomic virus in cytopathic cell culture is dem- onstrated by RT-PCR detection in cytopathic cells. | 8 | Cardiac protective role of a novel erythrocyte-derived depressing factor on rats and its Ca^2+ mechanism显示文摘The cardiac protective role of a novel erythro-cyte-derived depressing factor (EDDF) on spontaneous hy-pertensive rats (SHR), calcium overload (CaO) rats and Wistar rats and its mechanism was evaluated. Mean artery pressure (MAP), heart rate (HR) and LVdp/dtmax were measured by physiological recorder. The effect of EDDF on the Ca2+-ATPase activity in myocardial sarcoplasmic reticu-lum (SR) of CaO rats was determined by inorganic phos-phate assay. Calcium transport in myocytes was measured by 45Ca2+ radioactive isotope measurement. The phosphoryla-tion levels of extracellular signal-regulated protein kinases (ERK1/2) in myocardial tissue of SHR and CaO rats were measured by Western blot method. And the ultrastructures of cardiac muscle cells were observed with the transmission electron microscope. The results indicated that EDDF could significantly decrease MAP, HR and LVdp/dtmax in a dose dependent manner (P < 0.05). It seems that the mechanism might relate with activating the Ca2+-APTase, enhancing the uptake and release of Ca2+ from SR (P < 0.05), decreasing the phosphorylation levels of ERK1/2 of myocytes (P < 0.01) and lightening the ultrastructural lesion of cardiac muscle cells. In CaO rats, the Ca2+-ATPase activity decreased clearly com-pared to control (64.99 7.16 vs 94.48 7.68 nmol·min-1 ·mg-1 protein, P < 0.01), while EDDF (100 mg/mL) could significantly increase the activity (87.93 ?9.54 vs 64.99 ?7.16, P < 0.05, n = 7). Both uptake and release rate of Ca2+ (祄ol 45Ca2+/g protein/min) from myocardial SR of CaO rats re-markably decreased compared to control (32.40 ?2.70 and 15.46 ?1.49 vs 61.09 ?10.89 and 25.47 ?4.29, P < 0.05); EDDF (100 mg/mL) could significantly stimulate their activi-ties (50.48 6.76 and 21.76 2.75 vs 32.40 2.70 and 15.46 1.49, P < 0.05). EDDF could evidently down-regulate the phosphorylation of ERK1/2 in myocardial tissue from SHR and CaO rats (P < 0.01), lighten the ultrastructural lesion of cardiac muscle cells of SHR as well. | 9 | Comparison of K_(IC) Values for SiC Whisker Reinforced Ceramic Composites Obtained by Using Various Methods显示文摘The fracture toughness (KIC) values determined by indentation microfracture method (IM ) for SiC whisker reinforced Al2O3 and ZrO2 based composites were calculated with different IM equations and compared with those obtained by singte edge notched beam (SENB) technique. Experimental results show that the KIC (IM) values calculated with different equations are quite different one from another. For composites without phase transformable components the KIC (IM) and KIC (SENB) values are practically on the same level, but for composites with phase transformable components (partially stabilized zirconia) the KIC (SENB) values are always higher than KIC (IM). This is because that the IM method can not reveal sensitively the toughening effect due to dynamic t-m transformation of ZrO2 as the SENB method does. The accuracy of the IM method depends on the Suitability of the IM equations and was evaluated for the materials used in this investigation. | 12 | Studies on the Construction Parameter of an Artificial Occluded Cell for In-situ Inspection of the Propagation Rate of Localized Corrosion显示文摘An artificial localized corrosion system is assembled and some parameters related to the localized corrosion in active dissolution state (i.e., non-passive state) have been studied. The results showed that the developed electrochemical system can satisfactorily imitate a naturally formed localized corrosion and the coupling current can indicate the maximum localized propagating rate. In this artificial system, the anodic dissolution reaction followed the auto-catalytic mechanism. The localized corrosion current density was dependent on the area ratio R of the cathode to the occluded anode. While R was equal to or more than 6, the coupling current reached at a maximum value and did not alter with the increase in R-value. Therefore, R=7 is chosen as one of these optimum parameters used in constructing the system, with which the biggest galvanic current might be obtained. In contrast, the thickness of the polymer filler separating the occluded anode area from the bulk electrolyte solution and the volume of the occluded anode area did not affect the corrosion current obviously.
