1Drug resistance mechanisms and novel drug targets for tuberculosis therapy显示文摘Drug-resistant tuberculosis(TB) poses a significant challenge to the successful treatment and control of TB worldwide.Resistance to anti-TB drugs has existed since the beginning of the chemotherapy era.New insights into the resistant mechanisms of anti-TB drugs have been provided.Better understanding of drug resistance mechanisms helps in the development of new tools for the rapid diagnosis of drugresistant TB.There is also a pressing need in the development of new drugs with novel targets to improve the current treatment of TB and to prevent the emergence of drug resistance in Mycobacterium tuberculosis.This review summarizes the anti-TB drug resistance mechanisms,furnishes some possible novel drug targets in the development of new agents for TB therapy and discusses the usefulness using known targets to develop new anti-TB drugs.Whole genome sequencing is currently an advanced technology to uncover drug resistance mechanisms in M.tuberculosis.However,further research is required to unravel the significance of some newly discovered gene mutations in their contribution to drug resistance.Md Mahmudul Islam H.M. 2Stripe Rust Effector PstGSRE1 Disrupts Nuclear Localization of ROS-Promoting Transcription Factor TaLOL2 to Defeat ROS-Induced Defense in Wheat显示文摘Puccinia striiformis f.sp.tritici(Pst),a biotrophic plant pathogen,secretes numerous effectors to modulate host defense systems.Understanding the molecular mechanisms by which Pst effectors regulate wheat immunity is of great importance for the development of novel strategies for durable control of stripe rust.In this study,we identified a glycine-serine-rich effector gene,PstGSRE1,which is highly induced dur-ing early infection.Transgenic expression of PstGSRE1 RNAi constructs in wheat significantly reduced virulence of Pst and increased H2O2 accumulation in wheat.PstGSRE1 was shown to target the reactive ox-ygen species(ROS)-associated transcription factor TaLOL2,a positive regulator of wheat immunity.PstGSRE1 disrupted nuclear localization of TaLOL2 and suppressed ROS-mediated cell death induced by TaLOL2,thus compromising host immunity.This work reveals a previously unrecognized strategy whereby rust fungi exploit the PstGSRE1 effector to defeat ROS-associated plant defense by modulating the subcellular compartment of a host immune regulator and facilitate pathogen infection.Tuo Qi Jia Guo Peng Liu Fuxin He Cuiping Wan Md Ashraful Islam Brett M.Tyler Zhensheng Kang Jun Guo 2019Molecular Plant2019,12,12:11
3One-step synthesis of N-doped metal/biochar composite using NH_3-ambiance pyrolysis for efficient degradation and mineralization of Methylene Blue显示文摘A series of new biochar-supported composite based on the combination of biochar and metallic nanoparticles(NPs)were produced through single-step pyrolysis of FeCl_3–Ti(OBu)_4 laden agar biomass under NH_3 environment.The physiochemical properties of composites were characterized thoroughly.It has found that heating temperature and N-doping through NH_3-ambiance pyrolysis significantly influence the visible-light sensitivity and bandgap energy of composites.The catalytic activities of composites were measured by degradation of Methylene Blue(MB)in the presence or absence of H_2O_2 and visible-light irradiation.Our best catalyst(N–TiO_2–Fe_3O_4-biochar)exhibits rapid and high MB removal competency(99.99%)via synergism of adsorption,photodegradation,and Fenton-like reaction.Continuous production of O_2U^-and UOH radicles performs MB degradation and mineralization,confirmed by scavenging experiments and degradation product analysis.The local trap state Ti^(3+),Fe_3O_4,and N-carbon of the catalyst acted as active sites.It has suggested that the Ti^(3+)and N-doped dense carbon layer improve charge separation and shuttle that prolonged photo-Fenton like reaction.Moreover,the catalyst is highly stable,collectible,and recyclable up to 5 cycles with high MB degradation efficiency.This work provides a new insight into the synthesis of highly visible-light sensitized biocharsupported photocatalyst through NH_3-ambiance pyrolysis of NPs-laden biomass.Md Manik Mian Guijian Liu Balal Yousaf Biao Fu Rafay Ahmed Qumber Abbas Mehr Ahmed Mujtaba Munir Liu Ruijia 2019Journal of Environmental Sciences2019,31,4:8
4Physicochemical and Sensory Properties of japonica Rice Varied with Production Areas in China显示文摘Northeast of China and Jiangsu Province are major production areas of japonica rice in China.Rice from northeast of China is well-known for its good-eating and appearance quality,and that from Jiangsu Province is viewed as inferior.However,little is known concerning the difference in physicochemical and sensory properties of rice between the major two production areas.Analysis of 16 commercial rice samples showed marked differences in physicochemical properties,including chalky grain rate,contents of amylose and protein and pasting properties between the two main areas.Northeastern rice contained more shortchain amylopectin as compared with Jiangsu rice.However,Jiangsu rice is comparable to northeastern rice in terms of sensory quality including overall acceptability and textural properties of springiness,stickiness and hardness as evaluated by trained panel.Our results indicated the limitation of conventional index of physicochemical properties,and suggested the necessity of identification of new factors controlling rice sensory property.In addition,the taste analyzer from Japan demonstrates limitation in distinguishing the differences between northeastern and Jiangsu rice,and therefore needs localization to fit China.YANG Xiao-yu LIN Zhao-miao LIU Zheng-hui Md A Alim BI Jun-guo LI Gang-hua WANG Qiang-sheng WANG Shao-hua DING Yan-feng 2013Journal of Integrative Agriculture2013,12,10:8
5MicroRNA and mRNA profiling of cerebral cortex in a transgenic mouse model of Alzheimer’s disease by RNA sequencing显示文摘In a previous study,we found that long non-coding genes in Alzheimer’s disease(AD)are a result of endogenous gene disorders caused by the recruitment of microRNA(miRNA)and mRNA,and that miR-200a-3p and other representative miRNAs can mediate cognitive impairment and thus serve as new biomarkers for AD.In this study,we investigated the abnormal expression of miRNA and mRNA and the pathogenesis of AD at the epigenetic level.To this aim,we performed RNA sequencing and an integrative analysis of the cerebral cortex of the widely used amyloid precursor protein and presenilin-1 double transgenic mouse model of AD.Overall,129 mRNAs and 68 miRNAs were aberrantly expressed.Among these,eight down-regulated miRNAs and seven up-regulated miRNAs appeared as promising noninvasive biomarkers and therapeutic targets.The main enriched signaling pathways involved mitogen-activated kinase protein,phosphatidylinositol 3-kinase-protein kinase B,mechanistic target of rapamycin kinase,forkhead box O,and autophagy.An miRNA-mRNA network between dysregulated miRNAs and corresponding target genes connected with AD progression was also constructed.These miRNAs and mRNAs are potential biomarkers and therapeutic targets for new treatment strategies,early diagnosis,and prevention of AD.The present results provide a novel perspective on the role of miRNAs and mRNAs in AD.This study was approved by the Experimental Animal Care and Use Committee of Institute of Medicinal Biotechnology of Beijing,China(approval No.IMB-201909-D6)on September 6,2019.Li Zeng Hai-Lun Jiang Ghulam Md Ashraf Zhuo-Rong Li Rui Liu 2021Neural Regeneration Research2021,16,10:6
6Proteome analysis of alfalfa roots in response to water deficit stress显示文摘To evaluate the response of alfalfa to water deficit(WD) stress, WD-induced candidates were investigated through a proteomic approach. Alfalfa seedlings were exposed to WD stress for 12 and 15 days respectively, followed by 3 days re-watering. Water deficit increased H2O2 content, lipid peroxidation, DPPH(1,1-diphenyl-2-picrylhydrazyl)-radical scavenging activity, and the free proline level in alfalfa roots. Root proteins were extracted and separated by two-dimentional polyacrylamide gel electrophoresis(2-DE). A total of 49 WD-responsive proteins were identified in alfalfa roots; 25 proteins were reproducibly found to be up-regulated and 24 were down-regulated. Two proteins, namely cytosolic ascorbate peroxidase(APx2) and putative F-box protein were newly detected on 2-DE maps of WD-treated plants. We identified several proteins including agamous-like 65, albumin b-32, inward rectifying potassium channel, and auxin-independent growth promoter. The identified proteins are involved in a variety of cellular functions including calcium signaling, abacisic acid(ABA) biosynthesis, reactive oxygen species(ROS) regulation, transcription/ translation, antioxidant/detoxification/stress defense, energy metabolism, signal transduction, and storage. 7Putrescine, spermidine, and spermine play distinct roles in rice salt tolerance显示文摘Polyamines(PAs) play diverse roles in plant growth and development,as well as responses to environmental stimuli.In this study,the effects of PAs on rice salt tolerance were investigated.Salt stress resulted in the alteration of endogenous PAs levels in rice roots and leaves,where spermidine(Spd) and spermine(Spm) contents were increased,and putrescine(Put) content was decreased.RT-qPCR analysis revealed that PAs biosynthesis-related genes ADC1,ODC,and Arginase were significantly downregulated by salt treatment;however,SAMDC transcription was significantly upregulated.Exogenous Spm enhanced rice salt tolerance remarkably;however,exogenous Put and Spd undermined rice salt tolerance.Transgenic rice plants overexpressing SAMDC display a higher ratio of Spm/(Put+Spd) and enhanced salt tolerance.Salt stress also increased polyamine oxidase activities in rice,resulting in elevated reactive oxygen species(ROS) production.Our findings revealed that accumulation of Put and Spd substantially reduced salt tolerance in rice,likely by facilitating ROS production;whereas,conversion of Put and Spd to Spm contributes to rice salt tolerance.Md Azizul ISlam PANG Jin-huan MENG Fan-wei LI Ya-wen XU Ning YANG Chao LIU Jun 2020Journal of Integrative Agriculture2020,19,3:5
8Source identification, contamination status and health risk assessment of heavy metals from road dusts in Dhaka, Bangladesh显示文摘In this study, concentrations of Cr, Mn, Ni, Cu, Zn, Cd and Pb were determined in road dusts collected from different locations in Dhaka to assess source, contamination status and health risk. Energy-dispersive X-ray fluorescence spectroscopy and energy-dispersive X-ray spectroscopy were used to determine Cr, Mn, Ni, Cu, Zn, Cd and Pb and their mean concentrations were 162.27 ± 29.46, 721.18 ± 180.14, 35.65 ± 12.55, 104.56 ± 128.33, 515.32 ± 321.90,BDL, and 342.82 ± 591.20 mg/kg, respectively. Among the heavy metals, highest concentrations of Cu, Zn and Pb were found at urban sites-7(municipal waste dumping) and 8(medical waste incineration). Highest concentration of Cr followed by Cu and Zn was found at site-5(Tejgaon, urban). Principal component analysis revealed that anthropogenic activities are the potential sources for Cr, Ni, Cu, Zn and Pb while earth crust for Mn. Pollution index and pollution load index results suggested that all the sites were contaminated and/or degraded by Cr, Cu, Zn and Pb except sites-9(urban), 10(sub-urban), 11(rural) while sites-7 and 8(urban) were extremely degraded. For noncarcinogenic health risk, hazard quotient values for dermal were higher compared to that of inhalation/ingestion. Though hazard index values were less than 1 at all the sites, these were at least one order of magnitude higher for children group than that of adult group, thus the children group may face more noncarcinogenic health risk at sites -7 and 8. 9Flooding and its relationship with land cover change, population growth, and road density显示文摘Bangladesh experiences frequent hydro-climatic disasters such as flooding.These disasters are believed to be associated with land use changes and climate variability.However,identifying the factors that lead to flooding is challenging.This study mapped flood susceptibility in the northeast region of Bangladesh using Bayesian regularization back propagation(BRBP)neural network,classification and regression trees(CART),a statistical model(STM)using the evidence belief function(EBF),and their ensemble models(EMs)for three time periods(2000,2014,and 2017).The accuracy of machine learning algorithms(MLAs),STM,and EMs were assessed by considering the area under the curve—receiver operating characteristic(AUC-ROC).Evaluation of the accuracy levels of the aforementioned algorithms revealed that EM4(BRBP-CART-EBF)outperformed(AUC>90%)standalone and other ensemble models for the three time periods analyzed.Furthermore,this study investigated the relationships among land cover change(LCC),population growth(PG),road density(RD),and relative change of flooding(RCF)areas for the period between 2000 and 2017.The results showed that areas with very high susceptibility to flooding increased by 19.72%between 2000 and 2017,while the PG rate increased by 51.68%over the same period.The Pearson correlation coefficient for RCF and RD was calculated to be 0.496.These findings highlight the significant association between floods and causative factors.The study findings could be valuable to policymakers and resource managers as they can lead to improvements in flood management and reduction in flood damage and risks.Mahfuzur Rahman Chen Ningsheng Golam Iftekhar Mahmud Md Monirul Islam Hamid Reza Pourghasemi Hilal Ahmad Jules Maurice Habumugisha Rana Muhammad Ali Washakh Mehtab Alam Enlong Liu Zheng Han Huayong Ni Tian Shufeng Ashraf Dewan 2021Geoscience Frontiers2021,12,6:3
10Differences between soybean genotypes in physiological response to sequential soil drying and rewetting显示文摘Soybean genotypes show diverse physiological responses to drought, but specific physiological traits that can be used to evaluate drought tolerance have not been identified. In the present study we investigated physiological traits of soybean genotypes under progressive soil drying and rewetting, using a treatment mimicking field conditions.After a preliminary study with eight soybean genotypes, two drought-tolerant genotypes and one susceptible genotype were grown in the greenhouse and subjected to water restriction. Leaf expansion rate, gas exchange, water relation parameters, total chlorophyll(Chl), proline contents of leaves, and root xylem p H were monitored in a time course, and plant growth and root traits were measured at the end of the stress cycle. Drought-tolerant genotypes maintained higher leaf expansion rate, net photosynthetic rate(Pn), Chl content,instantaneous water use efficiency(WUEi), % relative water content(RWC), water potential(ψw), and turgor potential(ψp) during progressive soil drying and subsequent rewetting than the susceptible genotypes. By contrast, stomatal conductance(gs) and transpiration rate(Tr)of tolerant genotypes declined faster owing to dehydration and recovered more sharply after rehydration than the same parameters in susceptible ones. Water stress caused a significant increase in leaf proline level and root xylem sap p H of both genotypes but tolerant genotypes recovered to pre-stress levels more quickly after rehydration. Tolerant genotypes also produced longer roots with higher dry mass than susceptible genotypes. 11Effect of river plume on phytoplankton community structure in Zhujiang River estuary显示文摘To examine the phytoplankton assemblages and the effect of diluted waters on them,a research cruise was conducted from July 19 to August 7,2015 in the Zhujiang(Pearl)River estuary in the northern South China Sea(21°N–23.5°N,111°E–117°E).Samples were collected from 65 stations including one for time-series sampling.A total of 212 phytoplankton taxa were identified from 61 genera belonging to 4 phyla.Among them,122 species identified from 42 genera of Bacillariophyta and 83 species from 15 genera of Pyrrophyta.Chain-forming diatoms dominated the phytoplankton community where Pseudonitzschia delicatissima,Guinardia striata,Thalassionema nitzschioides,and P.pungens comprised about 52%of the total abundance.However,higher cell abundances concentrated on both sides of the estuary,because of low salinity and high nutrients brought by diluted water.In addition,Canonical Correspondence Analysis revealed that salinity and dissolved inorganic nitrogen shaped the species composition in the study area.Furthermore,the Jaccard similarity index showed prevailing high similarity in the distribution of species in low-salinity diluted waters,and the Bray-Curtis similarity depicted distinguished grouping for phytoplankton assemblages along the salinity gradient.However,phytoplankton diel vertical cycles showed maximum abundance occurred at 2:00 am,which was mainly contributed by benthic phytoplankton species Pseudo-nitzschia spp.and T.nitzschioides.Qi ZHONG Bing XUE Md Abu NOMAN Yuqiu WEI Haijiao LIU Hongbin LIU Liping ZHENG Hongmei JING Jun SUN 2021Journal of Oceanology and Limnology2021,39,2:2

14Advances in the development of molecular genetic tools for Mycobacterium tuberculosis显示文摘Mycobacterium tuberculosis, a Gram-positive bacterium of great clinical relevance, is a lethal pathogen owing to its complex physiological characteristics and development of drug resistance. Several molecular genetic tools have been developed in the past few decades to study this microorganism. These tools have been instrumental in understanding how M. tuberculosis became a successful pathogen. Advanced molecular genetic tools have played a significant role in exploring the complex pathways involved in M. tuberculosis pathogenesis. Here, we review various molecular genetic tools used in the study of M. tuberculosis. Further, we discuss the applications of clustered regularly interspaced short palindromic repeat interference(CRISPRi), a novel technology recently applied in M. tuberculosis research to study target gene functions.
