1Drug resistance and new therapies in colorectal cancer显示文摘Colorectal cancer(CRC) is often diagnosed at an advanced stage when tumor cell dissemination has taken place. Chemo-and targeted therapies provide only a limited increase of overall survival for these patients. The major reason for clinical outcome finds its origin in therapy resistance. Escape mechanisms to both chemo-and targeted therapy remain the main culprits. Here, we evaluate major resistant mechanisms and elaborate on potential new therapies. Amongst promising therapies is α-amanitin antibodydrug conjugate targeting hemizygous p53 loss. It becomes clear that a dynamic interaction with the tumor microenvironment exists and that this dictates therapeutic outcome. In addition, CRC displays a limited response to checkpoint inhibitors, as only a minority of patients with microsatellite instable high tumors is susceptible. 5Antibiotic Treatment Drives the Diversification of the Human Gut Resistome显示文摘Despite the documented antibiotic-induced disruption of the gut microbiota, the impact of antibiotic intake on strain-level dynamics, evolution of resistance genes, and factors influencing resistance dissemination potential remains poorly understood. To address this gap we analyzed public metagenomic datasets from 24 antibiotic treated subjects and controls, combined with an in-depth prospective functional study with two subjects investigating the bacterial community dynamics based on cultivation-dependent and independent methods. We observed that shortterm antibiotic treatment shifted and diversified the resistome composition, increased the average copy number of antibiotic resistance genes, and altered the dominant strain genotypes in an individual-specific manner. More than 30% of the resistance genes underwent strong differentiation at the single nucleotide level during antibiotic treatment. We found that the increased potential for horizontal gene transfer, due to antibiotic administration, was ~3-fold stronger in the differentiated resistance genes than the non-differentiated ones. 7Comparative studies on fouling of homogeneous anion exchange membranes by different structured organics in electrodialysis显示文摘Five negatively charged organic compounds with different structures, sodium methane sulfonate(MS), sodium benzene sulfonate(BS), sodium 6-hydroxynaphthalene-2-sulfonate(NSS), sodium dodecyl sulfate(SDS), and sodium dodecyl benzene sulfonate(SDBS), were used to examine the fouling of an anion exchange membrane(AEM) in electrodialysis(ED),to explore the effect of molecular characteristics on the fouling behavior on the AEM and changes in the surface and electrochemical properties of the AEM. Results indicated that the fouling degree of the AEM by the different organics followed the order:SDBS > SDS > NSS > BS > MS. SDBS and SDS formed a dense fouling layer on the surface of the AEM, which was the main factor in the much more severe membrane fouling, and completely restricted the transmembrane ion migration. The other three organics caused fouling of the AEM by adsorption on the surface and/or accumulation in the interlayer of the AEM, and exhibited almost no influence on the transmembrane ion migration. 8Perspectives of CRISPR/Cas-mediated cisengineering in horticulture:unlocking the neglected potential for crop improvement显示文摘Directed breeding of horticultural crops is essential for increasing yield,nutritional content,and consumer-valued characteristics such as shape and color of the produce.However,limited genetic diversity restricts the amount of crop improvement that can be achieved through conventional breeding approaches.Natural genetic changes in cisregulatory regions of genes play important roles in shaping phenotypic diversity by altering their expression.Utilization of CRISPR/Cas editing in crop species can accelerate crop improvement through the introduction of genetic variation in a targeted manner.The advent of CRISPR/Cas-mediated cis-regulatory region engineering(cis-engineering)provides a more refined method for modulating gene expression and creating phenotypic diversity to benefit crop improvement.Here,we focus on the current applications of CRISPR/Cas-mediated cis-engineering in horticultural crops.We describe strategies and limitations for its use in crop improvement,including de novo cis-regulatory element(CRE)discovery,precise genome editing,and transgene-free genome editing.In addition,we discuss the challenges and prospects regarding current technologies and achievements.CRISPR/Cas-mediated cis-engineering is a critical tool for generating horticultural crops that are better able to adapt to climate change and providing food for an increasing world population.Qiang Li Manoj Sapkota Esther van der Knaap 2020Horticulture Research2020,7,1:3

12Computer simulation of W-C-Co-V system diffusion couples显示文摘Diffusion couples,WC/Co-1at.%C,V8C7/Co,and WC/Co-0.3wt.%V8C7 were constructed and annealed at tem-peratures ranging from 1100 to 1300°C. The concentration profile of V and W atoms in these couples were measured by EPMA. Aided by Thermo-calc & DICTRA software package,the concentration profiles of W and V in established diffu-sion couples were calculated by the moving boundary model according to revised dynamic parameters and MOB database,respectively. Then the concentration profile of W in Co binder during sintering WC-Co cemented carbide was simulated by the Cell model. It can provide reference for composition and sintering process design of cemented carbides with high per-formance. 13Efficient photon coupling from a diamond nitrogen vacancy center by integration with silica fiber显示文摘A central goal in quantum information science is to efficiently interface photons with single optical modes for quantum networking and distributed quantum computing.Here,we introduce and experimentally demonstrate a compact and efficient method for the low-loss coupling of a solid-state qubit,the nitrogen vacancy(NV)center in diamond,with a single-mode optical fiber.In this approach,single-mode tapered diamond waveguides containing exactly one high quality NV memory are selected and integrated on tapered silica fibers.Numerical optimization of an adiabatic coupler indicates that near-unity-efficiency photon transfer is possible between the two modes.Experimentally,we find an overall collection efficiency between 16%and 37%and estimate a single photon count rate at saturation above 700 kHz.This integrated system enables robust,alignment-free,and efficient interfacing of single-mode optical fibers with single photon emitters and quantum memories in solids.Rishi N Patel Tim Schröder Noel Wan Luozhou Li Sara L Mouradian Edward H Chen Dirk R Englund 2016Light(Science & Applications)2016,5,1:2
14Post-hepatectomy liver regeneration in the context of bile acid homeostasis and the gut-liver signaling axis显示文摘Background:Liver regeneration following partial hepatectomy(PHx)is a complicated process involving multiple organs and several types of signaling networks.The bile acid-activated metabolic pathways occupy an auxiliary yet important chapter in the entire biochemical story.PHx is characterized by rapid but transient bile acid overload in the liver,which constitutes the first wave of proliferative signaling in the remnant hepatocytes.Bile acids trigger hepatocyte proliferation through activation of several nuclear receptors.Following biliary passage into the intestines,enterocytes reabsorb the bile acids,which results in the activation of farnesoid X receptor(FXR),the consequent excretion of fibroblast growth factor(FGF)19/FGF15,and its release into the enterohepatic circulation.FGF19/FGF15 subsequently binds to its cognate receptor,fibroblast growth factor receptor 4(FGFR4)complexed withβ-klotho,on the hepatocyte membrane,which initiates the second wave of proliferative signaling.Because some bile acids are toxic,the remnant hepatocytes must resolve the potentially detrimental state of bile acid excess.Therefore,the hepatocytes orchestrate a bile acid detoxification and elimination response as a protective mechanism in concurrence with the proliferative signaling.The response in part results in the excretion of(biotransformed)bile acids into the canalicular system,causing the bile acids to end up in the intestines.Relevance for patients:Recently,FXR agonists have been shown to promote regeneration via the gut-liver axis.This type of pharmacological intervention may prove beneficial for patients with hepatobiliary tumors undergoing PHx.In light of these developments,the review provides an in-depth account of the pathways that underlie post-PHx liver regeneration in the context of bile acid homeostasis in the liver and the gut-liver signaling axis.Lianne de Haan Sarah Jvan der Lely Anne-Loes K.Warps Quincy Hofsink Pim B.Olthof Mark J.de Keijzer Daniel A.Lionarons Lionel Mendes-Dias Bote G.Bruinsma Korkut Uygun Hartmut Jaeschke Geoffrey C.Farrell Narci Teoh Rowan Fvan Golen Tiangang Li Michal Heger 2018Journal of Clinical & Translational Research2018,4,1:2
