1Structure and regulatory function of plant transcription factors显示文摘The expression of inducible genes in plants is regulated by specific transcription factors at the transcrip-tional level. A typical transcription factor usually contains a DNA-binding domain, a transcription regulation domain, a dimerization site and a nuclear localization domain. These functional domains define the characteristic, localization and regulatory role of a transcription factor. Transcription factors recognize and bind to specific cis-acting elements or interact with other proteins, and then activate or repress the transcription of target genes by their functional domains. 2Regulation of embryonic stem cell self-renewal and differentiation by TGF-β family signaling显示文摘Embryonic stem (ES) cells are characterized by their ability to indefinitely self-renew and potential to differentiate into all the cell lineages of the body. ES cells are considered to have potential applications in regenerative medicine. In particular, the emergence of an ES cell analogue-induced pluripotent stem (iPS) cells via somatic cell reprogramming by co-expressing a limited number of critical stemness-related transcriptional factors has solved the problem of obtaining patient-specific pluripotent cells, encouraging researchers to develop more specific and functional cell lineages from ES or iPS cells for broad therapeutic applications. ES cell fate choice is delicately controlled by a core transcriptional network, epigenetic modification profiles and complex signaling cascades both intrinsically and extrinsically. Of these signals, transforming growth factor β (TGF-β) family members, including TGF-β, bone morphogenetic protein (BMP), Activin and Nodal, have been reported to influence cell self-renewal and a broad spectrum of lineage differentiation in ES cells, in accordance with the key roles of TGF-β family signaling in early embryo development. 3Plant protein kinase genes induced by drought, high salt and cold stresses显示文摘Drought, high salt and cold are three different kinds of environment stresses that severely influence the growth, development and productivity of crops. They all decrease the water state of plant cells, and consequently result in the harm of plant from water deficit. Several genes encoding protein kinases and induced by drought, high salt and low temperature have been isolated from Arabidopsis. These protein kinases include receptor protein kinase (RPK), MAP kinases, ribosomal-protein kinases and transcription-regulation protein kinase. 4Effects of disinhibition on spatiotemporal pattern of neuronal first recruitment in neuronal networks显示文摘The propagation of neuronal activities is a key feature to understanding information processing in networks. The analysis based on first-spikes of bursts in turn plays an important role in the research of neuronal activity propagation. Our focus here is to investigate how spatiotemporal patterns of neuronal first-spikes are affected by disinhibition. Multi-electrode arrays were used to record stimulation-evoked bursts of multiple neurons in randomly cultured neuronal networks. Both the precise timing of and the rank relationships between first-spikes were analyzed. Compared with evoked bursts in the network's native state, the precise first-spike latencies in its dis-inhibited state are more consistent and the propagation of its bursting activities is much faster. 5Cloning and sequence characteristics of the genomic gene of a rice metallothionein显示文摘Northern blot analysis showed that a metallothionein gene, ricMT is expressed strongly in the stem of rice with an expression level that could be more than 100-fold stronger than in leaf blades. The results suggest that the 5’ upstream region flanking the coding sequence of the ricMT may contain a fairly strong promoter To elucidatr its regulation and promoter structure, the genomic clones of ricMT were screened out from a rice genomic library and a fragment of about 4084 bp was sequenced. The fragment included a 5’ upstream region of ca. 2970 bp, a transcription region of ca. 690 bp and a 3’ downstream region of ca. 420 bp. Computer analysis of the sequence homology showed that the 5’ upstream region included a putative TATA box, a putative CAAT box, and a typical metal-responsive element TGCGCGCG. 6Short exposure to paclitaxel induces multipolar spindle formation and aneuploidy through promotion of acentrosomal pole assembly显示文摘Paclitaxel is a widely used microtubule drug and cancer medicine. Here we report that by short exposure to paclitaxel at a low dose, multipolar spindles were induced in mitotic cells without centrosome amplification. Both TPX2 depletion and Aurora-A overexpression antagonized the multipolarity. Live cell imaging showed that some paclitaxel-treated cells accomplished multipolar cell division and a portion of the daughter cells went on to the next round of mitosis. The surviving cells grew into clones with varied genome content. 7EFFECT OF DIFFERENT OPERATION METHODS ON METHANE HYDRATE FORMATION显示文摘Three experiments of static state storage method, low-temperature and constant-pressure storage method and low-temperature and constant-pressure storage method were carried out to investigate which method was best in gas hydrate. The relationships of hydrate rate, capacity and liquid temperature versus time were derived and three results were contrasted. The experimental results show lowtemperature and constant-pressure method is better than the other two methods because it's operation period is shorter and storage capacity is larger than the other two. Low-temperature and constant-pressure method is the best method. 8Advances in studies on hepatic stem cells显示文摘The question whether hepatic stem cells exist or not has been debated for several decades. Current researches confirm that there are hepatic stem cells in the liver. Oval cells, putative bipotential hepatic stem cells, are probably located within canals of Hering, portal tracts or branches of biliary trees. Bone marrow is a potential source of oval cells, indicating that there exists a close relationship between liver and hematopoiesis in adulthood. Hepatic stem cells are able to proliferate in vitro and can be induced to differentiate into hepatocytes. This will provide a promising approach of cell transplantation, tissue engineering and gene therapy for liver diseases. 18Immunoinhibitory effect of soluble HLA-G1 on the cytotoxicity of NK92 cells显示文摘HLA-G (human leukocyte antigen-G) is a non-classical HLA class I molecule, playing an important immuno-modulatory role in maintaining maternal immune tolerance of the semiallogenic fetus and organ transplantation. In this study, the cDNA sequence of extracellular domain of HLA-G1 was subcloned into the pET28a vector and a soluble 35 kD fusion protein (His-sHLA-G1) with six histine residues was obtained. In the 4 h 51Cr-release assay the fusion protein obviously inhibited the cytotoxicity of NK92 cells in a dose-dependent manner.
