1Rho/Rock signal transduction pathway is required for MSC tenogenic differentiation显示文摘Mesenchymal stem cell(MSC)-based treatments have shown promise for improving tendon healing and repair.MSCs have the potential to differentiate into multiple lineages in response to select chemical and physical stimuli, including into tenocytes. Cell elongation and cytoskeletal tension have been shown to be instrumental to the process of MSC differentiation. Previous studies have shown that inhibition of stress fiber formation leads MSCs to default toward an adipogenic lineage, which suggests that stress fibers are required for MSCs to sense the environmental factors that can induce differentiation into tenocytes. As the Rho/ROCK signal transduction pathway plays a critical role in both stress fiber formation and in cell sensation, we examined whether the activation of this pathway was required when inducing MSC tendon differentiation using rope-like silk scaffolds. To accomplish this, we employed a loss-of-function approach by knocking out ROCK,actin and myosin(two other components of the pathway) using the specific inhibitors Y-27632, Latrunculin A and blebbistatin, respectively. We demonstrated that independently disrupting the cytoskeleton and the Rho/ROCK pathway abolished the expression of tendon differentiation markers and led to a loss of spindle morphology. 3Instantaneous frequency and wave mode identification in a magnetosheath using few spatial points显示文摘Observational data such as those obtained from the magnetosheath in the downstream of Earth's bow shock have waveforms that differ from those of sinusoidal signals. In practice, they are not even aggregates of sinusoidal signals. Therefore, the frequency decomposition for the data requires technique that will account for the time-varying features of the data that will lead to deduction of physical meaning of the observations. The combination of empirical mode decomposition (EMD) and Hilbert transform has been used for extracting the various contributing oscillatory modes (EMDs) and the instantaneous frequency determination (Hilbert transform) of every physically meaningful mode called intrinsic mode function (IMF). The resulting instantaneous frequencies are used to determine instantaneous wave vectors. The combination of the instantaneous frequencies and wave vectors is useful in the identification of wave modes based on the characteristics of the waves. 11Alignment of the hindfoot following total knee arthroplasty: A systematic review显示文摘BACKGROUND There appears to be a close relationship between deformities at the knee joint and at the hindfoot in patients with knee osteoarthritis(OA).Despite this intrinsic link,there is a dearth of studies investigating alterations in hindfoot alignment following total knee arthroplasty(TKA)in patients with knee OA.AIM To evaluate changes in alignment of the hindfoot following TKA,foot and ankle clinical outcomes in terms of subjective clinical scoring tools following surgical intervention,and to analyse the level of evidence(LOE)and quality of evidence(QOE)of the included studies.METHODS MEDLINE,EMBASE and Cochrane Library databases were systematically reviewed in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines.Studies reporting changes in the postoperative alignment of the hindfoot following TKA were included.The level and QOE were recorded and assessed.RESULTS Eleven studies with a total of 1142 patients(1358 knees)met the inclusion/exclusion criteria.Six studies were of LOE II and 5 studies were of LOE III.Patients with preoperative varus knee deformity and valgus hindfoot deformity demonstrated improvement in hindfoot alignment post TKA.Patients with preoperative varus knee deformity and varus hindfoot deformity demonstrated no improvement in hindfoot alignment following TKA.Twelve different radiographic parameters were used to measure the alignment of the hindfoot across the included studies,with the tibio-calcaneal angle most frequently utilised(27.3%).CONCLUSION This systematic review demonstrated that the hindfoot may display compensatory changes in alignment following TKA in patients with knee OA.However,the marked heterogeneity between the included studies and poor QOE limits any meaningful cross sectional comparisons between studies.Further,well designed studies are necessary to determine the changes and outcomes of hindfoot alignment following TKA.James J Butler Nathaniel P Mercer Eoghan T Hurley Mohammad T Azam John G Kennedy 2021World Journal of Orthopedics2021,12,10:1
