Validation of a serum microRNA panel as biomarkers for early diagnosis of hepatocellular carcinoma post-hepatitis C infection in Egyptian patients显示文摘AIM To investigate the prospective importance of serum micro(mi)RNAs(mi R-125 b, mi R-138 b, mi R-1269, mi R-214-5p, mi R-494, mi R375 and mi R-145) as early biomarkers for the diagnosis of hepatitis C virus(HCV)-related hepatocellular carcinoma(HCC).METHODS Two-hundred and fifty HCV4 a patients, 224 HCV4 aHCC patients, and 84 healthy controls were enrolled in the study. Expression levels of mi R214-5p, mi R-125 b, mi R-1269 and mi R-375 were quantified using quantitative real-time PCR.RESULTS Expression of the selected mi RNAs in serum wassignificantly lower in HCC patients than in the healthy controls, except for mi R-1269 and mi R-494. There was a significant difference between HCC and HCV patients, in particular for HCC and late stage fibrosis, rather than HCV patients and early fibrosis. It is obvious that mi R-1269 was significantly upregulated in HCC cases compared to hepatic fibrosis cases. Each mi RNA can show HCC progression. Multivariate logistic regression analysis indicated that the tested panel of mi RNAs(mi R214-5p, mi R-125 b, mi R-1269 and mi R-375) represent accurate and specific indictors of HCC development.CONCLUSION This study presents a panel of mi RNAs with strong power as putative diagnostic and prognostic biomarkers for HCV-induced HCC. Moreover, mi R-214-5p and mi R-1269 could be considered as early biomarkers for tracking the progress of liver fibrosis to HCC. | Moustafa Nouh Elemeery Ahmed Noah Badr Marwa Anwar Mohamed Doaa Ahmed Ghareeb | 2017 | World Journal of Gastroenterology2017,23,21: | 20
Magnetic resonance imaging of the spinal marrow:Basic understanding of the normal marrow pattern and its variant显示文摘For now, magnetic resonance(MR) is the best noninvasive imaging modality to evaluate vertebral bone marrow thanks to its inherent soft-tissue contrast andnon-ionizing nature. A daily challenging scenario for every radiologist interpreting MR of the vertebral column is discerning the diseased from normal marrow. This requires the radiologist to be acquainted with the used MR techniques to judge the spinal marrow as well as its normal MR variants. Conventional sequences used basically to image marrow include T1 W, fat-suppressed T2 W and short tau inversion recovery(STIR) imaging provides gross morphological data. Interestingly, using non-routine MR sequences; such as opposed phase, diffusion weighted, MR spectroscopy and contrastedenhanced imaging; may elucidate the nature of bone marrow heterogeneities; by inferring cellular and chemical composition; and adding new functional prospects. Recalling the normal composition of bone marrow elements and the physiologic processes of spinal marrow conversion and reconversion eases basic understanding of spinal marrow imaging. Additionally, orientation with some common variants seen during spinal marrow MR imaging as hemangiomas and bone islands is a must. Moreover, awareness of the age-associated bone marrow changes as well as changes accompanying different variations of the subject's health state is essential for radiologists to avoid overrating normal MR marrow patterns as pathologic states and metigate unnecessary further work-up. | Mohamed Ragab Nouh Ahmed Fathi Eid | 2015 | World Journal of Radiology2015,7,12: | 4
MicroRNA signature in patients with hepatocellular carcinoma associated with type 2 diabetes显示文摘BACKGROUND Nonalcoholic steatohepatitis-related cirrhosis is one of the liver complications in type 2 diabetes mellitus(T2DM)and reported to be a risk factor for developing hepatocellular carcinoma(HCC).A reliable screening biomarker of liver cirrhosis(LC)and HCC among T2DM patients is important to reduce the morbidity and mortality of this disease.MicroRNA(miRNA)is considered a key player in HCC and T2DM,and it might be a hidden culprit in diabetes-associated HCC,making it a promising reliable prognostic tool.AIM To investigate the signature of serum miRNAs as early biomarkers for the screening of HCC among diabetic patients.METHODS Expression profiles of miRNAs in serum samples of diabetic LC and diabetic HCC patients were assessed using Illumina sequencing;then,RT-qPCR was used to validate significantly altered miRNAs between the two groups.Candidate miRNAs were tested in serum samples of 200 T2DM patients,270 LC patients,200 HCC patients, and 225 healthy control subjects.Additionally, receiver operating characteristic(ROC)analysis, with area under the curve(AUC),was performed to assess the diagnostic performance of the screened miRNAs for discriminating HCC from LC and nonmalignant patients(LC+T2DM).RESULTS Expression of the sequenced miRNAs in serum was different in HCC vs LCpositive T2DM patients. Two miRNAs(miR-34a,miR-221)were significantly upregulated and five miRNAs(miR-16,miR-23-3p,miR-122-5p,miR-198,miR-199a-3p)were significantly down-regulated in HCC compared to LC patients.Analysis of ROC curve demonstrated that the combination of these seven miRNAs can be used as a reliable biomarker for detection of HCC in diabetic patients,as it could identify HCC with high diagnostic accuracy in diabetic LC patients(AUC=0.993)and in diabetic nonmalignant patients(AUC=0.961).CONCLUSION This study validates a panel of serum miRNAs that can be used as a reliable noninvasive screening biomarker of HCC among T2DM cirrhotic and noncirrhotic patients. The study recommends further research to shed light on a possible role of c-Met in T2DM-associated HCC via the miRNA regulatory pathway. | Moustafa Nouh Elemeery Marwa Anwar Mohamed Marwa Ahmed Madkour Mohammed Mohammed Shamseya Noha Mahmoud Issa Ahmed Noah Badr Doaa Ahmed Ghareeb Cheol-Ho Pan | 2019 | World Journal of Gastroenterology2019,25,42: | 3
Simulation, Modeling, and Optimization of Intelligent Kidney Disease Predication Empowered with Computational Intelligence Approaches显示文摘Artificial intelligence(AI)is expanding its roots in medical diagnostics.Various acute and chronic diseases can be identified accurately at the initial level by using AI methods to prevent the progression of health complications.Kidney diseases are producing a high impact on global health and medical practitioners are suggested that the diagnosis at earlier stages is one of the foremost approaches to avert chronic kidney disease and renal failure.High blood pressure, diabetes mellitus, and glomerulonephritis are the root causes of kidney disease. Therefore, the present study is proposed a set of multiple techniques such as simulation, modeling, and optimization of intelligent kidney disease prediction(SMOIKD)which is based on computational intelligence approaches.Initially, seven parameters were used for the fuzzy logic system(FLS),and then twenty-five different attributes of the kidney dataset were used for the artificial neural network(ANN)and deep extreme machine learning(DEML).The expert system was proposed with the assistance of medical experts. For the quick and accurate evaluation of the proposed system,Matlab version 2019 was used. The proposed SMOIKD-FLSANN-DEML expert system has shown 94.16%accuracy.Hence this study concluded that SMOIKD-FLS-ANN-DEML system is effective to accurately diagnose kidney disease at initial levels. | Abdul Hannan Khan Muhammad Adnan Khan Sagheer Abbas Shahan Yamin Siddiqui Muhammad Aanwar Saeed Majed Alfayad Nouh Sabri Elmitwally | 2021 | Computers, Materials & Continua2021,,5: | 2
Spinal fusion-hardware construct: Basic concepts and imaging review显示文摘The interpretation of spinal images fixed with metallic hardware forms an increasing bulk of daily practice in a busy imaging department. Radiologists are required to be familiar with the instrumentation and operative options used in spinal fixation and fusion procedures, especially in his or her institute. This is critical in evaluating the position of implants and potential complications associated with the operative approaches and spinal fixation devices used. Thus, the radiologist can play an important role in patient care and outcome. This review outlines the advantages and disadvantages of commonly used imaging methods and reports on the best yield for each modality and how to overcome the problematic issues associated with the presence of metallic hardware during imaging. Baseline radiographs are essential as they are the baseline point for evaluation of future studies should patients develop symptoms suggesting possible complications. They may justify further imaging workup with computed tomography, magnetic resonance and/or nuclear medicine studies as the evaluation of a patient with a spinal implant involves a multi-modality approach. This review describes imaging features of potential complications associated with spinal fusion surgery as well as the instrumentation used. This basic knowledge aims to help radiologists approach everyday practice in clinical imaging. | Mohamed Ragab Nouh | 2012 | World Journal of Radiology2012,4,5: | 2
