| 1 | Scale dependence of direct shear tests显示文摘Direct shear test has been widely used to measure the shear strength of soils and other particulate materials in industry because of its simplicity. However, the results can be dependent on the specimen size. The ASTM (American Society for Testing and Materials) publications suggest that for testing soils the shear box should be at least ten times the diameter of the largest particle and the height of the box should be no more than half of its diameter. These guidelines are empirically based. A series of two-dimensional numerical direct shear tests are performed to investigate this scaling effect. By analyzing the bulk friction, particle translation and rotation, percentage of sliding, average volume (area) and shear strain and the evolution of the shear band, we find that the traditional guidelines for direct shear tests are questionable. Scaling dependency of bulk friction on the property of granular materials is clearly present. Our current analysis points out that the scaling effects can vary significantly depending on the particle properties other than their sizes. Of all the parameters we observed, particle rotation appears to have a decisive correlation with the bulk friction. Formation of a shear band is universal. As the shearing progresses, particle rotation begins to concentrate near the shear plane. By defining the width of a shear band as the standard deviation of the distribution of translational gradient or the standard deviation of the distribution of particle rotation, quantitative evolutions of shear band are presented. Both measures of the shear band width dropped rapidly during pre-failure stage. After peak stress both measures begin to approach steady state as the bulk friction stabilizes to the residual stage. | 3 | Prostate volume does not provide additional predictive value to prostate health index for prostate cancer or clinically significant prostate cancer:results from a multicenter study in China显示文摘To evaluate whether prostate volume(PV)would provide additional predictive utility to the prostate health index(phi)for predicting prostate cancer(PCa)or clinically significant prostate cancer,we designed a prospective,observational multicenter study in two prostate biopsy cohorts.Cohort 1 included 595 patients from three medical centers from 2012 to 2013,and Cohort 2 included 1025 patients from four medical centers from 2013 to 2014.Area under the receiver operating characteristic curves(AUC)and logistic regression models were used to evaluate the predictive performance of PV-based derivatives and models.Linear regression analysis showed that both total prostate-specific antigen(tPSA)and free PSA(fPSA)were significantly correlated with PV(all P<0.05).[-2]proPSA(p2PSA)was significantly correlated with PV in Cohort 2(P<0.001)but not in Cohort 1(P=0.309),while no significant association was observed between phi and PV.When combining phi with PV,phi density(PHID)and another phi derivative(PHIV,calculated as phi/PV°5)did not outperform phi for predicting PCa or clinically significant PCa in either Cohort 1 or Cohort 2.Logistic regression analysis also showed that phi and PV were independent predictors for both PCa and clinically significant PCa(all P<0.05);however,PV did not provide additional predictive value to phi when combining these derivatives in a regression model(all models vs phi were not statistically significant,all P>0.05).In conclusion,PV-based derivatives(both PHIV and PHID)and models incorporating PV did not improve the predictive abilities of phi for either PCa or clinically significant PCa. | Da Huang Yi-Shuo Wu Ding-Wei Ye Jun Qi Fang Liu Brian T Helfand Siqun L Zheng Qiang Ding Dan-Feng Xu Rong Na Jian-Feng Xu Ying-Hao Sun | 2020 | Asian Journal of Andrology2020,22,5: | 2 |

| 18 | Genome-wide association analysis reveals regulation of at-risk loci by DNA methylation in prostate cancer显示文摘Epigenetic changes are potentially important for the ontogeny and progression of tumors but are not usually studied because of the complexity of analyzing transcript regulation resulting from epigenetic alterations.Prostate cancer(PCa)is characterized by variable clinical manifestations and frequently unpredictable outcomes.We performed an expression quantitative trait loci(eQTL)analysis to identify the genomic regions that regulate gene expression in PCa and identified a relationship between DNA methylation and clinical information.Using multi-level information published in The Cancer Genome Atlas,we performed eQTL-based analyses on DNA methylation and gene expression.To better interpret these data,we correlated loci and clinical indexes to identify the important loci for both PCa development and progression.Our data demonstrated that although only a small proportion of genes are regulated via DNA methylation in PCa,these genes are enriched in important cancer-related groups.In addition,single nucleotide polymorphism analysis identified the locations of CpG sites and genes within at-risk loci,including the 19q13.2-q13.43 and 16q22.2-q23.1 loci.Further,an epigenetic association study of clinical indexes detected risk loci and pyrosequencing for site validation.Although DNA methylation-regulated genes across PCa samples are a small proportion,the associated genes play important roles in PCa carcinogenesis. | Qiang Liu Gang Liu Darryl T Martin Yu-Tong Xing Robert M Weiss Jun Qi Jian Kang | 2021 | Asian Journal of Andrology2021,23,5: | 1 |
