17组织染色法观察灰盖鬼伞子实体发育过程(英文)显示文摘灰盖鬼伞(Coprinopsis cinerea)是一种嗜粪模式食用菌,可在实验室条件下快速生长并结实。子实体发育从黑暗环境下形成初级菌丝结开始,随后在光照刺激下初级菌丝结紧密聚集形成二级菌丝结,然后二级菌丝结继续分化出菌柄和菌盖。在子实体形成培养条件下,前5天(P1-P5)为原基发育期,第6天担子及担孢子中发生核融合及减数分裂,伴随菌柄伸长和菌盖形成,子实体发育成熟。成熟的子实体在第7天自溶,含有孢子的液滴落地并释放出孢子。本文运用不同的组织染色技术观察原基组织切片,从而发现灰盖鬼伞子实体发育过程中不同的组织和细胞类型。前担子比其他组织更容易被乳酚蓝、迈尔士苏木精明矾、孔雀绿、曙红染色。菌盖皮最容易被孔雀绿和乳酚蓝染色。过碘酸雪夫氏染色(periodic acid Schiff,PAS)的菌盖皮和子实基层颜色比其他组织深,其中菌盖皮颜色很深,子实基层颜色稍深。所有切片还尝试了各种可能组合的双染。在PAS和苏木精明矾先后双染色下,子实基层和前胆子可以获得最佳区分。SUBBA Shanta WINKLER Marco KüES Ursula 2019食用菌学报2019,26,4:1
