1用硫化物沉淀法净化硫酸钴溶液的进展显示文摘在50年代初期,比利时卢万(Louvain)大学的布雷克波特(Breckpot)教授与霍博肯冶金总公司的试验室共同研究,发现用硫化镍去极化置换沉淀法从硫酸钻溶液中分离镍是可能的,后来以锌沉淀进行的这种反应发展成扎伊尔矿业总公司西土鲁和卢易卢(Shituru,Luilu Gecamine)钴厂的净(?)方法。由于对高质量产品不断增长的需求和降低生产成本的需要,对硫化镍和硫化锌沉淀的关键参数做了许多试验室和中间工厂研究,尤其是对用元素硫或硫氢化钠沉淀的钴、镍、锌硫化物的相对稳定性与pH值和氧化还原电位的关系做了试验,这些试验已导致生产实践的改善并给新的发展带来可能。同样,对硫化物沉淀的处理也作了考虑。A. 2Gelatin degradation assay reveals MMP-9 inhibitors and function of O-glycosylated domain显示文摘AIM: To establish a novel, sensitive and high-throughput gelatinolytic assay to define new inhibitors and compare domain deletion mutants of gelatinase B/matrix metalloproteinase (MMP)-9. METHODS: Fluorogenic Dye-quenched (DQ)TM-gelatin was used as a substrate and biochemical parameters (substrate and enzyme concentrations, DMSO solvent concentrations) were optimized to establish a highthroughput assay system. Various small-sized libraries (ChemDiv, InterBioScreen and ChemBridge) of hetero-cyclic, drug-like substances were tested and compared with prototypic inhibitors. RESULTS: First, we designed a test system with gelatin as a natural substrate. Second, the assay was validated by selecting a novel pyrimidine-2,4,6-trione (barbitu- rate) inhibitor. Third, and in line with present structural data on collagenolysis, it was found that deletion of the O-glycosylated region significantly decreased gelatinolytic activity (kcat/kM ± 40% less than full-length MMP-9). CONCLUSION: The DQTM-gelatin assay is useful in high-throughput drug screening and exosite targeting. 3Review on modelling of corrosion under droplet electrolyte for predicting atmospheric corrosion rate显示文摘Atmospheric corrosion of metals is the most common type of corrosion which has a significant impact on the environment and operational safety in various situations of everyday life.Some of the common examples can be observed in land,water and air transportation systems,electronic circuit boards,urban and offshore infrastructures.The dew drops formed on metal surface due to condensation of atmospheric moisture facilitates corrosion as an electrolyte.The corrosion mechanisms under these droplets are different from classically known bulk electrolyte corrosion.Due to thin and non-uniform geometric thickness of the droplet electrolyte,the atmospheric oxygen requires a shorter diffusion path to reach the metal surface.The corrosion under a droplet is driven by the depletion of oxygen in the center of the droplet compared to the edge,known as differential aeration.In case of a larger droplet,differential aeration leads to preferential cathodic activity at the edge and is controlled by the droplet geometry.Whereas,for a smaller droplet,the oxygen concentration remains uniform and hence cathodic activity is not controlled by droplet geometry.The geometry of condensed droplets varies dynamically with changing environmental parameters,influencing corrosion mechanisms as the droplets evolve in size.In this review,various modelling approaches used to simulate the corrosion under droplet electrolytes are presented.In the efforts of developing a comprehensive model to estimate corrosion rates,it has been noted from this review that the influence of geometric evolution of the droplet due to condensation/evaporation processes on corrosion mechanisms are yet to be modelled.Dynamically varying external factors like environmental temperature,relative humidity,presence of hygroscopic salts and pollutants influence the evolution of droplet electrolyte,making it a complex phenomenon to investigate.Therefore,an overview of available dropwise condensation and evaporation models which describes the formation and the evolution of droplet geometry are also presented from an atmo s pheric corrosion viewpoint.Bangalore Gangadharacharya Koushik Nils Van den Steen Mesfin Haile Mamme Yves Van Ingelgem Herman Terryn 2021Journal of Materials Science & Technology2021,,3:3
4Better to be alone than in bad company:The antagonistic effect of cisplatin and crizotinib combination therapy in non-small cell lung cancer显示文摘AIM To investigate the potential benefit of combining the cMET inhibitor crizotinib and cisplatin we performed in vitro combination studies.METHODS We tested three different treatment schemes in four non-small cell lung cancer(NSCLC) cell lines with a different cMET/epidermal growth factor receptor genetic background by means of the sulforhodamine B assay and performed analysis with Calcusyn.RESULTS All treatment schemes showed an antagonistic effect in all cell lines,independent of the cMET status.Despite their different genetic backgrounds,all cell lines(EBC-1,HCC827,H1975 and LUDLU-1) showed antagonistic combination indexes ranging from 1.3-2.7.These results were independent of the treatment schedule.CONCLUSION These results discourage further efforts to combine cMET inhibition with cisplatin chemotherapy in NSCLC.Nele Van Der Steen Christophe Deben Vanessa Deschoolmeester An Wouters Filip Lardon Christian Rolfo Paul Germonpré Elisa Giovannetti Godefridus J Peters Patrick Pauwels 2016World Journal of Clinical Oncology2016,7,6:2
5Bioengineering bacterial encapsulin nanocompartments as targeted drug delivery system显示文摘The development of Drug Delivery Systems(DDS)has led to increasingly efficient therapies for the treatment and detection of various diseases.DDS use a range of nanoscale delivery platforms produced from polymeric of inorganic materials,such as micelles,and metal and polymeric nanoparticles,but their variant chemical composition make alterations to their size,shape,or structures inherently complex.Genetically encoded protein nanocages are highly promising DDS candidates because of their modular composition,ease of recombinant production in a range of hosts,control over assembly and loading of cargo molecules and biodegradability.One example of naturally occurring nanocompartments are encapsulins,recently discovered bacterial organelles that have been shown to be reprogrammable as nanobioreactors and vaccine candidates.Here we report the design and application of a targeted DDS platform based on the Thermotoga maritima encapsulin reprogrammed to display an antibody mimic protein called Designed Ankyrin repeat protein(DARPin)on the outer surface and to encapsulate a cytotoxic payload.The DARPin9.29 chosen in this study specifically binds to human epidermal growth factor receptor 2(HER2)on breast cancer cells,as demonstrated in an in vitro cell culture model.The encapsulin-based DDS is assembled in one step in vivo by co-expressing the encapsulin-DARPin9.29 fusion protein with an engineered flavin-binding protein mini-singlet oxygen generator(MiniSOG),from a single plasmid in Escherichia coli.Purified encapsulin-DARPin_miniSOG nanocompartments bind specifically to HER2 positive breast cancer cells and trigger apoptosis,indicating that the system is functional and specific.The DDS is modular and has the potential to form the basis of a multi-receptor targeted system by utilising the DARPin screening libraries,allowing use of new DARPins of known specificities,and through the proven flexibility of the encapsulin cargo loading mechanism,allowing selection of cargo proteins of choice.Alexander Van de Steen Rana Khalife Noelle Colant Hasan Mustafa Khan Matas Deveikis Saverio Charalambous Clare M.Robinson Rupali Dabas Sofia Esteban Serna Diana A.Catana Konstantin Pildish Vladimir Kalinovskiy Kenth Gustafsson Stefanie Frank 2021Synthetic and Systems Biotechnology2021,6,3:2
