2Experimental study of“Tong Xia”purgative method in ameliorating lung injury in acute necrotizing pancreatitis显示文摘AIM To investigate the role of tumor necrosisfactor(TNF)in lung injury during acutenecrotizing pancreatitis(ANP),and thetherapeutic effect of'Tong Xia'purgativemethod in minimizing the severity of lung injury.METHODS Fourteen canines were randomlydivided into 3 groups:the'Tong Xia'treatmentgroup(n = 5)using Dachengqitang;salinecontrol group(n = 5),and the sham operationgroup(n = 4).TNF activity in serum and inbronchoalveolar lavage fluid(BALF),the serumendotoxin levels were measured,and theseverity of lung injury evaluated.RESULTS Elevation of TNF activity was moreprominent in BALF than in serum.TNF activity inserum at 6 and 12 hours and in BALI:wassignificantly decreased in the'Tong Xia'treatment group than in the saline control one(q=21.11,q=12.07,q=9.03,respectively,P<0.01)and the lung injury was significantlyalleviated at 12 hours as compared with that inthe saline group,manifested as amelioration otthe lung wet/ dry weight ratio,decrease inprotein concentration and neutrophils count inBALF,and improvement of pulmonaryinflammatory changes.A positive correlationwas demonstrated between serum TNF activity and endotoxin level.CONCLUSION Hypersecretion of TNF is shownto be one of the major causes of lung injuryduring ANP;'Tong Xia'purgative method couldalleviate the degree of lung injury mediated byTNF.Xia Q Jiang JM Gong X Chen GY Li L Huang ZW 2000World Journal of Gastroenterology2000,6,1:31

7Measurement and Prediction of Insertion Force for the Mosquito Fascicle Penetrating into Human Skin显示文摘Mosquitoes are exceptional in their ability to pierce into human skin with a natural ultimate painless microneedle, namedfascicle. Here the structure of the Aedes albopictus mosquito fascicle is obtained using a Scanning Electron Microscope (SEM),and the whole process of the fascicle inserting into human skin is observed using a high-speed video imaging technique. Directmeasurements of the insertion force for mosquito fascicle to penetrate into human skin are reported. Results show that themosquito uses a very low force (average 18 μN) to penetrate into the skin. This force is at least three orders of magnitude smallerthan the reported lowest insertion force for an artificial microneedle with an ultra sharp tip to insert into the human skin. In orderto understand the piercing mechanism of mosquito fascicle tip into human multilayer skin tissue, a numerical simulation isconducted to analyze the insertion process using a nonlinear finite element method. 11Novel functional proteins interact with midkine in hepatic cancer cells显示文摘BACKGROUND: Midkine is a heparin-binding growth factor that promotes the proliferation, survival, migration and differentiation of various target cells. Midkine plays an important role in tumorigenesis and tumor progression, and is overexpressed in many human malignant tumors. Patients with high tumor midkine expression frequently have a worse prognosis than those with low expression. The present study was designed to investigate the interaction network of midkine in hepatic cancer cells, and to elucidate its role in hepatocellular carcinoma. METHODS: DNA encoding full-length midkine was cloned into pDBLeu vector to serve as bait in yeast two-hybrid screening to identify interacting proteins. Candidate proteins were examined on SC-Leu-Trp-His+3-AT (20 mmol/L) plates and assayed for X-gal activity, then sequenced and classified according to the GenBank. Finally, identified proteins were expressed by the in vitro expression system pCMVTnT, and protein interactions were confirmed by co-immunoprecipitation. RESULTS: Using the yeast two-hybrid system, we found 6 proteins that interacted with midkine: NK-kappa-B inhibitor alpha (I-κ-B-α), Dvl-binding protein naked cuticle 2, granulin, latent active TGF-β binding protein 3, latent active TGF-β binding protein 4, and phospholipid scramblase 1. 12桑叶多糖对早期断奶仔猪生长性能、腹泻、血液指标和肠道菌群的影响显示文摘研究旨在探讨日粮补充桑叶多糖(MLPs)对早期断奶仔猪生长性能、腹泻、血液生化指标和肠道菌群的影响。共选取(28±2)日龄、体重为(9.18±0.46)kg的'杜洛克×长白×约克夏'杂交断奶仔猪150头,随机分成5个处理组,每组6个重复,每个重复5头猪。饮食处理如下:对照组(CT):基础日粮(BD);低剂量桑叶多糖组(LT):0.3 g·kg-1MLPs+BD;中剂量桑叶多糖组(MT):0.6 g·kg-1MLPs+BD;高剂量桑叶多糖组(HT):0.9 g·kg-1MLPs+BD;抗生素处理组(AT):0.15 g·kg-1金霉素+BD。分别在第0、10、21天的早晨,每头猪空腹测量体重并记录采食量,这些数据用来计算平均日增重(ADG)、平均日采食量(ADFI)和料重比(F/G)。每天两次(上午和下午)对每头仔猪的腹泻状况进行检查并记录。在试验结束时,收集血液样本用于生化分析,并收集盲肠、结肠和直肠内容物进行识别和量化相关的肠道菌群。研究结果表明,MLPs处理组间的日增重、平均日采食量和料重比无显著差异(P>0.05),但低剂量组(LT)、中剂量组(MT)、高剂量组(HT)的平均日采食量高于对照组(CT)和抗生素组(AT)。并且,LT、MT和HT组的料重比低于CT和AT组。与CT和AT组相比,LT、MT和HT组仔猪的腹泻发病率也显著降低(P<0.05)。生化分析表明,MLPs各处理组仔猪的血糖(GLU)水平均显著低于CT或AT组(P<0.05)。MT和HT组血液尿素氮(BUN)水平与CT和AT组存在显著差异(P<0.05),HT组T3水平显著高于CT和AT组(P<0.05)。虽然MLPs各处理组的T4水平与AT组并没有显著差异(P>0.05),但MT组的胰岛素样生长因子Ⅰ(IGF-Ⅰ)和生长激素(GH)水平较CT或AT组均显著升高(P<0.05)。LT、MT和HT 3组在抑制肠道大肠杆菌生长和促进肠道乳酸杆菌和双歧杆菌生长的整体效果上均优于CT和AT组。值得注意的是,与CT和AT组相比,HT组(0.9 g·kg-1MLPs)对抑制肠道大肠杆菌以及促进肠道有益菌群具有更显著效果(P<0.05)。结果表明,日粮添加桑叶多糖可以改善早期断奶仔猪肠道菌群的生态环境,降低腹泻率,提高整体生长性能。Zhao X J Lin L Luo Q L Ye M Q Luo G Q Kuang Z S 2015饲料博览2015,0,7:7
13Improvement of Fracture Toughness Lanthanum Zirconate显示文摘La2Zr2O7 (LZ) is a promising thermal barrier coating material for the high temperature applications. The fracture toughness and microhardness of nanocrystalline LZ (n-LZ), microcrystalline LZ (m-LZ) and LZ-5mol%8YSZ (LZ-5-8YSZ) composite (8YSZ for zirconia stabilized by 8 mol% ytrria) were studied. The n-LZ had a thermal expansion coefficient of (9.6±0.4)×10 -6 K -1 (200~1000℃) and fracture toughness of (1.98±0.07) MPa·m 1/2 which are obviously higher than those of the m-LZ ( (9.1±0.4)×10 -6 K -1 and (1.40±0.23) MPa·m 1/2, respectively), indicating that nanofication was an efficient way to increase the toughness and thermal expansion coefficient of LZ. 17笋芽顶端分生组织在竹秆横切面形态形成中的作用与机制研究显示文摘竹秆的横切面主要由表皮、皮层、维管组织、基本组织及髓腔(早期为髓组织)组成。这些组织均由笋芽茎顶端分生组织分化发育而来,且其在生长发育过程中促使了竹秆横切面形态(本文特指:竹秆横切面外轮廓形状、围度及壁-腔结构)的形成。然而,人们对于这一生长发育过程的生物学机制知之甚少。本研究利用解剖学、数学模拟等方法,对 20 种具不同秆径竹种的笋芽顶端分生组织进行了分析。研究结果发现,笋芽顶端分生组织指状形态的维持是竹子产生圆柱形竹秆的基础;在顶端分生组织细胞数目增加的同时,线性增加原体细胞数目应是竹子在演化过程中产生大径竹秆且保持规则壁-腔结构的方式。进一步通过研究一个竹壁增厚且壁-腔架构紊乱的稳定变异体———实肚竹( Phyllostachys nidularia f. farcta)发现,其笋芽顶端分生组织细胞数目减少且原体/原套细胞数比值降低使其产生了高比例的壁组分组织(指基本分生组织与肋状分生组织)与低比例的髓组织。上述 2 种组织的比例失衡,使得实肚竹笋芽在后续发育中壁组分组织随机挤入到了髓组织中,最终形成了实肚竹异常的竹秆横切面形态(竹壁多样化增厚且壁-腔结构紊乱)。此外,相对较小的顶端分生组织,使得实肚竹笋芽产生了较少的内源激素,如其生长素含量仅约为其原种篌竹的 67. 5%,各细胞分裂素含量也显著低于篌竹笋芽。转录组分析进一步发现,在实肚竹笋芽中与激素及代谢相关的功能基因呈显著下调表达。这些生理与分子变化最终导致了实肚竹竹秆矮化、秆径减小且生物量降低。本论文的研究结果为研究竹秆横切面发育提供了一个重要的视角,同时也为实肚竹厚壁茎秆的形成提出了一个合理的机制。Wang Y Sun X Ding Y Fei Z Jiao C Fan M Yao B Xin P Chu J Wei Q 王雨珺 魏强 2019世界竹藤通讯2019,17,3:5
