Hepatitis B virus,HBx mutants and their role in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is one of the leading causes of death induced by cancer in the modern world and majority of the cases are related to chronic hepatitis B virus(HBV)infection.HBV-encoded X protein(HBx)is known to play a pivotal role in the pathogenesis of viral induced HCC.HBx is a multifunctional protein of17 kDa which modulates several cellular processes by direct or indirect interaction with a repertoire of host factors resulting in HCC.HBX might interfere with several cellular processes such as oxidative stress,DNA repair, signal transduction, transcription, protein degradation, cell cycle progression and apoptosis.A number of reports have indicated that HBx is one of the most common viral ORFs that is often integrated into the host genome and its sequence variants play a crucial role in HCC.By mutational or deletion analysis it was shown that carboxy terminal of HBx has a likely role in protein-protein interactions, transcriptional transactivation,DNA repair, cell, signaling and pathogenesis of HCC.The accumulated evidence thus far suggests that it is difficult to understand the mechanistic nature of HBx associated HCC,and HBx mediated transcriptional transactivation and signaling pathways may be a major determinant. This article addresses the role of HBx in the development of HCC with particular emphasis on HBx mutants and their putative targets. | Ashraf Ali Hany Abdel-Hafiz Mohd Suhail Amany Al-Mars Mohammad Khalid Zakaria Kaneez Fatima Sultan Ahmad Esam Azhar Adeel Chaudhary Ishtiaq Qadri | 2014 | World Journal of Gastroenterology2014,20,30: | 29
Human umbilical cord mesenchymal stem cells as treatment of adjuvant rheumatoid arthritis in a rat model显示文摘AIM:To investigate the effect of human umbilical cord stem cells, both mesenchymal and hematopoietic(CD34+),in the treatment of arthritis.METHODS:Mesenchymal stem cells(MSCs) and hematopoietic(CD34+) stem cells(HSC) were isolated from human umbilical cord blood obtained from the umbilical cord of healthy pregnant donors undergoing fullterm normal vaginal delivery.MSC,HSC,methotrexate(MTX) and sterile saline were injected intra-articularly into the rat hindpaw with complete freunds adjuvant(CFA) induced arthritis after the onset of disease(day 34),when arthritis had become well established(arthritis score ≥ 2).Arthritic indices were evaluated and the levels of interleukin(IL)-1,tumor necrosis factor(TNF)-α and interferon(IFN)-γ and anti-inflammatory cytokine IL-10 in serum were determined using enzyme-linked immunosorbent assay.Animals of all groups were sacrificed 34 d after beginning treatment, except positive control(PC) which was sacrificed at 10,21 and 34 d for microscopic observation of disease progression. We used hematoxylin, eosin and Masson's trichrome stains for histopathological examination of cartilage and synovium.RESULTS:The mean arthritis scores were similar in all groups at 12 and 34 d post immunization, with no statistical significant difference.Upon the injection of stem cells(hematopoietic and mesenchymal),the overall arthritis signs were significantly improved around 21 d after receiving the injection and totally disappeared at day 34 post treatment in MSC group.Mean hindpaw diameter(mm) in the MSC rats was about half that of the PC and MTX groups(P = 0.007 and P = 0.021,respectively) and 0.6 mm less than the HSC group(P = 0.047),as indicated by paw swelling.Associated with these findings, serum levels of TNF-α,IFN-γ and IL-1 decreased significantly in HSC and MSC groups compared to PC and MTX groups(P < 0.05),while the expression of IL-10 was increased.Histopathological examination with H and E stain revealed that the MTX treated group showed significant reduction of leucocytic infiltrate and hypertrophy of the synovial tissue with moderate obliteration of the joint cavity.Stem cells treated groups(both hematopoietic CD34+ and mesenchymal),showed significant reduction in leucocytic infiltrate and hypertrophy of the synovial tissue with mild obliteration of the joint cavity. With Masson's trichrome, stain sections from the PC group showed evidence of vascular edema of almost all vessels within the synovium in nearly all arthritic rats.Vacuoles were also visible in the outer vessel wall. The vessel became hemorrhagic and finally necrotic. In addition, there was extensive fibrosis completely obliterating the joint cavity. The mean color area percentage of collagen in this group was 0.324 ± 0.096,which was significantly increased when compared to the negative control group. The mean color area percentage of collagen in hematopoietic CD34+ and mesenchymal groups was 0.176 ± 0.0137 and 0.174 ± 0.0197 respectively, which showed a marked decrement compared to the PC group, denoting a mild increase in synovial tissue collagen fibers.CONCLUSION:MSC enhance the efficacy of CFAinduced arthritis treatment, most likely through the modulation of the expression of cytokines and amelioration of pathological changes in joints. | Sahar Greish Noha Abogresha Zeinab Abdel-Hady Eman Zakaria Mona Ghaly Mohamed Hefny | 2012 | World Journal of Stem Cells2012,4,10: | 17
Piper sarmentosum as an antioxidant on oxidative stress in human umbilical vein endothelial cells induced by hydrogen peroxide显示文摘Endothelial cell death due to increased reactive oxygen species(ROS) may contribute to the initial endothelial injury, which promotes atherosclerotic lesion formation.Piper sarmentosum(PS),a natural product, has been shown to have an antioxidant property, which is hypothesized to inhibit production of ROS and prevent cell injury. Thus, the present study was designed to determine the effects of PS on the hydrogen peroxide(H2O2)-induced oxidative cell damage in cultured human umbilical vein endothelial cells(HUVECs).In this experiment,HUVECs were obtained by collagenase perfusion of the large vein in the umbilical cord and cultured in medium M200 supplemented with low serum growth supplementation(LSGS).HUVECs were treated with various concentrations of H2O2(0-1000 μmol/L) and it was observed that 180 μmol/L H2O2 reduced cell viability by 50% as denoted by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay.Using the above concentration as the positive control, the H2O2-induced HUVECs were concomitantly treated with various concentrations(100,150,250 and 300 μg/ml) of three different extracts(aqueous, methanol and hexane) of PS.Malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT) and glutathione peroxidase(GPX) levels showed a significant increase(P<0.05) in HUVECs compared to the negative control. However,PS extracts showed a protective effect on HUVECs from H2O2-induced cell apoptosis with a significant reduction in MDA,SOD,CAT and GPX levels(P<0.05).Furthermore,PS had exhibited ferric reducing antioxidant power with its high phenolic content.Hence,it was concluded that PS plays a beneficial role in reducing oxidative stress in H2O2-induced HUVECs. | Abdul Hamid HAFIZAH Zakaria ZAITON Amom ZULKHAIRI Adenan MOHD ILHAM Megat Mohd Nordin NOR ANITA Abdullah Mahdy ZALEHA | 2010 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,5: | 13
Biochar:An Emerging Panacea for Remediation of Soil Contaminants from Mining,Industry and Sewage Wastes显示文摘Mine tailings, waste rock piles, acid mine drainage, industrial wastewater, and sewage sludge have contaminated a vast area of cultivable and fallow lands, with a consequence of deterioration of soil and water quality and watercourses due to the erosion of contaminated soils for absence of vegetative cover. High concentrations of toxic elements, organic contaminants, acidic soils, and harsh climatic conditions have made it difficult to re-establish vegetation and produce crops there. Recently, a significant body of work has focussed on the suitability and potentiality of biochar as a soil remediation tool that increases seed emergence, soil and crop productivity, above ground biomass, and vegetation cover on mine tailings, waste rock piles, and industrial and sewage wastecontaminated soils by increasing soil nutrients and water-holding capacity, amelioration of soil acidity, and stimulation of microbial diversity and functions. This review addresses: i) the functional properties of biochar, and microbial cycling of nutrients in soil; ii)bioremediation, especially phytoremediation of mine tailings, industrial waste, sewage sludge, and contaminated soil using biochar; iii)impact of biochar on reduction of acid production, acid mine drainage treatment, and geochemical dynamics in mine tailings; and iv)treatment of metal and organic contaminants in soils using biochar, and restoration of degraded land. | Hossain M.ANAWAR Farjana AKTER Zakaria M.SOLAIMAN Vladimir STREZOV | 2015 | Pedosphere2015,25,5: | 13
Risk assessment for the daily intake of polycyclic aromatic hydrocarbons from the ingestion of cockle (Anadara granosa) and exposure to contaminated water and sediments along the west coast of Peninsular Malaysia显示文摘The concentration of carcinogenic polycyclic aromatic hydrocarbons (c-PAHs) present in the sediment and water of Peninsular Malaysia as well as in the cockle Anadara granosa was investigated.Samples were extracted and analysed with gas chromatographymass spectrometry. The concentrations of total carcinogenic polycyclic aromatic hydrocarbons (t-PAHs) were measured between 0.80 ± 0.04 to 162.96 ± 14.74 ng/g wet weight (ww) in sediment, between 21.85 ± 2.18 to 76.2 ± 10.82 ng/L in water samples and between 3.34 ± 0.77 to 46.85 ± 5.50 ng/g ww in the cockle tissue. The risk assessment of probable human carcinogens in the Group B2 PAHs was calculated and assessed in accordance with the standards of the United States Environmental Protection Agency (US EPA).Case I in the toxicity assessment analysed the cancer risk to consumers of Malaysian blood cockle.Case II assessed the risk of cancer from exposure to PAHs from multiple pathways. The average cancer risk of case I and case II were found to be classifiable as unsafe according to the US EPA standard. The cancer risk due to c-PAHs acquired by the ingestion of blood cockle was (8.82 ± 0.54) × 10-6 to (2.67 ± 0.06) × 10-2,higher than the US EPA risk management criterion. The non-cancer risks associated with multiple pathways in Kuala Gula,Kuala Juru and Kuala Perlis were higher than the US EPA safe level, but the non-cancer risk for eating blood cockle was below the level of US EPA concern. | Seiedeh Aghileh Mirsadeghi Mohamad Pauzi Zakaria Chee Kong Yap Azadeh Shahbazi | 2011 | Journal of Environmental Sciences2011,23,2: | 11
The development of MOFs-based nanomaterials in heterogeneous organocatalysis显示文摘Metal-organic framework(MOF) is a class of inorganic-organic hybrid material assembled periodically with metal ions and organic ligands. MOFs have always been the focuses in a variety of frontier fields owing to the advantageous properties, such as large BET surface areas, tunable porosity and easyfunctionalized surface structure. Among the various application areas, catalysis is one of the earliest application fields of MOFs-based materials and is one of the fastest-growing topics. In this review, the main roles of MOFs in heterogeneous organocatalysis have been systematically summarized, including used as support materials(or hosts), independent catalysts, and sacrificial templates. Moreover, the application prospects of MOFs in photocatalysis and electrocatalysis frontiers were also mentioned. Finally, the key issues that should be conquered in future were briefly sketched in the final parts of each item. We hope our perspectives could be beneficial for the readers to better understand these topics and issues, and could also provide a direction for the future exploration of some novel types of MOFs-based nanocatalysts with stable structures and functions for heterogeneous catalysis. | Xiaomei Liu Bing Tang Jilan Long Wei Zhang Xiaohong Liu Zakaria Mirza | 2018 | Science Bulletin2018,63,8: | 10
Mismatch repair genes (hMLH1, hPMS1, hPMS2, GTBP/hMSH6,HMSH2) in the pathogenesis of hepatocellular carcinoma显示文摘AIM: DNA misrnatch repair (MMR) is an impotent mechanism for maintaining fidelity of genomic DNA. Abnormalities in one or more MMR genes are implicated in the development of many cancers. We investigated the role of expression of MMR genes (hMLH1, hPMS1, hPMS2, GTBP/hMSH6, hMSH2) in hepatocellular carcinogenesis. METHODS: We evaluated the expression level of MMR genes in 33 hepatocellular carcinoma (HCC) cases using the multiplex reverse transcription (RT) PCR assays, as well as in 16 cases of normal adjacent hepatic tissues. Β-actin gene was used as an internal control and calibrator for quantification of gene expression. RESULTS: Out of the 33 studied cases, 25 were HCV positiveand 30 (90.9%) showed reduced expression in one or more of the studied MMR genes. Reduced expression was found in hMSH2 (71.9%), hMLH1 (53.3%), GTBP (51.1%), hPMS2 (33.3%) and hPMS1 (6%). A significant correlation was found between reduced expression of hPMS2(P= 0.0069) and GTBP(P= 0.0034), hPMS2and non-cirrhosis (P= 0.0197),hMLH1 and high grade. On the other hand, 57.1%, 50%, 20%, 18.8%, and 6% of the normal tissues distant to tumors showed reduced expression of hMSH2, hMLH1, GTBP, hPMS2, and hPMS1 respectively. Multivariate analysis revealed a significant correlation between the expression level of hMSH2(P= 0.008), hMLH1 (P= 0.001) and GTBP (P = 0.032)and HCC, between hPMS2, GTBP and HCVassociated HCC (P<0.001, 0.002).CONCLUSION: Reduced expression of MMR genes seemsto play an important role in HCV-associated HCC. hPMS2 islikely involved at an early stage of hepatocarcinogenesis since it was detected in normal adjacent tissues. Reduced expression of hPMS2 provides a growth advantage and stimulates proliferation which encourages malignant transformation in non-cirrhotic HCV-infected patients via acquisition of more genetic damages. | Abdel-Rahman N. | 9 | Application of Biochars for Soil Constraints:Challenges and Solutions显示文摘Biochar addition to soil is currently being considered as a means to sequester carbon while simultaneously improving soil health, soil fertility and agronomic benefits. The focus of this special issue is on current research on the effects of biochar application to soil for overcoming diverse soil constraints and recommending further research relating to biochar application to soil. The biochar research has progressed considerably with important key findings on agronomic benefits, carbon sequestration, greenhouse gas emissions, soil acidity, soil fertility, soil health, soil salinity, etc., but more research is required before definitive recommendations can be made to end-users regarding the effects of biochar application across a range of soils, climates and land management practices. | Zakaria M.SOLAIMAN Hossain M.ANAWAR | 2015 | Pedosphere2015,25,5: | 10
Rice Aroma: A Natural Gift Comes with Price and the Way Forward显示文摘Aromatic rice belongs to a small but important sub-group of rice, which is highly regarded for its excellent aroma and superior grain quality.Aromatic rice, especially Basmati-and Jasmine-type rice,is being traded at a high price in the local and global markets.Genetically, rice aroma is a phenotypical expression of spontaneous recessive mutations of the OsBadh2 gene(also known as fgr/badh2/osbadh2/os2AP gene). These mutations inhibit the flow ofγ-aminobutyraldehyde(GAB-ald)toγ-aminobutyric acid(GABA),and consequently, the accumulated GAB-ald is diverted to a potent flavour component 2-acetyl-1-pyrroline(2AP)by a non-enzymatic reaction with methylglyoxal. The natural incidence of non-functional osbadh2 mutation along with selection and nursing by the farmer from the ancient time makes rice aroma as a prominent natural gift.As GABA and methylglyoxal play significant roles in stress tolerance, and their biosynthesis is strictly regulated in rice plants, the accumulation of 2AP in aromatic rice depends on the interaction of various genetic and environmental factors, and its production may come at some costs of sacrificing tolerance. This review focused on some potential underlying genes in the 2AP and GABA biosynthesis pathways, and analyzed most aspects of aroma formation in rice, and summarized the molecular mechanism of aroma production together with its genetic and non-genetic influencing factors. The present review also stated approaches to produce high-quality aromatic rice via developing novel cultivars and with good agronomic knowledge-based practice. | Zakaria Hossain PRODHAN SHU Qingyao | 2020 | Rice science2020,27,2: | 8
Effects of Enriched Biochars Containing Magnetic Iron Nanoparticles on Mycorrhizal Colonisation,Plant Growth,Nutrient Uptake and Soil Quality Improvement显示文摘At present, there is little commercial sale of biochar, since farmers find they can not gain a return on their investment in this amendment in the first few years after its application, because of the high cost associated with large application rates. To overcome this constraint, development of artificially aged enriched biochar-mineral complexes(BMCs), having a higher mineral content, surface functionality, exchangeable cations, high concentration of magnetic iron(Fe) nanoparticles, and higher water-extractable organic compounds has been undertaken by a combined team of researchers and a commercial company. Two biochars produced under different pyrolysis conditions were activated with a phosphoric acid treatment. A mixture of clay, chicken litter, and minerals were added to the biochar, and then this composite was torrefied at either 180 or 220?C. In this study a pot experiment was carried out in glasshouse conditions to determine the effects of four different BMCs, with different formulations applied at rates of 100 and 200 kg ha-1, on the mycorrhizal colonisation, wheat growth and nutrient uptake, and soil quality improvement. It was found that the phosphorus(P) and nitrogen uptake in wheat shoots were significantly greater for a low application rate of BMCs(100 kg ha-1). The present formulation of BMC was effective in enhancing growth of wheat at low application rate(100 kg ha-1). The increase in growth appeared due to an increase in P uptake in the plants that could be partly attributed to an increase in mycorrhizal colonisation and partly due to the properties of the BMC. | Stephen JOSEPH Hossain M.ANAWAR Paul STORER Paul BLACKWELL Chee CHIA Yun LIN Paul MUNROE Scott DONNE Josip HORVAT Jianli WANG Zakaria M.SOLAIMAN | 2015 | Pedosphere2015,25,5: | 7

Trauma patterns in patients attending the Emergency Department of Jazan General Hospital, Saudi Arabia显示文摘BACKGROUND: Modern civilization and the sharp rise in living standards have led to dramatic changes in trauma pattern in Saudi Arabia. This study aimed to describe the different patterns of injuries of patients attending the Emergency Department of Jazan General Hospital(JGH) in the southwest corner of Saudi Arabia.METHODS: A total number of 1 050 patients were enrolled in the study. A pre-organized data sheet was prepared for each patient attended the Emergency Department of JGH from February 2012 to January 2013. It contains data about socio-demographics, trauma data, clinical evaluation results, investigations as well as treatment strategies.RESULTS: The mean age of the patients was 25.3±16.8 years. Most(45.1%) of the patients were at age of 18–30 years. Males(64.3%) were affected by trauma more common than females. More than half(60.6%) of the patients were from urban areas. The commonest kind of injury was minor injury(60%), followed by blunt trauma(30.9%) and then penetrating trauma(9.1%). The mean time from the incident to arrival at hospital was 41.3±79.8 minutes. The majority(48.2%) of the patients were discharged after management of trivial trauma, whereas 2.3% were admitted to ICU, 7.7% transferred to inpatient wards, and 17.7% observed and subsequently discharged. The mortality rate of the patients was 2.6%.CONCLUSION: Trauma is a major health problem, especially in the young population in Saudi Arabia. Blunt trauma is more frequent than penetrating trauma, with road traffic accidents accounting for the majority. | Emad Hokkam Abdelaziz Gonna Ossama Zakaria Amany El-shemally | 2015 | World Journal of Emergency Medicine2015,6,1: | 7
Influence of potassium fertilization and foliar application of zinc and phosphorus on growth, yield components, yield and fiber properties of Egyptian cotton(Gossypium barbadense L.)显示文摘Aims Supplying optimal quantities of mineral nutrients to growing crop plants is one way to improve crop yields.Nutrients need to be used rationally in order to avoid a negative ecological impact and undesirable effects on the sustainability of agricultural production systems.Excessive application of nutrients also affects the farmer’s economy. In order to calculate the amount of fertilizer to be applied to crops,it is necessary to develop recommendation programmers that adjust nutrient rates to crop requirements. Methods Experiments in two successive seasons were conducted to investigate the effect of K fertilization and foliar application of Zn and P on yield and fiber properties of cotton cv.Giza 86.Potassium(0.0 and 47.4 kg of K ha^(-1))was soil applied, while chelated zinc(0.0 and 57.6 g of Zn ha^(-1),applied twice at 70 and 85 days after sowing‘DAS’)and phosphorus(0.0,576,1152 and 1728 g of P ha^(-1),applied twice at 80 and 95 DAS)were applied to the foliage.Important findings Dry matter yield, total chlorophyll concentration,K,Zn and P uptake per plant, number of opened bolls per plant, boll weight, seed index, lint index, seed cotton yield per plant, seed cotton and lint yield ha^(-1) and earliness of harvest increased with the application of K,Zn and P.Treatments generally had no significant effect on lint percentage and fiber properties, with exceptions, for micronaire reading and flat bundle strength, and uniformity ratio, where the mean values of these characters were significantly increased over the untreated control by applying K,and for the micronaire reading in the first season, when applying P at 1728 g ha^(-1),and uniformity ratio in the second season, when applying Pat 1152 and 1728 g ha^(-1),where the mean values of these characters were significantly increased over the untreated control by applying P.Under the conditions of this study, applying K fertilization at 47.4 kg ha^(-1) combined with spraying cotton plants with zinc at 57.6 g ha^(-1) and also with P at 1728 g ha^(-1) improved growth and yield of Egyptian cotton. | Zakaria M.Sawan Mahmoud H.Mahmoud Amal H.El-Guibali | 2008 | Journal of Plant Ecology2008,1,4: | 6

Effects of human disturbance on liana community diversity and structure in a tropical rainforest, Malaysia: implication for conservation显示文摘Aim Due to the important role of lianas in the functioning of forest ecosystem, knowledge of the factors that affect them are important in the management of forests.Currently, there are conflicting reports on the response of liana communities to disturbance, calling for more research in the area. The present study was carried out to investigate the response of liana diversity and structure to human disturbance within two major forests in the Penang National Park,Malaysia. The study also looked at the implication of the findings for conservation. Methods A total of 15 40×40-m^(2)(or 40-m×40-m)plots each were randomly located across a range of habitats in a primary forest and disturbed secondary forest.Trees with diameter at breast height≥10 cm were examined for lianas with diameter≥2 cm.Both lianas and trees were enumerated and compared between the two forests.Diversity and structural variables of lianas were compared between the two forests using the t-test analysis.Tree abundance was also compared between the two forests with t-test,while linear regression analysis was run to determine the effects of tree abundance on liana abundance.Important Findings A total of 46 liana species belonging to 27 genera and 15 families were identified in the study.Human disturbance significantly reduced liana species richness and species diversity in the secondary forest.Liana abundance remained the same in both forests whereas liana basal area was;7 times higher in the primary forest.Twiners and hook climbers were significantly more abundant in the primary and secondary forest, respectively.Large diameter lianas were more abundant in the primary forest compared with the secondary forest. The diameter distribution of most families in the primary forest followed the inverted J-shaped curve whereas only a few of the families in the secondary forest did so.Tree abundance was significantly higher in the primary forest. The abundance of lianas significantly depended on tree abundance in all the forests. The study has provided evidence of negative effects of human disturbance on liana diversity and structure that does not auger well for biodiversity in the forest. In view of the critical role of lianas in maintaining biodiversity in the forest ecosystem, lianas in the national park should be protected from further exploitation. | Patrick Addo-Fordjour Zakaria B.Rahmad A.M.S.Shahrul | 2012 | Journal of Plant Ecology2012,5,4: | 6
Pharmacological activity, phytochemical analysis and toxicity of methanol extract of Etlingera elatior(torch ginger) flowers显示文摘Objective:To elucidate its pharmacological activities and medicinal potential of extract of Etlingera elatior(E.elatior).Methods:Phytochemical screening of the flower extract was done to determine the phytochemical in the extract. The pharmacological study included the determination of antimicrobial activity and minimum inhibitory concentration(MIC) of metabolic flower extract. The antimicrobial activity of the extract was tested against medically important bacterial, yeast and fungal strains.Apart from that, the methanolic extract of E.elatior flower was further tested in vivo toxicity using the brine shrimp lethality test. Moreover, the flower extract was qualitatively screened for their free radical scavenging activity by 2,2-diphenyl-1- picrylhydrazyl radical(DPPH) assay. Results:The extract was effective on tested microorganisms and MIC values were in the range of 1.563-50.000 mg/mL.The brine shrimp lethality test exhibited no significant toxicity(LC50= 2.52 mg/mL) against Artemia salina. The E.elatior flower extract with high LC50 value signified that this plant is not toxic to humans. While the phytochemical screening of the flower extract revealed the presence of the following compounds: flavonoids, terpenoids, saponin, tannins and carbohydrates whereas, alkaloids, anthraquinone and reducing sugars were absent. The concentration of the flower extract required for 50% inhibition of DPPH radical scavenging effect(IC50) were 9.14 mg/mL and 8.08 mg/mL for butylated hydroxytoluene 8.08 mg/mL.Conclusions:These findings indicate that the extract of E.elatior flower possesses pharmacological properties and potential to develop natural products based pharmaceuticals products. | Subramanion Jo Thy Lachumy Sreenivasan Sasidharan Velio Sumathy Zakaria Zuraini | 2010 | Asian Pacific Journal of Tropical Medicine2010,3,10: | 5
p53 antibodies,metallothioneins,and oxidative stress markers in chronic ulcerative colitis with dysplasia显示文摘AIM:To investigate the role of p53 antibodies (p53Abs),metallothioneins (MTs) and oxidative stress markers in the early detection of dysplasia in chronic ulcerative colitis (UC).METHODS:The study included 30 UC patients,15 without dysplasia (group Ⅱ) and 15 with dysplasia (group Ⅲ),in addition to 15 healthy volunteers (group Ⅰ,control subjects).The enzyme-linked immunosorbent assay technique was used to measure serum p53Abs and MTs,while advanced oxidation protein products (AOPPs),and reduced glutathione (GSH) levels were measured by spectrophotometric method in all subjects.RESULTS:In group Ⅱ and group Ⅲ compared to group Ⅰ,there were significant increases in serum levels of AOPPs (145.94 ± 29.86 μmol/L and 192.21 ± 46.71 μmol/L vs 128.95 ± 3.06 μmol/L,P < 0.002 and P <0.001,respectively),MTs (8.18 ± 0.35 μg/mL and 9.20 ± 0.58 μg/mL vs 6.12 ± 0.25 μg/mL,P < 0.05 and P < 0.05,respectively),and p53Abs (20.19 ± 3.20 U/mL and 34.66 ± 1.34 U/mL vs 9.42 ± 1.64 U/mL,P < 0.001 and P < 0.001,respectively).There were significantly higher levels of AOPPs (P < 0.05) and p53Abs (P < 0.001) in UC patients with dysplasia compared to those without dysplasia,while MTs showed no significant difference between the 2 groups (P > 0.096).In contrast,GSH levels showed a significant decrease in both patients' groups (1.87 ± 0.02 μmol/mL and 1.37 ± 0.09 μmol/mL vs 2.49 ± 0.10 μmol/mL,P < 0.05 and P < 0.05 in groups Ⅱ and Ⅲ,respectively) compared with group Ⅰ,and the levels were significantly lower in group Ⅲ than group Ⅱ (P < 0.05).There was a positive correlation between AOPPs and both MTs (r=0.678,P < 0.001) and p53Abs (r=0.547,P < 0.001),and also between p53Abs and MTs (r=0.739,P < 0.001).There was a negative correlation between AOPPs and GSH (r =-0.385,P < 0.001),and also between GSH and both MTs (r=-0.662,P < 0.001) and p53Abs (r=-0.923,P < 0.001).CONCLUSION:Oxidative stress and oxidative cellular damage play an important role in the pathogenesis of chronic UC and the associated carcinogenetic process.p53Abs levels could help in early detection of dysplasia in these conditions. | Hala E Hamouda Soha S Zakaria Saber A Ismail Mahmoud A Khedr Wael W Mayah | 2011 | World Journal of Gastroenterology2011,17,19: | 5
