2Comparison of chemiluminescence enzyme immunoassay based on magnetic microparticles with traditional colorimetric ELISA for the detection of serum α-fetoprotein显示文摘A chemiluminescence enzyme immunoassay based on magnetic microparticles (MmPs-CLEIA) was developed to evaluate serum a-fetoprotein (AFP) in parallel with traditional colorimetric enzyme-linked immunosorbent assay (ELISA).A systematic comparison between the MmPs-CLEIA and colorimetric ELISA concluded that the MPs-CLEIA exhibited fewer dosages of immunoreagents,less total assay time,and better linearity,recovery,precision,sensitivity and validity.AFP was detected in forty human serum samples by the proposed MPs-CLEIA and ELISA,and the results were compared with commercial electrochemiluminescence immunoassay (ECLIA) kit.The correlation coefficient between MPs-CLEIA and ELISA was obtained with R 2 0.6703;however,the correlation between MPs-CLEIA and ECLIA (R 2 0.9582) was obviously better than that between colorimetric ELISA and ECLIA (R 2 0.6866).Qian-Yun Zhang a,b,Hui Chen a,Zhen Lin a,Jin-Ming Lin a a Beijing Key Laboratory of Microanalytical Methods and Instrumentation,Department of Chemistry,Tsinghua University,Beijing 100029,China b Institute of Biophysics,Chinese Academy of Sciences,Beijing 100101,China 2012Journal of Pharmaceutical Analysis2012,2,2:5
3Zinc protects against cadmium-induced toxicity in neonatal murine engineered cardiac tissues via metallothionein-dependent and independent mechanisms显示文摘Cadmium(Cd)is a nonessential heavy metal and a prevalent environmental toxin that has been shown to induce significant cardiomyocyte apoptosis in neonatal murine engineered cardiac tissues(ECTs).In contrast,zinc(Zn)is a potent metallothionein(MT)inducer,which plays an important role in protection against Cd toxicity.In this study,we investigated the protective effects of Zn against Cd toxicity in ECTs and explore the underlying mechanisms.ECTs were constructed from neonatal ventricular cells of wild-type(WT)mice and mice with global MT gene deletion(MT-KO).In WT-ECTs,Cd(5−20μM)caused a dose-dependent toxicity that was detected within 8 h evidenced by suppressed beating,apoptosis,and LDH release;Zn(50−200μM)dose-dependently induced MT expression in ECTs without causing ECT toxicity;co-treatment of ECT with Zn(50µM)prevented Cd-induced toxicity.In MT-KO ECTs,Cd toxicity was enhanced;but unexpectedly,cotreatment with Zn provided partial protection against Cd toxicity.Furthermore,Cd,but not Zn,significantly activated Nrf2 and its downstream targets,including HO-1;inhibition of HO-1 by a specific HO-1 inhibitor,ZnPP(10µM),significantly increased Cd-induced toxicity,but did not inhibit Zn protection against Cd injury,suggesting that Nrf2-mediated HO-1 activation was not required for Zn protective effect.Finally,the ability of Zn to reduce Cd uptake provided an additional MT-independent mechanism for reducing Cd toxicity.Thus,Zn exerts protective effects against Cd toxicity for murine ECTs that are partially MT-mediated.Further studies are required to translate thesefindings towards clinical trials.Hai-tao Yu Juan Zhen Jian-xiang Xu Lu Cai Ji-yan Leng Hong-lei Ji Bradley B Keller 2020Acta Pharmacologica Sinica2020,41,5:3
4Nanoscopy reveals surface-metallic black phosphorus显示文摘Black phosphorus(BP)is an emerging two-dimensional material with intriguing physical properties.It is highly anisotropic and highly tunable by means of both the number of monolayers and surface doping.Here,we experimentally investigate and theoretically interpret the near-field properties of a-few-atomic-monolayer nanoflakes of BP.We discover near-field patterns of bright outside fringes and a high surface polarizability of nanofilm BP consistent with its surface-metallic,plasmonic behavior at mid-infrared frequencies o1176 cm−1.We conclude that these fringes are caused by the formation of a highly polarizable layer at the BP surface.This layer has a thickness of~1 nm and exhibits plasmonic behavior.We estimate that it contains free carriers in a concentration of n≈1.1×10^(20) cm^(−3).Surface plasmonic behavior is observed for 10–40 nm BP thicknesses but absent for a 4-nm BP thickness.This discovery opens up a new field of research and potential applications in nanoelectronics,plasmonics and optoelectronics.Yohannes Abate Sampath Gamage Zhen Li Viktoriia Babicheva Mohammad H Javani Han Wang Stephen B Cronin Mark I Stockman 2016Light(Science & Applications)2016,5,1:2
5Laser patterning for the study of MSC cardiogenic differentiation at the single-cell level显示文摘Mesenchymal stem cells(MSCs)have been cited as contributors to heart repair through cardiogenic differentiation and multiple cellular interactions,including the paracrine effect,cell fusion,and mechanical and electrical couplings.Due to heart–muscle complexity,progress in the development of knowledge concerning the role of MSCs in cardiac repair is heavily based on MSC–cardiomyocyte coculture.In conventional coculture systems,however,the in vivo cardiac muscle structure,in which rod-shaped cells are connected end-to-end,is not sustained;instead,irregularly shaped cells spread randomly,resulting in randomly distributed cell junctions.Consequently,contact-mediated cell–cell interactions(e.g.,the electrical triggering signal and the mechanical contraction wave that propagate through MSC–cardiomyocyte junctions)occur randomly.Thus,the data generated on the beneficial effects of MSCs may be irrelevant to in vivo biological processes.In this study,we explored whether cardiomyocyte alignment,the most important phenotype,is relevant to stem cell cardiogenic differentiation.Here,we report(i)the construction of a laser-patterned,biochip-based,stem cell–cardiomyocyte coculture model with controlled cell alignment;and(ii)single-cell-level data on stem cell cardiogenic differentiation under in vivo-like cardiomyocyte alignment conditions.Zhen Ma Qiuying Liu Huaxiao Yang Raymond B Runyan Carol A Eisenberg Meifeng Xu Thomas K Borg Roger Markwald Yifei Wang Bruce Z Gao 2013Light(Science & Applications)2013,2,1:2
