| 2 | Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. | 3 | Effect of carbon source on the denitrification in constructed wetlands显示文摘The ability of constructed wetlands with different plants in nitrate removal were investigated. The factors promoting the rates of denitrification including organic carbon, nitrate load, plants in wetlands, pH and water temperature in field were systematically investigated. The results showed that the additional carbon source (glucose) can remarkably improve the nitrate removal ability of the constructed wetland. It demonstrated that the nitrate removal rate can increase from 20% to more than 50% in summer and from 10% to 30% in winter, when the nitrate concentration was 30–40 mg/L, the retention time was 24 h and 25 mg/L dissolved organic carbon (DOC) was ploughed into the constructed wetland. However, the nitrite in the constructed wetland accumulated a little with the supply of the additional carbon source in summer and winter, and it increased from 0.15 to 2 mg/L in the effuent. It was also found that the abilities of plant in adjusting pH and temperature can result in an increase of denitrification in wetlands. | 4 | The influence of Enteral Nutrition in postoperative patients with poor liver function显示文摘AIM: To investigate the safety, rationality and the practicality of enteral nutritional (EN) support in the postoperative patients with damaged liver function and the protective effect of EN on the gut barrier.METHODS: 135 patients with liver function of Child B or C grade were randomly allocated to enteral nutrition group (EN, 65 cases), total parenteral nutrition group (TPN, 40cases) and control group (CON, 30 cases). Nutritional parameters, hepatic and kidney function indexes were measured at the day before operation, 5th and 10th day after the operation respectively. Comparison was made to evaluate the efficacy of different nutritional support. Urinary concentrations of lactulose(L) and mannitol(M) were measured by pulsed electrochemical detection(HPLC-PED)and the L/M ratio calculated to evaluate their effectiveness on protection of gut barrier.RESULTS: No significant damages in hepatic and kidney function were observed in both EN and TPN groups between pre- and postoperatively. | 6 | S-wave velocity and Poisson's ratio structure of crust in Yunnan and its implication显示文摘Receiver function of body wave under the 23 stations in Yunnan was extracted from 3-component broadband digital recording of teleseismic event. Thus, the S-wave velocity structure and distribution characteristics of Poisson's ratio in crust of Yunnan are obtained by inversion.The results show that the crustal thickness is gradually thinned from north to south. The crustal thickness in Zhongdian of northwest reaches as many as 62.0 km and the one in Jinghong of further south end is only 30.2 km. What should be especially noted is that there exists a Moho upheaval running in NS in the Chuxiong region and a Moho concave is generally parallel to it in Dongchuan. In addition, there exists an obvious transversal inhomogeneity for the S-wave veIocity structure in upper mantle and crust in the Yunnan region. The low velocity layer exists not only in 10.0-15.0 km in upper crust in some regions, but also in 30.0-40.0 km in lower crust.Generally, the Poisson's ratio is on the high side, however it has a better corresponding relation to the crustal velocity structure. An obvious block distribution feature is still shown on such a high background of Poisson's ratio. It is discovered by synthetically analyzing the velocity structure and Poisson's ratio distribution that there are high Poisson's ratio and complicated crust-mantle velocity structure feature in the Sichuan-Yunnan Diamond Block with Xiaojiang fault to be the east boundary and Yulong Snow Mountain fault to be the west boundary besides the frequent seismicity. | 7 | Micro-mechanical analysis of dynamic processes of nanomanipulation显示文摘In this study, atomic force microscope (AFM) tips are used as tools to cut and manipulate carbon nanotubes on various surfaces. The lateral forces acting on AFM tips during manipulation are also recorded and analyzed from the perspective of micro-mechanics. It is found that differences in surface conditions can lead to obvious increase in micro-friction between nanotube and substrate. And also due to rehybridization, carbon nanotubes present excellent resilience when undergoing different degrees of strain. | 12 | Effect of myosin heavy chain composition of muscles on meat quality in Laiwu pigs and Duroc显示文摘In order to explain the mechanism of high meat quality in Laiwu pigs and investigate the relation between myosin heavy chains (MyHC) composition and meat quality, meat quality analysis was conducted and mRNA expression of MyHC I, IIa, IIx, IIb was quantified by real-time fluorescence PCR in longissimus muscle (LM) and semimembranous muscle of Laiwu pigs and Duroc. The result indicated that, compared with Duroc, mRNA expression of MyHC IIa, IIx in LM and semimembranous muscle of Laiwu pigs was significantly increased, mRNA expression of MyHC IIb was dramatically decreased. However, the expression of MyHC I was not significantly affected by breeds. The correlation between mRNA expression of MyHC I, IIa, IIx in LM and meat color, pH value, marbling, intramuscular fat content was positive, but shear value of LM was negative. The relation between MyHC IIb mRNA expression and marbling, intramuscular fat content was dramatically negative, whereas shear value was strikingly positive, as well as fiber diameter, but without reaching statistical significance. | 14 | Biofeedback therapy for chronic pelvic pain syndrome显示文摘Aim: To evaluate the efficacy of biofeedback therapy in patients with chronic pelvic pain syndrome(CPPS). Methods: From November 2001 to April 2002, patients visiting the Urological Outpatient Clinic of this Hospital were evaluated by means of the National Institute of Health Chronic Prostatitis Symptom Index (NIH-CPSI)and classified by the NIH classification standard. Sixty-two patients of CPPS category Ⅲ were involved in this study. All patients had been treated by conventional approaches such as antibiotics and alpha-blockers for more than half a year without any improvement. The expressed prostatic secretion results were as follows: WBC 5 to 9/high power field, lipid +-+++ and bacterial culture negative. Their NIH-CPSI were 12-40. All the 62 cases complained of micturitional irritation (frequency, urgency, splitted stream and sense of residual urine), 32 cases, of pain or discomfort at the testicular, penile, scrotal, pelvic or rectal region and 13 cases, of white secretion-dripping. The patients were treated by the Urostym Biofeedback equipment (Laborie Co., Canada) 5 times a week for 2 weeks with a stimulus intensity of 15 mA-23 mA and duration of 20 minutes. Results: Sixty patients were significantly improved or cured, while no significant improvement in the remaining 2. No apparent side effect was observed. The NIH-CPSI dropped to 6 to 14 with an average reduction of 21 (P<0.01). In the 60 improved cases, pain was relieved after 2-3treatment courses and other symptoms disappeared after 4-5 courses. Conclusion: Biofeedback therapy is a safe and effective treatment for CPPS. Large randomized clinical trials are needed to confirm its efficacy and to explore the mechanism of action. | 15 | Effects of Soil Water Content on Cotton Root Growth and Distribution Under Mulched Drip Irrigation显示文摘The relation between soil water content and the growth of cotton root was studied for the scheme of field water and cotton yield under mulched drip irrigation. Based on the field experiments, three treatments of soil water content were conducted with 90%θf, 75%θf, and 60%θf (θf is field water capacity). Cotton roots and root-shoot ratio were studied with digging method, and the soil moisture was observed with TDR (time domain reflector), and cotton yield was measured. The results indicated that the growth of cotton root accorded with Logistic growth curve in the three treatments, the cotton root grew quickly and its weight was very high under 75%θf because of the suitable soil water condition, while grew slowly and its weight was lower under 90%θf due to water moisture beyond the suitable condition, and the root weight was in between under 60%θf. For the three water treatments, the cotton root weight decreased with soil depth, and decreased more significantly in deeper soil layer with the soil moisture increasing. And the ratio of cotton root weight in 0-30 cm soil layer to the total root weight was the highest under 75%θf. The cotton root system was distributed mainly in the soil of narrow row and wide row mulched with plastic film, and little in the soil outside plastic film. The weight of cotton root was the highest in the soil of narrow row or wide row mulched with plastic film under 75%θf. Root-shoot ratio decreased with the soil moisture increasing. The soil water content affected cotton yields, and cotton yield was the highest under 75%θf. | 16 | Establishment of an untransfected human corneal epithelial cell line and its biocompatibility with denuded amniotic membrane显示文摘AIM: To establish an untransfected human corneal epithelial (HCEP) cell line and characterize its biocompatibility with denuded amniotic membrane (dAM).METHODS: The torn HCEP pieces were primarily cultured in DMEM/F12 media (pH 7.2) supplemented with 20% fetal bovine serum and other necessary factors,yielding an HCEP cell line which was its growth performance,chromosome morphology,tumorigenicity and expression of marker proteins analyzed.In addition,the biocompatibility of HCEP cells with dAM was evaluated through histological and immunocytochemistry analyses and with light,electron and slit-lamp microscopies.RESULTS: HCEP cells proliferated to confluence in 3 weeks,which have been subcultured to passage 160.A continuous untransfected HCEP cell line,designated as utHCEPC01,was established with a population doubling time of 45.42 hours as was determined at passage 100.The cells retained HCEP cell properties as were approved by chromosomal morphology and the expression of keratin 3.They,with no tumorigenicity,formed a multilayer epithelium-like structure on dAMs through proliferation and differentiation during air-liquid interface culture,maintained expression of marker proteins including keratin 3 and integrin β1 and attached tightly to dAMs.The reconstructed HCEP was highly transparent and morphologically and structurally similar to the original.CONCLUSION: An untransfected and non-tumorigenic HCEP cell line was established in this study.The cells maintained expression of marker proteins.The cell line was biocompatible with dAM.It holds the potential of being used for in vitro reconstruction of tissue-engineered HCEP,promising for the treatment of diseases caused by corneal epithelial disorders. | Ting-Jun Fan Bin Xu Jun Zhao Hong-Shou Yang Rui-Xin Wang and Xiu-Zhong Hu | 2011 | International Journal of Ophthalmology(English edition)2011,4,3: | 22 |

A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. This study analyzes the ribosomal ITS1 sequence (~520 bp) to distinguish genetic difference of B. tabaci from representative different geographical populations in China.
