1Mapping of Fertility Restoring Gene for Aegilops kotschyi Cytoplasmic Male Sterility in Wheat Using SSR Markers显示文摘LK783 was found to be a good fertility restorer for K-type male sterility of wheat. Microsatellite markers were employed to map the major restoring gene in LK783. Maintainer and restorer DNA pools were established using the extreme sterile and fertile plants among (KJ5418A//911289/LK783)F1 population,respectively. Seventy-nine sets of SSR primers were screened for polymorphism between the two pools, 6 of which were found polymorphic. Linkage analysis showed that Xgwm11, Xgwm18 , Xgwm264a and Xgwm273were linked to the restoring gene in LK783, while Xgwm11, Xgwm18 and Xgwm273 were co-segregated. The distance between the Rf gene in LK783 and the three co-segregated markers was 6.54±4.37 cM, the distance between Rf gene and Xgwm264a was 5.71±4.10 cM. The four SSR markers were located on chromosome 1BS by amplifying the DNA of nulli-tetrasomics and ditelosomics of CS with the 4 sets of primers, indicating that the major restoring gene in LK783 was located on 1BS, but the relative location of the gene was different from Rfv1, allelism of the two genes should be further investigated. 2Ectopic expression of a hyacinth AGL6 homolog caused earlier flowering and homeotic conversion in Arabidopsis显示文摘MADS-box genes are involved in floral organ development. Here we report that an AGL6(Agamous-like 6)-like MADS-box gene, HoAGL6, was isolated from Hyacinthus orientalis L. Expression pattern analy-sis demonstrated that HoAGL6 transcript was detected in inflorescence buds, tepals, carpels and ovules, but not in stamina, leaves or scales. Transgenic Arabidopsis plants ectopically expressing HoAGL6 exhibited novel phenotypes of significantly reduced plant size, extremely early flowering, and losing inflorescence indeterminacy. In addition, wide homeotic conversion of sepals, petals, and leaves into carpel-like or ovary structures, and disappearance or number reduction of stamens in 35S::HoAGL6 Arabidopsis plants were also observed. RT-PCR analysis indicated that the expressions of flowering time gene SOC1 and flower meristem identity gene LFY were significantly up-regulated in 35S::HoAGL6 transgenic Arabidopsis plants, and the expression levels of floral organ identity genes AG and SEP1 in leaves were also elevated. 3RAPD-PCR Analysis on Genetic Relationships Between Cultivars of Tree Peony显示文摘Random amplified polymorphic DNA (RAPD) was used to analyze genetic polymophism of 35Tree Peony cultivars with 7 different color groups. Thirty four primers amplified 418 DNA fragments and 337polymorphic bands (80.6%), including specific DNA markers for 18 cultivars that could be used to differentiate cultivars. The UPGMA method was used to analyze the genetic relationship among cultivars. The results showed that 35 Peony cultivars could be divided into 2 cluster groups when using similarity criteria of 1.5, and into 4 cluster groups when using similarity criteria of 1.0. The result confirmed that the flower color has no relation to the genetic clusters and the Tree Peony cultivars originated from the same area has close genetic relationship. Therefore, genetic background has no large effect on the genetic relationship. 6Effect of Nitrogen on Apple Fruit Development in Different Loads显示文摘The experiments including soil application N, thinning and priming leaf treatment, using eight -year-old apple trees (M. domestica Borkh. cv. Red Fuji/M. hupenensis Rhed) as materials were carried out to study the effect of N on fruit development. The main results were as follows: on heavily thinned trees, SS activity was independent of N and priming leaves treatments. The results show that the carbohydrate restriction of source to sink activity did not exist, and N application did not stimulate fruit growth rates relative to those on nonfertilized trees; however, N fertilization resulted in a longer fruit development period and increased the growth potential of individual fruit by 20.8% (fresh weight) and 14.1% (dry weight) vs. controls; in unthinned trees, SS activity was increased by N fertilization but decreased by priming leaves treatment, so the carbohydrate restriction of source to sink activity existed, N fertilization increased the average single fruit weight both by extending the fruit development period and by increasing fruit growth rate, and the increasing rates were 28.2% (fresh weight) and 19.4 % (dry weight) compared to the unthinned nonfertilized controls. 8Polymorphisms in the 5 ′ regulatory region of my-ostatin gene are associated with early growth traits in Yorkshire pigs显示文摘Myostatin is a negative regulator of skeletal muscle mass. The present study cloned the 5′ regulatory region of porcine myostatin gene, screened its polymorphisms and analyzed their associations with early growth traits in Yorkshire pigs. The results indicated that a fragment length polymorphism and a polymorphism concerning two nucleotide changes exist in the 5′ regulatory region of porcine my-ostatin gene. At sites 435 and 447, allele A and allele B have the haplotypes of A-G and G-A, respec-tively. The allelic frequency of B is 0.475 in Yorkshire pigs. No homozygous BB genotype was detected in 9 Laiwu Black pigs. Allele B was found to have positive effect on body weight on day 21 (BW21) (P<0.01), body weight on day 28 (BW28) (P<0.05), body weight on day 70 (BW70) (P<0.05), average daily gain from birth to 21 d (ADG1) (P<0.01), average daily gain from birth to 28 d (ADG2) (P<0.05) and av-erage daily gain from 21 d to 70 d (ADG3) (P<0.01), respectively. 9Immobilization of an enzyme from a Fusarium fungus WZ-I for chlorpyrifos degradation显示文摘The free enzyme extracted from WZ-I,which was identified as Fusarium LK.ex Fx,could effectively degrade chlorpyrifos,an organophosphate insecticide.The methods of immobilizing this free enzyme and determined its degradation-related characteristics were investigated.The properties of the immobilized enzyme were compared with those of the free enzyme.The optimal immobilization of the enzyme was achieved in a solution of 30 g/L sodium alginate at 4°C for 4-12 hr.The immobilized enzyme showed the maximal activity at pH 8.0,45°C.The maximum initial rate and the substrate concentration of the immobilized enzyme were less than that of the free enzyme.The immobilized enzyme,therefore,had a higher capacity to withstand a broader range of temperatures and pH conditions than the free enzyme.With varying pH and temperatures,the immobilized enzyme was more active than the free enzyme in the degradation reaction.In addition,the immobilized enzyme exhibited only a slight loss in its initial activity,even after three repeated uses.The results showed that the immobilized enzyme was more resistant to different environmental conditions,suggesting that it was viable for future practical use.Hui Xie,Lusheng Zhu,Tingting Ma,Jun Wang,Jinhua Wang,Jun Su,Bo Shao College of Resources and Environment,Shandong Agricultural University,Taian 271018,China 2010Journal of Environmental Sciences2010,22,12:3
10Study on the Pathogenicity of Chinese Strains of Subgroup J Avian Leukosis Viruses显示文摘The pathogenicity of 4 Chinese strains of subgroup J avian leukosis viruses (ALV-J), SD9901,SD9902, YZ9901 and YZ9902, was studied. The results showed that only SD9902 among the 4 strains induced mortality from myeloid leukosis (ML). In the 12 meat-type chickens inoculated with SD9902 at 1-day-old, 9died between 22 days and 38 days after inoculation. No death or ML was found in chickens inoculated with the other 3 strains during the period of 6 months. These results suggested the SD9902 strain of ALV-J was an acute transforming virus, but SD9901, YZ9901 and YZ9902 were non-transforming viruses. 11Effect of Nano Red Elemental Selenium on GPx Activity of Broiler Chick Kidney Cells in vitro显示文摘A new selenium source, Nano red elemental selenium (Nano-Se) was used to study the effect on the GPx activity of broiler chick kidney cells (BCKC) in vitro, Sodium selenite (Na_ 2 SeO_ 3 ) and seleno-1-methionine (Se-Met) were used as the controls. The results showed that the effects of three kinds of Se forms on the GPx activity of BCKC were accordant(p>0.05) compared with each other at 0.01,0.05 and 0.10 μmol/L Se concentrations treatments. In the range of 0.00-0.10 μmol/L Se concentrations, the GPx activity increased with elevation of Se concentrations in medium. For the three kinds of Se forms, the GPx activity reached the climax at 0.10 μmol/L Se concentration. At 0.20 and 0.30 μmol/L Se concentrations, the influnces of three kinds of Se forms were not accordant with one another. For Nano-Se, the GPx activity at 0.20 and 0.30 μmol/L Se concentrations remained the same as that at 0.10 μmol/L Se concentration treatment. For Se-Met, the GPx activity at 0.20 μmol/L Se concentration treatment remained the same with 0.10 μmol/L treatment; the GPx activity at 0.30 μmol/L Se concentration treatment was declined significantly(p<0.05) compared with 0.10 or 0.20 μmol/L treatment. For Na_ 2 SeO_ 3 , the GPx activity falled gradually with Se concentration increasing from 0.10 μmol/L to 0.30 μmol/L, and at 0.30 μmol/L Se concentration treatment, the GPx activity was less than the original of BCKC. 12The Comparison of Sugar Components in the Developing Grains of Sweet Corn and Normal Corn显示文摘The sugar components and their dynamic variation in the developing grains of sweet corn(Zeamays L. seccharata Sturt)and normal corn (Zea mays L. indentata Sturt) were compared. There are WSP(water-soluble polysaccharides), sucrose, fructose, glucose, mannitol and sorbitol in both sweet corn and nor-mal corn, but no maltose. Two components with different degrees of polymerization (D. P. N) were detected inthe sweet corn; only one of them was detected in the normal corn 20 days after pollination. With the develop-ment of grains, the total soluble sugar content(TSS)in sweet corn increased, but in normal corn it decreased.The dynamic variation of WSP, sucrose, glucose, fructose, mannitol and sorbitol in sweet and normal corngrains are different. The contents of sugar components in the sweet corn grains are higher than that in the nor-mal corn. 13Effect of Different Fertilizing Methods on Zinc Nutrition in Zn-Deficiency Apple Trees显示文摘Red Fuji apple trees of Zn-deficiency were used as material in the experiment. The effect of dif-ferent Zn application methods on the regularity of Zn transportation and nutrition was studied. It was foundthat the storage of Zn in the branches was greatly increased by foliar application of the solution of 15%as well as between Zn and P and K, which increased Zn transportation and reusability. Zn content in roots in-creased when Zn was used in soil in autumn. However, the absorbed Zn did not transport to the top of thegrowth enhanced Zn content of the branch. However, the effect was kept for a short time. 14Effects of Soil Aeration on Sweet Potato Yield and Its Physiological Mechanism显示文摘The effects of soil aeration on physiological characters and root tuber yield of Ipomoea batatas (L.) Lam. CV Lushu7 and Xushu18 were studied. The results showed that soil aeration improvement could increase ATP content and ATPase activity in functional leaves and root tubers and ABA content in root tubers.It also accelerated the transportation of 14C-photosynthate from leaves to root tubers and enhanced dry matter distribution in root tubers and thus root tuber yield was significantly raised. 15The Analysis of China’s Integrity Situation Based on Big Data显示文摘In order to study deeply the prominent problems faced by China’s clean government work,and put forward effective coping strategies,this article analyzes the network information of anti-corruption related news events,which is based on big data technology.In this study,we take the news report from the website of the Communist Party of China(CPC)Central Commission for Discipline Inspection(CCDI)as the source of data.Firstly,the obtained text data is converted to word segmentation and stop words under preprocessing,and then the pre-processed data is improved by vectorization and text clustering,finally,after text clustering,the key words of clean government work is derived from visualization analysis.According to the results of this study,it shows that China’s clean government work should focus on‘the four forms of decadence’issue,and related departments must strictly crack down five categories of phenomena,such as“illegal payment of subsidies or benefits,illegal delivery of gifts and cash gift,illegal use of official vehicles,banquets using public funds,extravagant wedding ceremonies and funeral”.The results of this study are consistent with the official data released by the CCDI’s website,which also suggests that the method is feasible and effective.Wangdong Jiang Taian Yang Guang Sun Yucai Li Yixuan Tang Hongzhang Lv Wenqian Xiang 2019Journal on Big Data2019,1,3:1

20The Changes of Photosynthetic Properties and Cell Microstructure in Peanut Leaves during Leaf Senescence显示文摘The changes of photosynthetic properties and cell microstructure in peanut leaves during leaf senescence were studied with two high-yielding peanut cultivars (cv. Luhua11 and Fu8707). The main results showed that during the whole process of leaf growth and senescence, changes in the photosynthesis rate (Pn)and contents of chlorophyll in leaves, could be described with a parabolic function, y = A + Bx + Cx2 (where y refers to the values of the above parameters and x to the days after leaf unfolding). During peanut leaf senescence, the shape of chloroplast changed gradually from long ellipses to circles. The starch globule in chloroplast altered gradually from more and larger sizes to fewer and smaller, but the oil globule from fewer and smaller to more and larger.
