2INTERFACE REACTION IN MAGNETIC MULTILAYERS显示文摘Ta/NiO/NiFe/Ta multilayers were prepared by rf reactive and dc magnetron sputtering. The exchange coupling field (Hex) between NiO and NiFe reached 120Oe. The composition and chemical states at the interface region of NiO/NiFe were studied using the x-ray photoelectron spectroscopy (XPS) and peak decomposition technique. The results show that there are two thermodynamically favorable reactions at NiO/NiFe interface: NiO+Fe=Ni+FeO and 3NiO+2Fe = 3Ni+Fe2O3. The thickness of the chemical reaction as estimated by angle-resolved XPS was about 1-1.5nm. These interface reaction products are magnetic defects, and we believe that the Hex and the coercivity (Hc) of NiO/NiFe are affected by these defects. Moreover, the results also show that there is an 'intermixing layer' at the Ta/NiO (and NiO/Ta) interface due to a thermodynamically favorable reaction: 2Ta+5NiO=5Ni+ Ta2O5. 5高分辨电感耦合等离子质谱测定黄连中的微量元素显示文摘文章采用高分辨电感耦合等离子体质谱(HR-ICP-MS)的方法,测定了中草药中四种黄连——味连、雅连、马尾连和胡黄连中的微量元素。结果表明:在所有16种稀土元素中,La,Ce,Nd,Gd,Sm,Sc含量较高,胡黄连的稀土总量为5.18μg·g^(-1),是所有四种黄连的最高者,其次是雅连和马尾连,最低的是味连(1.26μg·g^(-1))。而对于与清热解毒有关的金属无素如Mg,Mn,Zn,Ca和Fe元素,雅连中Mg,Mn,Zn的含量最高。高Fe和Ca的含量存在于马尾连和胡连之中。在测定不同提取方法的提取物中发现,只有部分的稀土元素能被醇和水所提取,它们分别是31.6%和4.1%。清热解毒元素的提取率明显高于稀土元素。王小逸 Zhu W Witkamp G T 曾衍钧 2003光谱学与光谱分析2003,23,6:20

8Exogenous abscisic acid significantly affects proteome in teaplant (Camellia sinensis) exposed to drought stress显示文摘Tea[Camellia sinensis(L.)O.Kuntze]is an important economic crop,and drought is the most important abiotic stress affecting yield and quality.Abscisic acid(ABA)is an important phytohormone responsible for activating drought resistance.Increased understanding of ABA effects on tea plant under drought stress is essential to develop drought-tolerant tea genotypes,along with crop management practices that can mitigate drought stress.The objective of the present investigation is evaluation of effects of exogenous ABA on the leaf proteome in tea plant exposed to drought stress.Leaf protein patterns of tea plants under simulated drought stress[(polyethylene glycol(PEG)-treated]and exogenous ABA treatment were analyzed in a time-course experiment using two-dimensional electrophoresis(2-DE),followed by matrix-assisted laser desorption/ionization time-of-flight(MALDI-TOF)mass spectrometry(MS).Among the 72 protein spots identified by MALDI-TOF MS,16 proteins were downregulated and two were upregulated by exogenous ABA.The upregulated proteins have roles in glycolysis and photosystem II stabilization.Twenty-one protein spots were responsive to drought stress and most participate in carbohydrate and nitrogen metabolism,control of reactive oxygen species(ROS),defense,signaling or nucleic acid metabolism.The combined treatments of exogenous ABA and drought showed upregulation of 10 protein spots at 12 h and upregulation of 11 proteins at 72 h after initiation of drought stress.The results support the importance of the role that ABA plays in the tea plant during drought stress,by improving protein transport,carbon metabolism and expression of resistance proteins.Lin Zhou Hui Xu Sue Mischke Lyndel W Meinhardt Dapeng Zhang Xujun Zhu Xinghui Li Wanping Fang 2014Horticulture Research2014,1,1:12

11Cultivation-Independent Analysis of the Development of the Lactobacillus spp.Community in the Intestinal Tract of Newborn Piglets显示文摘Molecular diversity and development of the Lactobacillus community in the intestinal tract, as influenced by age and intestinal compartment, were studied in one litter of 12 conventionally raised piglets. Piglets were euthanized at each week (3 animals per time). Digesta and tissue samples from stomach, duodenum, jejunum, ileum, caecum, colon, and rectum were collected and analysed by using 16S ribosomal RNA-based methods. DGGE (denaturing gradient gel electrophoresis) profiles revealed that the Lactobacillus communities throughout the GI tract from duodenum to rectum showed good stability at same age. This indicates that fecal Lactobacillus communities can effectively represent the intestinal community. Two dominant bands were found in tissue samples of the small intestine, suggesting that the lactobacilli can adhere to the small intestinal wall. The Lactobacillus communities in different GI tract compartments developed over time. A successional change of Lactobacillus communities was observed from birth, through creep feeding to one week after weaning, showing a trend from simple to complex and back to simple. Furthermore, a clone library of Lactobacillus spp. 16S rRNA gene sequences were generated from jejunal and colonic chymes. Six dominant DGGE bands generated from jejunal chymes were matched with sequences that show 94-98% similarity to the bands derived from L. reuteri, L. delbrueckii, and L. crispatus. Seven dominant DGGE bands generated from colon chymes were matched with sequences that show 88-99% similarity to those derived from L. reuteri, L. delbrueckii, L. amylovorus/L. sobrius, and L. acidophilus. Amplicons related to L. reuteri were found in all DGGE fingerprints from jejunal digesta of age of weeks 1, 3, and 4. Amplicons related to L. amylovorus/L. sobrius were present in all DGGE fingerprints from colonic digesta of age of week 1, 3, and 4. Amplicons related to L. delbrueckii were found before weaning, L. crispatus after creep feeding before weaning and L. acidophilus after weaning. 15Development of a laser capture microscope-based single-cell-type proteomics tool for studying proteomes of individual cell layers of plant roots显示文摘Single-cell-type proteomics provides the capability to revealing the genomic and proteomics information at cell-level resolution.However,the methodology for this type of research has not been well-developed.This paper reports developing a workflow of laser capture microdissection(LCM)followed by gel-liquid chromatography-tandem mass spectrometry(GeLC-MS/MS)-based proteomics analysis for the identification of proteomes contained in individual cell layers of tomato roots.Thin-sections(~10-μm thick,10 sections per root tip)were prepared for root tips of tomato germinating seedlings.Epidermal and cortical cells(5000–7000 cells per tissue type)were isolated under a LCM microscope.Proteins were isolated and then separated by SDS–polyacrylamide gel electrophoresis followed by in-gel-tryptic digestion.The MS and MS/MS spectra generated using nanoLC-MS/MS analysis of the tryptic peptides were searched against ITAG2.4 tomato protein database to identify proteins contained in each single-cell-type sample.Based on the biological functions,proteins with proven functions in root hair development were identified in epidermal cells but not in the cortical cells.Several of these proteins were found in Al-treated roots only.The results demonstrated that the cell-type-specific proteome is relevant for tissue-specific functions in tomato roots.Increasing the coverage of proteomes and reducing the inevitable cross-contamination from adjacent cell layers,in both vertical and cross directions when cells are isolated from slides prepared using intact root tips,are the major challenges using the technology in proteomics analysis of plant roots.Yingde Zhu Hui Li Sarabjit Bhatti Suping Zhou Yong Yang Tara Fish Theodore W Thannhauser 2016Horticulture Research2016,3,1:2
