1厚朴酚对高脂饲料-STZ 诱导的糖尿病大鼠血糖血脂及肝脏氧化应激损伤的影响 (英文)显示文摘本文研究了厚朴酚对高脂饲料-链脲佐菌素诱导的2型糖尿病大鼠高血糖、高血脂、肝脏氧化应激和CYP2E1活性的影响。与糖尿病模型组相比,连续给予厚朴酚(25、50和100 mg·kg-1·d-1,灌胃)10周后,其血糖、血脂(TC、TG和LDL-C)及肝脏CYP2E1活性和MDA含量显著降低,口服糖耐量和肝脏抗氧化酶(CAT和GSH-Px)活性显著增加。研究结果不仅证明厚朴酚对高脂饲料-链脲佐菌素诱导的2型糖尿病大鼠的高血糖、高血脂、肝脏氧化应激损伤有明显治疗作用,而且厚朴酚对糖尿病大鼠肝脏CYP2E1活性的抑制作用可能是其拮抗糖尿病所致肝脏氧化损伤及胰岛素抵抗的重要机制之一。王俊俊 赵容 梁继超 陈勇 2014药学学报2014,49,4:11

3In vitro and in vivo antioxidative and hepatoprotective activity of aqueous extract of Cortex Dictamni显示文摘AIM To investigate the antioxidant and hepatoprotective effects of Cortex Dictamni aqueous extract(CDAE) in carbon tetrachloride(CCl4)-induced liver damage in rats.METHODS The in vitro antioxidant effect of CDAE was investigated using α,α-diphenyl-β-picrylhydrazyl(DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS), β-carotene bleaching, reducing power, and thiobarbituric acid reactive substance assays. A linoleic acid system, including ferric thiocyanate(FTC) and thiobarbituric acid(TBA) assays, was used to evaluate the inhibition of lipid peroxidation. The in vivo hepatoprotective and antioxidant effects of CDAEagainst CCl4-induced liver damage were evaluated in Sprague-Dawley rats. Silymarin was used as a positive control. Liver damage was assessed by determining hepatic histopathology and liver marker enzymes in serum. Enzyme and non-enzyme antioxidant levels and lipid peroxide content were measured in the liver. Cytochrome P450 2E1(CYP2E1) protein expression was measured via immunohistochemical staining. Nuclear factor E2-related factor(Nrf2), heme oxygenase-1(HO-1), NAD(P)H quinine oxidoreductase 1(NQO1), and γ-glutamylcysteine synthetase catalytic subunit(γ-GCSc) protein expression was measured by Western blot.RESULTS Our results showed that CDAE exhibited a strong antioxidant activity in vitro. CDAE scavenged DPPH and ABTS radicals in a dose-dependent manner. CDAE inhibited lipid peroxidation with a lipid peroxide inhibition rate of 40.6% ± 5.2%. In the FTC and TBA assays, CDAE significantly inhibited lipid peroxidation(P < 0.01). In vivo histopathological studies indicated that CCl4-induced liver injury was alleviated following CDAE treatment in rats of both sexes. CDAE(160 and 320 mg/kg) significantly prevented CCl4-induced elevations of alkaline phosphatase, glutamate pyruvate transaminase, aspartate aminotransferase, and total bilirubin levels in rats of both sexes(P < 0.05, 0.01, or 0.001). Moreover, CDAE restored the decreased activities of hepatic antioxidant enzymes, including superoxide dismutase, catalase, and glutathione peroxidase, as well as non-enzyme antioxidant glutathione, which were induced by CCl4 treatment. CDAE significantly suppressed the up-regulation of CYP2E1 and promoted Nrf2, HO-1, NQO1, and γ-GCSc protein expression.CONCLUSION CDAE exhibits good antioxidant performance in vitro, with marked radical-scavenging and anti-lipid peroxidation activities. CDAE is effective in preventing CCl4-induced hepatic damage in rats of both sexes. 5茶多酚对乙醇所致原代肝细胞损伤的保护作用研究显示文摘目的研究乙醇对原代肝细胞的氧化损伤作用,同时探讨茶多酚对乙醇所致的原代肝细胞损伤模型的保护作用。方法分离小鼠原代肝细胞,直接以不同浓度乙醇作用于肝细胞,或以茶多酚预处理后再用乙醇作用于肝细胞,测定各组细胞存活率、抗氧化酶及丙氨酸氨基转移酶(ALT)活性的变化。结果随着乙醇浓度的增加,小鼠原代肝细胞存活率逐渐下降,抗氧化酶活性降低,转氨酶升高;而茶多酚的预处理对于乙醇所致的小鼠原代肝细胞损伤具有明显的保护作用,表现为相同乙醇浓度作用下,随茶多酚浓度的升高,细胞存活率逐渐升高,抗氧化酶活性增加,转氨酶下降,且差异有统计学意义(P<0.05)。结论乙醇可以诱导小鼠原代肝细胞的氧化损伤,而茶多酚对其氧化损伤具有明显的保护作用。张栗 冉云 王玲 陈晨 张遵真 2009营养学报2009,31,4:4
6Antidiabetic and Anti-oxidative Effects of Honokiol on Diabetic Rats Induced by High-fat Diet and Streptozotocin显示文摘Objective To study the antidiabetic and anti-oxidative effects of honokiol(Hon)in Magnolia officinalis and its underlying molecular mechanism in diabetic rats induced by high-fat diet(HFD)and streptozotocin(STZ).Methods After ig administration with Hon[25,50,and 100 mg/(kg·d)]to diabetic rats for consecutive 10 weeks,the levels of blood glucose(BG),oral glucose tolerance(OGT),blood lipids including total cholesterol(TC),triglyceride(TG),high density lipoprotein-cholesterol(HDL-C),and low density lipoprotein-cholesterol(LDL-C),hepatic oxidative stress including the activities of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px),methane dicarboxylic aldehyde(MDA),and cytochrome P4502E1(CYP2E1)in diabetic rats were measured.Results Compared to the diabetic control rats,ig administration of Hon resulted in significant decrease in BG,TC,TG,and LDL-C levels in serum,as well as hepatic CYP2E1 activity and MDA content in diabetic rats,whereas the level of OGT and activities of hepatic CAT,SOD,and GSH-Px in diabetic rats were significantly increased.Conclusion Hon could alleviate hyperglycemia,hyperlipemia,hepatic oxidative damage,and insulin resistance in diabetic rats by inhibiting hepatic CYP2E1 activity.Jun-jun Wang Rong Zhao Ji-chao Liang Yong Chen 2014Chinese Herbal Medicines2014,6,1:4
7一次性大剂量乙醇摄入对小鼠肝脏损伤作用的研究显示文摘目的研究一次性大剂量乙醇摄入对小鼠肝脏功能的损害作用。方法将健康SPF级雄性昆明小鼠24只随机分为对照组(蒸馏水)和乙醇(50%,V/V)组,每组12只。采用一次性灌胃方式进行染毒,染毒剂量为12ml/kg。染毒16h后,称重,观察并记录各组小鼠的一般情况。迅速取出肝脏,称重,计算肝脏系数。测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、葡萄糖(Glu)、总蛋白(TP)含量,通过检测肝脏中甘油三酯(TG)、游离脂肪酸(FFA)的含量和病理学观察来反映肝脏中脂肪的蓄积,通过测定钙离子诱导的线粒体渗透性转变(MPT)来检测线粒体的功能,通过测定肝组织和线粒体中的丙二醛(MDA)和还原性谷胱甘肽(GSH)的含量反映肝脏的脂质过氧化水平,测定肝微粒体细胞色素P450(CYP)2E1、1A2和3A的活性和蛋白表达量。结果一次性大剂量乙醇摄入后,小鼠逐渐出现反应迟钝、步态不稳、翻正反射消失等醉酒症状;约3h后,翻正反射恢复。与对照组相比,乙醇组小鼠肝脏系数较高,血清ALT、AST活力较高,Glu和TP含量较低,肝脏中TG和FFA含量较高,肝组织匀浆和肝细胞线粒体中MDA的含量较高,肝组织匀浆中GSH含量较低,肝脏微粒体CYP2E1活性和蛋白表达量增加,差异均有统计学意义(P<0.05或P<0.01);而CYP1A2和3A未见明显改变(P>0.05)。乙醇组小鼠线粒体加入Ca2+后A值高于对照组,差异有统计学意义(P<0.05或P<0.01)。乙醇组小鼠肝脏可见大量的肝细胞发生脂肪变性。结论一次性大剂量乙醇摄入可导致小鼠肝脏脂质代谢障碍、线粒体功能紊乱、肝组织和线粒体中脂质过氧化反应的增强和CYP2E1的激活。曾涛 张翠丽 宋福永 赵秀兰 于丽华 朱振平 谢克勤 2009环境与健康杂志2009,26,7:3

10Determination of platelet-activating factor by reverse phase high-performance liquid chromatography and its application in viral hepatitis显示文摘AIM: To detect the platelet-activating factor (PAF) and the plasma or serum levels of tumor necrosis factor-α (TNF-α) malondialdehyde (MDA), endotoxin (ET) and to discuss their significance in various types of viral hepatitis.METHODS: PAF, TNF-α, MDA, and ET levels in 60 controls, 16 cases of acute viral hepatitis, 71 cases of chronic viral hepatitis, 19 cases of severe viral hepatitis were detected by reverse phase high-performance liquid chromatography (rHPLC), bio-assay, ELISA, thiobarbituric acid (TBA), and limulus lysate test (LLT), respectively.RESULTS: The rHPLC was more sensitive and specific than bio-assay (r = 0.912, P<0.01). 11乙醇染毒对果蝇寿命及某些生化指标的影响显示文摘目的探讨不同浓度乙醇对果蝇寿命以及果蝇体内超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力、以及丙二醛(MDA)含量的影响。方法收集8小时内羽化未交配雌雄果蝇7200只,雌雄各半。取其中2400只,随机分为6组,每组雌雄果蝇各200只,给予不同浓度乙醇染毒,每4天更换1次培养基,每天记录死亡数,用生存实验检测乙醇对果蝇寿命的影响。另取4800只,随机分为4组,每组雌雄各600只,给予含不同浓度乙醇的培养基喂养,分别于喂养0天、10天、20天和30天测定SOD和GSH-Px活力,同时检测MDA的含量。结果随着乙醇剂量的增加,雌雄果蝇的寿命均呈下降趋势,高剂量乙醇染毒组果蝇寿命显著降低(P<0.05)。在相同染毒剂量下,随着染毒时间的增加,果蝇体内SOD、GSH-Px活性逐渐降低,MDA含量逐渐升高;当染毒20天和30天时,90mmol/L、270mmol/L剂量组SOD、GSH-Px活性及MDA含量与对照组相比差异有显著性(P<0.05)。结论乙醇暴露能降低果蝇寿命,并导致果蝇体内SOD和GSH-Px活力的降低和MDA含量的增高。张栗 冉云 吴媚 王玲 刘蜀坤 张遵真 2009卫生研究2009,38,2:0
