| 1 | Ameliorating effects of traditional Chinese medicine preparation, Chinese materia medica and active compounds on ischemia/reperfusion-induced cerebral microcirculatory disturbances and neuron damage显示文摘Ischemic stroke and ischemia/reperfusion(I/R) injury induced by thrombolytic therapy are conditions with high mortality and serious long-term physical and cognitive disabilities. They have a major impact on global public health. These disorders are associated with multiple insults to the cerebral microcirculation, including reactive oxygen species(ROS) overproduction, leukocyte adhesion and infiltration, brain blood barrier(BBB) disruption, and capillary hypoperfusion, ultimately resulting in tissue edema, hemorrhage, brain injury and delayed neuron damage. Traditional Chinese medicine(TCM)has been used in China, Korea, Japan and other Asian countries for treatment of a wide range of diseases.In China, the usage of compound TCM preparation to treat cerebrovascular diseases dates back to the Han Dynasty. Even thousands of years earlier, the medical formulary recorded many classical prescriptions for treating cerebral I/R-related diseases. | 3 | Over-expression of VEGF in Marrow Stromal Cells Promotes Angiogenesis in Rats with Cerebral Infarction via the Synergistic Effects of VEGF and Ang-2显示文摘This study explored whether the transplantation of modified marrow stromal cells (MSCs) has angiogenic effects in a left middle cerebral artery occlusion infarction/reperfusion (MCAO I/R) rat model and preliminarily examined the mechanism of angiogenesis following cerebral infarction.MSCs were isolated by using a direct adherent method and cultured.Vascular endothelial growth factor (VEGF) was transfected into MSCs by employing the liposome transfection.The transfection efficiency was measured by the optical density method.The protein expression of VEGF gene before and after transfection was measured by Western blotting.SD rat model of transient occlusion of the left middle cerebral artery was established by using an approach of intra-luminal occlusion.Tetrazolium (TTC) and HE staining were performed to observe the cerebral infarction.ELISAs were used to measure the levels of VEGF in the rat cerebral tissues.The expression patterns of angiopoietin-2 (Ang-2) and CD34 in cells surrounding the area of infarction were immunohistochemistrically oserved.Ang-2 protein expression in the tissue surrounding the area of infarction was measured by Western blotting.VEGF expression in the MSCs increased after transfection at a rate of approximately 28%±3.4%.ELISA showed that the expression of VEGF in the cerebral tissue was significantly increased after induction of infarction,peaking on the 4th day and decreasing to the levels of the sham surgery group (normal) within 7 to 10 days.The VEGF level was significantly higher at each time point in the VEGF-MSC and MSC groups compared to the model group.Moreover,the VEGF level was higher in the VEGF-MSC group than in the MSC group and stayed relatively high until the 10th day.The immunohistochemical results showed that 10 days after the infarction,the number of Ang-2 and CD34-expressing cells in the area surrounding the infarction was significantly higher in the VEGF-MSC group and the MSC group compared to the model group.Moreover,the VEGF level was higher in the VEGF-MSC group than the MSC group.A similar trend in Ang-2 protein expression was revealed by Western blotting.In the MCAO rat model transfected with modified MSCs over-expressing VEGF,compared to the MSC transplantation group,the concentration of VEGF was significantly increased in the brain tissue after cerebral infarction.In addition,the level of Ang-2 was up-regulated,with angiogenesis promoted,the blood supply to the areas surrounding the cerebral infarction increased,and neurological function improved.We are led to speculate that the synergistic effects of VEGF and Ang-2 may be responsible for the angiogenesis following cerebral infarction. | 赖天宝 李嫚 郑丽芳 宋艳玲 徐小丽 郭远瑾 张远 张宗胜 梅元武 | 2012 | Journal of Huazhong University of Science and Technology(Medical Sciences)2012,32,5: | 26 |
| 4 | 基于网络药理学研究补阳还五汤干预缺血性脑卒中神经血管单元的作用机制显示文摘目的利用网络药理学方法研究补阳还五汤治疗缺血性脑卒中涉及的重要信号通路和保护神经血管单元的作用机制。方法使用TCMSP、上海有机所中药与化学成分数据库和相关文献获取并筛选补阳还五汤的活性成分及作用靶点;使用DrugBank数据库、药物靶标数据库(TTD)、在线人类孟德尔遗传数据库(OMIM)和GeneCards数据库获取缺血性脑卒中靶点。使用Cytoscape 3.7.2软件建立活性成分-治疗靶点网络,分析网络获取关键靶点。使用Metascape数据库对治疗靶点进行GO富集分析和KEGG信号通路富集分析。使用AlzData数据库分析治疗靶点的基因表达情况。结果通过网络分析,获得了补阳还五汤治疗缺血性脑卒中的关键靶点,包括前列腺素内过氧化物合酶2、前列腺素内过氧化物合酶1、毒蕈碱型乙酰胆碱受体M1、β2肾上腺素能受体等;通过富集分析获取了补阳还五汤治疗缺血性脑卒中的多条关键信号通路,包括PI3K-Akt信号通路、钙信号通路、IL-17信号通路、血小板活化、Apelin信号通路、NF-κB信号通路、AMPK信号通路和花生四烯酸代谢通路等;通过基因表达分析了补阳还五汤对不同组织细胞的作用,发现治疗靶点在内皮细胞、星形胶质细胞、小胶质细胞、少突胶质细胞、少突胶质细胞前体细胞和神经元中调控多种生物过程、细胞组成和分子功能。结论补阳还五汤治疗缺血性脑卒中具有多成分、多靶点、多通路、联系复杂的特点,在基因的细胞表达层面与神经血管单元密切相关,治疗作用机制包括神经保护、神经发生、抗血栓、促血管再生、胶质细胞调控等。 | 韩沛凇 马重阳 赵晖 | 2021 | 国际中医中药杂志2021,43,3: | 18 |
| 5 | Buyang Huanwu Decoction ameliorates ischemic stroke by modulating multiple targets with multiple components: In vitro evidences显示文摘Buyang Huanwu Decoction(BYHWD) is a well-known traditional Chinese medicine prescription which is used to treat ischaemic stroke and stroke-induced disabilities. However, the exact mechanism underlying BYHWD's amelioration of ischaemic stroke and its effective constituents remain unclear. The present study aimed to identify the effective constituents of BYHWD and to further explore its action mechanisms in the amelioration of ischaemic stroke by testing the activities of 15 absorbable chemical constituents of BYHWD with the same methods under the same conditions. The following actions of these 15 compounds were revealed: 1) Ferulic acid, calycosin, formononetin, astrapterocarpan-3-O-β-D-glucoside, paeonol, calycosin-7-O-β-D-glucoside, astraisoflavan-7-O-β-D-glucoside, ligustrazine, and propyl gallate significantly suppressed concanavalin A(Con A)-induced T lymphocyte proliferation; 2) Propyl gallate, calycosin-7-O-β-D-glucoside, paeonol, and ferulic acid markedly inhibited LPS-induced apoptosis in RAW264.7 cells; 3) Propyl gallate and formononetin significantly inhibited LPS-induced NO release; 4) Hydroxysafflor yellow A and inosine protected PC12 cells against the injuries caused by glutamate; and 5) Formononetin, astragaloside IV, astraisoflavan-7-O-β-Dglucoside, inosine, paeoniflorin, ononin, paeonol, propyl gallate, ligustrazine, and ferulic acid significantly suppressed the constriction of the thoracic aorta induced by KCl in rats. | 6 | 补阳还五汤联合骨髓间充质干细胞移植脑缺血再灌注大鼠脑组织紧密连接蛋白的表达显示文摘背景:急性脑缺血时内皮细胞间的紧密连接受到破坏,紧密连接蛋白转录或表达发生改变,引起血脑屏障通透性增加而形成脑水肿。目的:旨在从紧密连接蛋白的调控方面,探讨补阳还五汤联合骨髓间充质干细胞移植对脑缺血再灌注大鼠血脑屏障修复的作用机制。方法:80只大鼠(山东省动物实验中心提供)随机分为假手术组(n=16),模型组(n=16),骨髓间充质干细胞组(n=16),补阳还五汤组(n=16),补阳还五汤联合骨髓间充质干细胞组(联合组,n=16)。利用改良线栓法制备大脑中动脉栓塞缺血再灌注大鼠模型,造模前3 d开始,补阳还五汤组、联合组给予补阳还五汤15 mL/kg灌胃,假手术组、模型组、骨髓间充质干细胞组给予等量生理盐水灌胃;造模后1d时,骨髓间充质干细胞组、联合组尾静脉注射含1×10~6个骨髓间充质干细胞悬液1m L,其他组给予等量生理盐水。在造模后3,5d取缺血侧脑组织,采用Western Blot法检测脑组织中Occludin、Claudin-5、JAM-A、ZO-1等紧密连接相关蛋白的表达。结果与结论:(1)与假手术组相比,模型组大鼠缺血脑组织中Occludin、Claudin-5、JAM-A、ZO-1的蛋白表达显著下降(P<0.01);(2)与模型组相比,补阳还五汤组、骨髓间充质干细胞组和联合组缺血脑组织中Occludin、Claudin-5、JAM-A、ZO-1的蛋白表达量上升(P <0.05),以联合组上调最为明显(P <0.01);(3)结果表明,补阳还五汤联合骨髓间充质干细胞对血脑屏障的修复具有协同作用,可能是通过上调缺血脑组织中Occludin、Claudin、JAM-A、ZO-1的蛋白表达来维持血脑屏障紧密连接的完整性,以实现脑保护的作用。 | 张运克 车志英 李可 | 2019 | 中国组织工程研究2019,23,1: | 16 |
| 7 | 补阳还五汤联合骨髓间充质干细胞对脑缺血大鼠血脑屏障通透性的影响显示文摘目的:为了研究补阳还五汤联合骨髓间充质干细胞(MSCs)对脑缺血再灌注大鼠血脑屏障(blood brain barrier, BBB)通透性及其影响因素AQP4的表达。方法:将大鼠80只随机分为假手术组、模型组、补阳还五汤组、MSCs组、补阳还五汤联合MSCs组(联合组),每组各16只。利用改良线栓法制备大脑中动脉栓塞(middle cerebral artery obstruction,MCAO)缺血再灌注大鼠模型,在3、5 d运用干湿重法测定缺血脑组织含水量,采用伊文思蓝(Evans blue, EB)法检测血脑屏障的通透性,用透射电镜观察血脑屏障超微结构的变化,采用Western Blot法检测缺血脑组织中AQP4的表达。结果:与假手术组相比,在造模后3、5 d时间点,MCAO模型组大鼠脑组织含水量、EB含量显著升高,AQP4的蛋白表达水平上升,具有极显著性统计学意义(P<0.01);与模型组相比,补阳还五汤组、MSCs组、联合组中缺血脑组织含水量、EB含量及AQP4的蛋白表达水平显著下降,各组间有显著统计学意义(P<0.05)。联合组的作用在第5天更为明显,与MSCs组及补阳还五汤组相比,具有显著性差异(P<0.05)。结论:补阳还五汤联合MSCs可以降低脑组织的含水量及血脑屏障的通透性,维持BBB结构的完整性,其作用机制与水通道蛋白AQP4的调控密切相关。 | 张运克 车志英 李可 | 2019 | 中华中医药学刊2019,37,7: | 16 |

| 10 | 补阳还五汤联合骨髓间充质干细胞移植对大脑中动脉栓塞模型整合素相关因子的影响显示文摘背景:前期研究发现,补阳还五汤联合骨髓间充质干细胞移植可发挥协同抗脑缺血损伤作用。目的:分析骨髓间充质干细胞移植联合补阳还五汤促进脑缺血血管新生的作用。方法:将96只SD大鼠随机分为4组,均制作大脑中动脉栓塞模型。联合组于造模前3 d灌胃给予补阳还五汤10 mL/kg,1次/d,造模成功后2 h灌胃1次,以后每12 h灌胃1次;局灶脑缺血2 h再灌注2 h后,干细胞组、联合组向脑室中注入骨髓间充质干细胞悬液,30 min后再注射抗CD34、CD45抗体;抗体组注射抗CD34、CD45抗体;模型组注射生理盐水。联合组于再灌注后12,24,36,48 h进行血清αVβ3水平及脑组织免疫组织化学、Q-PCR、免疫印迹检测,其余3组均于再灌注后36 h检测上述指标。结果与结论:(1)血清αVβ3水平:抗体组血清αVβ3水平低于模型组(P<0.05),干细胞组、联合组高于抗体组(P<0.05),且联合组再灌注不同时间点的血清αVβ3水平高于干细胞组(P<0.01);(2)免疫组织化学:抗体组CD34阳性细胞数量少于模型组(P<0.05),干细胞组、联合组高于抗体组(P<0.05);(3)Q-PCR与免疫印迹检测:与模型组相比,抗体组、干细胞组p FAK蛋白表达水平降低(P<0.05),FAK基因及蛋白表达变化不大;与其他组比较,联合组FAK蛋白(自再灌注后24 h开始)和p FAK蛋白(自再灌注12 h开始)显著升高(P<0.05,P<0.01),且随着再灌注时间延长有逐渐升高趋势;(4)结果表明:骨髓间充质干细胞移植联合补阳还五汤可逆转抗CD34+CD45抗体引起的血管内皮细胞数量减少、整合素αVβ3及其下游信号分子表达下降,促进血管新生。 | 张运克 杨俊红 高峰 段枫阳 | 2017 | 中国组织工程研究2017,21,5: | 11 |

| 12 | 补阳还五汤联合骨髓间充质干细胞对大脑中动脉闭塞再灌注治疗的作用机制显示文摘背景:急性脑缺血时基质金属蛋白酶通过降解细胞外基质,破坏内皮细胞间的紧密连接而增加微血管的通透性,破坏血脑屏障,导致脑水肿的发生。目的:旨在从基质金属蛋白酶及细胞外基质方面,探讨补阳还五汤联合骨髓间充质干细胞移植对脑缺血再灌注大鼠的治疗作用机制。方法:将96只SD大鼠随机分为模型组,组织型基质金属蛋白酶抑制剂1组,组织型基质金属蛋白酶抑制剂1+骨髓间充质干细胞组(简称骨髓间充质干细胞组),组织型基质金属蛋白酶抑制剂1+补阳还五汤联合骨髓间充质干细胞组(简称联合组,分12,24,36,48 h 4个亚组)。构建大鼠脑缺血再灌注模型,在立体定位仪下采用微量进样器脑部给予基质金属酶抑制剂及培养的骨髓间充质干细胞;联合组术前7 d开始给予补阳还五汤(10 m L/kg)灌胃,其余各组给予等剂量的生理盐水。给药10 d后采集实验动物血清和脑组织,采用ELISA法测定血清中基质金属蛋白酶2、基质金属蛋白酶9水平,明胶酶谱法分析脑组织基质金属蛋白酶9活性。结果与结论:与模型组相比,各组血清基质金属蛋白酶2、基质金属蛋白酶9水平和脑组织基质金属蛋白酶9的活性均有不同程度的下降,其中基质金属蛋白酶9下降明显(P<0.05);与骨髓间充质干细胞组相比,联合组36 h和联合组48 h血清基质金属蛋白酶2、基质金属蛋白酶9水平和脑组织基质金属蛋白酶9、基质金属蛋白酶9前体活性下降最为明显(P<0.01)。结果表明补阳还五汤联合骨髓间充质干细胞可与组织型基质金属蛋白酶抑制剂1协同作用,从而抑制因基质金属蛋白酶2、基质金属蛋白酶9所致的细胞外基质降解,修复受损的血脑屏障及防治缺血后脑水肿的形成。 | 张运克 宋军营 张振强 | 2015 | 中国组织工程研究2015,19,50: | 10 |

After 28 days, nestin and neuron-specific enolase were expressed in the cytoplasm. Reverse transcription-PCR and western blot analyses showed that nestin and neuron-specific enolase mRNA and protein expression was greater in the active principle region group compared with the original formula group. | 17 | 加味补阳还五汤对七氟烷麻醉处理后早期糖尿病肾病大鼠肾足细胞影响实验研究显示文摘目的:探讨加味补阳还五汤(MDIY)对七氟烷麻醉处理后的早期糖尿病肾病(DN)大鼠肾足细胞的影响。方法:将早期DN大鼠随机分为4组(每组n=16):MDIY组(A组),罗格列酮组(B组),MDIY+罗格列酮组(C组)和模型对照组(D组),另设正常大鼠(E组)为空白对照组。连续治疗7周,每天收集大鼠死亡数和葡萄糖(GLU)、肌酐(CR)、尿素氮(BUN)和尿微量白蛋白(UAL)。七氟烷麻醉处理后采集标本,通过苏木精和伊红染色(H&E)收集肾脏和肾病理变化检查。应用实RT-PCR和western blotting检测评估肾足突细胞各m-RNA的表达水平,包括nephrin、podocin、α-SMA和desmin的表达。结果:在治疗周期中,A组、B组和C组的GLU和UAL水平均低于D组(P<0.01),且7周后3组的GLU、CR和BUN水平也都远低于D组(P<0.05)。其中,C组的上述生化指标远低于A组和B组(P<0.05)。相比于正常SD大鼠,通过H&E染色,观察各组的肾小球疾病、慢性肾小管间质损伤和肾小球足细胞损伤的发病率。然而,只有C组显示更能优化改善上述损伤的发病率。在A组、B组和C组与D组比较,α-SMA和desmin的表达减少,而nephrin和podocin的表达明显增加(P<0.05);C组不仅nephrin和podocin mRNA表达(P<0.05),而且在蛋白表达方面均观察有协同效应。结论:MDIY对早期DN引起的肾功能损害是一种保护作用,可以大大提高与罗格列酮相结合的治疗效果,其机制是通过增加nephrin、podocin的表达和减少α-SMA、desmin的表达,从而推测MDIY具有显著的协同治疗DN作用。七氟烷预处理能改善糖尿病肾病足细胞结构和功能,对围术期外科快速康复有利。 | 林华阳 刘志宏 曾忱 林斐翎 刘国平 田芳曦 陈明芳 郑京 邱水生 | 2019 | 辽宁中医药大学学报2019,21,8: | 5 |
