1苏丹红Ⅰ、Ⅲ和Ⅳ对HepG-2细胞和SGC-7901细胞增殖的影响显示文摘研究苏丹红Ⅰ、Ⅲ和Ⅳ对人肝癌细胞HepG-2,胃癌细胞SGC-7901增殖的影响.利用激光共聚焦显微镜及三维图像分析HepG-2细胞受到苏丹红Ⅰ、Ⅲ和Ⅳ不同剂量作用后DNA和RNA含量的比值,流式细胞仪测定SGC-7901细胞DNA含量的变化,分析细胞周期的变化.结果表明,对照组细胞DNA/RNA比值为1.223 2±0.084 4,苏丹红Ⅰ、Ⅲ和Ⅳ处理后的HepG-2细胞DNA荧光染色强度明显高于RNA,以低剂量组的作用显著(p<0.01),其DNA/RNA比值分别为1.609 6±0.199 0,1.445 5±0.163 3,1.708 1±0.109 0.三维图表明,HepG-2细胞DNA荧光多聚集于细胞核区域,较RNA浓集、荧光强度大;可促进SGC-7901细胞由G1期进入S期和G2期,表现为G1期细胞平均DNA含量下降,S期DNA含量上升,G2期DNA含量上升,细胞增殖指数(PI)增加,S期细胞(SPF)指数增加.苏丹红Ⅰ、Ⅲ、Ⅳ可以通过影响细胞周期,从而调节细胞的增殖活动,促进细胞分化增殖.季宇彬 汲晨锋 高世勇 郎朗 于蕾 2006环境科学2006,27,6:20
2Gene expression profiles of hepatoma cell line HLE显示文摘AIM: To investigate the global gene expression of cancer related genes in hepatoma cell line HLE using Atlas Human Cancer Array membranes with 588 well-characterized human genes related with cancer and tumor biology.METHODS: Hybridization of cDNA blotting membrane was performed with 32P-labeled cDNA probes synthesized from RNA isolated from Human hepatoma cell line HLE and noncirrhotic normal liver which was liver transplantation donor.AtlasImage, a software specific to array, was used to analyze the result. The expression pattern of some genes identified by Atlas arrays hybridization was confirmed by reverse transcription polymerase chain reaction (RT-PCR)in 24 pairs of specimens and Northern blot of 4 pairs of specimens.RESULTS: The differential expression of cell cycle/growth regulator in hepatocellular carcinoma (HCC) showed a stronger tendency toward cell proliferation with more than 1.5-fold up-regulation of Cyclin C, ERK5, ERK6, E2F-3, TFDP2 and CK4. The anti-apoptotic factors such as Akt-1 were up-regulated, whereas the promotive genes of apoptosis such as ABL2 were down-regulated. Among oncogene/tumors suppressors, SKY was down-regulated. Some genes such as Integrin beta 8, Integrin beta 7, DNA-PK, CSPCP,byglycan, Tenacin and DNA Topo were up-regulated. A number of genes, including LAR, MEK1, eps15, TDGF1,ARHGDIA were down-regulated. In general, expression of the cancer progression genes was up-regulated, while expression of anti-cancer progression genes was downregulated. These differentially expressed genes tested with RT-PCR were in consistent with cDNA array findings.CONCLUSION: Investigation of these genes in HCC is helpful in disclosing molecular mechanism of pathogenesis and progression of HCC. For the first time few genes were discovered in HCC. 4激光共聚焦显微重建鲜红斑痣三维结构的初步研究显示文摘目的:探讨利用激光共聚焦显微重建鲜红斑痣三维结构可行性,初步建立利用激光共聚焦显微重建鲜红斑痣三维结构的方法。方法:鲜红斑痣石蜡标本切取30μm厚的切片,用鼠抗人anti-cd34、羊抗鼠FITC标记二抗免疫荧光染色,再用激光共聚焦显微镜观察,重建鲜红斑痣三维立体结构图像。结果:利用激光共聚焦显微完成了对10μm及30μm厚鲜红斑痣组织的三维结构重建,获得了鲜红斑痣三维结构的高清图形。结论:利用激光共聚焦显微镜重建鲜红斑痣三维结构组织图像是可行的,对进一步了解鲜红斑痣的结构及治疗有一定的意义。田克斌 韩丽林 江银华 周国瑜 2011温州医学院学报2011,41,5:2
5A study of the effect of Sudan I, III, and IV on the DNA/RNA ratio and 3D structure of HepG-2 using LCM显示文摘To explore the effect of sudan I, III, and IV on the DNA/RNA ratio and changes in the 3D structure of HepG-2. LCM and 3D images are used to detect the DNA/RNA ratio and changes in the 3D structure of HepG-2 when treated with different dosages of sudan I, III, and IV. The DNA/RNA ratio of the control group is 1.223 2±0.084 4, while the fluorescence intensity of DNA in HepG-2 treated with sudan I, III, or IV is markedly greater than that of RNA, with the low-dosage group showing significant effect (P<0.01), yielding DNA/RNA ratios of 1.609 6±0.199 0, 1.445 5±0.163 3, 1.708 1±0.109 0 respectively; 3D images show that DNA fluorescence in HepG-2 is mostly concentrated in the nuclear region, and is denser and stronger than RNA fluorescence. 6利用整体染色与透明技术观察棉花胚囊方法显示文摘目的:探索一种观察棉花胚囊的方法。方法:用整体染色与透明技术对棉花胚囊进行处理,利用激光扫描共聚焦显微技术进行观察。首先在解剖镜下将内外珠被剥掉,取出胚囊;然后水解过程需要反复实验得到合适的时间,否则荧光染料很难进入细胞;最后材料在激光扫描共聚焦显微镜上用488nm激光激发观察并摄像。结果:可以清晰地观察到棉花胚囊组织的各组成部分及其细胞结构。结论:利用激光扫描共聚焦显微镜的重组功能构建胚囊的立体结构,有助于了解胚囊各组成细胞的空间结构,为观察棉花及其他较大子房或胚珠提供了一种简单快捷的方法。李建欣 代西梅 张佳佳 张永山 2014实验技术与管理2014,31,11:0
