| 1 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 3 | Hepatitis B virus reactivation in patients with hepatocellular carcinoma undergoing anti-cancer therapy显示文摘Patients with hepatocellular carcinoma(HCC)often experience hepatic morbidity.Hepatitis B virus(HBV)reactivation is well documented as a serious hepatic morbidity during anti-cancer therapy.Reported rates of HBV reactivation in chronic carriers with HCC undergoing chemotherapy range from 4%-67%.Apart from chemotherapy,HBV reactivation has been increasingly identified in settings of hepatectomy and local ablation therapies.The rates of HBV reactivation vary with different levels of immunosuppression and depend on treatment,viral factors,and patient characteristics.The principal concern relating to reactivation is that a substantial proportion of patients with reactivation suffer from liver dysfunction during therapy,which often leads to disruption of planned,potentially life-prolonging treatments,adversely affecting the patients’final outcome.The first step in the management of HBV reactivation is identification of patients at risk of reactivation by testing for HBV serology prior to commencing anticancer therapy.Although it is a serious complication,HBV reactivation is preventable with prophylactic antiHBV drugs.Multiple publications have shown the benefit of prophylactic or preemptive antiviral therapy in this setting and justified such an approach before the start of therapy.Given the tumors and underlying cirrhosis,long-term use of antivirals with high potency and low risk of resistance is recommended in patients with HCC.This topic review will summarize the epidemiology,pathogenesis,and clinical issues related to HBV reactivation in HCC patients,and will discuss proper management against HBV reactivation during anti-cancer therapy for HCC. | Jeong Won Jang | 2014 | World Journal of Gastroenterology2014,20,24: | 18 |

| 6 | Effect of activation agents on the surface chemical properties and desulphurization performance of activated carbon显示文摘Flue gas pollution is a serious environmental problem that needs to be solved for the sustainable development of China.The surface chemical properties of carbon have great influence on its desulphurization performance.A series of activated carbons (ACs) were prepared using HNO3,H2O2,NH3·H2O and steam as activation agents with the aim to introduce functional groups to carbon surface in the ACs preparation process.The ACs were physically and chemically characterized by iodine and SO2 adsorption,ultimate analysis,Boehm titration,and temperature-programmed reduction (TPR).Results showed that the iodine number and desulphurization capacity of NH3·H2O activated carbon (AC-NH3) increase with both activation time,and its desulphurization capacity also increases with the concentration of activation agent.However,HNO3 activated carbon (AC-HNO3) and H2O2 activated carbon (AC-H2O2) exhibit more complex behavior.Only their iodine numbers increase monotonously with activation time.Compared with steam activated AC (AC-H2O),the nitrogen content increases 0.232% in AC-NH3 and 0.077% in AC-HNO3.The amount of total basic site on AC-HNO3 is 0.19 mmol·g-1 higher than that on AC-H2O.H2O2 activation introduces an additional 0.08 mmol·g-1 carboxyl groups to AC surface than that introduced by steam activation.The desulphurization capacity of ACs in simulate flue gas desulphurization decreases as follows: AC-NH3 > AC-HNO3 > AC-H2O2 > AC-H2O.This sequence is in accord with the SO2 catalytic oxidation/oxidation ratio in the absence of oxygen and the oxidation property reflected by TPR.In the presence of oxygen,all adsorbed SO2 on ACs can be oxidized into SO3.The desulphurization capacity increases differently according to the activation agents;the desulphurization capacity of AC-NH3 and AC-HNO3 improves by 4.8 times,yet AC-H2O increases only by 2.62 as compared with the desulphurization of corresponding ACs in absence of oxygen. | ZHANG XiangLan,ZHANG Yan,WANG ShuangSheng,ZHANG JingJing & ZHOU Wei School of Chemical and Environmental Engineering,China University of Mining and Technology (Beijing),Beijing 100083,China | 2010 | Science China(Technological Sciences)2010,53,9: | 14 |
| 7 | A methylated-DNA-binding complex required for plant development mediates transcriptional activation of promoter methylated genes显示文摘Although the mechanism of DNA methylationmediated gene silencing is extensively studied, relatively little is known about how promoter methylated genes are protected from transcriptional silencing. SUVH1, an Arabidopsis Su(var)3-9 homolog, was previously shown to be required for the expression of a few promoter methylated genes. By chromatin immunoprecipitation combined with sequencing, we demonstrate that SUVH1 binds to methylated genomic loci targeted by RNA-directed DNA methylation. SUVH1 and its homolog SUVH3 function partially redundantly and interact with three DNAJ domain-containing homologs, SDJ1, SDJ2, and SDJ3, thus forming a complex which we named SUVH-SDJ. The SUVH-SDJ complex components are co-localized in a large number of methylated promoters and are required for the expression of a subset of promoter methylated genes. We demonstrate that the SUVHSDJ complex components have transcriptional activation activity. SUVH1 and SUVH3 function synergistically with SDJ1,SDJ2, and SDJ3 and are required for plant viability. | 11 | Kinetics of indium dissolution from marmatite with high indium content in pressure acid leaching显示文摘The kinetics of oxygen pressure acid leaching marmatite with high indium content was studied. The effects of particle size, agitation rate, temperature, H_2SO_4 concentration, and oxygen partial pressure on leaching rate of indium were investigated. The results show that when the agitation rate is above 600 r-min^(-1), its influence on indium leaching rate is insignificant. It is determined that the leaching rates increase with the increase in sulfuric acid concentration, temperature, partial oxygen pressure, and the decrease in particle size. Moreover, the results demonstrate that the process of indium leaching is controlled by interface chemical reaction, with apparent activation energy of 65.7 k J-mol^(-1). The apparent reaction orders of sulfuric acid and oxygen partial pressure are determined to be 0.749 and 1.260, respectively. The leaching reaction process follows shrinking unreacted core model. | 12 | Plant immune signaling: Advancing on two frontiers显示文摘Plants have evolved multiple defense strategies to cope with pathogens, among which plant immune signaling that relies on cell-surface localized and intracellular receptors takes fundamental roles. Exciting breakthroughs were made recently on the signaling mechanisms of pattern recognition receptors(PRRs) and intracellular nucleotide-binding site(NBS) and leucine-rich repeat(LRR)domain receptors(NLRs). This review summarizes the current view of PRRs activation, emphasizing the most recent discoveries about PRRs’ dynamic regulation and signaling mechanisms directly leading to downstream molecular events including mitogen-activated protein kinase(MAPK) activation and calcium(Ca2+) burst. Plants also have evolved intracellular NLRs to perceive the presence of specific pathogen effectors and trigger more robust immune responses. | 15 | Treg/Th17 cell balance and phytohaemagglutinin activation of T lymphocytes in peripheral blood of systemic sclerosis patients显示文摘AIM To investigate T-cell activation, the percentage of peripheral T regulatory cells(Tregs), Th17 cells and the circulating cytokine profile in systemic sclerosis(SSc).METHODS We enrolled a total of 24 SSc patients and 16 healthy controls in the study and divided the patients as having diffuse cutaneous SSc(dc SSc, n = 13) or limited cutaneous SSc(lc SSc, n = 11). We performed a further subdivision of the patients regarding the stage of the disease-early, intermediate or late. Peripheral venous blood samples were collected from all subjects. We performed flow cytometric analysis of the activationcapacity of T-lymphocytes upon stimulation with PHA-M and of the percentage of peripheral Tregs and Th17 cells in both patients and healthy controls. We used ELISA to quantitate serum levels of human interleukin(IL)-6, IL-10, tissue growth factor-β1(TGF-β1), and IL-17 A.RESULTS We identified a decreased percentage of CD3+CD69+ cells in PHA-stimulated samples from SSc patients in comparison with healthy controls(13.35% ± 2.90% vs 37.03% ± 2.33%, P < 0.001). However, we did not establish a correlation between the down-regulated CD3+CD69+ cells and the clinical subset, nor regarding the stage of the disease. The activated CD4+CD25+ peripheral lymphocytes were represented in decreased percentage in patients when compared to controls(6.30% ± 0.68% vs 9.36% ± 1.08%, P = 0.016). Regarding the forms of the disease, dc SSc patients demonstrated lower frequency of CD4+CD25+ T cells against healthy subjects(5.95% ± 0.89% vs 9.36% ± 1.08%, P = 0.025). With regard to Th17 cells, our patients demonstrated increased percentage in comparison with controls(18.13% ± 1.55% vs 13.73% ± 1.21%, P = 0.031). We detected up-regulated Th17 cells within the lc SSc subset against controls(20.46% ± 2.41% vs 13.73% ± 1.21%, P = 0.025), nevertheless no difference was found between dc SSc and lc SSc patients. Flow cytometric analysis revealed an increased percentage of CD4+CD25-Foxp3+ in dc SSc patients compared to controls(10.94% ± 1.65% vs 6.88% ± 0.91, P = 0.032). Regarding the peripheral cytokine profile, we detected raised levels of IL-6 [2.10(1.05-4.60) pg/m L vs 0.00 pg/m L, P < 0.001], TGF-β1(19.94 ± 3.35 ng/m L vs 10.03 ± 2.25 ng/m L, P = 0.02), IL-10(2.83 ± 0.44 pg/m L vs 0.68 ± 0.51 pg/m L, P = 0.008), and IL-17 A [6.30(2.50-15.60) pg/m L vs 0(0.00-0.05) pg/m L, P < 0.001] in patients when compared to healthy controls. Furthermore, we found increased circulating IL-10, TGF-β, IL-6 and IL-17 A in the lc SSc subset vs control subjects, as it follows: IL-10(3.32 ± 0.59 pg/m L vs 0.68 ± 0.51 pg/m L, P = 0.003), TGF-β1(22.82 ± 4.99 ng/m L vs 10.03 ± 2.25 ng/m L, P = 0.031), IL-6 [2.08(1.51-4.69) pg/m L vs 0.00 pg/m L, P < 0.001], and IL-17 A [14.50(8.55-41.65) pg/m L vs 0.00(0.00-0.05) pg/m L, P < 0.001]. Furthermore, circulating IL-17 A was higher in lc SSc as opposed to dc SSc subset(31.99 ± 13.29 pg/m L vs 7.14 ± 3.01 pg/m L, P = 0.008). Within the dc SSc subset, raised levels of IL-17 A and IL-6 were detected vs healthy controls: IL-17 A [2.60(0.45-9.80) pg/m L vs 0.00(0.00-0.05) pg/m L, P < 0.001], IL-6 [2.80(1.03-7.23) pg/m L vs 0.00 pg/m L, P < 0.001]. | 17 | Molecular mechanisms of triggering,amplifying and targeting RANK signaling in osteoclasts显示文摘Osteoclast differentiation depends on receptor activator of nuclear factor-κB(RANK) signaling,which can be divided into triggering,amplifying and targeting phases based on how active the master regulator nuclear factor of activated T-cells cytoplasmic 1(NFATc1) is. The triggering phase is characterized by immediateearly RANK signaling induced by RANK ligand(RANKL) stimulation mediated by three adaptor proteins,tumor necrosis factor receptor-associated factor 6,Grb-2-associated binder-2 and phospholipase C(PLC)γ2,leading to activation of IκB kinase,mitogen-activated protein kinases and the transcription factors nuclear factor(NF)-κB and activator protein-1(AP-1). Mice lacking NF-κB p50/p52 or the AP-1 subunit c-Fos(encoded by Fos) exhibit severe osteopetrosis due to a differentiation block in the osteoclast lineage. The amplification phase occurs about 24 h later in a RANKLinduced osteoclastogenic culture when Ca2+ oscillation starts and the transcription factor NFATc1 is abundantly produced. Osteoclast precursors lacking PLCγ2,inositol-1,4,5-trisphosphate receptors,regulator of G-protein signaling 10,or NFATc1 show an impaired transition from the triggering to amplifying phases. The final targeting phase is mediated by activation of numerous NFATc1 target genes responsible for cell-cell fusion and regulation of bone-resorptive function. In this study, we aimed to evaluate whether fluoxetine attenuates early brain injury(EBI) after SAH. We demonstrated that intraperitoneal injection of fluoxetine(10 mg/kg per day) significantly attenuated brain edema and blood-brain barrier(BBB) disruption, microglial activation, and neuronal apoptosis in EBI after experimental SAH, as evidenced by the reduction of brain water content and Evans blue dye extravasation, prevention of disruption of the tight junction proteins zonula occludens-1, claudin-5, and occludin, a decrease of cells staining positive for Iba-1, ED-1, and TUNEL and a decline in IL-1 b, IL-6, TNF-a, MDA, 3-nitrotyrosine, and 8-OHDG levels. Moreover, fluoxetine significantly improved the neurological deficits of EBI and long-term sensorimotor behavioral deficits following SAH in a rat model.
