| 1 | Aryl hydrocarbon receptor expression in serum,peripheral blood mononuclear cells,and skin lesions of patients with atopic dermatitis and its correlation with disease severity显示文摘Background:The aryl hydrocarbon receptor(AhR)is a ligand-activated transcription factor,which is critically involved in the pathogenesis of a variety of skin diseases.The aim of this study was to detect AhR and its downstream regulators including cytochrome P450(CYP1A1),AhR nuclear translocation(ARNT),and aryl hydrocarbon receptor repressor(AhRR)in serum,peripheral blood mononuclear cells(PBMCs),and skin lesions in patients with atopic dermatitis(AD).Methods:Twenty-nine AD patients defined according to the criteria of Hanifin and Rajka and Chinese criteria of AD were included.Subjects without allergic and chronic diseases were recruited as controls.Patients and controls were selected from the dermatology outpatient clinic of Peking University People’s Hospital from August 1 to December 31 in 2018.Enzyme-linked immunosorbent assay was performed to detect serum AhR level.ThemRNA of AhR,AhRR,ARNT,and CYP1A1 in PBMCs were measured by realtime quantitative polymerase chain reaction.AhR expression in skin lesions was measured by immunohistochemistry.Results:AhR was significantly higher expressed in serum(41.26±4.52 vs.33.73±2.49 pmol/L,t=6.507,P<0.001)and skin lesions(0.191±0.041 vs.0.087±0.017,t=10.036,P<0.001)of AD patients compared with those of controls.The mRNA levels of AhR(1.572±0.392 vs.1.000±0.173,t=6.819,P<0.001),AhRR(2.402±1.716 vs.1.000±0.788,t=3.722,P<0.001),CYP1A1(2.258±1.598 vs.1.000±0.796,t=3.400,P=0.002)in PBMCs of AD patients were higher compared with those of controls.The difference in mRNA levels of ARNT was not statistically significant between the patients and controls(1.383±0.842 vs.1.000±0.586,t=1.653,P=0.105).AhR mRNA levels in PBMCs positively correlated with eczema area and severity index score and serum interleukin-6 levels.Conclusion:AhR and its downstream regulators were highly expressed in serum,PBMCs,and skin of AD patients,which might contribute to the pathogenesis of AD. | Yu-Qing Hu Ping Liu Zhang-Lei Mu Jian-Zhong Zhang | 2020 | Chinese Medical Journal2020,,2: | 6 |

| 6 | 2,3,7,8-Tetrachlorodibenzo-p-dioxin promotes migration ability of primary cultured rat astrocytes via aryl hydrocarbon receptor显示文摘Emerging evidence showed that 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD) could induce expression of certain reactivation-associated genes in astrocytes, however, the consequent cellular effects and molecular mechanisms are still unclear. During the process of astrocyte reactivation, migration is a critical cellular event. In the present study, we employed woundhealing assay and Transwell? motility assay to explore the effects of TCDD on cell migration in primary cultured rat cortical astrocytes. We found that upon TCDD treatments at relative low concentrations(10^(-10) and/or 10^(-9) mol/L), the ability of primary astrocytes to migrate horizontally and vertically was promoted. In line with this cellular effect, the mR NA expression of two promigratory genes, including cell division cycle 42(CDC42) and matrix metalloproteinase 2(MMP2)was induced by TCDD treatment. Dioxin exerts its toxic effects mainly through aryl hydrocarbon receptor(AhR) pathway. So the role of AhR pathway in the pro-migratory effects of TCDD was examined using an AhR antagonist, CH223191. We found that application of CH223191 significantly reversed the pro-migratory effects of TCDD. Interestingly, the basal ability of horizontal migration as well as basal levels of CDC42 and MMP2 expression were dramatically reduced suggesting a possible physiological role of AhR in maintaining the endogenous migration ability of the primary astrocytes. | 10 | Intestinal inflammation and the diet:Is food friend or foe?显示文摘Inflammatory bowel disease(IBD) is a chronic intestinal illness of autoimmune origin affecting millions across the globe. The most common subtypes include ulcerative colitis(UC) and Crohn's disease. While many medical treatments for IBD exist,none come without the risk of significant immunosuppression and in general do not have benign side effect profiles. Surgical intervention exists only as radical resection for medically refractory UC. There exists a dire need for novel treatments that target the inherent pathophysiologic disturbances of IBD,rather than global immune suppression. One avenue of investigation that could provide such an agent is the interaction between certain dietary elements and the aryl hydrocarbon receptor(AHR). The AHR is a cytosolic transcription factor with a rich history in environmental toxicant handling,however,recently a role has emerged for the AHR as a modulator of the gastrointestinal immune system. Studies have come to elucidate these effects to include the enhancement of Th cell subset differentiation,interactions between enteric flora and the luminal wall,and modulation of inflammatory interleukin and cytokine signaling. This review highlights advancements in our understanding of AHR activity in the digestive tract and how this stimulation may be wrought by certain dietary 'micronutriceuticals',namely indole-3-carbinol(I3C) and its derivatives. | 11 | Synthesis and Characterization of Aryl Acylguanidines and Their Copper(Ⅱ) Complexes显示文摘A series of eight N-mono-substituted aryl acylguanidines L1~8 were synthesized from the reactions of their parent acylthioureas and respective primary amines, including five R1C(O)N=C(NHR2)2 with balanced arms(denoted as R1–R2–R2: Ph–Ar’–Ar’, L1(1);Ph–Ar’’–Ar’’, L2(2);1-Naph–Ar’–Ar’, L3(3);1-Naph–Ar’’–Ar’’, L4(4);2-Naph–Ar’’–Ar’’, L[5(5);Ar’ = 2,6-Me2C6H3, Ar’’ = 2,6-iPr2C6H3) and three R1C(O)N=C(NHR2)NHR3 with unbalanced arms(denoted as R1–R2–R3: Ph–Ar’–Ph, L6(6);Ph–Ar’ –Ph, L7(7);Ph–Ar’ –Ar’, L8(8)). Treatment of L1~5(1~5) with copper acetate in a molar ratio of 2:1 in dichloromethane led to the formation of homoleptic Cu(Ⅱ) complexes trans-L2n’Cu(9~13), in which the Cu(Ⅱ) center was stabilized by deprotonated ligands Ln’(n = 1~5) in the O,N-bidentate mode. When similar reactions of L6(Ph–Ar’–Ph)(6) and L7(Ph–Ar’’–Ph)(7) that contain unbalanced arms were investigated, the Cu(Ⅱ) products trans-L26’Cu(14) and trans-L27’Cu(15) were formed by deprotonating the bulky amine groups in Ln(-NHAr’ for L6 and-NHAr’’ for L7) rather than the-NHPh group. In sharp contrast, trans-L28’Cu(16) was obtained from two-arm unbalanced L8(Ph–Ar’’–Ar’)(8) not by deprotonating the more bulky-NHAr’’ group but the relatively less bulky-NHAr’ group. In a further exploration, the reaction of 1:1 mixed ligands Ln and Lm with copper acetate was found to give only homoleptic Cu(Ⅱ) complex L2n’Cu instead of the heteroleptic one Ln’CuLm’.
