1Molecular cloning and functional characterization of a DREB1/CBF-like gene (GhDREB1L) from cotton显示文摘The transcription factors DREB1s/CBFs play important roles in the regulation of plant resistance to environmental stresses and are quite useful for generating transgenic plants tolerant to these stresses. In the present work, a cDNA encoding DREB1/CBF-like protein (GhDREB1L) from cotton was isolated, and its sequence features, DNA binding preference, and expression patterns of the transcripts were also characterized. GhDREB1L contained one conserved AP2/ERF domain and its amino acid sequence was similar to the DREB1/CBF group of the DREB family from other plants. The DNA-binding domain of GhDREB1L was successfully expressed as a fusion protein in Escherichia coli BL21 (DE3) and purified by Ni-NTA affinity chromatography. Electrophoretic mobility shift assay revealed that the purified GhDREB1L fusion protein had a specific binding activity with the previously characterized DRE ele-ment (core sequence, ACCGAC) and also with the DRE-like sequence (core sequence, GCCGAC) in the promoter of the dehydration-responsive late embryogenesis-abundant gene LEA D113. Semi-quantita- tive RT-PCR showed that GhDREB1L was induced in the cotton cotyledons by low temperature, as well as drought and NaCl treatments. 2Molecular mapping of Verticillium wilt resistance QTL clustered on chromosomes D7 and D9 in upland cotton显示文摘Verticillium wilt is a destructive disease with international consequences for cotton production. Breeding broad-spectrum resistant cultivars is considered to be one of the most effective means for reducing crop losses. A resistant cotton cultivar,60182,was crossed with a susceptible cultivar,Jun-mian 1,to identify markers for Verticillium resistance genes and validate the mode of its inheritance. Genetic segregation analysis for Verticillium wilt resistance was evaluated based upon infected leaf percentage in the seedling stage using major gene-polygene mixed inheritance models and joint analysis of P1,P2,F1,B1,B2 and F2 populations obtained from the cultivar cross. We found that resistance of upland cotton cultivar 60182 to isolates BP2,VD8 and T9,and their isoconcentration mixture was controlled by two major genes with additive-dominance-epistatic effects,and the inheritance of the major gene was dominant. Furthermore,a genetic linkage map was constructed using F2 segregating population and resistance phenotypic data were obtained using F2:3 families inoculated with different isolates and detected in different developmental stages. The genetic linkage map with 139 loci was comprised of 31 linkage groups covering 1165 cM,with an average distance of 8.38 cM between two markers,or 25.89% of the cotton genome length. From 60182,we found 4 QTL on chromosome D7 and 4 QTL on D9 for BP2,5 QTL on D7 and 9 QTL on D9 for VD8,4 QTL on D7 and 5 QTL on D9 for T9 and 3 QTL on D7 and 7 QTL on D7 for mixed pathogens. The QTL mapping results revealed that QTL clusters with high contribution rates were screened simultaneously on chromosomes D9 and D7 by multiple interval mapping (CIM),whether from resistance phenotypic data from different developmental stages or for different isolates. The result is consistent with the genetic model of two major genes in 60182 and suggests broad-spectrum resistance to both defoliating isolates of V. dahliae and nondefoliating isolates. 3Effects of planting patterns on yield, quality, and defoliation in machine-harvested cotton显示文摘The aim of this study was to elucidate the effects of different machine-harvested cotton-planting patterns on defoliation,yield,and fiber quality in cotton and to provide support for improving the quality of machine-harvested cotton.In the 2015 and 2016 growing seasons,the Xinluzao 45(XLZ45)and Xinluzao 62(XLZ62)cultivars,which are primarily cultivated in northern Xinjiang,were used as study materials.Conventional wide-narrow row(WNR),wide and ultra-narrow row(UNR),wide-row spacing with high density(HWR),and wide-row spacing with low density(LWR)planting patterns were used to assess the effects of planting patterns on defoliation,yield,and fiber quality.Compared with WNR,the seed cotton yields were significantly decreased by 2.06–5.48%for UNR and by 2.50–6.99%for LWR,respectively.The main cause of reduced yield was a reduction in bolls per unit area.The variation in HWR yield was–1.07–1.07%with reduced bolls per unit area and increased boll weight,thus demonstrating stable production.In terms of fiber quality indicators,the planting patterns only showed significant effects on the micronaire value,with wide-row spacing patterns showing an increase in the micronaire values.The defoliation and boll-opening results showed that the number of leaves and dried leaves in HWR was the lowest among the four planting patterns.Prior to the application of defoliating agent and before machine-harvesting,the numbers of leaves per individual plant in HWR were decreased by 14.45 and 25.00%on average,respectively,compared with WNR,while the number of leaves per unit area was decreased by 27.44 and 36.21%on average,respectively.The rates of boll-opening and defoliation in HWR were the highest.Specifically,the boll-opening rate before defoliation and machine-harvesting in HWR was 44.54 and 5.94%higher on average than in WNR,while the defoliation rate prior to machine-harvesting was 3.45%higher on average than in WNR.The numbers of ineffective defoliated leaves and leaf trash in HWR were the lowest,decreased by 33.40 and 32.43%,respectively,compared with WNR.In conclusion,the HWR planting pattern is associated with a high and stable yield,does not affect fiber quality,promotes early maturation,and can effectively decrease the amount of leaf trash in machine-picked seed cotton,and thus its use is able to improve the quality of machine-harvested cotton.WANG Fang-yong HAN Huan-yong LIN Hai CHEN Bing KONG Xian-hui NING Xin-zhu WANG Xu-wen YU Yu LIU Jing-de 2019Journal of Integrative Agriculture2019,18,9:14
4Identification of RAPD marker linked with fertility-restoring gene of cytoplasmic male sterile lines in upland cotton显示文摘Bulked segregant analysis was employed to construct two mixed DNA pools to screen the RAPd marker linked with the fertilityrestoring gene(\%Rf\-i)\% of upland cotton. A total of 425 arbitrary 10mer oligonucleotide primers were screened on two DNA pools, bulked male fertile and sterile DNAs isolated from BC\-3 segregating population of (06132R×Simian No. 3). Three primers produced repeatable polymorphisms between the paired bulks and their parents. DNA was extracted and amplified with these three primers for 92 plants of (Zhong 12A1×06132R)F\-2. 6Effects of soil moisture on cotton root length density and yield under drip irrigation with plastic mulch in Aksu Oasis farmland显示文摘Effects of soil moisture on cotton root length density (total root length per unit soil volume) and yield under drip irrigation with plastic mulch were studied through field experiments. The results indicate that spatial distributions of root length density of cotton under various water treatments were basically similar. Horizontally, both root length densities of cotton in wide and narrow rows were similar, and higher than that between mulches. Vertically, root length density of cotton decreased with increasing soil depth. The distribution of root length density is different under different irrigation treatments. In conditions of over-irrigation, the root length density of cotton between mulches would increase. However, it would decrease in both the wide rows and narrow rows. The mean root length density of cotton increased with increasing irrigation water. Water stress caused the root length density to increase in lower soil layers. There is a significant correlation between root length density and yields of cotton at the flower-boll and wadding stages. The regression between irrigation amount and yield of cotton can be expressed as y = -0.0026x2+18.015x-24845 (R2 = 0.959). It showed that the irrigation volume of 3,464.4 m3/hm2 led to op-timal root length density. 7A decade of Bt cotton in Chinese fields: Assessing the direct effects and indirect externalities of Bt cotton adoption in China显示文摘The objective of this study is to examine whether or not the gains from reduced spraying for bollworms are being sustained more than one decade after the initial adoption in 2007. Based on farm-level data collected by the authors in 1999–2007 in 16 villages from 4 provinces, this study shows that insecticides applied for controlling bollworms have declined. This analysis supports Chinese policy makers’ decision to not require refuges of non-Bt cotton fields. 8Invasion biology of the cotton mealybug, Phenacoccus solenopsis Tinsley: Current knowledge and future directions显示文摘The cotton mealybug Phenacoccus solenopsis Tinsley(Hemiptera: Pseudococcidae), which is native to North America, emerged as a major invasive insect pest of multiple crops in Asia at the beginning of the 21 st century. Considering the economic significance of this insect and its rapid worldwide spread, we examined the possible factors driving its invasions. In this paper, we summarize the life history traits of P. solenopsis conceivably related to population development in invaded regions; these traits include its use of diverse host plants, reproductive capacity and mode, adaptation to temperature, response to food shortage, and insecticidal resistance. Then, focusing on the multiple trophic interactions that may promote or hinder invasion, we review the mutualistic relationship of this mealybug with ants and predation and parasitism by natural enemies. Finally, we suggest topics for future research and provide our perspective on the biological invasions of this mealybug. 9Cotton bollworm resistance to Bt transgenic cotton: A case analysis显示文摘Cotton bollworm (Helicoverpa armigera) is one of the most serious insect pests of cotton. Transgenic cotton expressing Cry toxins derived from a soil bacterium, Bacillus thuringiensis (Bt), has been produced to target this pest. Bt cotton has been widely planted around the world, and this has resulted in efficient control of bollworm populations with reduced use of synthetic insecticides. However, evolution of resistance by this pest threatens the continued success of Bt cotton. To date, no field populations of bollworm have evolved significant levels of resistance; however, several laboratory-selected Cry-resistant strains of H. armigera have been obtained, which suggests that bollworm has the capacity to evolve resistance to Bt. The development of resistance to Bt is of great concern, and there is a vast body of research in this area aimed at ensuring the continued success of Bt cotton. Here, we review studies on the evolution of Bt resistance in H. armigera, focusing on the biochemical and molecular basis of Bt resistance. 11Optimization and test for spraying parameters of cotton defoliant sprayer显示文摘Boom sprayer is widely used in large farm crops because of its high working efficiency and favorable spraying effect.But there are still some problems in cotton defoliant spraying in Xinjiang,China.Cotton is planted in a high density in Xinjiang,the row space is(10+66)cm,leaves in two adjacent rows are seriously overlapped,the lower leavers are poorly sprayed,so the defoliation effect is poor,and the cotton quality is degraded.To solve this problem and improve the defoliant droplets coverage on the cotton canopy,the original boom spraying was modified,and the spraying pardameters was optimized by the central combination test and design concept of Box-Behnken based on a single-factor test.A quadratic polynomial model of droplets coverage was created by using working parameters including horizontal spraying boom height,hang boom height and nozzles angle as the influential factors and the mean droplets coverage on cotton canopy as the target function,and the effectiveness of mode and interaction of factors were analyzed.The model was optimized and analyzed using the regression analysis method and response surface analysis method of software Design-Expert 7.0.0,and the optimal combination of spraying parameters was obtained.The results showed that the droplets coverage on cotton canopy were influenced by boom height,sprayer height and angled nozzles sequentially from large to small,and the optimal combination of spraying parameters was under horizontal spraying boom height of 134 cm,hang spraying boom height of 27.5 cm and nozzles angle of 21°.The mean droplets coverage of experimental value and predicted value on cotton canopy were 19.6%and 20.43%respectively in such conditions,and the relative error to the estimated value on the model was–4.25%.The research result can provide a reference for further optimizing the spraying parameters of cotton defoliant sprayer.Qin Weicai Xue Xinyu Cui Longfei Zhou Qingqing Xu Zhufeng Chang Feilong 2016International Journal of Agricultural and Biological Engineering2016,9,4:9
12Biological mechanism of controlling cotton aphid (Homoptera: aphididae) by the marginal alfalfa zone surrounding cotton field显示文摘At the beginning stage of cotton aphids (Aphis gossypii Glover) invading the cotton field, large numbers of insects have accumulated in the marginal alfalfa (Medicago sativa L.) zone surrounding it. They are 6.94 times (excluded natural enemies) as many as the aphids in cotton field. Most of these insects are Therioaphid maculata (Buckton), and occur in fields 10-15 d earlier than cotton aphids. Consequently, abundant natural enemies have bred in alfalfa zone when cotton aphids increase rapidly in cotton fields in the middle of June in Xinhe County, Xinjiang Autonomous Region. The total number of the major natural enemies, including Coccinellids, Chrysopids and Syphid flies in alfalfa zone, is 13.65 times that in cotton field within the same acreage of fields. 13Automatic image segmentation method for cotton leaves with disease under natural environment显示文摘In order to improve the image segmentation performance of cotton leaves in natural environment, an automatic segmentation model of diseased leaf with active gradient and local information is proposed. Firstly, a segmented monotone decreasing edge composite function is proposed to accelerate the evolution of the level set curve in the gradient smooth region. Secondly, canny edge detection operator gradient is introduced into the model as the global information. In the process of the evolution of the level set function, the guidance information of the energy function is used to guide the curve evolution according to the local information of the image, and the smooth contour curve is obtained. And the main direction of the evolution of the level set curve is controlled according to the global gradient information, which effectively overcomes the local minima in the process of the evolution of the level set function. Finally, the Heaviside function is introduced into the energy function to smooth the contours of the motion and to increase the penalty function Φ(x) to calibrate the deviation of the level set function so that the level set is smooth and closed. The results showed that the model of cotton leaf edge profile curve could be obtained in the model of cotton leaf covered by bare soil, straw mulching and plastic film mulching, and the ideal edge of the ROI could be realized when the light was not uniform. In the complex background, the model can segment the leaves of the cotton with uneven illumination, shadow and weed background, and it is better to realize the ideal extraction of the edge of the blade. Compared with the Geodesic Active Contour(GAC) algorithm, Chan-Vese(C-V) algorithm and Local Binary Fitting(LBF) algorithm, it is found that the model has the advantages of segmentation accuracy and running time when processing seven kinds of cotton disease leaves images, including uneven lighting, leaf disease spot blur, adhesive diseased leaf, shadow, complex background, unclear diseased leaf edges, and staggered condition. 14Goosegrass (Eleusine indica) density effects on cotton (Gossypium hirsutum)显示文摘Goosegrass is one of the worst agricultural weeds on a worldwide basis. Understanding of its interference impact in crop field will provide useful information for weed control programs. Field experiments were conducted during 2010–2012 to determine the influence of goosegrass density on cotton growth at the weed densities of 0, 0.125, 0.25, 0.5, 1, 2, and 4 plants m–1 of row. Seed cotton yield tended to decrease with the increase in weed density, and goosegrass at a density of 4 plants m–1 of row significantly reduced cotton yields by 20 to 27%. A density of 11.6–19.2 goosegrass plant m–1 of row would result in a 50% cotton yield loss from the maximum yield according to the hyperbolic decay regression model. Boll production was not affected in the early growing season. But boll numbers per plant were reduced about 25% at the density of 4 plants m–1 of row in the late growing season. Both cotton boll weight and seed numbers per boll were significantly reduced(8%) at 4 goosegrass plants m–1 of row. Cotton plant height, stem diameter and sympodial branch number were not affected as much as cotton yields by goosegrass competition. Seed index, lint percentage and lint fiber properties were unaffected by weed competition. Intraspecific competition resulted in density-dependent effects on weed biomass per plant, 142–387 g dry weight by harvest. Goosegrass biomass m–2 tended to increase with increasing weed density as indicated by a quadratic response. 15Leaf surface factors of transgenic Bt cotton associated with the feeding behaviors of cotton aphids:A case study on non-target effects显示文摘The present paper reports case study results of the risk assessment of transgenic Bt cotton on a non-target pest, cotton aphid, Aphis gossypii. Several types of techniques, i.e., electrical penetration graph (EPG), light and electron microscopy, bioassays and chemical analysis, were applied to investigate physical and chemical leaf factors of 2 transgenic Bt cotton lines (GK12 and GK19) and their pa-rental non-Bt cotton line (Simian3) associated with searching and feeding behaviors of cotton aphids on leaves or leaf extracts of cotton plants. EPG results showed that there were some differences among behaviors of cotton aphids on 2 Bt cotton and 1 non-Bt cotton lines. Cotton aphids performed similarly to leaf surface extracts from 3 cotton lines; and leaf surface chemicals, mainly volatiles and waxes, were almost identical in the components and concentrations among the cotton lines. However, three cotton lines were quite different from each other in the densities of certain kinds of covering trichomes. Therefore, the relationships between the physical characteristics and the searching behaviors of cotton aphids on the three cotton lines were constructed as the regression equations. Glandular trichomes and covering trichomes with 5 branches influenced the cotton aphids' searching behaviors effectively; and other trichomes with other branches affected aphids in varying ways. These results demonstrated that leaf surface physical factors of transgenic Bt cotton lines different from their parental non-Bt line could affect the penetration behaviors of non-target cotton aphids. 16Influence of potassium fertilization and foliar application of zinc and phosphorus on growth,yield components,yield and fiber properties of Egyptian cotton(Gossypium barbadense L.)显示文摘Aims Supplying optimal quantities of mineral nutrients to growing crop plants is one way to improve crop yields.Nutrients need to be used rationally in order to avoid a negative ecological impact and undesirable effects on the sustainability of agricultural production systems.Excessive application of nutrients also affects the farmer’s economy.In order to calculate the amount of fertilizer to be applied to crops,it is necessary to develop recommendation programmers that adjust nutrient rates to crop requirements.Methods Experiments in two successive seasons were conducted to investigate the effect of K fertilization and foliar application of Zn and P on yield and fiber properties of cotton cv.Giza 86.Potassium(0.0 and 47.4 kg of K ha^(-1))was soil applied,while chelated zinc(0.0 and 57.6 g of Zn ha^(-1),applied twice at 70 and 85 days after sowing‘DAS’)and phosphorus(0.0,576,1152 and 1728 g of P ha^(-1),applied twice at 80 and 95 DAS)were applied to the foliage.Important findings Dry matter yield,total chlorophyll concentration,K,Zn and P uptake per plant,number of opened bolls per plant,boll weight,seed index,lint index,seed cotton yield per plant,seed cotton and lint yield ha^(-1) and earliness of harvest increased with the application of K,Zn and P.Treatments generally had no significant effect on lint percentage and fiber properties,with exceptions,for micronaire reading and flat bundle strength,and uniformity ratio,where the mean values of these characters were significantly increased over the untreated control by applying K,and for the micronaire reading in the first season,when applying P at 1728 g ha^(-1),and uniformity ratio in the second season,when applying Pat 1152 and 1728 g ha^(-1),where the mean values of these characters were significantly increased over the untreated control by applying P.Under the conditions of this study,applying K fertilization at 47.4 kg ha^(-1) combined with spraying cotton plants with zinc at 57.6 g ha^(-1) and also with P at 1728 g ha^(-1) improved growth and yield of Egyptian cotton.Zakaria M.Sawan Mahmoud H.Mahmoud Amal H.El-Guibali 2008Journal of Plant Ecology2008,1,4:6
17Inheritance of the delayed gland morphogenesis trait in Australian wild species of Gossypium显示文摘Five Australian wild cotton species with the delayed gland morphogenesis trait, as well as G. arboreum, G. davidsonii and four different gland genotypes of G. hirsutum, Gl2Gl2Gl3Gl3, Gl2Gl2gl3gl3, gl2gl2Gl3Gl3, and gl2gl2gl3gl3, were used in this experiment and 10 interspecific hybrids were obtained by the crossing among them. 19One-time fertilization at first flowering improves lint yield and dry matter partitioning in late planted short-season cotton显示文摘Cotton producers have substantially reduced their inputs(labor,nutrients,and management)mainly by adopting a shortseason cropping management that is characterized by late sowing,high density,and reduced fertilization with one-time application at the first bloom stage without lint yield reduction.However,it has been hypothesized that one-time fertilization at an earlier growth stage could be a more effective and economic management practice.A two-year field experiment was conducted by applying five fertilizer one-time fertilization at 0(FT1),5(FT2),10(FT3),15(FT4),and 20(FT5)days after the first flower appeared in the field and one three-split fertilizer application taken as the conventional control(FT6),making six treatments altogether.Cotton growth period,biomass accumulation,yield,and its formation were quantified.The results showed that the one-time fertilization did not affect the cotton growth progress as compared to FT6,however,the total crop cycles for FT3–FT5 were 3 days shorter.FT1 produced the highest cotton lint yield(1396 kg ha–1),which was similar to the FT6 but higher than the other treatments,and could be attributed to more bolls per unit area and higher lint percentage.Cotton yield was positively correlated with cotton plant biomass accumulated.FT1 had both the highest average(VT)(193.7 kg ha–1 d–1)and the highest maximum(VM)(220.9 kg ha–1 d–1)rates during the fast biomass accumulation period.These results suggest that one-time fertilizer application at the first flower stage might be an adjustment that is more effective than at first bloom,and allowed for easier decision making for application date due to non counting of plants with flowers is needed.LUO Hong-hai WANG Qiang ZHANG Jie-kun WANG Lei-shan LI Ya-bing YANG Guo-zheng 2020Journal of Integrative Agriculture2020,19,2:6
20Impact of agricultural residual plastic film on the growth and yield of drip-irrigated cotton in arid region of Xinjiang, China显示文摘Long-term and widespread use of plastic mulching has led to the accumulation of residual plastic film(RPF)in farmland soils of Xinjiang,China.However,there is no specific quantitative basis for RPF pollution or a clear understanding of the influence of residual film on crop growth.The aim of this study was to investigate the effect of RPF on the growth of cotton,an important cash crop of Xinjiang.Based on the field conditions and previous reports,various amount of residual film was applied in 0-30 cm soil layer.The growth index including emergence rate,dry matter,and yield of cotton was examined at different growth stages under different soil residual film levels.Results demonstrated a significant effect of RPF on soil moisture distribution and movement.Plastic residues had a significant effect on cotton growth at levels above 200 kg/hm2,and the yield decreased as the RPF amount increased.Based on these findings,200 kg/hm2 was suggested as a threshold level to determine the effects of RPF on cotton.This study provided a basis to rate RPF pollution in farmland soils and help understand the impact of pollution on crop productivity.Can Hu Xufeng Wang Shiguo Wang Bing Lu Wensong Guo Chaoji Liu Xiuying Tang 2020International Journal of Agricultural and Biological Engineering2020,13,1:5
