| 4 | Plant diversity reduces the effect of multiple heavy metal pollution on soil enzyme activities and microbial community structure显示文摘It is unclear whether certain plant species and plant diversity could reduce the impacts of multiple heavy metal pollution on soil microbial structure and soil enzyme activities. Random amplified polymorphic DNA (RAPD) was used to analyze the genetic diversity and microbial similarity in planted and unplanted soil under combined cadmium (Cd) and lead (Pb) pollution. A metal hyper- accumulator, Brassica juncea, and a common plant, Festuca arundinacea Schreb, were used in this research. The results showed that microorganism quantity in planted soil significantly increased, compared with that in unplanted soil with Cd and Pb pollution. The order of microbial community sensitivity in response to Cd and Pb stress was as follows: actinomycetes 〉 bacteria 〉 fungi. Respiration, phosphatase, urease and dehydrogenase activity were significantly inhibited due to Cd and Pb stress. Compared with unplanted soil, planted soils have frequently been reported to have higher rates of microbial activity due to the presence of additional surfaces for microbial colonization and organic compounds released by the plant roots. Two coexisting plants could increase microbe population and the activity of phosphatases, dehydrogenases and, in particular, ureases. Soil enzyme activity was higher in B. juncea phytoremediated soil than in F. arundinacea planted soil in this study. Heavy metal pollution decreased the richness of the soil microbial community, but plant diversity increased DNA sequence diversity and maintained DNA sequence diversity at highlevels. | 6 | Expression of liver cancer associated gene HCCA3显示文摘AIM: To study and clone a novel liver cancer reisted gene,and to explore the molecular basis of liver cancer genesis.METHODS: Using mRNA differential display polymerasechain reaction (DDPCR), we investigated the difference of mRNA in human hepatocellular carcinoma (HCC) and paired surrounding liver tissues, and got a gene probe. By screening a human placenta cDNA library and genomic homologous extend, we obtained a full-length cDNA named HCCA3. We analyzed the expression of this novel gene in 42pairs of HCC and the surrounding liver tissues, and distribution in human normal tissues by means of Northern blot assay.RESULTS: A full-length cDNA of liver cancer associated gene HCCA3 has been submitted to the GeneBank nucleotide sequence databases ( Accession No. AF276707 ). The positive expression rate of this gene was 78.6% (33/42) in HCC tissues, and the clinical pathological data showed that the HCCA3 was closely associated with the invasion of tumor capsule ( P = 0.023) and adjacant small metastasis satellite nodules lesions ( P= 0.041). The HCCA3 was widely distributed in the human normal tissues, which was intensively expressed in lungs, brain and colon tissues,while lowly expressed in the liver tissues.CONCLUSION: A novel full-length cDNA was cloned and differentiated, which was highly expressed in liver cancer tissues. | 9 | Differential expression of a novel colorectal cancer differentiation-related gene in colorectal cancer显示文摘AIM To investigate SBA2 expression in CRC cell lines snd surgical specimens of CRC and sutologous healthy mucosa.METHODS Reverse transcription-polymerase chain reaction (RT-PCR) was used for relative quantification of SBA2 mRNA levels in 4 human CRC cell lines with different grades of differentiation and 30 clinical samples.Normalization of the results was achieved by simultaneous amplification of β-actin as an internal control.RESULTS In the exponential range of amplification, fairly good linearity demonstrated identical amplification efficiency for SBA2 and β-actin (82%). Markedly lower levels of SBA2 mRNA were detectable in tumors, as compared with the coupled normal counterparts ( P < 0.01 ). SBA2 expression was significantly (0.01 < P <0.05) correlated with the grade of differentiation in CRC,with relatively higher levels in well-differentiated samples and lower in poorly-differentiated cases. | 10 | HBV DNA阳性的中晚期肝癌预后因素分析及抗病毒治疗对生存期的影响显示文摘目的探讨HBV DNA阳性的中晚期原发性肝癌(PHC)预后的影响因素及抗病毒治疗对患者生存的影响。方法选择HBV DNA阳性的93例中晚期肝癌患者临床指标和随访资料,采用Kaplan-Meier法及Cox比例风险回归模型对可能影响其预后的因素进行单变量和多变量回归分析,重点分析病毒载量及核苷(酸)类似物抗病毒治疗对肝癌预后的影响。结果除GGT、Child-Pugh分级、门脉癌栓、治疗方式、治疗次数外,抗病毒治疗也是影响患者预后的重要因素,而病毒载量对预后无统计学意义(P>0.05)。结论虽病毒载量并不影响HBV DNA阳性患者的生存期,但抗病毒治疗仍是影响生存的重要因素,对于HBV DNA阳性的中晚期肝癌应积极抗病毒治疗,以期改善患者的生存。 | 臧允红 丁明权 何登明 | 2011 | 临床肝胆病杂志2011,27,6: | 6 |

| 12 | Copper transportion of WD protein in hepatocytes from Wilson disease patients in vitro显示文摘AIM: To study the effect of copper transporting P-type ATPase in copper metabolism of hepatocyte and pathogenesis of Wilson disease (WD).METHODS: WD copper transporting properties in some organelles of the cultured hepatocytes were studied from WD patients and normal controls. These cultured hepatocytes were incubated in the media of copper 15mg.L-1 only, copper 15 mg. L-1 with vincristine (agonist of P-type ArPase) 0.5mg. L-1, or copper 15 mg. L-1 withvanadate (antagonist of P-type ATPase) 18.39 mg. L-1separately. Microsome (endoplasmic reticulum and Golgi apparatus), lysosome, mitochondria, and cytosol were isolated by differential centrifugation. Copper contents in these organelles were measured with atomic absorption spectrophotometer, and the influence in copper transportion of these organelles by vanadate and vincristine were comparatively analyzed between WD patients and controls.WD copper transporting P-type ATPase was detected by SDS-PAGE in conjunction with Western blot in liver samples of WD patients and controls.RESULTS: The specific WD proteins (Mr155 000 lanes) were expressed in human hepatocytes, including the control and WD patients. After incubation with medium containing copper for 2 h or 24 h, the microsome copper concentration in WD patients was obviously lower than that of controls,and the addtion of vanadate or vincristine would change the copper transporting of microsomes obviously. When incubated with vincristine, levels of copper in microsome were significantly increased, while incubated with vanadate,the copper concentrations in microsome were obviously decreased. The results indicated that there were Wdproteins, the copper transportion P-type ATPase in the microsome of hepatocytes. | 14 | Analysis of Mitochondrial DNA from the Ancient Tombs of Turfan显示文摘MtDNA was successfully extracted from ten individual bones (femurs) in the tombs of ancient Jushi in Turfan basin, dated back to the year about 3 000-2 500 years ago. By means of four overlapping primers, we got nucleotide sequence of the 218bp length. Ancient mtDNA was analyzed by the sequencing of hypervariable region Ⅰ of the mtDNA control region. The result shows that 9 haplotypes with 24 polymorphic sites were obtained. The phylogenetic analysis indicated that Mongolians and Altai are the population genetically closest to the Jushi groups and Jushi mtDNA pool being an admixture of eastern Asian and European lineages. | 17 | Mechanism of intram olecular charge transfer in DNA helix as probed by the use of the fluorescent 2-aminopurine显示文摘As a structural analogue of adenine, 2-aminopurine (2Ap) is often used as a fluorescent probe to study the intramolecular charge transfer reaction in DNA. We have designed and synthesized a series of model DNA helix with the variation in the distance between the 2Ap probe and the GGG sequence, and have investigated, by means of picosecond time-resolved fluorescence spectroscopy, the effect of the length of the bridge (consisting of a number of inosines, I) separating the electron donor (???GGG???) and the acceptor (2Ap) on the charge transfer dynamics. The fluorescence dynamics of 2Ap exhibited three exponential decay components, the one with a time constant of a few hundred picoseconds is assigned to the intramolecular charge transfer from GGG to 2Ap. Within 2.4 nm of the donor-acceptor separation, the rate of charge transfer decreased exponentially upon increasing the separation, from which the decay factor β is determined to be 1.3 nrrf-1. | 19 | Preparation of PLL-PEG-PLL and its application to DNA encapsulation显示文摘PLL-PEG-PLL was synthesized by deprotection of the amino group of PLL(Z)-PEG-PLL(Z). The properties of the polyion complex (PIC) micelles comprising of PLL-PEG-PLL and DNA were investigated. The results showed that the core of PIC was formed by PLL and DNA through electrostatic interactions, and PEG was shell of PIC, so PIC micelles in aque- ous medium were homogenous with no precipitation even under the neutral condition, and PIC had the characteristic of stability, which were very important in gene therapy. At the same time, the results showed PLL-PEG-PLL could condense DNA, longer PLL blocks were generally more effective at condensation, and the nuclease resistance of DNA was increased remarkably. PIC was spherical with diameter of 10―35 nm, which could be easily taken up by cells.
