1Clinical value of preoperative methylated septin 9 in Chinese colorectal cancer patients显示文摘BACKGROUND The methylated septin 9(mSEPT9) assay was the first blood-based test approved by the United States Food and Drug Administration as a colorectal screening test.However, the diagnostic and prognostic role of preoperative mSEPT9 for colorectal cancer(CRC) in Chinese patients is still unknown.AIM To improve the understanding of diagnostic and prognostic factors, serum m SEPT9 was detected in Chinese CRC patients.METHODS A retrospective analysis of 354 cases, of which 300 had CRC and 54 were normal,was performed in China. Patients' characteristics, treatments, and laboratory data, including age, the date of surgery, Union for International Cancer Control(UICC) stages, distant metastasis(M), and so on, were collected. Methylation levels of SEPT9 were quantified by quantitative, methylation-specific polymerase chain reaction before surgery. In addition, the effects of mSEPT9 on the occurrence and prognosis of 330 CRC cases from The Cancer Genome Atlas(TCGA) database were evaluated using bioinformatics analyses. Potential prognostic factors for overall survival(OS) and progression-free survival(PFS)were evaluated by Kaplan-Meier univariate analysis.RESULTSIn Chinese CRC patients, positive mSEPT9 was strongly associated with advanced UICC stages, deeper invasion by the primary tumor, and more distant metastasis. Methylation levels of SEPT9 were stage-dependent and showed a stepwise increase in UICC stages(I–IV), primary tumor categories(T1–T4),regional node categories(N0–N2), and distant metastasis categories(M0–M1).The patients with positive mSEPT9 showed a tendency toward lower PFS. After analyzing TCGA clinical data, the high mSEPT9 group was found to be obviously correlated only with more distant metastasis. The patients with high mSEPT9 levels showed a tendency toward lower OS. Besides, nine meaningful mSEPT9 sites were found to provide guidance for the follow-up studies.CONCLUSION MSEPT9 analysis may add valuable information to current tumor staging. 2A methylated-DNA-binding complex required for plant development mediates transcriptional activation of promoter methylated genes显示文摘Although the mechanism of DNA methylationmediated gene silencing is extensively studied, relatively little is known about how promoter methylated genes are protected from transcriptional silencing. SUVH1, an Arabidopsis Su(var)3-9 homolog, was previously shown to be required for the expression of a few promoter methylated genes. By chromatin immunoprecipitation combined with sequencing, we demonstrate that SUVH1 binds to methylated genomic loci targeted by RNA-directed DNA methylation. SUVH1 and its homolog SUVH3 function partially redundantly and interact with three DNAJ domain-containing homologs, SDJ1, SDJ2, and SDJ3, thus forming a complex which we named SUVH-SDJ. The SUVH-SDJ complex components are co-localized in a large number of methylated promoters and are required for the expression of a subset of promoter methylated genes. We demonstrate that the SUVHSDJ complex components have transcriptional activation activity. SUVH1 and SUVH3 function synergistically with SDJ1,SDJ2, and SDJ3 and are required for plant viability. 3The relationship between methylated chromans and maturity of organic matter in the source rocks from Jianghan hypersaline basin显示文摘The distribution,the relative composition,and their variation of methylated chromans(MTTC) with maturation of organic matter in the source rocks from Mingjia 1 well,Jianghan hypersaline basin,have been analyzed and studied. There is a close relationship between the distribution of methylatd chromans and paleosalinity of depositional environment. In the Eq1-Eq3 source rocks deposited under hypersaline environment with lower Pr/Ph ratio,higher gammacerane index and abundant organic sulfur-containing compounds,methyl MTTC and dimethyl MTTC are main components,but in the Eq4 source rocks deposited non-hypersaline environment with relatively higher Pr/Ph ratio,lower gammacerane index and organic sulfur-containing compounds,trimethyl MTTC is a major compound in methylate chromans. Obviously,the distributions and the compositions of methylated chromans in the source rocks are in harmony with biomarker indicators indicating the paleaosalinity and redox of depositional environment. The relationship between the ratio of 5,8-dimethyl MTTC to 7,8-dimethyl MTTC(dimethyl MTTC ratio) and the maturity of organic matter in the source rocks from Mingjia 1 well has been discussed. The results show that dimethyl MTTC ratio increased with the burial depth and the maturation of organic matter in the source rocks,especially in immature level. It is noteworthy that this ratio is very susceptible to small variation in the maturity of organic matter in the source rocks when vitrinite reflectance Ro is less than 0.65%. 4Genome-wide differences in DNA methylation changes in caprine ovaries between oestrous and dioestrous phases显示文摘Background: DNA methylation plays a vital role in reproduction.Entire genome DNA methylation changes during the oestrous phase(ES) and dioestrous phase(DS) in the ovaries of Guanzhong dairy goats were investigated using bisulphite sequencing to understand the molecular biological mechanisms of these goats' oestrous cycle.Results: We discovered distinct genome-wide DNA methylation patterns in ES and DS ovaries.A total of 26,910 differentially methylated regions were upregulated and 21,453 differentially methylated regions were downregulated in the ES samples compared with the DS samples(P-values ≤0.05 and fold change of methylation ratios ≥2).Differentially methylated region analysis showed hypomethylation in the gene body regions and hypermethylation in the joining region between upstream regions and gene bodies.The methylation ratios of the STAR,FGF2,FGF12,BMP5 and SMAD6 genes in the ES samples were lower than those of the DS samples(P-values≤0.05 and fold change of methylation ratios ≥2).Conversely,the methylation ratios of the EGFR,TGFBR2,IGF2 BP1 and MMD2 genes increased in the ES samples compared with the DS samples.In addition,223 differentially methylated genes were found in the GnRH signalling pathway(KO04912),ovarian steroidogenesis pathway(KO04913),oestrogen signalling pathway(KO04915),oxytocin signalling pathway(KO04921),insulin secretion pathway(KO04911) and MAPK signalling pathway(KO04010).Conclusions: This study is the first large-scale comparison of the high-resolution DNA methylation landscapes of oestrous and dioestrous ovaries from dairy goats.Previous studies and our investigations have shown that the NR5 A2,STAR,FGF2 and BMP5 genes might have potential application value in regulating caprine oestrus.Xiaopeng An Haidong Ma Peng Han Chao Zhu Binyun Cao Yueyu Bai 2019Journal of Animal Science and Biotechnology2019,10,1:1
