| 1 | Dissection of genetic overlap of salt tolerance QTLs at the seedling and tillering stages using backcross introgression lines in rice显示文摘QTLs for salt-tolerance(ST)related traits at the seedling and tillering stages were identified using 99 BC2F8 introgression lines(IL)derived from a cross between IR64(indica)as a recurrent parent and Binam(japonica)from Iran as the donor parent.Thirteen QTLs affecting survival days of seedlings(SDS), score of salt toxicity of leaves(SST),shoot K + concentration(SKC)and shoot Na + concentration(SNC) at the seedling stage and 22 QTLs underlying fresh weight of shoots(FW),tiller number per plant(TN) and plant height(PH)at the tillering stage were identified.Most QTLs detected at the tillering stage showed obvious differential expression to salt stress and were classified into three types based on their differential behaviors.Type I included 11 QTLs which were expressed only under the non-stress condition.Type II included five QTLs expressed in the control and the salt stress conditions,and three of them(QPh5,QPh8 and QTn9)had similar quantity and the same direction of gene effect,suggesting their expression was less influenced by salt stress.Type III included six QTLs which were detectable only under salt stress,suggesting that these QTLs were apparently induced by the stress.Thirteen QTLs affecting trait difference or trait stability of ILs between the stress and non-stress conditions were identified and the Binam alleles at all loci except QPh4,QTn2 and QFw2a decreased trait difference.The three QTLs less influenced by the stress and 13 QTLs affecting trait stability were considered as ST QTLs which contributed to ST.Comparing the distribution of QTLs detected at the seedling and tillering stages,most(69%)of them were genetically independent.Only four were the same or adjacent regions on chromosomes 1,2,8 and 11 harboring ST QTLs detected at the two stages,suggesting that partial genetic overlap of ST across the two stages occurs.It is likely,therefore,to develop ST rice variety for both stages by pyramiding of ST QTLs of different stages or selection against the overlapping QTLs between the two stages via marker-assisted selection(MAS). | ZANG JinPing 1 ,SUN Yong 1 ,WANG Yun 1 ,YANG Jing 1 ,LI Fang 1 ,ZHOU YongLi 1 ,ZHU LingHua 1 , Reys JESSICA2,Fotokian MOHAMMADHOSEIN 2,XU JianLong 1&LI ZhiKang 1,2 1Institute of Crop Sciences/National Key Facility for Crop Gene Resources&Genetic Improvement,Chinese Academy of Agricul- tural Sciences,Beijing 100081,China 2International Rice Research Institute,DAPO Box 7777,Metro Manila,Philippines | 2008 | Science China(Life Sciences)2008,51,7: | 32 |
| 2 | Endothelial function and T-lymphocyte subsets in patients with overlap syndrome of chronic obstructive pulmonary disease and obstructive sleep apnea显示文摘Background:The coexistence of chronic obstructive pulmonary disease (COPD) and obstructive sleep apnea (OSA) is termed overlap syndrome (OS).COPD and OSA both have increased risks of developing cardiovascular diseases.This study aimed to explore if patients with OS exhibited a higher prevalence of cardiovascular complications,and if patients with OS exhibited vascular endothelial dysfunction and abnormalities in the cellular immune function of T lymphocytes.Methods:Totally 25 patients with stable COPD (COPD group),25 patients with OSA (OSA group),25 patients with OS (OS group),and 20 healthy adults (control group) were enrolled between January 2017 and December 2017 from the Respiratory Department of Tianjin Medical University General Hospital.The clinical characteristics of the four groups were collected and the expression levels of soluble vascular cell adhesion molecule-1 (sVCAM-1),tumor necrosis factor-α(TNF-α),and T-lymphocyte subsets were detected.One-way analysis of variance,x^2 test and Pearson correlation were used to manage the data.Results:The prevalence of hypertension and coronary heart disease was significantly higher in the OS group than in the control,OSA,and COPD groups (x^2 =20.69,P < 0.05 and x^2 =11.03,P < 0.05,respectively).The levels of sVCAM-1 and TNF-α were significantly higher in the OS group than in other groups (F =127.40,P < 0.05 and F =846.77,P < 0.05,respectively).The percentage of CD4+ lymphocytes and CD4+/CD8+ were both significantly lower in the OS group than in any other group (F =25.40,P < 0.05 and F =75.08,P < 0.05,respectively).There were significantly negative correlations in the levels of sVCAM-1 and TNF-α with CD4^+/CD8^+ lymphocytes (r =-0.77,P < 0.05 and r =-0.83,P < 0.05,respectively).Conclusions:The prevalence of hypertension and coronary heart disease was higher in patients with OS than in patients with either OSA or COPD alone.Patients with OS exhibited more severe vascular endothelial injury,stronger inflammatory response,and lower cellular immune function. | Juan Wang Xin Li Wan-Ju Hou Li-Xia Dong Jie Cao | 2019 | Chinese Medical Journal2019,,14: | 18 |
| 3 | Performance of transient elastography in assessing liver fibrosis in patients with autoimmune hepatitis-primary biliary cholangitis overlap syndrome显示文摘AIM To investigate the performance of transient elastography(TE) for diagnosis of fibrosis in patients with autoimmune hepatitis-primary biliary cholangitis(AIHPBC) overlap syndrome.METHODS A total of 70 patients with biopsy-proven AIH-PBC overlap syndrome were included. Spearman correlation test was used to analyze the correlation of liver stiffness measurement(LSM) and fibrosis stage. Independent samples Student's t-test or one-way analysis of variance was used to compare quantitative variables. Receiver operating characteristics(ROC) curve was used to calculate the optimal cut-off values of LSM for predicting individual fibrosis stages. A comparison on the diagnostic accuracy for severe fibrosis was made between LSM and other serological scores.RESULTS Patients with AIH-PBC overlap syndrome had higher median LSM than healthy controls(11.3 ± 6.4 k Pa vs 4.3 ± 1.4 k Pa, P < 0.01). LSM was significantly correlated with fibrosis stage(r = 0.756, P < 0.01). LSM values increased gradually with an increased fibrosis stage. The areas under the ROC curves of LSM for stages F ≥ 2, F ≥ 3, and F4 were 0.837(95%CI: 0.729-0.914), 0.910(0.817-0.965), and 0.966(0.893-0.995), respectively. The optimal cut-off values of LSM for fibrosis stages F ≥ 2, F ≥ 3, and F4 were 6.55, 10.50, and 14.45 k Pa, respectively. | 5 | 洋葱伯克霍尔德菌脂肪酶的基因改造及其在毕赤酵母中组成型和诱导型的表达显示文摘【目的】克隆洋葱伯克霍尔德菌(Burkholderia cepacia)脂肪酶基因,实现其在毕赤酵母中快速、安全和稳定性的大量表达。【方法】首先设计引物扩增B.cepacia脂肪酶基因,然后应用生物信息学方法分析B.cepacia和毕赤酵母整体密码子使用情况、脂肪酶基因信号肽及密码子偏好性。在此基础上,运用overlap PCR对脂肪酶基因中低使用频率密码子进行改造并同时降低基因的G+C含量,获得优化的脂肪酶基因。再分别把原始和优化的脂肪酶基因导入载体pGAPZα和pPIC9K中,获得组成型表达载体pGAPlipW、pGAPlipO和诱导型表达载体pPIClipW、pPIClipO。分别将所得4种载体转入GS115中,得到一系列工程菌。经发酵和NTA树脂纯化后,对脂肪酶的酶学性质进行了初步研究。【结果】4种工程菌的脂肪酶活力分别为pPIClipW37.8U/mL,pPIClipO129.5U/mL,pGAPlipW40.2U/mL和pGAPlipO184.3U/mL。改造后脂肪酶活力比原始脂肪酶提高了4.6倍。酶学性质研究表明,脂肪酶在60℃时活力最高,在40℃-65℃范围内非常稳定;脂肪酶最适pH值为9.0,在pH6.0-pH10.0范围均表现很好的稳定性。【结论】通过overlap PCR改造后的脂肪酶显著提高了其在毕赤酵母中的表达效率,且GAP启动子比AOX1启动子更适合于B.cepacia脂肪酶的表达。大量表达的重组脂肪酶的性质与野生脂肪酶的性质相同,符合生产要求。 | 贾彬 刘文山 杨江科 叶才伟 徐莉 闫云君 | 2010 | 微生物学报2010,50,9: | 10 |

| 7 | Inverse-designed photonic fibers and metasurfaces for nonlinear frequency conversion[Invited]显示文摘Typically, photonic waveguides designed for nonlinear frequency conversion rely on intuitive and established principles, including index guiding and bandgap engineering, and are based on simple shapes with high degrees of symmetry. We show that recently developed inverse-design techniques can be applied to discover new kinds of microstructured fibers and metasurfaces designed to achieve large nonlinear frequency-conversion efficiencies. As a proof of principle, we demonstrate complex, wavelength-scale chalcogenide glass fibers and gallium phosphide three-dimensional metasurfaces exhibiting some of the largest nonlinear conversion efficiencies predicted thus far,e.g., lowering the power requirement for third-harmonic generation by 104 and enhancing second-harmonic generation conversion efficiency by 107. | 8 | 腹腔镜全胃切除术食管空肠经辅助切口圆形吻合与腔内吻合的安全性和有效性分析显示文摘目的分析比较腹腔镜全胃切除术食管空肠经辅助切口圆形吻合与腔内吻合的安全性和有效性。方法采用回顾性队列研究方法,收集2018年10月至2021年6月期间,在南方医科大学南方医院普通外科行腹腔镜全胃切除、D2淋巴结清扫、并完成食管空肠Roux-en-Y吻合术、具备术后6个月完整随访资料的261例胃癌患者的临床资料。根据患者食管空肠吻合方式不同进行分组,采用经辅助切口圆形吻合139例(辅助切口圆吻组),腔内吻合122例(腔内吻合组),腔内吻合组中OrVil^(TM)吻合43例(OrVil^(TM)吻合组),Overlap吻合79例(Overlap吻合组)。与辅助切口圆吻组的基线资料比较,腔内吻合组患者的体质指数偏大,肿瘤直径偏小,肿瘤T分期和M分期偏早,差异均有统计学意义(均P<0.05);与Overlap吻合组基线资料比较,OrVil^(TM)吻合组术前合并胃癌所致梗阻比例以及侵犯食管的比例均较高,T分期较晚,差异均有统计学意义(均P<0.05);两组其他基线资料的比较,差异均无统计学意义(均P>0.05)。主要观察指标为比较辅助切口圆吻组与腔内吻合组以及OrVil^(TM)吻合与Overlap吻合两个亚组的术后6个月内并发症发生情况;次要观察指标为上述各组手术情况、术中并发症发生情况以及术后恢复情况的比较。符合正态分布的连续变量采用±s表示,两组间比较采用t检验;非正态分布的连续变量用M(IQR)表示,两组间比较采用Mann-Whiney U检验;分类变量用例(%)表示,组间比较采用χ^(2)检验、连续校正χ^(2)检验或Fisher精确检验。结果辅助切口圆吻组与腔内吻合组手术情况比较,腔内吻合组手术切口更小[5.0(1.0)cm比8.0(1.0)cm,Z=-10.931,P=0.001]、联合脏器切除比例更低[0.8%(1/122)比7.9%(11/139),χ^(2)=7.454,P=0.006]、R0切除率更高[94.3%(115/122)比84.9(118/139),χ^(2)=5.957,P=0.015],差异均有统计学意义(均P<0.05);辅助切口圆吻组和腔内吻合组术中并发症发生率分别为2.9%(4/139)和4.1%(5/122),两组差异无统计学意义(χ^(2)=0.040,P=0.842)。在术后恢复方面,两组的首次下地活动时间、首次排气时间和腹腔引流管拔除时间比较,差异均无统计学意义(均P>0.05),但是辅助切口圆吻组比腔内吻合组更早恢复全流食和半流食[分别(5.1±2.4)d比(5.9±3.6)d,t=-2.268,P=0.024;(7.3±3.7)d比(8.8±6.5)d,t=-2.227,P=0.027]、术后更快出院[(10.5±5.1)d比(12.2±7.7)d,t=-2.108,P=0.036],两组比较,差异均有统计学意义(均P<0.05)。辅助切口圆吻组与腔内吻合组术后6个月并发症发生率分别为25.9%(36/139)和31.1%(38/122),食管空肠吻合口并发症发生率分别为5.0%(7/139)和8.2%(10/122),两组差异均无统计学意义(均P>0.05);两组术后并发症严重程度的差异亦无统计学意义(P>0.05)。腔内吻合组中,与Overlap吻合组比较,OrVil^(TM)吻合组术中出血量较多[100.0(100.0)ml比50.0(50.0)ml,Z=-2.992,P=0.003],手术切口较长[6.0(1.0)cm比5.0(1.0)cm,Z=-3.428,P=0.001],术后拔除腹腔引流管时间更久[(9.5±6.1)d比(7.2±3.1)d,t=2.367,P=0.022],差异均有统计学意义(均P<0.05);与OrVil^(TM)吻合组比较,Overlap吻合组术中并发症发生率低[2.5%(2/79)比7.0%(3/43)],术后6个月并发症发生率也低[27.8%(22/79)比37.2%(16/43)],并发症严重程度较轻[Ⅲ~Ⅳ级:7.6%(6/79)比14.0%(6/43)],但差异均无统计学意义(均P>0.05)。结论经辅助切口食管空肠吻合与腔内食管空肠吻合在安全性方面相当。而腔内Overlap食管空肠吻合较OrVil^(TM)吻合更显安全、更具开展潜力,值得进一步探索优化。 | 陈新华 胡彦锋 林填 赵明利 陈韬 陈豪 麦锦胜 梁延锐 刘浩 赵丽瑛 李国新 余江 | 2022 | 中华胃肠外科杂志2022,25,5: | 5 |

| 12 | Efficient expression of codon-adapted human acetaldehyde dehydrogenase 2 cDNA with 6×His tag in Pichia pastoris显示文摘Human mitochondrial acetaldehyde dehydrogenase 2 (ALDH2) catalyzes the oxidation of acetaldehyde to acetic acid. Therefore, ALDH2 has therapeutic potential in detoxification of acetaldehyde. Further-more, ALDH2 catalyzes nitroglycerin to nitrate and 1, 2-glyceryldinitrate during therapy for angina pectoris, myocardial infarction, and heart failure. Large quantities of ALDH2 will be needed for potential clinical practice. In this study, Pichia pastoris was used as a platform for expression of human ALDH2. Based on the ALDH2*1 cDNA sequence, we designed ALDH2 cDNA by choosing the P. pastoris preferred codons and by decreasing the G + C content level. The sequence was synthesized using the overlap extension PCR method. The cDNA and 6×His tags were subcloned into the plasmid pPIC9K. The recombinant protein was expressed in P. pastoris GS115 and purified using Ni2+-Sepharose affinity chromatography. The amount of secreted protein in the culture was 80 mg/L in shake-flask cultivation and 260 mg/L in high-density bioreactor fermentation. Secreted ALDH2 was easily purified from the culture supernatant by using Ni2+-Sepharose affinity chromatography. After purification of the fermentation supernatant, the enzyme had a specific activity of 1.2 U/mg protein. | 13 | Optimizational production of phenyllactic acid by a Lactobacillus buchneri strain via uniform design with overlay sampling methodology显示文摘A Lactobacillus buchneri GBS3 strain isolated from the traditional Chinese pickles was used for the production of 3-phenyllactic acid(PLA), an important compound with antimicrobial activities against a wide species of grampositive and gram-negative bacteria and some fungi. The growth performance of this strain in the de Man, Rogosa and Sharpe(MRS) medium, the production of metabolites of valuable organic acids, and the biosynthesis of PLA using this strain as the whole-cell biocatalyst and phenylpyruvic acid(PPA) as the precursor, were investigated experimentally. The uniform design method with overlay sampling was developed for the optimization of the biotransformation conditions. The results showed that although it produced naturally lactic acid with the maximum concentration of 1.84 g·L^(-1) and PLA with the concentration of 0.015 g·L^(-1) after 66 to 72 h cultivation in MRS broth by fermentation, the present strain displayed an effective utilization ability by transforming PPA to PLA. By the uniform design method with overlay sampling for the design and optimization of transformation conditions, a maximum yield of 10.93 g·L^(-1) PLA with the mole conversion ratio of 83.07% from PPA to PLA was achieved under the optimized condition, i.e., 20 g·L^(-1) glucose, 270 g·L^(-1) cells, 13 g·L^(-1) PPA, pH 8.0 and the reaction time of 15 h, indicating that Lactobacillus buchneri GBS3 was an interesting strain for the biosynthesis of PLA via the microbial transformation. The prediction of PLA yield under different conditions was achieved successfully based on the limited information of only a small number of experiments by the uniform design with overlay sampling. | 14 | Brucellosis and tuberculosis:Clinical overlap and pitfalls显示文摘Objective:To identify characteristic features of tuberculosis in patients with culture proven brucellosis.Methods:A retrospective analysis was performed on patients diagnosed with culture proven brucellosis between January and December 2011,based on review of their medical records.Patients with demonstrable co-infection with tuberculosis were excluded.Clinical features,laboratory parameters and tissue histopathology reports where available were noted.Results:Thirty-two patients with brucellosis were included in the study.Twenty-one(65.63%)patients had chronic fever,thirteen(40.63%)had a productive cough,while significant weight loss,evening rise of temperature and night sweats were reported by eight(25.00%),eleven(34.38%)and five(15.63%)patients respectively.Nine(28.13%)patients had at least three of these symptoms.Lymphadenopathy,hepatomegaly and splenomegaly were noted on examination in seven(21.88%),fifteen(46.88%)and twelve(37.50%)patients respectively.Eight(25.00%)patients had hepatosplenomegaly,of these only two had associated significant lymphadenopathy.Respiratory examination was normal in all patients.Elevated ESR greater than 50 mm/hr was seen in eight(25.00%),it was greater than 100 mm/hr in five(15.63%)patients.Hypergammaglobulinemia was seen in eight(25.00%)cases.Bone marrow biopsy showed non-caseating granulomas in three(9.38%)cases,lymph node biopsy showed granulomas in one case.Overall,three(9.38%)patients had known risk factors for tuberculosis,while six(18.75%)had risk factors for brucellosis.Conclusions:There is a clear overlap between brucellosis and tuberculosis both in terms of clinical presentation and laboratory parameters.It is essential to c-arefully rule out tuberculosis in all cases of suspected or proven brucellosis before initiating antimicrobial therapy,in order to forestall development of drug-resistant tuberculosis. | Sowjanya Dasari Kushal Naha Mukhyaprana Prabhu | 2013 | Asian Pacific Journal of Tropical Medicine2013,6,10: | 2 |

| 17 | A new perspective on functional redundancy and phylogenetic niche conservatism in soil microbial communities显示文摘Functional redundancy in soil microbial communities seems to contradict the notion that individual species have distinct metabolic niches in multi-species communities.All soil microbiota have the metabolic capacity for'basic'functions(e.g.,respiration and nitrogen and phosphorus cycling),but only a few soil microbiota participate in'rare'functions(e.g.,methanogenesis and mineralization of recalcitrant organic pollutants).The objective of this perspective paper is to use the phylogenetic niche conservatism theory as an explanation for the functional redundancy of soil microbiota.Phylogenetic niche conservatism is defined as the tendency for lineages to retain ancestral functional characteristics through evolutionary time-scales.The present-day soil microbiota is the result of a community assembly process that started when prokaryotes first appeared on Earth.For billions of years,microbiota have retained a highly conserved set of core genes that control the essential redox and biogeochemical reactions for life on Earth.These genes are passed from microbe to microbe,which contributes to functional redundancy in soil microbiota at the planetary scale.The assembly of microbial communities during soil formation is consistent with phylogenetic niche conservatism.Within a specific soil,the heterogeneous matrix provides an infinite number of sets of diverse environmental conditions,i.e.,niches that lead to the divergence of microbial species.The phylogenetic niche conservatism theory predicts that two or more microbial species diverging from the same clade will have an overlap in their niches,implying that they are functionally redundant in some of their metabolic processes.The endogenous genetic factors that constrain the adaptation of individuals and,thus,populations to changing environmental conditions constitute the core process of phylogenetic niche conservatism.Furthermore,the degree of functional redundancy in a particular soil is proportional to the complexity of the considered function.We conclude with a conceptual model that identifies six patterns of functional redundancy in soil microbial communities,consistent with the phylogenetic niche conservatism theory. | Yu JIA Joann K.WHALEN | 2020 | Pedosphere2020,30,1: | 2 |
