2Effect of Fuzhenghuayu decoction on vascular endothelial growth factor secretion in hepatic stellate cells显示文摘Objective: To investigate the effect of Fuzhenghuayudecoction on autocrine activation of hepatic stellatecell (HSC).Methods: The drug serum containing Fuzhenghuayudecoction was collected from normal rats, and cul-tured with activated HSC in vitro. The conditionedmedium from the drug serum treated HSC was addedto primary cultured quiescent HSC. Cell prolifera-tion was assayed by tetrazolium colorimetric test,and the contents of type Ⅰ collagen and vascular endo-thelial growth factor (VEGF) in the supernatantwere measured with ELISA.Results: The conditioned medium from activatedHSC could stimulate the quiescent HSC proliferationand type Ⅰ collagen secretion. 3Islet cell transplantation as a cure for insulin dependent diabetes: current improvements in preserving islet cell mass and function显示文摘OBJECTIVE: To review the current progress of islet cell transplantation in patients withinsulin-dependent diabetes, emphasizing on the difficulties with recovering and preserving islet cellmass and function, 30% of which is lost during the peri-transplantation period.RESULTS: The islet-cell isolation technique is perfected, but improvements are still progressing in twomajor directions: preservation of islet cells and tolerance induction. Optimum islet cell viability andfunction depends on appropriate revascularization of the islet graft and blockade of thrombus formation aswell as cytokine and free radical release. Conditioning the islet cells in-vitro prior to transplantation toeither upregulate VEGF expression or downregulate NF-kappa B transcription factor has proven to improverevascularization and to prevent islet cell apoptosis and cytokine-mediated damage. Tolerance induction iscurrently being best achieved by selecting and combining immunosuppressive agents such as monoclonalantibodies which target the major signaling molecules during immune activation, but which are least toxicto islet cells.CONCLUSIONS: Patients with insulin-dependent diabetes will greatly benefit from current developmentsin effective approaches to protect islets during the peritransplant period. 4Thermal evolution and its significance of I-A type granitoid complex ——The Laoshan Granitoid as an example显示文摘To reconstruct the cooling process of compound granites in different units (phases) with Laoshan Granitoid as an example, the relationships between formation, emplacement of I A type granitoid and variation in tectonic environment and uplift of the orogenic belt are discussed from the viewpoint of thermal evolution. Now it has been pointed out that the cooling rates presented by the whole spectral series from I type to A type granites tend to be gradually faster and faster. 5Interference of Y-27632 on the signal transduction of transforming growth factor beta type 1 in ocular Tenon capsule fibroblasts显示文摘AIM: To investigate the interfering effect of Y-27632, a ROCK-I selective inhibitor, on the signal transduction pathway of transforming growth factor-β1 (TGF-β1) in ocular Tenon capsule fibroblasts (OTFS) in vitro . METHODS: After OTFS from passages 4 to 6 in vitro were induced by TGF-β1 and then treated by Y-27632, the changes of the OTFS cell cycles were analyzed via flow cytometry, and the proteins expression of the α-smooth muscular actin (α-SMA), connective tissue growth factor (CTGF), collagen I were calculated by Western blot. After OTFS treated by the different concentrations of Y-27632, the expression levels of the α-SMA, CTGF and collagen I mRNA were assayed by RT-PCR.RESULTS: Y-27632 had no markedly effect on the OTFS cell cycles. After treated by TGF-β1, OTFS in G1 period significantly increased. The cell cycles distribution by both TGF-β1 and Y-27632 had no remarkable difference from that in control group. Y-27632 significantly inhibited the proteins expressions of both α-SMA and CTGF, while to some extent inhibited that of collagen I. TGF-β1 significantly promoted the proteins expressions of α-SMA, CTGF and collagen I. After OTFS treated by both TGF-β1 and Y-27632, of α-SMA, the protein expression was similar with that in control group (P=0.066>0.05), but the protein expression of CTGF or collagen I, respectively, was significantly different from that in control group (P=0.000<0.01). The differences of expressions of the α-SMA, CTGF and collagen I mRNA in 30, 150, 750μmol/L Y-27632 group were statistically significant, compared with those in control group, respectively (α-SMA, P=0.002, 0.000, 0.000; CTGF, P=0.014, 0.002, 0.001; collagen I, P=0.003, 0.002, 0.000).CONCLUSION: Blocking the Rho/ROCK signaling pathway by using of Y-27632 could inhibit the cellular proliferation and the expression of both CTGF and α-SMA whatever OTFS induced by TGF-β1 or not. 6Proteomic analysis of human serum from diabetic retinopathy显示文摘· AIM: To establish and compare serum proteomic of diabetic retinopathy(DR) patients in various phases and discuss pathogenesis of DR so as to find out possible serum specific molecular markers for early diagnosis of DR. · METHODS: Thirty-two subjects were divided into four groups: one group of eight type 2 diabetes mellitus (T2DM) patients without apparent DR (No-DR, NDR), one group of eight T2DM patients with non-proliferative diabetic retinopathy(NPDR), one group of eight T2DM patients with proliferative diabetic retinopathy(PDR) and one group of eight healthy volunteer participants. Two dimensional fluorescence difference gel electrophoresis (2D-DIGE) was applied to establish differential protein expression profiles in four groups. Matrix-assisted laser desorption/ionization time of flight tandem mass spectrometry (MALDI-TOF-TOF MS) was applied to identify mass spectrometry of differential proteins and analyze follow-up bioinformatics. · RESULTS: 2D-DIGE maps of serum protein were satisfactory obtained from NDR, NPDR, PDR and normal control groups. Twenty-six different proteins spots were screened(the volume ratio was >1.5 based on DeCyder software analysis). Twenty-four of them were verified and two of them were not. Fifteen proteins were verified. Most of them were high-abundant proteins in serum. The four relatively low-abundant ones were beta 2-glycoprotein I (β2-GPI), alpha2-HS-glycoprotein(AHSG), alpha1-acid glycoprotein(α1-AGP) and apolipoprotein A-1(apo A-1). β2-GPI expression was gradually increased in the development of DR but unrelated to the severity of DR. The volume ratio of β2-GPI is 1.54, 2.43, and 2.84 in NDR, NPDR and PDR group respectively compared with normal control group. · CONCLUSION: Serum proteomic analysis of 2D-DIGE combined with MALDI-TOF-TOF MS is feasible to be applied in the study of DR. β2-GPI probably takes part in the process of DR occurrence and development and it could be a candidate biomarker on DR diagnosis in early phase. ·Yin-Ping Liu, Xiao-He Lu 2011International Journal of Ophthalmology(English edition)2011,4,6:4
7The heterozygous EDNRB mutation in a Chinese family with Waardenburg syndrome type Ⅰ显示文摘The genovariation of endothelin receptor type B(EDNRB) was identified in a Chinese family with Waardenburg syndrome type I(WS1) in the present study. WS1 was diagnosed in a 19-year-old young man, his older sister and aunt according to WS consortium criteria. After extracting genomic DNA from the peripheral blood samples, the coding exons and intronic regions of EDNRB were sequenced. A missense heterozygous mutation was found in the coding region of exon 2 in the EDNRB gene on chormosome 13 q22.3 of the proband. The same mutation was detected in the proband’s afflicted paternal aunt and first older sister. Subsequent polyphen analysis and three-dimensional modeling confirmed that the c.469 A>G heterozygous mutation in EDNRB was possibly pathogenic. 8Novel electrospun poly(ε-caprolactone)/type Ⅰ collagen nanofiber conduits for repair of peripheral nerve injury显示文摘Recent studies have shown the potential of artificially synthesized conduits in the repair of peripheral nerve injury.Natural biopolymers have received much attention because of their biocompatibility.To investigate the effects of novel electrospun absorbable poly(ε-caprolactone)/type I collagen nanofiber conduits(biopolymer nanofiber conduits)on the repair of peripheral nerve injury,we bridged 10-mm-long sciatic nerve defects with electrospun absorbable biopolymer nanofiber conduits,poly(ε-caprolactone)or silicone conduits in Sprague-Dawley rats.Rat neurologica1 function was weekly evaluated using sciatic function index within 8 weeks after repair.Eight weeks after repair,sciatic nerve myelin sheaths and axon morphology were observed by osmium tetroxide staining,hematoxylin-eosin staining,and transmission electron microscopy.S-100(Schwann cell marker)and CD4(inflammatory marker)immunoreactivities in sciatic nerve were detected by immunohistochemistry.In rats subjected to repair with electrospun absorbable biopolymer nanofiber conduits,no serious inflammatory reactions were observed in rat hind limbs,the morphology of myelin sheaths in the injured sciatic nerve was close to normal.CD4 immunoreactivity was obviously weaker in rats subjected to repair with electrospun absorbable biopolymer nanofiber conduits than in those subjected to repair with poly(ε-caprolactone)or silicone.Rats subjected to repair with electrospun absorbable biopolymer nanofiber conduits tended to have greater sciatic nerve function recovery than those receiving poly(ε-caprolactone)or silicone repair.These results suggest that electrospun absorbable poly(ε-caprolactone)/type I collagen nanofiber conduits have the potential of repairing sciatic nerve defects and exhibit good biocompatibility.All experimental procedures were approved by Institutional Animal Care and Use Committee of Taichung Veteran General Hospital,Taiwan,China(La-1031218)on October 2,2014.Chun-Ming Yen Chiung-Chyi Shen Yi-Chin Yang Bai-Shuan Liu Hsu-Tung Lee Meei-Ling Sheu Meng-Hsiun Tsai Wen-Yu Cheng 2019Neural Regeneration Research2019,14,9:1
9Manipulating the Interferon Signaling Pathway: Implications for HIV Infection显示文摘During human immunodeficiency virus(HIV) infection, type I interferon(IFN-I) signaling induces an antiviral state that includes the production of restriction factors that inhibit virus replication, thereby limiting the infection. As seen in other viral infections, type I IFN can also increase systemic immune activation which, in HIV disease, is one of the strongest predictors of disease progression to acquired immune deficiency syndrome(AIDS) and non-AIDS morbidity and mortality.Moreover, IFN-I is associated with CD4 T cell depletion and attenuation of antigen-specific T cell responses. Therefore,therapeutic manipulation of IFN-I signaling to improve HIV disease outcome is a source of much interest and debate in thefield. 10Caspase-3 Mediates Apoptosis of Striatal Cells in GA Ⅰ Rat Model显示文摘In previous study,glutaric acid (GA) induced apoptosis of primary striatal neuron in vitro.In order to investigate the neurotoxic effects of GA on neonatal rat corpus striatum and the possible mechanism,34 male pups were randomly assigned to NS group,low dose GA (LGA,5 μmol GA/g body weight) group and high dose GA (HGA,10 μmol GA/g body weight) group.These pups were subcutaneously administered with three injections from postnatal day 3 to 22 at 7:30 am,15:00 pm and 22:30 pm and killed 12 h after the last injection.Microscopic pathology in corpus striatum was evalu-ated by HE staining.The apoptotic cells were identified by TUNEL staining.The transcript levels of caspase-3,8,9,Bax,Bcl-2 were detected by using real-time PCR and the protein levels of procaspase-3 and the active fraction were evaluated by Western blotting.In LGA and HGA groups,ventricle collapse,cortical atrophy by a macroscope and interstitial edema,vacuolations,widened perivascular space of bi-lateral striatum by a microscope were observed.TUNEL assay revealed that the apoptotic cells were in-creased in LGA and HGA groups.The transcript of caspase-3 was up-regulated to 2.5 fold,accompanied by the up-regulation of caspase-9,Bax and down-regulation of Bcl-2.The protein levels of procaspase-3 and the active fraction were up-regulated in LGA and HGA groups.The rat model for GA Ⅰ showed mitochondrial apoptotic pathway may be involved in the GA-induced striatal lesion.Further studies should be taken to investigate the underlying mechanisms.田凤艳 付溪 高金枝 张猜 宁琴 罗小平 2012Journal of Huazhong University of Science and Technology(Medical Sciences)2012,32,1:1
11Role of SR-BI in atherosclerosis, malignancies, and infectious diseases显示文摘Scavenger receptor class B type Ⅰ(SR-BI) is a high-affinity receptor for high-density lipoprotein(HDL). The primary role of this receptor is the selective uptake of HDLs in the liver through reverse cholesterol transport. SR-BI interacts with HDL to regulate lipid metabolism and affect various vascular cell functions involved in atherosclerosis(As). In addition, SRBI is involved in the development of malignant tumors and infectious diseases. 12High expression of type I inositol 1,4,5-trisphosphate receptor in the kidney of rats with hepatorenal syndrome显示文摘AIM To detect the expression of typeⅠ inositol 1,4,5-trisphosphate receptor(IP3 RI) in the kidney of rats with hepatorenal syndrome(HRS).METHODS One hundred and twenty-five Sprague-Dawley rats were randomly divided into four groups to receive an intravenous injection of D-galactosamine(D-Gal N) plus lipopolysaccharide(LPS; group G/L, n = 50), D-Gal N alone(group G, n = 25), LPS alone(group L, n = 25), and normal saline(group NS, n = 25), respectively.At 3, 6, 9, 12, and 24 h after injection, blood, liver, and kidney samples were collected. Hematoxylineosin staining of liver tissue was performed to assess hepatocyte necrosis. Electron microscopy was used to observe ultrastructural changes in the kidney. Western blot analysis and real-time PCR were performed to detect the expression of IP3 RI protein and m RNA in the kidney, respectively.RESULTS Hepatocyte necrosis was aggravated gradually, which was most significant at 12 h after treatment with D-galactosamine/lipopolysaccharide, and was characterized by massive hepatocyte necrosis. At the same time, serum levels of biochemical indicators including liver and kidney function indexes were all significantly changed. The structure of the renal glomerulus and tubules was normal at all time points. Western blot analysis indicated that IP3 RI protein expression began to rise at 3 h(P < 0.05) and peaked at 12 h(P < 0.01). 13Comparison of metastatic potentiality and features related to metastasis in human colorectal carcinoma cell lines显示文摘Objective To compare metastatic potentiality and features related to metastasis in two kinds ofhuman colorectal carcinorna (HCC) cell lines. Methods: Experimental metastatic potentiality of two kinds ofcell lines, HT29 and LoVo cells were compared by using nude mice model for human colorectal carcinoma liver metastasis. The expressions of epithelial-cadherin (E-cad), laminin receptor(LNR) and type Ⅳcollagenase(Ⅳ col) in two kinds of HCC cell lines, HT29 and LoVo cells, were observed quantitatively. The intracellular CD44V6 expressed in mRNA and in protein of the HCC cell lines were detected with in situ hybridization and immunohistochemistry. Results: 100% of nude mice had liver metastasis, which demonstrated thatLoVo is a high metastatic HCC cell line. However, HT29 is a relatively low metastatic HCC cell line becauseonly 42. 9 % of nude mice transplanted HT29 cells had liver metastasis. The expressions of E--cad, LNR andIVcol of HT29 and LoVo cells in vitro were detected quantitatively. HT29 cells expressed E-cad with weakpositive on cell membrane. and LoVo cells almost expressed none. LoVo cells expressed higher level of LNRand tV col than HT29 cells did. The expression of CD44V6 in LoVo cells whether in mRNA or in protein werehigher than that of HT29 cells. Conclusion: LoVo is a high metastatic HCC cell line while HT29 is a relatively low metastatic HCC cell line. 14Energy Conditions and Conservation Laws in LRS Bianchi Type Ⅰ Spacetimes via Noether Symmetries显示文摘In this paper, we have completely classified the locally rotationally symmetric(LRS) Bianchi Type Ⅰ spacetimes via Noether symmetries(NS). The usual Lagrangian corresponding to LRS Bianchi Type Ⅰ metric is used to find the set of determining equations. To achieve a complete classification, these determining equations are generally integrated to find the components of NS vector field and the metric coefficients. During this procedure, several cases arise which give different Noether algebras of dimension 5,..., 9, 11, and 17. A comparison is established between the obtained NS and the Killing and homothetic vectors. Corresponding to all NS generators, the conservation laws are stated by using Noether's theorem. 15Promotion of minTBP-1-PRGDN on the attachment,proliferation and collagen I synthesis of human keratocyte on titanium显示文摘AIM: To investigate the influence of minTBP-1-PRGDN on the attachment,proliferation and collagen I synthesis of human keratocyte on titanium(Ti) surface. · METHODS: The chimeric peptide RKLPDAPRGDN(minTBP-1-PRGDN) was synthesized by connecting RKLPDA(minTBP-1) to the N-terminal of PRGDN,the influence of minTBP-1-PRGDN on the attachment,proliferation and collagen I synthesis of human keratocyte on Ti surface were tested using PRGDN and minTBP-1 as controls. The keratocytes attached to the surface of Ti were either stained with FITC-labeled phalloidin and viewed with fluorescence microscope or quantified with alamar Blue method. The proliferation of keratocytes on Ti were quantified with 3-(4,5-dim-ethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide uptaking methods. The secretion of type I collagen was determined using an ELISA kit. ·RESULTS: The results showed that minTBP-1-PRGDN at a concentration of 100ng/mL was the most potent peptide to enhance the attachment of human keratocytes to the surface of Ti(1.40±0.03 folds,P =0.003),to promote the proliferation(1.26 ±0.05 folds,P =0.014) and the synthesis of type I collagen(1.530 ±0.128,P =0.008). 16Studying dynamic process of photosensitization by ESR method显示文摘By kinetic methods, functional relation of TAN radical, produced in the process of TEMPONE trapping O2, vs. time during photosensitization was established. Accordingly relative rate constants of generating all kinds of active intermediates through types I and II mechanism of photosensitization can be calculated.
