| 4 | Comprehensive analysis of coding and non-coding RNA transcriptomes related to hypoxic adaptation in Tibetan chickens显示文摘Background:Tibetan chickens,a unique native breed in the Qinghai-Tibet Plateau of China,possess a suite of adaptive features that enable them to tolerate the high-altitude hypoxic environment.Increasing evidence suggests that long non-coding RNAs(lncRNAs)and microRNAs(miRNAs)play roles in the hypoxic adaptation of high-altitude animals,although their exact involvement remains unclear.Results:This study aimed to elucidate the global landscape of mRNAs,lncRNAs,and miRNAs using transcriptome sequencing to construct a regulatory network of competing endogenous RNAs(ceRNAs)and thus provide insights into the hypoxic adaptation of Tibetan chicken embryos.In total,354 differentially expressed genes(DE genes),389 differentially expressed lncRNAs(DE lncRNAs),and 73 differentially expressed miRNAs(DE miRNAs)were identified between Tibetan chickens(TC)and control Chahua chickens(CH).GO and KEGG enrichment analysis revealed that several important DE miRNAs and their target DE lncRNAs and DE genes are involved in angiogenesis(including blood vessel development and blood circulation)and energy metabolism(including glucose,carbohydrate,and lipid metabolism).The ceRNA network was then constructed with the predicted DE gene-DE miRNA-DE lncRNA interactions,which further revealed the regulatory roles of these differentially expressed RNAs during hypoxic adaptation of Tibetan chickens.Conclusions:Analysis of transcriptomic data revealed several key candidate ceRNAs that may play high-priority roles in the hypoxic adaptation of Tibetan chickens by regulating angiogenesis and energy metabolism.These results provide insights into the molecular mechanisms of hypoxic adaptation regulatory networks from the perspective of coding and non-coding RNAs. | Ying Zhang Woyu Su Bo Zhang Yao Ling Woo Kyun Kim Hao Zhang | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 6 |

| 9 | Genome-wide profiling of long noncoding RNA expression patterns and CeRNA analysis in mouse cortical neurons infected with different strains of borna disease virus显示文摘Borna disease virus 1(BoDV-1)is neurotropic prototype of Bornaviruses causing neurological diseases and maintaining persistent infection in brain cells of mammalian species.Long non-coding RNA(lncRNA)is transcript of more than 200 nucleotides without proteincoding function regulating various biological processes as proliferation,apoptosis,cell migration and viral infection.However,regulatory of lncRNAs in BoDV-1 infection remains unknown.To identify differential expression profiles and predict functions of lncRNA in BoDV-1 infection,microarray data showed that 3528 lncRNAs and 2661 lncRNAs were differentially expressed in Strain V and Hu-H1 BoDV-infected groups compared with control groups,respectively.Gene Ontology(GO)and pathway analysis suggested that differential lncRNAs may be involved in regulation of metabolic,biological regulation,cellular process,endocytosis,viral infections and cell adhesion processes,cancer in both BoDV-infected strains.ENSMUST00000128469 was found down-regulated in both BoDV-infected groups compared with control groups consistent with microarray(p<0.05).ceRNA analysis indicated possible interaction networks as ENSMUST00000128469/miR-22-5p,miR-206-3p,miR-302b-5p,miR-302c-3p,miR-1a-3p/Igf1.Igf1 was found up-regulated in both BoDV-infected groups compared with control groups(p<0.05).Possible functions of predicted target mRNAs and miRNAs of ENSMUST00000128469 were involved in cell proliferation,transcriptional misregulation and proteoglycan pathways enriched in cancer.lncRNA may be involved in regulation of Hu-H1 inhibited cell proliferation and promoted apoptosis through NF-kB,JNK/MAPK signaling,BCL2 and CDK6/E2F1 pathways different from Strain V.Possible interaction networks as ENSMUST00000128469/miR-22-5p,miR-206-3p,miR-302b-5p,miR-302c-3p,miR-1a-3p/Igf1 may involve in regulation of cell proliferation,apoptosis,and cancer. | Lin Sun Yujie Guo Peng He Xiaoyan Xu Xiong Zhang Haiyang Wang Tian Tang Wei Zhou Ping Xu Peng Xie | 2019 | Genes & Diseases2019,6,2: | 5 |
| 10 | Down-regulating Circular RNA Prkcsh suppresses the inflammatory response after spinal cord injury显示文摘Circular RNAs(circRNAs)are a class of conserved,endogenous non-coding RNAs that are involved in transcriptional and post-transcriptional gene regulation and are highly enriched in the nervous system.They participate in the survival and differentiation of multiple nerve cells,and may even promote the recovery of neurological function after stroke.However,their role in the inflammatory response after spinal cord injury remains unclear.In the present study,we established a mouse model of T9 spinal cord injury using the modified Allen’s impact method,and identified 16,013 circRNAs and 960 miRNAs that were differentially expressed after spinal cord injury.Of these,the expression levels of circPrkcsh were significantly different between injured and sham-treated mice.We then treated astrocytes with tumor necrosis factor-αin vitro to simulate the inflammatory response after spinal cord injury.Our results revealed an elevated expression of circPrkcsh with a concurrent decrease in miR-488 expression in injured cells.We also found that circPrkcsh regulated the expression of the inflammationrelated gene Ccl2.Furthermore,in tumor necrosis factor-α-treated astrocytes,circPrkcsh knockdown decreased the expression of Ccl2 by upregulating miR-488 expression,and reduced the secretion of inflammatory cytokines in vitro.These findings suggest that differentially expressed circRNAs participate in the inflammatory response after spinal cord injury and act as the regulators of certain microRNAs.Furthermore,circPrkcsh may be used as an miR-488 sponge to regulate Ccl2 expression,which might provide a new potential therapy for SCI.The study was approved by the Animal Ethics Committee of Shandong University of China(approval No.KYLL-20170303)on March 3,2017. | Jia-Nan Chen Yi-Ning Zhang Li-Ge Tian Ying Zhang Xin-Yu Li Bin Ning | 2022 | Neural Regeneration Research2022,17,1: | 4 |

| 12 | 长链非编码RNA SOX21-AS1调控miR-31-5p/MMP-16轴对胰腺癌细胞活性与增殖的影响显示文摘背景与目的:胰腺癌是一种常见的消化系统恶性肿瘤,也是癌症死亡的常见病因。长链非编码RNA(lncRNA)已在多种恶性肿瘤中被鉴定为调节因子和特异度生物标志物。最新研究表明,lncRNA SOX21-AS1在多种恶性肿瘤的发生、发展中起重要作用。但其在胰腺癌中的作用尚未明确。因此,本研究探讨SOX21-AS1在胰腺癌中的表达及其功能。方法:用qRT-PCR检测SOX21-AS1在20对胰腺癌与邻近癌旁正常组织,以及胰腺癌细胞系(PANC-1、CAPAN2、CFPAC-1、BXPC3、SW1990)与人胰管上皮细胞系(HPDE)中的表达。通过GEPIA数据库分析SOX21-AS1表达与胰腺癌患者预后的关系。采用MTT实验与集落形成实验分析SOX21-AS1沉默后胰腺癌细胞活力与增殖能力的变化。通过DIANA数据库预测SOX21-AS1的靶微小RNA(miRNA)及其下游的靶mRNA,随后进一步通过系列功能实验、双荧光素酶实验、拯救实验以及相关性分析明确它们之间的关系。结果:SOX21-AS1在胰腺癌组织和细胞系中表达均上调,其高表达患者预后差(均P<0.05)。SOX21-AS1沉默后,胰腺癌细胞的活力与增殖能力均明显降低(均P<0.05)。生物信息学分析显示,miR-31-5p与SOX21-AS1及MMP-16 mRNA均存在靶向结合序列,功能实验、荧光素酶实验及挽救实验结果均验证了三者之间的互作关系。此外,相关性分析结果显示,SOX21-AS1的表达与miR-31-5p的表达呈负相关(r^(2)=0.2571,P<0.05),miR-31-5p的表达与MMP-16 mRNA的表达呈负相关(r^(2)=0.3113,P=0.0106)。结论:SOX21-AS1的表达上调与胰腺癌细胞活性、增殖能力增强及胰腺癌患者预后不良相关。机制上,SOX21-AS1可能作为竞争性内源RNA(ceRNA)与miR-31-5p发生竞争性结合,调节MMP-16的表达,从而促进胰腺癌的进展。SOX21-AS1可以作为胰腺癌的潜在预后生物标志物和诊治的靶点。 | 臧龙军 陈东杰 高文哲 黄珲 朱红伟 余枭 | 2022 | 中国普通外科杂志2022,31,3: | 4 |
