| 1 | A Liutex based definition and identification of vortex core center lines显示文摘Six core issues for vortex definition and identification concern with (1) the absolute strength,(2) the relative strength,(3) the rotational axis,(4) the vortex core center,(5) the vortex core size, and (6) the vortex boundary (Liu C. 2019). However, most of the currently popular vortex identification methods, including the Q criterion, the criterion and the Acj criterion etc., are Eulerian local region-type vortex identification criteria and can only approximately identify the vortex boundary by somewhat arbitrary threshold. On the other hand, the existing Eulerian local line-type methods, which seek to extract line-type features such as vortex core line, are not entirely satisfactory since most of these methods are based on vorticity or pressure minimum that will fail in many cases. The key issue is the lack of a reasonable mathematical definition for vortex core center. To address this issue, a Liutex (previously named Rortex) based definition of vortex core center is proposed in this paper. The vortex core center, also called vortex rotation axis line here, is defined as a line where the Liutex magnitude gradient vector is aligned with the Liutex vector, which mathematically implies that the cross product of the Liutex magnitude gradient vector and the Liutex vector on the line is equal to zero. Based on this definition, a novel three-step method for extracting vortex rotation axis lines is presented. Two test cases, namely the Burgers vortex and hairpin vortices, are examined to justify the proposed method. | 6 | Development of Chinese Near-isogenic Line of Rice and Their Differentiating Ability of Pathogenic Races of Blast Fungus显示文摘The rice( Oryza L. ) variety, Lijiangxintuanheigu(LTH), is a universally susceptible variety. A set of near-isogenic lines (NILs) of rice which has single resistance gene for each line was developed by use of LTH as recurrent parent and Kusabue,Tsuyuake, K1, PiNo. 4, K60 and BL1, which are from Kiyosawa's differential varieties(KDV), as donor parents. Differentiating ability of these NILs to Philippine isolates was compared with that of their donor parents, KDV and IRRI' s NILs. The results of comparative studies were as follows: (1) Differentiating ability of Chinese NILs to Philippine isolates was much stronger than that of their donor parents and that of KDV. (2) Chinese NILs had similar ability to differentiate races to IRRI's NILs or higher differentiating ability than them. But IRRI's NILs almost had no differentiating ability to isolates from japonica rice-growing regions. They can not be used to differentiate races of isolates from japonica ricegrowing regions. Whereas, Chinese NILs can be used to differentiate races of isolates from both japonica ricegrowing regions and indica rice-growing regions. | 9 | Characterization and evaluation of rice blast resistance of Chinese indica hybrid rice parental lines显示文摘The development of resistant varieties and hybrid combinations has been the most effective and economical strategy to control blast disease caused by Magnaporthe oryzae.However, the distribution of major R genes and blast resistance characterization in hybrid rice parents has not been well investigated, resulting in their limited use in hybrid rice blast-resistance breeding. In the present study, 88 elite indica hybrid rice parental lines were evaluated with 30 isolates of M. oryzae collected from the main planting area of indica hybrid rice in China and were characterized for the presence of 11 major resistance genes using molecular markers. The pathogenicity assays showed that four types of hybrid rice parent line showed some resistance to M. oryzae. However, the proportions of highly resistant lines and the mean resistance frequency(RF) varied among the four types, with resistance in decreasing order shown by three-line restorer lines, three-line maintainer lines, two-line sterile lines, and two-line restorer lines. All 88 hybrid rice parental lines carried more than one R gene, but none carried the R genes Pi1 and Pi2. Although Pid3 and Pi9 were present only in three-line restorer lines and Pigm only in three-line maintainer lines, the remaining six R genes(Pib, Pid2, Pi5, Pia, Pi54, and Pita) were present in the four types of hybrid rice parent with significantly different distribution frequencies. The correlation between R genes and resistance reactions was investigated. | 10 | Review on the applications and developments of drag reducing polymer in turbulent pipe flow显示文摘Drag reduction phenomenon in pipelines has received lots of attention during the past decades due to its potential engineering applications, especially in fluid transporting industries. Various methods to enhance drag reduction have been developed throughout the years and divided into two categories;non-additives method and additives method. Both categories have different types of methods, with different formulations and applications which will generally be discussed in this review. Among all the methods discussed, drag reduction using polymer additive is as one of the most enticing and desirable methods. It has been the subject of research in this field and has been studied extensively for quite some time. It is due to its ability to reduce drag up to 80% when added in minute concentrations. Reducing drag in the pipe will require less pumping power thus offering economic relieves to the industries. So, this paper will be focusing more on the use of polymer additives as drag reducing agent, the general formulations of the additives, major issues involving the use of drag reducing polymers, and the potential applications of it. However, despite the extensive works of drag reduction polymer, there are still no models that accurately explain the mechanism of drag reduction. More studies needed to be done to have a better understanding of the phenomenon. | 12 | KAI1 inhibits HGF-induced invasion of pancreatic cancer by sphingosine kinase activity显示文摘BACKGROUND:KAI1/CD82 has been reported to attenuate the process of metastases in a variety of tumors;however,its mechanism of action in invasion has not been fully elucidated. The present study aimed to investigate the importance of KAI1 in invasion and its correlation with activation of sphingosine kinase(SPK)in human pancreatic cancer PANC1 and Miapaca-2 cell lines. METHODS:The expression of KAI1 in PANC1 and Miapaca-2 cells,which was mediated by recombinant adenovirus(Ad-KAI1), was assessed by a flow cytometer and Western blotting.After successful infection was established,in vitro growth curve and invasive ability in Boyden Chamber assay were studied.The presence of KAI1 correlating with c-Met and SPK was detected by co-immunoprecipitation and[γ-32P]ATP incorporation. RESULTS:KAI1 genes had no significant effects on the curve representing cell growth.After infection with the KAI1 gene,decreased invasive ability in the Boyden Chamber assay was observed in PANC1 and Miapaca-2 cells that were induced by hepatocyte growth factor.Over-expression of KAI1 in the cells led to the deactivation of SPK and a decreased level of intracellular sphingosine-1-phosphate.No correlation was observed between c-Met and KAI1 during co-immunoprecipitation. | 14 | Cloning of UGT1A9 cDNA from liver tissues and its expression in CHL cells显示文摘AIM: To clone the cDNA of UGT1 A9 from a Chinese human liver and establish the Chinese hamster lung (CHL) cell line expressing human UGTI A9.METHODS: cDNA of UGT1A9 was transcripted from mRNA by reverse transcriptase-ploymerase chain reaction, and was cloned into the pGEM-T vector which was amplified in the host bacteric E. CoIl DH5α. The inserted fragment, verified by DNA sequencing, wes subcloned into the Hind III/Not I site of a mammalian expression vector pREP9 to construct the plasmid termed pREP9-UGT1A9. CHL cells were transfected with the resultant recombinants, pREP9- UGT1A9, and selected by G418 (400 mg. L-1) for one month. The surviving clone (CHL-UGT1A9) was harvested as a pool and sub-cultured in medium containing G418 to obtain samples for UGT1 A9 assays. The enzyme activity of CHL-UGT1A9 towards propranolol in S9 protein of the cell was determined by HPLC.RESULTS: The sequence of the cDNA segment cloned,which was 1666 bp in length, was identical to that released by Gene Bank (GenBank accession number: AF056188) in coding region. The recombinant constructed, pREP9- UGT1 A9, contains the entire coding region, along with 18bp of the 5' and 55 bp of the 3′ untranslated region of the UGT1A9 cDNA, respectively. The cell lines established expressed the protein of UGT1 A9, and the enzyme activity towards propranolol in S9 protein was found to be 101 ± 24pmol.min-1 .mg-1 protein (n = 3), but was not detectable in parental CHL cells.CONCLUSION: The cDNA of UGT1A9 was successfully cloned from a Chinese human liver and transfected into CHL ceils. | 19 | RETROVIRAL MEDIATED EFFICIENT TRANSFER ANDEXPRESSION OF MULTIPLE DRUG RESISTANCE GENE TO HUMAN LEUKEMIC CELLS显示文摘Objective: To investigate retroviral-mediated transfer and expression of human multidrug resistance (MDR) gene MDRI in leukemic cells. Methods: Human myeloid cells, K562 and NB4, were infected by MDR retrovirus from the producer PA317/HaMDR, and the resistant cells were selected with cytotoxic drug. The transfer and expression of MDRI gene was analyzed by using polymerasc chain reaction (PeR), now cytometry (FCM) and scmisolid colonies cultivation. Results: The resistant cells, K562/MDR and NB4/MDR, in which integration of the exogenous MDRI gene was confirmed by PCR analysis, displayed a typical MDR phenotype. The expression of MDR1 transgenc was detected on truncated as well as full-length transcripts. Moreover, the resistant cells were P-glycoprotein positive at 78.0% to 98.7% analyzed with FCM. The transduction efficieny in K562 cells was studied on suspension cultures and single-cell colonies. The transduction was more efficient in coculture system (67.9%-72.5%) than in supernatant system (33.1%-46.8%), while growth factors may improve the efficiency.
