1Hyperbaric oxygen preconditioning induces neuroprotection against ischemia in transient not permanent middle cerebral artery occlusion rat model显示文摘This study was designed to determine if repeated hyperbaric oxygen (HBO) exposure induces ischemic tolerance in focal cerebral ischemia Methods Sixty male SD rats were used in this study Thirty animals underwent transient middle cerebral artery occlusion (MCAO) and the other thirty permanent MCAO model The rats were randomly allocated to 3 sub-groups: control group (n=10), HBO-3 group (n=10), and HBO-5 group (n=10) The animals in HBO-3 and HBO-5 groups received 1*!hour hyperbaric oxygenation at 2 5 atmosphere absolute (ATA) in 100% oxygen every day for 3 and 5 days, respectively The animals in the control group received sham treatments 24*!hours after the last HBO, transient MCAO (120 min) and permanent MCAO were induced by introducing a 3-0 nylon monofilament suture through internal carotid artery based on the Koizumi technique The neurological outcome was evaluated until 24*!hours after reperfusion in transient MCAO rats and ischemia in permanent MCAO rats The infarct volume was then assessed by TTC staining Results In transient MCAO rats, the neurological outcome in both the HBO-3 and HBO-5 groups was better than that of the control group ( P <0 05 and 0 001) The infarct volume decreased from 171 5±113*!mm 3 to 40 6±49 9*!mm 3 ( P <0 05) in the HBO-3 group and 16 2±28 8*!mm 3 ( P <0 01) in the HBO-5 group There were no significant differences in neurological outcome and infarct volume among the three groups in permanent MCAO rats Conclusions The present study demonstrated that HBO preconditioning can induce ischemic tolerance in transient not permanent MCAO rats in a “dose-dependent'熊利泽 朱正华 董海龙 胡文能 候立朝 陈绍洋 2000Chinese Medical Journal2000,,9:60
2Transplantation of human hepatocytes into tolerized genetically immunocompetent rats显示文摘AIM To determine whether normal geneticallyirnmunocornpetent rodent hosts could bemanipulated to accept human hepatocytetransplants with long term survival withoutirnrnunosuppression.METHODS Tolerance towards humanhepatocytes was established by injection ofprimary human hepatocytes or Huh7 humanhepatoma cells into the peritoneal cavities offetal rats. Corresponding cells weresubsequently transplanted into newborn rats viaintrasplenic injection within 24 h after birth.RESULTS Mixed lymphocyte assays showedthat spleen cells from non-tolerized rats werestimulated to proliferate when exposed to humanhepatocytes, while cells from tolerized ratswere not. Injections made between 15 d and 17 dof gestation produced optimal tolerizaton.Transplanted human hepatocytes in rat liverswere visualized by immunohistochemicalstaining of human albumin. By dot blotting ofgenomic DNA in livers of tolerized rats 16 weeksafter hepatocyte transplantation, it was foundthat approximately 2.5 × 105 human hepatocytessurvived per rat liver. 3Toxicity of cadmium and its competition with mineral nutrients for uptake by plants:A review显示文摘Cadmium(Cd)is a toxic heavy metal occurring in the environment naturally and is also generated through various anthropogenic sources and acts as a pollutant.Human health is affected by Cd pollution in farmland soils because food is the main source of Cd intake in the non-smoking population.For crops,Cd toxicity may result from a disturbance in uptake and translocation of mineral nutrients and disturbance in plant metabolism,inhibiting plant growth and development.However,plants have Cd tolerance mechanisms,including restricted Cd uptake,decreased Cd root-to-shoot translocation,enhanced antioxidant enzyme activities,and increased production of phytochelatins.Furthermore,optimal supply of mineral nutrients is one of the strategies to alleviate the damaging effects of Cd on plants and to avoid its entry into the food chain.The emerging molecular knowledge contributes to understanding Cd uptake,translocation,and remobilization in plants.In this review,Cd toxicity and tolerance mechanisms,agricultural practices to minimize Cd accumulation,Cd competition with essential elements(calcium,copper,iron,zinc,and manganese),and genes associated with Cd uptake are discussed in detail,especially regarding how these mineral nutrients and genes play a role in decreasing Cd uptake and accumulation in crop plants.Shiyu QIN Hongen LIU Zhaojun NIE Zed RENGEL Wei GAO Chang LI Peng ZHAO 2020Pedosphere2020,30,2:20
4QTL analysis of the rice seedling cold tolerance in a double haploid population derived from anther culture of a hybrid between indica and japonica rice显示文摘A doubled haploid population, derived from anther culture of F hybrid between a typical indica cv. and a japonica cv. has been used to investigate the seedling cold tolerance (SCT) in growth cabinet. By dynamically analyzing every day’s survival percentages of the parents and DH lines under 7-d cold plus 9-d normal temperature condition, the guantitative trait loci (QTLs) for SCT have been mapped based on a molecular linkage map constructed from this population. The results show that two parents had significant differences in SCT and the segregation of SCT in DH lines was basically a continuous distribution with most serious injury on the 6th d of the cold treatment. A total of 4 QTLs for SCT have been identified on chromosomes 1, 2, 3 and 4 respectively. The additive effects of qSCT-1, qSCT-2 and qSCT-3 have been contributed by the japonica ev JX17, but that of qSCT-4 has been contributed by the indica cv ZYQ8. 5Piriformospora indica confers drought tolerance on Zea mays L.through enhancement of antioxidant activity and expression of drought-related genes显示文摘Drought stress is one of the most severe environmental constraints to plant growth and crop productivity. Plant growth is greatly affected by drought stress, and plants, to survive,adapt to this stress by invoking different pathways. Piriformospora indica, a root-colonizing endophytic fungus of Sebacinales, promotes plant growth and confers resistance to biotic and abiotic stresses, including drought stress, by affecting the physiological properties of the host plant. The fungus strongly colonizes the roots of maize(Zea mays L.) and promotes shoot and root growth under both normal growth conditions and drought stress. We used polyethylene glycol(PEG-6000) to mimic drought stress and found that root fresh and dry weight, leaf area, SPAD value, and leaf number were increased in P. indica-colonized plants.The antioxidative activities of catalases and superoxide dismutases were upregulated within 24h in the leaves of P. indica-colonized plants. Drought-related genes DREB2A, CBL1,ANAC072, and RD29A were upregulated in drought-stressed leaves of P. indica-colonized plants. Furthermore, after drought treatment, proline content increased, whereas accumulation of malondialdehyde(MDA), an indicator of membrane damage, decreased in P. indica-colonized maize. 7Curcumin attenuates Nrf2 signaling defect, oxidative stress in muscle and glucose intolerance in high fat diet-fed mice显示文摘AIM: To investigate the signaling mechanism of antioxidative action by curcumin and its impact on glucose disposal. METHODS: Male C57BL/6J mice were fed with either a normal diet (n = 10) or a high fat diet (HFD) (n = 20) to induce obesity and insulin resistance. After 16 wk, 10 HFD-fed mice were further treated with daily curcumin oral gavage at the dose of 50 mg/kg body weight (BW) (HFD + curcumin group). After 15 d of the curcumin supplementation, an intraperitoneal glucose tolerance test was performed. Fasting blood samples were also collected for insulin and glucose measurements. Insulin-sensitive tissues, including muscle, adipose tissue and the liver, were isolated for the assessments of malondialdehyde (MDA), reactive oxygen species (ROS)and nuclear factor erythroid-2-related factor-2 (Nrf2) signaling. RESULTS: We show here that in a HFD mouse model, short-term curcumin gavage attenuated glucose intolerance without affecting HFD-induced BW gain. Curcumin also attenuated HFD-induced elevations of MDA and ROS in the skeletal muscle, particularly in its mitochondrial fraction, but it had no such an effect in either adipose tissue or the liver of HFD-fed mice. Correspondingly, in skeletal muscle, the levels of total or nuclear content of Nrf2, as well as its downstream target, heme oxygenase-1, were reduced by HFD-feeding. Curcumin intervention dramatically reversed these defects in Nrf2 signaling. Further analysis of the relationship of oxidative stress with glucose level by a regression analysis showed a positive and significant correlation between the area under the curve of a glucose tolerance test with MDA levels either in muscle or muscular mitochondria. 9NaCl胁迫对甘薯试管苗生长和离子含量的影响(英文)显示文摘[Objective] This study aimed to investigate the salt-tolerance mechanism of sweet potato.[Method]Two sweet potato varieties of Xu 25-2(salt-tolerant cultivar)and Triumph 100(salt-sensitive cultivar)were treated by sodium chloride with the concentration of 0 mmol/L and 100 mmol/L.After 20 days,Na+ content and Na+/K+ ratio in the roots,shoots and leave were determined by the flame photometer,while dry weight and fresh weight of roots,shoots and leave in different varieties were also studied.[Result]The growth of two sweet potato varieties was inhibited under salt stress,so the plant became shorter,leaf and root became fewer,dry weight of roots and leave decreased,but seedlings of Xu 25-2 were inhibited slightly.Furthermore,Na+ content and Na+/K+ ratio in roots,shoots and leaves of two sweet potato varieties increased.Na+ content of salt-tolerant Xu 25-2 was low in roots,shoots and leaves,while Na+ content of salt-sensitive Triumph 100 was high in shoots and leave of seedlings,but the change range of Xu 25-2 was less than that of Triumph 100.[Conclusion]The lower Na+ content and Na+/K+ ratio in leaves under salt stress were the most important characteristics for salt-tolerance of sweet potato varieties.高叶 赵术珍 陈敏 宋晓征 王宝山 2008Agricultural Science & Technology2008,9,5:13
10Targeted therapies in breast cancer:New challenges to fight against resistance显示文摘Breast cancer is the most common type of cancer found in women and today represents a significant challenge to public health. With the latest breakthroughs in molecular biology and immunotherapy, very specific targeted therapies have been tailored to the specific pathophysiology of different types of breast cancers. These recent developments have contributed to a more efficient and specific treatment protocol in breast cancer patients. However, the main challenge to be further investigated still remains the emergence of therapeutic resistance mechanisms, which develop soon after the onset of therapy and need urgent attention and further elucidation. What are the recent emerging molecular resistance mechanisms in breast cancer targeted therapy and what are the best strategies to apply in order to circumvent this important obstacle? 11Impact of Nitrogen, Phosphorus and Potassium on Brown Planthopper and Tolerance of Its Host Rice Plants显示文摘The brown planthopper(BPH),Nilaparvata lugens(St?l),appeared as a devastating pest of rice in Asia. Experiments were conducted to study the effects of three nutrients,nitrogen(N),phosphorus(P) and potassium(K),on BPH and its host rice plants. Biochemical constituents of BPH and rice plants with varying nutrient levels at different growth stages,and changes in relative water content(RWC) of rice plants were determined in the laboratory. Feeding of BPH and the tolerance of rice plants to BPH with different nutrient levels were determined in the nethouse. Concentrations of N and P were found much higher in the BPH body than in its host rice plants,and this elemental mismatch is an inherent constraint on meeting nutritional requirements of BPH. Nitrogen was found as a more limiting element for BPH than other nutrients in rice plants. Application of N fertilizers to the rice plants increased the N concentrations both in rice plants and BPH while application of P and K fertilizers increased their concentrations in plant tissues only but not in BPH. Nitrogen application also increased the level of soluble proteins and decreased silicon content in rice plants,which resulted in increased feeding of BPH with sharp reduction of RWC in rice plants ultimately caused susceptible to the pest. P fertilization increased the concentration of P in rice plant tissues but not changed N,K,Si,free sugar and soluble protein contents,which indicated little importance of P to the feeding of BPH and tolerance of plant against BPH. 12Salt Tolerance in Rice: Focus on Mechanisms and Approaches显示文摘Salt tolerance is an important constrain for rice, which is generally categorized as a typical glycophyte. Soil salinity is one of the major constraints affecting rice production worldwide, especially in the coastal areas. Susceptibility or tolerance of rice plants to high salinity is a coordinated action of multiple stress responsive genes, which also interacts with other components of stress signal transduction pathways. Salt tolerant varieties can be produced by marker-assisted selection or genetic engineering by introducing salt-tolerance genes. In this review, we have updated on mechanisms and genes which can help in transferring of the salt tolerance into high-yielding rice varieties. 13Triptolide prolonged allogeneic islet graft survival in chemically induced and spontaneously diabetic mice without impairment of islet function显示文摘BACKGROUND:Triptolide(TPT)is a diterpenoid triepoxide extracted from the Chinese herb Tripterygium wilfordii Hook.F.It exhibits potent immunosuppressive and anti-inflammatory properties.This study was undertaken to investigate its effects on prolongation of islet allograft survival in rodents.Additionally,we investigated whether TPT would be toxic to islet function in vivo.METHODS:We transplanted BALB/c islets to either chemically induced diabetic C57BL/6 mice or spontaneously diabetic nonobese diabetic(NOD)mice.TPT was injected within 2 weeks or continuously,until rejection,in the two combinations.Then, we evaluated the toxicity of TPT on islet function by daily injection to naive BALB/c or diabetic BALB/c that was cured by syngeneic islet transplantation under the kidney capsule.Mice injected with cyclosporine A(CsA)or vehicle served as controls.Intraperitoneal glucose tolerance tests(IPGTTs)performed at 4 and 8 weeks in the na?ve BALB/c group,and at 2,4,6,and 8 weeks in the syngeneic transplanted group.RESULTS:The medium survival time of islets allograft from TPT treated C57BL/6 and NOD recipients were 28.5 days(range 24-30 days,n=10)and 33.0 days(range 15-47 days,n=6), respectively,and they were significantly different from those of the vehicle treated controls,which were 14.0 days(range 13-16 days,n=6)and 5.0 days(range 4-10 days,n=6),respectively(all P<0.0001).The IPGTT demonstrated that there was no difference between the TPT treated and vehicle treated groups, either in the normal or syngeneic transplanted islet BALB/c mice.However,CsA injection impaired islet function in both normal and syngeneic transplanted mice as early as 4 weeks.CONCLUSION:TPT prolonged islets allograft survival in a chemically induced diabetic or an autoimmune diabetic murine model without impairment of islet function.Xin, Ming-Jun Cui, Shi-Hua Liu, Shuang Sun, Hai-Chen Li, Fei Sun, Jia-Bang Luo, Bin 2010Hepatobiliary & Pancreatic Diseases International2010,9,3:11
14Physiological Mechanism of Salicylic Acid for Alleviation of Salt Stress in Rice显示文摘Soil salinity is one of the most important problems of crop production in estuarine and coastal zones. Improvement in salt tolerance of major food crops is an important way for the economic utilization of coastal zones. This study proved that the application of salicylic acid(SA) improved the growth and yield under salt stress conditions and investigated its physiological mechanisms for salt tolerance. The investigation on the effect of SA for salt tolerance during germination showed that the decreased rates of germination and growth(in terms of shoot and root lengths) by the salt stress were significantly increased by the SA application(SA + NaCl). The treatment of SA to the high and low saline soils enhanced the growth, yield and nutrient values of rice. The effects of SA on Na^+, K^+ and Cl~– ionic accumulation were traced under salt stress condition by inductively coupled plasma optical emission spectrometry and ion chromatography. It was revealed that the increased accumulation of Na^+ and Clˉ ions by the salt stress were reduced by SA application. An increased concentration of endogenous SA level was detected from the SA-treated rice varieties(ASD16 and BR26) by liquid chromatography electrospray Ionization-tandem mass spectrometry. The activities of antioxidant enzymes such as superoxide dismutase, catalase and peroxidase were increased by salt stress whereas decreased by the SA application. The study proved that the application of SA could alleviate the adverse effects of salt stress by the regulation of physiological mechanism in rice plants. 15Overexpression of GmDREB1 improves salt tolerance in transgenic wheat and leaf protein response to high salinity显示文摘The transcription factor dehydration-responsive element binding protein(DREB)is able to improve tolerance to abiotic stress in plants by regulating the expression of downstream genes involved in environmental stress resistance.The objectives of this study were to evaluate the salt tolerance of GmDREB1 transgenic wheat(Triticum aestivum L.)and to evaluate its physiological and protein responses to salt stress.Compared with the wild type,the transgenic lines overexpressing GmDREB1 showed longer coleoptiles and radicles and a greater radicle number at the germination stage,as well as greater root length,fresh weight,and tiller number per plant at the seedling stage.The yield-related traits of transgenic lines were also improved compared with the wild type,indicating enhanced salt tolerance in transgenic lines overexpressing GmDREB1.Proteomics analysis revealed that osmotic-and oxidative-stressrelated proteins were up-regulated in transgenic wheat leaves under salt stress conditions.Transgenic wheat had higher levels of proline and betaine and lower levels of malondialdehyde and relative electrolyte leakage than the wild type.These results suggest that GmDREB1 regulates the expression of osmotic-and oxidative-stress-related proteins that reduce the occurrence of cell injury caused by high salinity,thus improving the salt tolerance of transgenic wheat.Qiyan Jiang Zheng Hu Hui Zhang Youzhi Ma 2014The Crop Journal2014,2,Z1:11
16Effects of lactic acid bacteria isolated from fermented mustard on lowering cholesterol显示文摘Objective:To evaluate the ability of lactic acid bacteria(LAB)strains isolated from fermented mustard to lower the cholesterol in vitro.Methods:The ability of 50 LAB strains isolated from fermented mustard on lowering cholesterol in vitro was determined by modified o-phtshalaldehyde method.The LAB isolates were analyzed for their resistance to acid and bile salt.Strains with lowering cholesterol activity,were determined adherence to Caco-2 cells.Results:Strain B0007,B0006 and B0022 assimilated more cholesterol than BCRC10474 and BCRC17010.The isolated strains showed tolerance to pH 3.0 for 3h despite variations in the degree of viability and bile-tolerant strains,with more than 10~s CFU/mL after incubation for 24 h at 1%oxigall in MRS.In addition,strain B0007 and B0022 identified as Lactobacillus plantarum with 16S rDNA sequences were able to adhere to the Caco-2 cell lines.Conclusions:These strains B0007 and B0022 may be potential functional sources for cholesterollowering activities as well as adhering to Caco-2 cell lines.Shu Chen Wang Chen Kai Chang Shu Chang Chan Jiunn Shiuh Shieh Chih Kwang Chiu Pin-Der Duh 2014Asian Pacific Journal of Tropical Biomedicine2014,4,7:10
17Immunological inhibition of transplanted liver allografts by adeno-associated virus vector encoding CTLA4Ig in rats显示文摘BACKGROUND: Blockade interaction between CD28 and B7 with CTLA4Ig has been shown to induce experimental transplantation tolerance. In order to prolong the inhibitory effect of CTLA4Ig, a recombinant adeno- associated virus vector pSNAV expressing CTLA4Ig was constructed, and its effects on transplanted liver allografts were investigated. METHODS: The pSNAV-CTLA4Ig construct was infused into partial liver allografts of rats via the portal vein during transplantation. CTLA4Ig expression in the transplanted livers was detected with reverse transcriptase- polymerase chain reaction (RT-PCR) analysis and immunohistochemistry. Furthermore, real-time quantita- tive PCR was used to measure the expression of IL-2, IFN-γ, IL-4 and IL-10 in the allografts. RESULTS: The expression of CTLA4Ig in the partial allograft was detected successfully and pSNAV-CTLA4Ig improved the survival rate of rats after liver transplantation. Agarose gel analysis of RT-PCR products indicated the presence of CTLA4Ig in the pSNAV-CTLA4Ig treatment group. Cytokines expressed in allografts on day 7 after orthotopic liver transplantation showed that IL-2, IFN-γ, IL-4 and IL-10 mRNA levels decreased in transplant recipients treated with pSNAV-CTLA4Ig compared with those treated with pSNAV-LacZ (1.62±0.09, 1.52±0.11, 1.50± 0.07 and 1.43±0.07 versus 1.29±0.09, 1.32±0.07, 1.34±0.06 and 1.35±0.04, respectively).CONCLUSIONS: pSNAV-CTLA4Ig effectively expressed CTLA4Ig in liver allografts. CTLA4Ig improved the pathological findings after liver transplantation. CTLA4Ig induced immune tolerance of liver transplantation, and the mechanism involved induced alteration of Th1 and Th2 cytokine transcripts. 18Effect of down-regulation of voltage-gated sodium channel Nav1.7 on activation of astrocytes and microglia in DRG in rats with cancer pain显示文摘Objective:To evaluate the effect of down-regulation of Nav1.7 on the activation of astrocytes and microglia in DRG of rats with cancer pain,and explore the transmission of the nociceptive information.Methods:Lentiviral vector harboring RNAi sequence targeting the Navl.7 gene was constructed,and Walker 256 breast cancer cell and morphine was injected to build the bone cancer pain model and morphine tolerance model in rats.Lentiviral vector was injected.Rats in each model were divided into 4 groups:model group,PBS group,vehicle group and LV-Nav1.7 group.The expression levels of GFAP and OX42 in dorsal root ganglia(DRG) were measured.Results:After the animal model was built,the level of Navl.7,GFAP and OX42 was improved obviously with the time prolonged,which was statistically significant(P<0.05).The expression level of GFAP and OX42 in the DRG in the LV-Navl.7 group declined obviously compared to the model group,PBS group and vehicle group(P<0.05).Conclusions:Intrathecal injection of Navl.7 shRNA lentiviral vector can reduce the expression of Nav1.7and inhibit the activation of astrocytes and microglia in DRG.The effort is also effective in morphine tolerance bone cancer pain model rats.Jun Pan Xiang-Jin Lin Zhi-Heng Ling You-Zhi Cai 2015Asian Pacific Journal of Tropical Medicine2015,8,5:9
19Anti-diabetic property of ethanolic extract of Andrographis paniculata in streptozotocin-diabetic rats显示文摘AIM: To investigate the anti-diabetic effect of a crude ethanolic extract of Andrographis paniculata in normal and streptozotocin ( STZ )-induced diabetic rats. METHODS & RESULTS: Oral administration of the extract at different doses (0.1, 0.2, and 0.4 g/body weight) significantly reduced the fasting serum glucose level in STZ-diabetic rats compared to the vehicle (distilled water), but not in normal rats. This effect was dose-dependent. A similar result was seen with met-formin (0.5 g/body weight). In the glucose tolerance test, an oral administration of the extract at the same doses suppressed the elevated glucose level in normal and diabetic rats, as did metformin. The effects were also dose-respondent. In the long-term experiment, the extract (0.4 g/body weight), metformin (0.5 g/body weight), and vehicle were given twice daily to diabetic rats for 14 d. 20Clinical operational tolerance in liver transplantation:state-of-the-art perspective and future prospects显示文摘BACKGROUND:Liver transplantation is the definite treatment for end-stage liver diseases with satisfactory results.However,untoward effects of life-long immunosuppression prevent the development of alternative strategies to achieve better longterm outcome.Achieving clinical operational tolerance is the ultimate goal.DATA SOURCES:A PubMed and Google Scholar search using terms:'immune tolerance','liver transplantation','clinical trial','operational tolerance' and 'immunosuppression withdrawal' was performed,and relevant articles published in English in the past decade were reviewed.Full-text publications relevant to the field were selected and relevant articles from reference lists were also included.Priority was given to those articles which are relevant to the review.RESULTS:Because of the inherent tolerogenic property,around 20%-30% of liver transplantation recipients develop spontaneous operational tolerance after immunosuppression withdrawal,and the percentage may be even higher in pediatric living donor liver transplantation recipients.Several natural killer and γδT cell related markers have been identified to be associated with the tolerant state in liver transplantation patients.Despite the progress,clinical operational tolerance is still rare in liver transplantation.Reprogramming the recipient immune system by creating chimerism and regulatory cell therapies is among newer promising means to achieve clinical liver transplantation tolerance in the future.CONCLUSION:Although clinical operational tolerance is still rare in liver transplantation recipients,ongoing basic research and collaborative clinical trials may help to decipher the mystery of transplantation tolerance and extend the potential benefits of drug withdrawal to an increasing number of patients in a more predictable fashion.Xi-Qiang Liu Zhi-Qiu Hu Yao-Fei Pei Ran Tao 2013Hepatobiliary & Pancreatic Diseases International2013,12,1:9
