| 2 | In Vitro Probiotic Effect of Pseudostellaria heterophylla Fibrous Roots on Feed Bacillus subtilis and Preliminary Study on Its Solid Fermentation显示文摘Traditional Chinese medicine Pseudostellaria heterophylla fibrous roots have been widely studied and applied in non-antibiotic clinical breeding of livestock and poultry. In vitro probiotic effect of P. heterophylla fibrous roots extract on feed probiotics Bacillus subtilis BD-K010 was studied, and the feasibility of solid fermentation of P. heterophylla fibrous roots was preliminarily evaluated. For in vitro probiotic effect, the increased concentration of P. heterophylla fibrous roots extract dependently increased the total biomass of B. subtilis BD-K010;1.0% P. heterophylla fibrous roots extract received the best effect, and the final p H of 1.0% experimental group was closer to neutral. Meantime, B. subtilis BD-K010 with optimum concentration of P. heterophylla fibrous roots showed significantly higher in vitro antibacterial effect than the control group(P<0.01),and the antibacterial effects on Escherichia coli and Staphylococcus aureus were improved by 51.99% and 63.16%, but it was ineffective to Salmonella. For solid fermentation, the profile of substrate complex and appendage flocculent structure on substrate surface at the end of fermenta-tion in experimental groups added with B. subtilis BD-K010 and cellulase plus BD-K010 were more complex;the live bacteria number, polysaccharide content and saponin content at fermentation end-point in two experimental groups were extremely higher than those in the control group(P<0.01).P. heterophylla fibrous roots extract had good in vitro probiotic effect on B. subtilis BD-K010 and promoted its antibacterial effect, and it is feasible to use probiotics for solid fermentation of P. heterophylla fibrous roots and to improve effective components. | 3 | Comparison of DPPH Scavenging Ability of Flavonoid and Polysaccharide from Dandelion ( Taraxacum mongolicum)显示文摘The paper was to study DPPH(1,1-diphenyl-2-trinitrobenzene hydrazine) scavenging ability of different concentrations of flavonoid and polysaccharide from dandelion. Flavonoids were extracted from dandelion using alcohol precipitation ultrasonic extraction method and polysaccharide was extracted using water ultrasonic extraction method. DPPH free radical scavenging ability of flavonoid and polysaccharide was determined using ultraviolet spectrophotometer method.The difference in DPPH free radical scavenging ability between flavonoids and polysaccharide was analyzed statistically. The results showed that DPPH scavenging ability was the best when the concentration of flavonoid was 1. 6 mg/m L; the scavenge rate was up to 96. 11% and IC50(median inhibitory concentration of inhibitors) was 0. 38 mg/m L. DPPH scavenging ability was the best when the concentration of polysaccharide was 1. 4 mg/m L; the scavenge rate was 89. 33%and IC50 was 0. 51 mg/m L. Statistical results showed that there were significant differences in DPPH free radical scavenging ability between dandelion flavonoid and polysaccharide( P < 0. 05). | 5 | Analysis on Nutrient Compositions and Nutritional Quality of Lota lota( Linnaeus) Muscle显示文摘In order to evaluate the nutritional quality of Lota lota(Linnaeus) muscle,the common nutrient components,amino acid composition and fatty acid composition in muscle of wild Lota lota(captured in Hailang reaches of Mudan River) were determined by routine methods. The results showed that the contents of moisture,crude protein,crude fat and ash were 81. 82%,15. 93%,0. 44% and 1. 06% in fresh muscle of Lota lota,respectively. Totally 17 amino acids,including seven kinds of essential amino acids(EAA),were detected in dry muscle of Lota lota(Linnaeus),accounting for 72. 43% of total dry muscle. The content of EAA was 29. 82%,accounting for 41. 17% of TAA,meeting the amino acid composition of high-quality protein standard required by Food and Agriculture Organization /Word Health Organization(FAO /WHO). There were four delicious amino acids(DAA) in dry muscle,accounting for 28. 34% of total dry weight.The percentage of DAA to TAA was 39. 13% in dry muscle. The contents of C20:5n- 3(EPA) and C22:6n- 3(DHA) in dry muscle were 8. 65% and 20. 32%respectively,which were significantly higher than that of the artificially cultured fish species,such as Oxyeleotris marmorata(EPA:2. 24%,DHA:4. 74%),Spinibarbus sinensis(EPA:0. 87%,DHA:3. 08%) and Glossogobius giuris(EPA: 3. 94%,DHA: 8. 10%). Based on the amino acids score(AAS) and chemical score(CS),the first limited amino acid in Lota lota(Linnaeus) was sulfur-containing amino acid(methionine + cysteine). The essential amino acids index(EAAI) of Lota lota(Linnaeus) was 62.58. It is concluded that Lota lota(Linnaeus) is abundant in amino acids and high-unsaturated fatty acids,and rational in composition of amino acids. | 7 | Sources and Accumulation Risk of Heavy Metal in Major Animal Manure显示文摘Feed and corresponding manure samples were collected from 27 scale pig farms and 7 scale chicken farms in Jiaxing City,Zhejiang Province,and their heavy metal contents were analyzed. The weighted average values of Cr,Cu,Zn,Pb and As in pig feed were 4. 6,123. 4,370. 7,0. 69 and 6. 9 mg/kg,respectively. However,referred to Safe Use Specification of Feed Additives and MOA Announcement No. 1224,the exceeding standards rates of Cr,Cu,Zn,Pb and As in pig feed were 5. 36%,62. 5%,19. 64%,3. 57% and 37. 5%,respectively. The sequences of heavy metal content in pig feed successively were piglet > growing pig > big pig > sow,but the exceeding standard rates of heavy metal in feed were big pig > sow > growing pig > piglet,and the exceeding standard rate of big pig feed was more than 95%. The exceeding standard rates of Cu,Zn and As in chicken feed were 46. 7%,11. 1% and 33. 3% respectively. Based on Organic Fertilizer( NY525-2012) and Sludge Agricultural Pollution Control Standard( GB4284-84),Cu,Zn and As in pig manure and chicken manure exceeded the standard; the exceeding standard rates of pig manure were 71. 9%,78. 9% and 42. 1%,while that of chicken manure were 12. 5%,25. 0% and 44. 4%,respectively. The exceeding standard rates of Cu and Zn in piglet manure reached 100%. There were very significant positive correlations in Cu,Zn and As between pig feed and corresponding manure,or Hg,Cu and As between chicken feed and corresponding manure,and the correlation coefficients r^2 were greater than 0. 8. Besides,the correlation coefficient r^2 of Pb between pig feed and corresponding manure was above 0. 6. | 9 | Optimization on Production Key Factors of Microbial Fermented Feed and Analysis on Composition Variation During Fermentation显示文摘[Objective] In order to study the production key factors of microbial fermented feed and the composition variation during fermentation. [Methods]Water,temperature,energy and protein were selected to study the effects on microbial fermented feed. And the fermentation conditions were optimized. Changes on nutrient composition during fermentation were determined and analyzed. [Result] Test results showed that feed formula with high sugar content,low protein,34%-36% water content,high fermentation temperature was more conducive to the production and actual production needs of fermentation strains. During the fermentation,strains showed interactions,test tended to be completed at the 144^(th)h after the fermentation,and number of lactic acid bacteria reached the peak of 1. 86 × 10~9 strain/g,contents of lactic acid could even reach 0. 89%. As time prolonged,contents of total energy,fiber and isothiocyanate gradually reduced,while contents of water and protein increased slightly. | 11 | Ultrastructure Observation of Cysticercus Pisiformis and Its Function Analysis显示文摘[Objective]The aim of the study was to observe the ultrastructure of Cysticercus pisiformis under different state. [Method] The Cysticercus pisiformis were obtained in the procedure of slaughter inspection form meat-packing plant. The Cysticercus pisiformis was taken out by cutting cyst directly, or cultured with physiologic saline containing 10% bile until evagination. Then it was observed and recorded by scanning electron microscopy. [Result]Observed from top of scolex of Cysticercus pisiformis locate in cyst, the rostellum looked like an umbrella. The cutaneous muscular columns with hamula covered the front end of scolex. Viewed from the side, a row of antlers-liking hamula adhered to the scolex. Four suckers were looked like more width and body segments less fime, in which there were lots of wrinkles in their surface respectively. The Cysticercus pisiformis cultured was divided obviously into scolex, neck, body segment and zone of cyst. When cutaneous muscular column of rostellum contracted, the antler-liking hamula extended to periphery that the ring-liking contraction appeared in neck and the lengthways contraction in body segments. Bacterial genome DNA of separated L. acidophilus in fermented sample was taken as template,and L. acidophilus in fermented material was conducted the quantitative determination by real-time quantitative PCR( RT-PCR). Analysis on RT-PCR results shown that contents of L. acidophilus in the test sample reached 1. 5 billion CFU / g. The trial lasted 50 d.[Result] The final weight, daily gain and feed intake of experimental group were significantly higher than those of control group(P<0.05), but the feed/gain ratio of experimental group was markedly lower than that of control group(P<0.05). | 16 | Effects of Microencapsulated Compound Acidifier on Acidity and Development of Gastrointestinal Tract in Weaning Piglets显示文摘The effects of different types of compound acidifiers (encapsulated and non-encapsulated) in maize-soybean basic diets on acidity and development of the gastrointestinal tract in weaning piglets were investigated in this study. 64 28-day-old weaned Landrace×Yorkshire hybrid piglets with average weight of (7.00±0.10) kg were selected and grouped into four treatments with four pigs (half boars and half sows) in each repeat of four repeats in each treatment based on single-factor test design principles, and the pre-test period was 3 days but the test period was 35 days. The results showed that compared with acid-free diet group, encapsulated compound acidifier could reduce pH of stomach and intestinal in weaning piglets significantly (P<0.01), while Test group 1 could also increase the relative weight of stomach and intestinal in piglets significantly (P<0.05). Compared with non-encapsulated acidifier, encapsulated compound acidifier could greatly decrease pH in jejunum and ileum of weaning piglets (P<0.05) or relative weight of stomach in piglets (P<0.05), while Test group 1 could also enhance the relative weight of intestinal in piglets significantly (P<0.01). In addition, encapsulated compound acidifiers significantly increased the ratio between the villus height and crypt depth of jejunum (P<0.01). | 19 | Protective Effect of Bacillus subtilis Peptidoglycan (PG) on β-conglycinin-induced Intestinal Epithelial Cells Damage of Juvenile Carp (Cyprinus carpio)显示文摘[Objective] The paper was to investigate the protective effects of different concentrations of Bacillus subtilis peptidoglycan(PG) on β-conglycinin-induced inflammatory injury in intestinal epithelial cells of juvenile carp(Cyprinus carpio).[Method] In 24-cell microplates, the intestinal epithelial cells(IECs) of juvenile carp were primarily cultured for 72 h at 26°C and 6% CO2, and then the IECs were randomly divided into6 groups with 4 replicates per group. One of the six groups was set as negative control group, and the other groups were all supplemented with 1.0 mg/mL β-conglycinin in culture medium to establish inflammatory injury. At 24 h post induction, the culture media were changed into B. subtilis PG culture media with the concentrations of 0(positive control group), 0.15, 0.30, 0.45 and 0.60 mg/mL, respectively. The samples were collected to measure the antioxidant and anti-inflammatory indices at 12, 24 and 36 h post culture.[Result]β-conglycinin exposure significantly decreased the activity of ASA, AHR, SOD, CAT, GPx, and increased the PC content and the mRNA expression of inflammatory cytokines(IL-1β, IL-8, TNF-α1,IL-10 and TGF-β). At 12, 24 and 36 h post PG treatment, the activities of ASA, AHR, SOD, CAT, GPx and the content of PC in cells decreased in a dose-dependent manner;the mRNA levels of IL-1β, IL-8 and TNF-α1 were down-regulated and those of IL-10 and TGF-β were up-regulated.[Conclusion] Different concentrations of B. subtilis PG could protect IECs oxidative damage induced by β-conglycinin and improve the antioxidant capacity of IECs. | 20 | Isolation and Identification of High-Quality Lactic Acid Bacteria in Forage Corn显示文摘[Objective] To screen suitable lactic acid bacterium strains from forage corn which can be used as silage additives.[Method] The lactic acid bacterium strains were isolated by inoculation on MRS solid medium containing calcium carbonate,and they were preliminarily identified through morphological,physiological and biochemical experiments.The acid production efficiency was determined.Twelve strains having strong acid-production ability were selected,and their salt tolerance and acid tolerance were detected.The sequences of their 16 S rDNA were also analyzed.[Result] A total of 44 lactic acid bacterium strains were isolated from the forage corn.As evidenced by the physiological and biochemical experiments,the twelve strains having strong acid-production ability belonged to Leuconostoc,Lactobacillus and Enterococcus,respectively,and they had strong salt tolerance and acid tolerance.According to the sequences of 16 S rDNA,A4,B9,B11,B12 and B14 were Lactobacillus plantarum;A1,A2,A7,A11 and B8 were Leuconostoc mesenteroides dextran subspecies;and A8 and A9 were Enterococcus hirae.[Conclusion] The lactic acid bacterium strains with strong acid-production ability isolated from forage corn can be developed into silage additives. | Mairemunisa.Aimaier Abudukeyoumu.Maimaiti Wusiman.Yimiti | 2011 | Animal Husbandry and Feed Science2011,3,1: | 3 |
