1美国优质紫花苜蓿在宁夏中部干旱带的适应性研究显示文摘以宁夏盐池县本地苜蓿XM及宁夏益科农业科技有限公司引进的15个美国紫花苜蓿品种为研究对象,综合考虑生育期、株高、密度、产草量、茎叶比、越冬率等指标,开展苜蓿引种适应性研究.结果表明,以本地种XM为对照,15个外引种紫花苜蓿在生长发育特性上存在显著差异.其中,以品种ZHM,E3006,D4,D2表现较优,其返青早,青绿期长,鲜草产量分别是对照的1.52,1.45,1.28,1.25倍,干草产量分别是对照的1.46,1.25,1.22,1.19倍,且抗旱性强,品质优良,适宜在宁夏盐池县等中部干旱带大面积推广应用.高婷 张晓刚 纪立东 Orson Boyce Stanford A.Young 朱建宁 彭文栋 2009宁夏大学学报(自然科学版)2009,30,3:6
2Enhancer variants reveal a conserved transcription factor network governed by PU.1 during osteoclast differentiation显示文摘Genome-wide association studies(GWASs) have been instrumental in understanding complex phenotypic traits. However, they have rarely been used to understand lineage-specific pathways and functions that contribute to the trait. In this study, by integrating lineage-specific enhancers from mesenchymal and myeloid compartments with bone mineral density loci, we were able to segregate osteoblast-and osteoclast(OC)-specific functions. Specifically, in OCs, a PU.1-dependent transcription factor(TF)network was revealed. Deletion of PU.1 in OCs in mice resulted in severe osteopetrosis. Functional genomic analysis indicated PU.1 and MITF orchestrated a TF network essential for OC differentiation. Several of these TFs were regulated by cooperative binding of PU.1 with BRD4 to form superenhancers. Further, PU.1 is essential for conformational changes in the superenhancer region of Nfatc1. 4Treatment with a JAK1/2 inhibitor ameliorates murine autoimmune cholangitis induced by IFN overexpression显示文摘The interferon(IFN)signaling pathways are major immunological checkpoints with clinical significance in the pathogenesis of autoimmunity.We have generated a unique murine model named ARE-Del,with chronic overexpression of IFNγ,by altering IFNγmetabolism.Importantly,these mice develop an immunologic and clinical profile similar to patients with primary biliary cholangitis,including high titers of autoantibodies and portal inflammation.We hypothesized that the downregulation of IFN signaling pathways with a JAK1/2 inhibitor would inhibit the development and progression of cholangitis.To study this hypothesis,ARE-Del^(+/−)mice were treated with the JAK1/2 inhibitor ruxolitinib and serially studied.JAK inhibition resulted in a significant reduction in portal inflammation and bile duct damage,associated with a significant reduction in splenic and hepatic CD4^(+)T cells and CD8^(+)T cells.Functionally,ruxolitinib inhibited the secretion of the proinflammatory cytokines IFNγand TNF from splenic CD4^(+)T cells.Additionally,ruxolitinib treatment also decreased the frequencies of germinal center B(GC B)cells and T follicular helper(Tfh)cells and led to lower serological AMA levels.Of note,liver and peritoneal macrophages were sharply decreased and polarized from M1 to M2 with a higher level of IRF4 expression after ruxolitinib treatment.Mechanistically,ruxolitinib inhibited the secretion of IL-6,TNF and MCP1 and the expression of STAT1 but promoted the expression of STAT6 in macrophages in vitro,indicating that M1 macrophage polarization to M2 occurred through activation of the STAT6-IRF4 pathway.Our data highlight the significance,both immunologically and clinically,of the JAK/STAT signaling pathway in autoimmune cholangitis.Tihong Shao Patrick S.C.Leung Weici Zhang Koichi Tsuneyama William M.Ridgway Howard A.Young Zongwen Shuai Aftab A.Ansari M.Eric Gershwi 2022Cellular & Molecular Immunology2022,19,10:1

14宁夏中部风沙区草场表层土壤储水分布特征研究显示文摘为了从涵养水分角度验证牧草引种适应性效果,试验采用实时定位、动态监测方法对宁夏中部风沙区天然草场、美国优质牧草引种试验区0-40 cm层次土壤储水量分布特征进行对比探讨。结果表明:宁夏中部风沙区天然草场土壤水分与大气降水关系密切,土壤储水量以5—7月份为显著失墒期,8—9月份为明显增墒期,10—11月份为缓慢退墒期,11月份—翌年4月份为保墒期。美国优质牧草引种改良区年内及翌年土壤储水量均低于未改良区,而引种3年的牧草地土壤储水量显著高于未改良区,说明宁夏中部风沙区优质牧草需要持续一定年限的引种示范,才能使外引种真正演变为本地适应性优良的土生种,年限至少在3年以上。高婷 张晓刚 纪立东 Orson Boyce Stanford A.Young 朱建宁 彭文栋 2011黑龙江畜牧兽医2011,,4:1

16A Novel Atlas-Based Strategy for Understanding Cardiac Dysfunction in Patients with Congenital Heart Disease显示文摘Tetralogy of Fallot(TOF)is the most common form of cyanotic congenital heart disease.Infants diagnosed with TOF require surgical interventions to survive into adulthood.However,as a result of postoperative structural malformations and long-term ventricular remodeling,further interventions are often required later in life.To help identify those at risk of disease progression,serial cardiac magnetic resonance(CMR)imaging is used to monitor these patients.However,most of the detailed information on cardiac shape and biomechanics contained in these large four-dimensional(4D)data sets goes unused in clinical practice for lack of efficient and comprehensive quantitative analysis tools.While current global metrics of cardiac size and function,such as indexed ventricular mass and volumes,can identify patients at risk of further complications,they are not adequate to explain the underlying mechanisms causing the postoperative malfunctions,and help cardiologists plan optimal personalized treatments.We are proposing a novel approach that uses 4D ventricular shape models derived from CMR imaging exams to generate statistical atlases of ventricular shape and finite-element models of ventricular biomechanics to identify specific features of cardiac shape and biomechanical properties that explain variations in ventricular function.This study has the potential to discover novel biomarkers that precede adverse ventricular remodeling and dysfunction.Sara Salehyar Nickolas Forsch Kathleen Gilbert Alistair A.Young James C.Perry Sanjeet Hegde Jeffrey H.Omens Andrew D.McCulloch 2019Molecular & Cellular Biomechanics2019,16,3:0
