1Gallbladder cancer epidemiology, pathogenesis and molecular genetics: Recent update显示文摘Gallbladder cancer is a malignancy of biliary tract which is infrequent in developed countries but common in some specific geographical regions of developing countries. Late diagnosis and deprived prognosis are major problems for treatment of gallbladder carcinoma. The dramatic associations of this orphan cancer with various genetic and environmental factors are responsible for its poorly defined pathogenesis. An understanding to the relationship between epidemiology, molecular genetics and pathogenesis of gallbladder cancer can add new insights to its undetermined pathophysiology. Present review article provides a recent update regarding epidemiology, pathogenesis, and molecular genetics of gallbladder cancer. We systematically reviewed published literature on gallbladder cancer from online search engine Pub Med(http://www.ncbi.nlm.nih.gov/pubmed). Various keywords used for retrieval of articles were Gallbladder, cancer Epidemiology, molecular genetics and bullion operators like AND, OR, NOT. Cross references were manually searched from various online search engines(http://www.ncbi.nlm.nih.gov/pubmed,https://scholar.google.co.in/, http://www.medline.com/home.jsp). 2Efficacy of fermented milk and whey proteins in Helicobacter pylori eradication:A review显示文摘Helicobacter pylori(H.pylori)eradication is considered a necessary step in the management of peptic ulcer disease,chronic gastritis,gastric adenocarcinoma and mucosa associated lymphoid tissue lymphoma.Standard triple therapy eradication regimens are inconvenient and achieve unpredictable and often poor results.Eradication rates are decreasing over time with increase in antibiotic resistance.Fermented milk and several of its component whey proteins have emerged as candidates for complementary therapy.In this context the current review seeks to summarize the current evidence available on their role in H.pylori eradication.Pertinent narrative/systematic reviews,clinical trials and laboratory studies on individual components including fermented milk,yogurt,whey proteins,lactoferrin,α-lactalbumin(α-LA),glycomacropeptide and immunoglobulin were comprehensively searched and retrieved from Medline,Embase,Scopus,Cochrane Controlled Trials Register and abstracts/proceedings of conferences up to May 2013.A preponderance of the evidence available on fermented milk-based probiotic preparations and bovine lactoferrin suggests a beneficial effect in Helicobacter eradication.Evidence forα-LA and immunoglobulins is promising while that for glycomacropeptide is preliminary and requires substantiation.The magnitude of the potential benefit documented so far is small and the precise clinical settings are ill defined.This restricts the potential use of this group as a complementary therapy in a nutraceutical setting hinging on better patient acceptability/compliance.Further work is necessary to identify the optimal substrate,fermentation process,dose and the ideal clinical setting(prevention/treatment,first line therapy/recurrence,symptomatic/asymptomatic,gastritis/ulcer diseases etc.).The potential of this group in high antibiotic resistance or treatment failure settings presents interesting possibilities and deserves further exploration.Aarti Sachdeva Swapnil Rawat Jitender Nagpal 2014World Journal of Gastroenterology2014,20,3:7
3Neuroprotective Effect of Chrysophanol as a PI3K/AKT/mTOR Signaling Inhibitor in an Experimental Model of Autologous Blood-induced Intracerebral Hemorrhage显示文摘Objective Intracerebral hemorrhage(ICH)refers to predominant,sporadic,and non-traumatic bleeding in the brain parenchyma.The PI3K/AKT/mTOR signaling pathway is an important signal transduction pathway regulated by enzyme-linked receptors and has many biological functions in mammals.It plays a key role in neuronal metabolism,gene expression regulation,and tissue homeostasis in the healthy and diseased brain.Methods In the present study,the role of the PI3K/AKT/mTOR pathway inhibitor chrysophanol(CPH)(10 mg/kg and 20 mg/kg,orally)in the improvement of ICH-associated neurological defects in rats was investigated.Autologous blood(20µL/5 min/unilateral/intracerebroventricular)mimics ICH-like defects involving cellular and molecular dysfunction and neurotransmitter imbalance.The current study also included various behavioral assessments to examine cognition,memory,and motor and neuromuscular coordination.The protein expression levels of PI3K,AKT,and mTOR as well as myelin basic protein and apoptotic markers,such as Bax,Bcl-2,and caspase-3,were examined using ELISA kits.Furthermore,the levels of various neuroinflammatory cytokines and oxidative stress markers were assessed.Additionally,the neurological severity score,brain water content,gross brain pathology,and hematoma size were used to indicate neurological function and brain edema.Results CPH was found to be neuroprotective by restoring neurobehavioral alterations and significantly reducing the elevated PI3K,AKT,and mTOR protein levels,and modulating the apoptotic markers such as Bax,Bcl-2,and caspase-3 in rat brain homogenate.CPH substantially reduced the inflammatory cytokines like interleukin(IL)-1β,IL-6,and tumor necrosis factor-α.CPH administration restored the neurotransmitters GABA,glutamate,acetylcholine,dopamine,and various oxidative stress markers.Conclusion Our results show that CPH may be a promising therapeutic approach for overcoming neuronal damage caused by the overexpression of the PI3K/AKT/mTOR signaling pathway in ICH-induced neurological dysfunctions in rats.Kuldeep Singh JADAUN Sidharth MEHAN Aarti SHARMA Ehraz Mehmood SIDDIQUI Sumit KUMAR NaifALSUHAYMI 2022Current Medical Science2022,42,2:5
4Down syndrome and the molecular pathogenesis resulting from trisomy of human chromosome 21显示文摘Chromosome copy number aberrations, anueploidies, are common in the human population but generally lethal. However, trisomy of human chromosome 21 is compatible with life and people born with this form of aneuploidy manifest the features of Down syndrome, named after Langdon Down who was a 19th century British physician who first described a group of people with this disorder. Down syndrome includes learning and memory deficits in all cases, as well as many other features which vary in penetrance and expressivity in different people. While Down syndrome clearly has a genetic cause - the extra dose of genes on chromosome 21-we do not know which genes are important for which aspects of the syndrome, which biochemical pathways are disrupted, or, generally how design therapies to ameliorate the effects of these disruptions. Recently, with new insights gained from studying mouse models of Down syndrome, specific genes and pathways are being shown to be involved in the pathogenesis of the disorder. 5Anti-tubercular peptides:A quest of future therapeutic weapon to combat tuberculosis显示文摘Tuberculosis(TB) is a symbolic menace to mankind,infecting almost one third of the world's populace and causing over a million mortalities annually.Mycobacterium tuberculosis(M.tuberculosis) is the key pathogen of TB that invades and replicates inside the host's macrophage.With the emerging dilemma of multi-drug resistant tuberculosis(MDR-TB) and extensivelydrug resistant tuberculosis(XDR-TB),the exigency for developing new TB drugs is an obligation now for worldwide researchers.Among the propitious antimycobacterial agents examined in last few decades,anti-tubercular peptides have been substantiated to be persuasive with multiple advantages such as low immunogenicity,selective affinity to bacterial negatively charged cell envelopes and most importantly divergent mechanisms of action.In this review,we epitomized the current advances in the anti-tubercular peptides,focusing the sources and highlighting the mycobactericidal mechanisms of promising peptides.The review investigates the current anti-tubercular peptides exploited not only from human immune cells,human non-immune cells,bacteria and fungi but also from venoms,cyanobacteria,bacteriophages and several other unplumbed sources.The anti-tubercular peptides of those origins are also known to have unique second non-membrane targets within M.tuberculosis.The present context also describes the several cases that manifested the severe side effects of extant antiTB drugs.The downfall,failure to reach clinical trial phases,inept to MDR- or XDR-TB and severe complications of the currently available anti-tubercular drugs accentuate the imperative necessity to develop efficacious drugs from adequate anti-tubercular peptides.Keeping in view of the emerging trends of drug resistant M.tuberculosis globally and unexampled mycobactericidal characteristics of peptides,the anti-tubercular peptides of varied origins can be used as a potential weapon to eradicatc TB in future by developing new therapeutic drugs.Ameer Khusro Chirom Aarti Paul Agastian 2016Asian Pacific Journal of Tropical Medicine2016,9,11:3
6Application of Statistically Based Experimental Designs to Optimize Cellulase Production and Identification of Gene显示文摘A natural bacterial strain identified as Bacillus amyloliquefaciens MBAA3 using 16S rDNA partial genome sequencing has been studied for optimization of cellulase production.Statistical screening of media components for production of cellulase by B.amyloliquefaciens MBAA3 was carried out by Plackett–Burman design.Plackett–Burman design showed CMC,MgSO4 and pH as significant components influencing the cellulase production from the media components screened by Plackett-Burman fractional factorial design.The optimum concentrations of these significant parameters were determined employing the response surface central composite design,involving three factors and five levels was adopted to acquire the best medium for the production of cellulase enzyme revealed concentration of CMC(1.84 g),MgSO4(0.275 g),and pH(8.5)in media for highest enzyme production.Response surface counter plots revealed that middle level of MgSO4 and middle level of CMC,higher level of CMC and lower level of pH and higher level of MgSO4 with lower level of pH increase the production of cellulase.After optimization cellulase activity increased by 6.81 fold.Presence of cellulase gene in MBAA3 was conformed by the amplification of genomic DNA of MBAA3.A PCR product of cellulase gene of 1500 bp was successfully amplified.The amplified gene was conformed by sequencing the amplified product and sequence was deposited in the gene bank under the accession number KF929416.Aarti Thakkar Meenu Saraf 2014Natural Products and Bioprospecting2014,4,6:3

10Adenylate cyclase activator forskolin alleviates intracerebroventricular propionic acid-induced mitochondrial dysfunction of autistic rats显示文摘Neuronal mitochondrial dysfunction increases inflammatory mediators and leads to free radical generation and anti-oxidant enzymatic alterations,which are major neuropathological hallmarks responsible for autism.Mitochondrial dysfunction in autism is associated with decreased ATP levels due to reduced levels of cyclic adenosine monophosphate.Rat models of autism were established by intracerebroventricular injection of propionic acid.These rat models had memory dysfunction,decreased muscle coordination and gait imbalance.Biochemical estimation of propionic acid-treated rats showed changes in enzyme activity in neuronal mitochondrial electron transport chain complexes and increases in pro-inflammatory cytokines,oxidative stress and lipid biomarkers.Oral administration of 10,20 and 30 mg/kg adenylate cyclase activator forskolin for 15 days reversed these changes in a dose-dependent manner.These findings suggest that forskolin can alleviate neuronal mitochondrial dysfunction and improve neurological symptoms of rats with autism.This study was approved by the RITS/IAEC,SIRSA,HARYANA on March 3,2014(approval No.RITS/IAEC/2014/03/03).Sidharth Mehan Saloni Rahi Aarti Tiwari Tarun Kapoor Kajal Rajdev Ramit Sharma Himanshi Khera Sourabh Kosey Umesh Kukkar Rajesh Dudi 2020Neural Regeneration Research2020,15,6:1

16Annona muricata fruit extract protects against diethylnitrosamine-induced hepatocellular cancer in rats显示文摘Objective: To evaluate the anticancer potentials of Annona muricata fruit by in vitro and in vivo methods. Methods: The ethanolic extract of Annona muricata fruit was prepared by Soxhlet extraction method and further fractionated with petroleum ether, ethyl acetate and chloroform. The fractions were tested for cytotoxicity, apoptosis, scratch wound assay, and cell cycle analysis. IC50, apoptotic index and percentage cell migration were determined using HepG2 cells. For the in vivo studies, hepatocellular carcinoma was induced by administering 0.01% diethylnitrosamine(DEN) in drinking water in Wistar rats. In pre-treatment, rats were co-administered 200 mg/kg of fruit extract with DEN for 14 weeks. In post-treatment, the extract was co-administered after 8-weeks of DEN-induction for 14 weeks. Liver function test, haematological test, oxidative stress markers, relative liver weight, number of cancer nodules and histopathological parameters were determined.Results: Annona muricata fruit extract =significantly lowered cell proliferation counts. The chloroform-fraction possessed higher activity (IC50=(53.7±4.3) μg/mL)The chloroform fraction inhibited cell migration, which was significant compared to curcumin. Further investigations regarding the mode of anticancer activity revealed that the chloroform fraction induced apoptosis. The cell cycle analysis indicated that cells were being arrested at G0/G1. In the in vivo studies, the DEN-control group showed a significant decrease in body weights with increased mortality rate, hepatic nodules, and impairment of liver function compared to normal rats. The rats pre-treated and post-treated with the extract showed positive results with significant improvement in the parameters that were adversely affected by DEN.
