1Spatial-resolved metabolomics reveals tissue-specific metabolic reprogramming in diabetic nephropathy by using mass spectrometry imaging显示文摘Detailed knowledge on tissue-specific metabolic reprogramming in diabetic nephropathy(DN)is vital for more accurate understanding the molecular pathological signature and developing novel therapeutic strategies.In the present study,a spatial-resolved metabolomics approach based on air flowassisted desorption electrospray ionization(AFADESI)and matrix-assisted laser desorption ionization(MALDI)integrated mass spectrometry imaging(MSI)was proposed to investigate tissue-specific metabolic alterations in the kidneys of high-fat diet-fed and streptozotocin(STZ)-treated DN rats and the therapeutic effect of astragalosideⅣ,a potential anti-diabetic drug,against DN.As a result,a wide range of functional metabolites including sugars,amino acids,nucleotides and their derivatives,fatty acids,phospholipids,sphingolipids,glycerides,carnitine and its derivatives,vitamins,peptides,and metal ions associated with DN were identified and their unique distribution patterns in the rat kidney were visualized with high chemical specificity and high spatial resolution.These region-specific metabolic disturbances were ameliorated by repeated oral administration of astragaloside Ⅳ(100 mg/kg)for 12 weeks.This study provided more comprehensive and detailed information about the tissue-specific metabolic reprogramming and molecular pathological signature in the kidney of diabetic rats.These findings highlighted the promising potential of AFADESI and MALDI integrated MSI based metabolomics approach for application in metabolic kidney diseases.Zhonghua Wang Wenqing Fu Meiling Huo Bingshu He Yaqi Liu Lu Tian Wanfang Li Zhi Zhou Baili Wang Jianzhen Xia Yanhua Chen Jinfeng Wei Zeper Abliz 2021Acta Pharmaceutica Sinica B2021,11,11:7

3Rapid and sensitive liquid chromatography–tandem mass spectrometric method for the quantitative determination of potentially harmful substance 5,5′-oxydimethylenebis (2-furfural) in traditional Chinese medicine injections显示文摘With the rapid development and wide application of traditional Chinese medicine injection(TCMI), a number of adverse events of some TCMIs have incessantly been reported and have drawn broad attention in recent years. Establishing effective and practical analytical methods for safety evaluation and quality control of TCMI can help to improve the safety of TCMIs in clinical applications. In this study, a sensitive and rapid high-performance liquid chromatography–tandem mass spectrometry(HPLC–MS/MS)method has been developed and validated for the quantitative determination of potentially harmful substance5,5′-oxydimethylenebis(2-furfural, OMBF) in TCMI samples. Chromatographic separation was performed on a C18 reversed-phase column(150 mm × 2.1 mm, 5 μm) by gradient elution, using methanol–water containing 0.1% formic acid as mobile phase at the flow rate of 0.3 m L/min. MS/MS detection was performed on a triple quadrupole mass spectrometer with positive electrospray ionization in the multiple reaction-monitoring mode. The method was sensitive with a limit of quantification of 0.3 ng/m L and linear over the range of 0.3–30 ng/m L(r = 0.9998). Intra-and inter-day precision for analyte was o9.52% RSD withrecoveries in the range 88.0–109.67% at three concentration levels. The validated method was successfully applied to quantitatively determine the compound OMBF in TCMIs and glucose injections. 4疣状瓶霉致皮肤着色真菌病一例及致病菌的辅酶Q和DNA序列测定显示文摘目的报告1例疣状瓶霉致皮肤着色真菌病,并探讨其致病菌的实验室特征。方法皮损组织病理学检查、真菌学检查,分离株辅酶Q检测、DNA序列鉴定。结果该病表现为慢性疣状增殖性斑块,病程长,外伤后易发病。真皮层可见棕色厚壁孢子。菌落生长缓慢,需观察4周。小培养沙氏琼脂培养基无特殊结构,马铃薯葡萄糖琼脂见瓶梗多生长于短侧枝或气生菌丝顶端,玉米粉琼脂Ⅰ见瓶梗多侧生于菌丝,后两者其他结构相同。辅酶Q系统检测及LSUrDNA D1/D2区域碱基序列测定,与标准菌株比较完全相同。结论我国该病较少见,确诊需作真菌培养及组织病理,应同时采用沙氏琼脂培养基、马铃薯葡萄糖琼脂、玉米粉琼脂Ⅰ3种培养基培养。辅酶Q检测及分子生物学鉴定可排除表型不同的干扰,有利于进一步鉴定菌种。周冼苡 Paride Abliz 龙泽香代子 席丽艳 2005中华皮肤科杂志2005,38,2:3
5Human pathogenic fungus Trichophyton schoenleinii activates the NLRP3 inflammasome显示文摘The fungus Trichophyton schoenleinii(T.schoenleinii)is the causative agent of Trichophytosis and Tinea favosa of the scalp in certain regions of Eurasia and Africa.Hu-man innate immune system plays an important role in combating with various pathogens including fungi.The inflammasome is one of the most critical arms of host innate immunity,which is a protein complex controlling maturation of IL-1β.To clarify whether T.schoenleinii is able to activate the infl ammasome,we analyzed human monocytic cell line THP-1 for IL-1βproduction upon infec-tion with T.schoenleinii strain isolated from Tinea favosa patients,and rapid IL-1βsecretion from THP-1 cells was observed.Moreover,applying competitive inhibitors and gene specifi c silencing with shRNA,we found that T.sch-oenleinii induced IL-1βsecretion,ASC pyroptosome for-mation as well as caspase-1 activation were all dependent on NLRP3.Cathepsin B activity,ROS production and K+effl ux were required for the infl ammasome activation by T.schoenleinii.Our data thus reveal that the NLRP3 infl am-masome plays an important role in host defense against T.schoenleinii,and suggest that manipulating NLRP3 signaling can be a novel approach for control of diseases caused by T.schoenleinii infection.Hua Li Shuxian Wu Liming Mao Guowei Lei Liping Zhang Ailing Lu Liguo An Guiwen Yang Paride Abliz Guangxun Meng 2013Protein & Cell2013,4,7:3
6A high-performance bio-tissue imaging method using air flow-assisted desorption electrospray ionization coupled with a high-resolution mass spectrometer显示文摘Mass spectrometry imaging (MSI) technology can simultaneously obtain the spatial distribution of thousands of chemical compounds and has unique advantages compared to other techniques that allow mapping the surface of bio-tissue. Here, we combined an air flow-assisted desorption electrospray ionization (AFADESI) MSI device with a high-resolution mass spectrometer to optimize the system parameters and achieve more accurate spatial distribution characteristics for compounds of interest while investigating bio-tissue sections. The platform set-up, required instrumentation, sample pretreatment, parameter optimization and bio-tissue characterization are described and discussed.Finally, the parameter conditions that can provide optimal ionic intensity and enhanced resolution were confirmed. 7Spatiotemporal pharmacometabolomics based on ambient mass spectrometry imaging to evaluate the metabolism and hepatotoxicity of amiodarone in HepG2 spheroids显示文摘Three-dimensional(3D)cell spheroid models combined with mass spectrometry imaging(MSI)enables innovative investigation of in vivo-like biological processes under different physiological and pathological conditions.Herein,airflow-assisted desorption electrospray ionization-MSI(AFADESI-MSI)was coupled with 3D HepG2 spheroids to assess the metabolism and hepatotoxicity of amiodarone(AMI).High-coverage imaging of>1100 endogenous metabolites in hepatocyte spheroids was achieved using AFADESI-MSI.Following AMI treatment at different times,15 metabolites of AMI involved in Ndesethylation,hydroxylation,deiodination,and desaturation metabolic reactions were identified,and according to their spatiotemporal dynamics features,the metabolic pathways of AMI were proposed.Subsequently,the temporal and spatial changes in metabolic disturbance within spheroids caused by drug exposure were obtained via metabolomic analysis.The main dysregulated metabolic pathways included arachidonic acid and glycerophospholipid metabolism,providing considerable evidence for the mechanism of AMI hepatotoxicity.In addition,a biomarker group of eight fatty acids was selected that provided improved indication of cell viability and could characterize the hepatotoxicity of AMI.The combination of AFADESI-MSI and HepG2 spheroids can simultaneously obtain spatiotemporal information for drugs,drug metabolites,and endogenous metabolites after AMI treatment,providing an effective tool for in vitro drug hepatotoxicity evaluation.Limei Li Qingce Zang Xinzhu Li Ying Zhu Shanjing Wen Jiuming He Ruiping Zhang Zeper Abliz 2023Journal of Pharmaceutical Analysis2023,13,5:2
8A rapid and sensitive UPLC–MS/MS method for quantitative determination of arformoterol in rat plasma, lung and trachea tissues显示文摘A rapid and sensitive ultra-performance liquid chromatography-tandem mass spectrometry(UPLC–MS/MS) method was developed and fully validated for determination of arformoterol in rat plasma, lung and trachea tissues. The chromatographic separation was performed on an Agilent XDB C8 column with gradient elution by using acetonitrile and 0.1% formic acid water. The method presented high sensitivity(LLOQ of 1.83 pg/mL for plasma and 3.90 pg/mL for lung and trachea tissue homogenates) and great linearity over the range of 1.83–458 pg/mL for plasma, 13.9–1560 pg/mL for lung and trachea tissue homogenates. No carry over effect and matrix effect were observed. The intra-and inter-day precision/accuracy were within ±15% at three quality control concentration levels. This robust method was successfully applied to the pharmacokinetic, lung and trachea tissue distribution study after inhalation administration of arformoterol tartrate inhalation solution(50 m g/kg). 9Design and performance of air flow-assisted ionization imaging mass spectrometry system显示文摘The imaging mass spectrometry(IMS) technology has experienced a rapid development in recent years.A new IMS technology which is based on air flow assisted ionization(AFAI) was reported.It allows for the convenient pretreatment of the samples and can image a large area of sample in a single measurement with high sensitivity.The AFAI in DESI mode was used as the ion source in this paper.The new IMS method is named AFADESI-IMS.The adoption of assisted air flow makes the sample pretreatment easy and convenient.An optimization of the distance between the ion transport tube and MS orifice increases the sensitivity of the system.For data processing,a program based on MATLAB with the function of numerical analysis was developed.A theoretical imaging resolution of a few hundred microns can be achieved.The composite AFAI-IMS images of different target analytes were imaged with high sensitivity.A typical AFAI-IMS image of the whole-body section of a rat was obtained in a single analytical measurement.The ability to image a large area for relevant samples in a single measurement with high sensitivity and repeatability is a significant advantage.The method has enormous potentials in the MS imaging of large and complicated samples.Fei Tang Yi Chen Jiu-Ming He Zhi-Gang Luo Zeper Abliz Xiao-Hao Wang 2014Chinese Chemical Letters2014,25,5:2

Besides, it probes into the problems existing in the ecological migration, including the unscientific design of the moving project, the defects of the reclamation work, and the irrational treatment of the relation between ecological migration and regional development. 16A quick and effective multivariate statistical strategy for imaging mass spectrometry显示文摘A new multivariate statistical strategy for analyzing large datasets that are produced by imaging mass spectrometry(IMS) techniques is reported.The strategy divides the whole datacube of the sample into several subsets and analyses them one by one to obtain the results.Instead of analyzing the whole datacube at one time,the strategy makes the analysis easier and decreases the computation time greatly.In this report,the IMS data are produced by the air flow-assisted ionization IMS(AFAI-IMS).The strategy can be used in combination with most multivariate statistical analysis methods.In this paper,the strategy was combined with the principal component analysis(PCA) and partial least square analysis(PLS).It was proven to be effective by analyzing the handwriting sample.By using the strategy,the m/z corresponding to the specific lipids in rat brain tissue were distinguished successfully.Moreover the analysis time grew linearly instead of exponentially as the size of sample increased.The strategy developed in this study has enormous potential for searching for the mjz of potential biomarkers quickly and effectively.Fei Tang Yi Chen Tie-Gang Li Jiu-Ming He Zeper Abliz Gang Huang Xiao-Hao Wang 2014Chinese Chemical Letters2014,25,10:1
