| 1 | Phenolic composition and inhibitory activity of Mangifera indica and Mucuna urens seeds extracts against key enzymes linked to the pathology and complications of type 2 diabetes显示文摘Objective:To investigate the phenolic compounds composition and the inhibitory activity of Mangifera indica(M.indica) and Mucuna urens(M.urens) seeds extracts against some key enzymes(-amylase,-glucosidasc and aldose reductase) implicated in the pathology and complications of type 2 diabetes in vitro.Methods:Reverse phase chromatographic quantification of the major flavonoids and phenolic acids in the seeds extracts was carried out using high performance liquid chromatography coupled with diode array detection.The inhibitory activities of the seeds extracts against-amylase and-glucosidase were estimated using soluble starch and p-nitrophenylglucopyranosidc as their respective substrates.Inhibition of aldose reductase activity by the extracts was assayed using partially purified lens homogenate of normal male rat as source of enzyme;inhibition of Fe^(2+)-induced lipid peroxidation by extracts was tested in rat pancreas homogenate.Results:The chromatography result revealed that extracts of both seeds had appreciable levels of some major flavonoids and phenolic acids of pharmacological importance,including gallic acid,chlorogenic acid,caffeic acid,ellagic acid,catechin,rutin,quercitrin.quercetin and kaempferol.Extracts of both seeds effectively inhibited-amylase,-glucosidase and aldose reductase activities in a dose-dependent manner,having inhibitory preference for these enzymes in the order of aldose reductase>-glucosidase>-amylase.With lower half-maximal inhibitory concentrations(IC50)against-amylase,-glucosidase,and aldose reductase,M.indica had stronger inhibitory potency against these enzymes than M.urens.Extracts of both seeds also inhibited Fe2+-induced lipid peroxidation in a dose-dependent pattern,with M.indica being more potent than M.urens.Conclusions:The results obtained provide support for a possible use of M.indica and M.urens seeds in managing hyperglycemia and preventing the complications associated with it in type 2diabetes. | Emmanuel Anyachukwu Irondi Ganiyu Oboh Afolabi Akintunde Akindahunsi Aline Augusti Boligon Margareth Linde Athayde | 2014 | Asian Pacific Journal of Tropical Biomedicine2014,4,11: | 4 |
| 2 | Effect of reverse flotation on magnetic separation concentrates显示文摘Reverse flotation studies on magnetite samples have revealed that the use of starch as a depressant of Fe-oxides has a hydrophilic effect on the surface of Fe-bearing silicates and significantly decreases Fe in the silica-rich stream when used in combination with an amine (Lilaflot D817M). In this study, the effect of reverse flotation on the optimization of products obtained from magnetic separation was investigated. Two different magnetic samples, zones 1 and 2, were milled to <75 μm and then subjected to low intensity magnetic separation (LIMS). The LIMS test conducted on the <75 μm shown an upgrade of 46.40wt% Fe, 28.40wt% SiO 2 and 2.61wt% MnO for zone 1 and 47.60wt% Fe, 29.17wt% SiO 2 and 0.50wt% MnO for zone 2. Further milling of the ore to <25 μm resulted in a higher magnetic-rich product after magnetic separation. Reverse flotation tests were conducted on the agitated magnetic concentrate feed, and the result shows a significant upgrade of Fe compared to that obtained from the non-agitated feed. Iron concentrations greater than 69%, and SiO 2 concentrations less than 2% with overall magnetite recoveries greater than 67% and 71% were obtained for zones 1 and 2, respectively. | S.O. Bada A.S. | 3 | Homopterocarpin contributes to the restoration of gastric homeostasis by Pterocarpus erinaceus following indomethacin intoxication in rats显示文摘Objective:To investigate the restorative effect of Pterocarpus erinaceus(P.erinaceus) and homopterocarpin.an isoflavonoid isolated from it.on indomethacin-indueed disruption in gastric homeostasis in rats.Methods:Adult rats were dn ided into five groups and lasted for 48 h before treatment.Group I received olive oil(vehicle),group 2 received 25 ing/kg indomethaein while groups 3-5 received cimetidine(100 mg/kg).lioniopterocarpin(25 mg/kg) and P.erinaceus ethanolie stem hark extract(100 mg/kg) respectively.After 1 h.all the groups except group 2 were administered 25 mg/kg of indomethacin.One hour later,the rats were sacrificed and the ulcer index and other gaslroprotective indices were evaluated.Results:Indomethacin caused significant injury to the stomach of the rats as reflected in the ulcer indices(9.0±1.4) as compared with that of control(2.0±0.0).Equally,there were significant increases in gastric acid concentration and malondialdehvde level in the stomachs of the ulcerated animals compared with the control.However mucus content,reduced gluthatione level and gastric pH were significantly reduced in the ulcerated animals compared with the control.Pretreatment with either Pterocarpus bark extract or homopterocarpin reversed the effects of indomethaein on the evaluated parameters.Conclusions:These results indicate that both homopterocarpin and Plerocarpus extract offered gastroprotection against indomethacin-induced ulcer by antioxidative mechanism and the modulation of gastric homeostasis.The results also suggest that homopterocarpin might he responsible for.or contribute to the antiulcerogenic property of P.erinaceus. | M.Tolulope Olaleye Afolabi C.Akinmoladun Olamide O.Crown Katty E.Ahonsi A.O.Adetuyi | 2013 | Asian Pacific Journal of Tropical Medicine2013,6,3: | 2 |
| 4 | Chemical composition and antibacterial activity of Gongronema latifolium显示文摘Chemical composition of Gongronema latifolium leaves was determined using standard methods. Aqueous and methanol G. latifolium extracts were tested against thirteen pathogenic bacterial isolates. Crude protein, lipid extract, ash, crude fibre and nitrogen free extractives obtained are: 27.2%, 6.07%, 11.6%, 10.8% and 44.3% dry matter respectively. Potassium, sodium, calcium, phosphorus and cobalt contents are 332, 110, 115, 125 and 116 mg/kg respectively. Dominant essential amino acids are leucine, valine and phenylalanine. Aspartic acid, glutamic acid and glycine are 13.8%, 11.9% and 10.3% respectively of total amino acid. Saturated and unsaturated fatty acids are 50.2% and 39.4% of the oil respectively. Palmitic acid makes up 36% of the total fatty acid. Extracts show no activity against E. faecalis, Y. enterolytica, E. aerogenes, B. cereus and E. agglomerans. Methanol extracts were active against S. enteritidis, S. cholerasius ser typhimurium and P. aeruginosa (minimum inhibitory concentration (MIC) 1 mg; zone of growth inhibition 7, 6.5 and 7 mm respectively). Aqueous extracts show activity against E. coli (MIC 5 mg) and P. aeruginosa (MIC 1 mg) while methanol extracts are active against P. aeruginosa and L. monocytogenes. | 9 | Methanol extracts of Brachystegia eurycoma and Detarium microcarpum seeds flours inhibit some key enzymes linked to the pathology and complications of type 2 diabetes in vitro显示文摘The inhibitory effect of methanol extracts of Brachystegia eurycoma and Detarium microcarpum seeds flours on some key enzymes[α-amylase,α-glucosidase and aldose reductase(AR)]linked to the pathology and complications of type 2 diabetes(T2D);and their antioxidant properties were evaluated.The antioxidant properties evaluated were DPPH•and ABTS•^+scavenging abilities,reducing power,and antioxidant phytochemicals(total phenolics,tannins,total flavonoids and total saponins).Extracts of both flours inhibitedα-amylase,α-glucosidase and AR in a dose-dependent manner.The half-maximal inhibitory concentrations(IC50)of B.eurycoma onα-amylase,α-glucosidase,AR and lipid peroxidation were lower than those of D.microcarpum,indicating that it had stronger inhibitory potency than D.microcarpum.B.eurycoma also had significantly(P<0.05)higher DPPH•and ABTS•^+scavenging abilities,and reducing power than D.microcarpum.The antioxidant phytochemicals(total phenolics,tannins,total flavonoids and total saponins)were also significantly(P<0.05)higher in B.eurycoma than D.microcarpum.The inhibitory effect of B.eurycoma and D.microcarpum extracts onα-amylase,α-glucosidase and AR activities may be attributed to the combined action of their polyphenols and total saponins,and this may be a possible mechanism of action providing support for their use in managing hyperglycemia and the complications of T2D. | Emmanuel Anyachukwu Irondi Ganiyu Oboh Afolabi Akintunde Akindahunsi | 2015 | Food Science and Human Wellness2015,4,4: | 1 |
