5A New Ratio-Metric pH Probe,“ThiAKS Green”for Live-Cell pH Measurements显示文摘Deviation of the H+concentration from optimum values within the organelles is closely associated with irregular cellular functions that cause the onset of various diseases.Therefore,determining subcellular pH values in live cells and tissues is valuable for diagnostic purposes.In this study,we report a novel ratiometric fluorescence probe 1H-pyrazole-3-carboxylic acid,4-(benzo[d]thiazol-2-yl)-3-(2,4-dihydroxy-3-methylphenyl)-1H-pyrazole-5-carboxylicacid4-(2-benz othiazolyl)-5-(2,4-dihydroxy-3-methylphenyl),to which we will refer as ThiAKS Green(Thiazole AKyol shifting green),that is pH sensitive.The results presented here show that the probe can penetrate the celi membrane in iess than 30minutes and does not show any detectable toxicity.The measured color shifts up on pH change are linear and most significant around physiological pH(pKa=7.45),thus making this probe suitable for live-cell imaging and intracellular pH measurements.During the long-incubation periods following the application of the probe and the fluorescent microscopy measurements,it shows stable properties and is easy to detect in live cells.In conclusion,the results suggest that ThiAKS Green can be used to obtain precise information on the H+distribution at various compartments of the live cells.Ali AKYOL Doruk BAYKAL Akin AKDAG Özge SENSOY Çagdas Devrim SON 2023Photonic Sensors2023,13,1:0
6Protective effect of insulin and glucose at different concentrations on penicillin-induced astrocyte death on the primer astroglial cell line显示文摘Astrocytes perform many functions in the brain and spinal cord.Glucose metabolism is important for astroglial cells and astrocytes are the only cells with insulin receptors in the brain.The common antibiotic penicillin is also a chemical agent that causes degenerative effect on neuronal cell.The aim of this study is to show the effect of insulin and glucose at different concentrations on the astrocyte death induced by penicillin on primer astroglial cell line.It is well known that intracranial penicillin treatment causes neuronal cell death and it is used for experimental epilepsy model commonly.Previous studies showed that insulin and glucose might protect neuronal cell in case of proper concentrations.But,the present study is about the effect of insulin and glucose against astrocyte death induced by penicillin.For this purpose,newborn rat brain was extracted and then mechanically dissociated to astroglial cell suspension and finally grown in culture medium.Clutters were maintained for 2 weeks prior to being used in these experiments.Different concentrations of insulin(0,1,3 nM)and glucose(0,3,30 mM)were used in media without penicillin and with 2 500μM penicillin.Penicillin decreased the viability of astroglial cell seriously.The highest cell viability appeared in medium with 3 nM insulin and 3 mM glucose but without penicillin.However,in medium with penicillin,the best cell survival was in medium with 1 nM insulin but without glucose. 8Intrusion Detection System Through Deep Learning in Routing MANET Networks显示文摘Deep learning(DL)is a subdivision of machine learning(ML)that employs numerous algorithms,each of which provides various explanations of the data it consumes;mobile ad-hoc networks(MANET)are growing in promi-nence.For reasons including node mobility,due to MANET’s potential to provide small-cost solutions for real-world contact challenges,decentralized management,and restricted bandwidth,MANETs are more vulnerable to security threats.When protecting MANETs from attack,encryption and authentication schemes have their limits.However,deep learning(DL)approaches in intrusion detection sys-tems(IDS)can adapt to the changing environment of MANETs and allow a sys-tem to make intrusion decisions while learning about its mobility in the environment.IDSs are a secondary defiance system for mobile ad-hoc networks vs.attacks since they monitor network traffic and report anything unusual.Recently,many scientists have employed deep neural networks(DNNs)to address intrusion detection concerns.This paper used MANET to recognize com-plex patterns by focusing on security standards through efficiency determination and identifying malicious nodes,and mitigating network attacks using the three algorithms presented Cascading Back Propagation Neural Network(CBPNN),Feedforward-Neural-Network(FNN),and Cascading-Back-Propagation-Neural-Network(CBPNN)(FFNN).In addition to Convolutional-Neural-Network(CNN),these primary forms of deep neural network(DNN)building designs are widely used to improve the performance of intrusion detection systems(IDS)and the use of IDS in conjunction with machine learning(ML).Further-more,machine learning(ML)techniques than their statistical and logical methods provide MANET network learning capabilities and encourage adaptation to differ-ent environments.Compared with another current model,The proposed model has better average receiving packet(ARP)and end-to-end(E2E)performance.The results have been obtained from CBP,FFNN and CNN 74%,82%and 85%,respectively,by the time(27,18,and 17 s).Zainab Ali Abbood DoguÇagdaşAtilla Çagatay Aydin 2023Intelligent Automation & Soft Computing2023,,7:0
