1RLINl,encoding a putative coproporphyrinogen Ⅲ oxidase,is involved in lesion initiation in rice显示文摘Lesion mimic is necrotic lesions on plant leaf or stem in the absence of pathogenic infection,and its exact biological mechanism is varied.By a large-scale screening of our T-DNA mutant population,we identified a mutant rice lesion initiation 1(rlin1),which was controlled by a single nuclear recessive gene.Map-based cloning revealed that RLIN1 encoded a putative coproporphyrinogen Ⅲ oxidase in tetrapyrrole biosynthesis pathway.Sequencing results showed that a G to T substitution occurred in the second exon of RLIN1 and led to a missense mutation from Asp to Tyr.Ectopic expression of RLIN1 could rescue rlin1 lesion mimic phenotype.Histochemical analysis demonstrated that lesion formation in rlin1 was light-dependent accompanied by reactive oxygen species accumulated.These results suggest that tetrapyrrole participates in lesion formation in rice.Changhui Sun Linchuan Liu Jiuyou Tang Aihong Lin Fantao Zhang Jun Fang Genfa Zhang Chengcai Chu 2011Journal of Genetics and Genomics2011,38,1:31
2Experimental study on the mechanical properties of rocks at high temperature显示文摘The mechanical properties of marble, limestone, and sandstone as well as the stress-strain curve, the varying characteristics of the peak strength, the peak strain and elastic modulus were studied by using the MTS810 Rock Mechanics Servo-controlled Testing System under the action of temperatures rang- ing from room temperature to 800℃ . Results show that (1) the peak strength and elastic modulus of marble fluctuate at the temperature from normal to 400 ℃ ; and they decrease gradually over 400℃ . (2) With the rise of the temperature, the peak strength and elastic modulus of limestone show downward trend from normal temperature to 200 ℃ ; have little change from 200 ℃ to 600 ℃; and decrease sharply over 600 ℃ . (3) The peak strength of sandstone shows a downward trend while a little change for elastic modulus at normal temperature to 200 ℃ ; and from 200 ℃ to 600 ℃ , the peak strength of sandstone in- creases while a little change for elastic modulus; the peak strength and elastic modulus decrease rap- idly at the temperature over 600 ℃ . (4) The peak strain of limestone shows little change at normal tem- perature to 600 ℃ , however, the peak strain increases rapidly over 600 ℃; and for marble and sandstone, the peak strain decreases with the rise of the temperature from normal temperature to 200℃ , the peak strain increases rapidly over 200℃. 4Investigation of heavy metal partitioning influenced by flue gas moisture and chlorine content during waste incineration显示文摘The impact of moisture on the partitioning of the heavy metals including Pb,Zn,Cu and Cd in municipal solid waste (MSW) was studied in a laboratory tubular furnace.A thermodynamic investigation using CHEMKIN software was performed to compare the experimental results.Simulated waste,representative of typical MSW with and without chlorine compounds,was burned at the background temperature of 700 and 950°C,respectively.In the absence of chlorine,the moisture content has no evident effect on the volatility of Pb,Zn and Cu at either 700 or 950°C,however,as flue gas moisture increasing the Cd distribution in the bottom ash increased at 700°C and reduced at 950°C,respectively.In the presence of chlorine,the flue gas moisture reduced the volatility of Pb,Zn and Cu due to the transformation of the more volatile metal chlorides into less volatile metal oxides,and the reduction became significant as chlorine content increase.For Cd,the chlorine promotes its volatility through the formation of more volatile CdCl 2.As a result,the increased moisture content increases the Pb,Zn,Cu and Cd concentrations in the bottom ash,which limits the utilization of the bottom ash as a construction material.Therefore,in order to accumulate heavy metals into the fly ash,MSW should be dried before incineration.Qinghai Li,Aihong Meng,Jinyan Jia,Yanguo Zhang Department of Thermal Engineering,Key Laboratory for Thermal Science and Power Engineering of the Ministry of Education of China,Tsinghua University,Beijing 100084,China. 2010Journal of Environmental Sciences2010,22,5:24
5Effect of specimen size on energy dissipation characteristics of red sandstone under high strain rate显示文摘In this experiment, red sandstone specimens, having slenderness ratios of 0.5, 0.7, 0.9 and 1.1 respectively, were subjected to blow tests using a Split Hopkinson Pressure Bar(SHPB) system at a pressure of 0.4 atmospheres. In this paper, we have analyzed the effect of slenderness ratio on the mechanical properties and energy dissipation characteristics of red sandstone under high strain rates. The processes of compaction, elastic deformation and stress softening deformation of specimens contract with an increase in slenderness ratio, whilst the nonlinear deformation process extends correspondingly. In addition, degrees of damage of specimens reduced gradually and the type of destruction showed a transformation trend from stretching failure towards shear failure when the slenderness ratio increased. A model of dynamic damage evolution in red sandstone was established and the parameters of the constitutive model at different ratios of length to diameter were determined. By comparison with the experimental curve, the accuracy of the model, which could reflect the stress–strain dynamic characteristics of red sandstone, was verified. From the view of energy dissipation, an increase in slenderness ratio of a specimen decreased the proportion of energy dissipation and caused a gradual fall in the capability of energy dissipation during the specimen failure process. 6Behavior and dynamics of gecko’s locomotion:The effects of moving directions on a vertical surface显示文摘The study of the movement behavior of geckos on a vertical surface, including the measurement and recording of the reaction forces as they move in different directions, plays an important role in understanding the mechanics of the animals' locomotion. This study provides inspiration for the design of a control system for a bionics robot. The three-dimensional reaction forces of vertical surface-climbing geckos (Gekko gecko) were measured using a three-dimensional force-sensors-array. The behavior of gecko as it moved on a vertical surface was recorded with a high speed camera at 215 fps and the function of each foot of a gecko are discussed in this paper. The results showed that the gecko increased its velocity of movement mainly by increasing the stride frequency in the upward, downward and leftward direction and that the speed had no significant relationship to the attachment and detachment times. The feet above the center-of-mass play a key role in supporting the body, driving locomotion and balancing overturning etc. The movement behavior and foot function of geckos change correspondingly for different conditions, which results in safe and effective free vertical locomotion. 7Characterization and evaluation of rice blast resistance of Chinese indica hybrid rice parental lines显示文摘The development of resistant varieties and hybrid combinations has been the most effective and economical strategy to control blast disease caused by Magnaporthe oryzae.However, the distribution of major R genes and blast resistance characterization in hybrid rice parents has not been well investigated, resulting in their limited use in hybrid rice blast-resistance breeding. In the present study, 88 elite indica hybrid rice parental lines were evaluated with 30 isolates of M. oryzae collected from the main planting area of indica hybrid rice in China and were characterized for the presence of 11 major resistance genes using molecular markers. The pathogenicity assays showed that four types of hybrid rice parent line showed some resistance to M. oryzae. However, the proportions of highly resistant lines and the mean resistance frequency(RF) varied among the four types, with resistance in decreasing order shown by three-line restorer lines, three-line maintainer lines, two-line sterile lines, and two-line restorer lines. All 88 hybrid rice parental lines carried more than one R gene, but none carried the R genes Pi1 and Pi2. Although Pid3 and Pi9 were present only in three-line restorer lines and Pigm only in three-line maintainer lines, the remaining six R genes(Pib, Pid2, Pi5, Pia, Pi54, and Pita) were present in the four types of hybrid rice parent with significantly different distribution frequencies. The correlation between R genes and resistance reactions was investigated. 8Molecular Cloning and Expression Analysis of the ζ-Carotene Desaturase Gene in Chinese kale(Brassica oleracea var.alboglabra Bailey)显示文摘ζ-Carotene desaturase(ZDS)is an important enzyme in carotenoid biosynthesis.Here,the Brassica oleracea var.alboglabra ZDS(Boa ZDS)gene was cloned from Chinese kale via reverse transcription-polymerase chain reaction(RT-PCR)and deposited in Gen Bank(accession number KY662297).The Boa ZDS gene contains an open reading frame of 1 686 bp that encodes a 561-amino acid protein.Sequence analysis indicates that the ZDS protein is apparently conserved during plant evolution and is most closely related to B.oleracea var.capitata and B.rapa.The promoter sequence of the Boa ZDS gene was predicted to harbor several cis-acting elements that are related to light and phytohormone responses.Semiquantitative RT-PCR analysis showed that Boa ZDS expression varied among different developmental stages and organs.Relative ZDS expression remained stable during germination and seedling stages and rapidly increased at the mature leaf stage.The leaves showed the highest ZDS expression levels compared to the other organs.ZDS expression decreased in all flower tissues during blooming.The fused protein of Boa ZDS was obtained by prokaryotic expression.Heterologous expression of Boa ZDS in Escherichia coli confirmed that Boa ZDS encodes a functionalζ-carotene desaturase that increases β-carotene accumulation in E.coli cells harboring a β-carotene-producing plasmid.The findings of the present study provide a molecular basis for the elucidation of ZDS gene function in Chinese kale.SUN Bo ZHANG Fen XUE Shengling CHANG Jiaqi ZHENG Aihong JIANG Min MIAO Huiying WANG Qiaomei TANG Haoru 2018Horticultural Plant Journal2018,4,3:10
9Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. 10Sources, evolution and impacts of EC and OC in snow on sea ice: a measurement study in Barrow, Alaska显示文摘Based on the field measurements in Barrow, Alaska within the period of April-May 2015, we investigate the sources and variations of elemental carbon(EC) and organic carbon(OC) in the surface layer of snowpack on sea ice, and estimate their effects on the sea ice albedo. Results show that the snow OC in Barrow are from natural sources(e.g. terrestrial higher plants and micro-organisms) mainly, as well as biomass burning(e.g. forest fires and straw combustion) as an important part. Both EC and OC can accumulate at the snow surface with snow melt. The variations in EC and OC and liquid water content in the snow layer are well consistent during the snow-melting period. A higher rate of snow melt implied a more efficient enrichment of EC and OC. In the last phase of snow melt, the concentration increased to a maximum of 16.2 ng/g for EC and 128 ng/g for OC, which is ~10 times larger than those before snow melt onset. Except for the dominant influence of melt amplification mechanism, the variation in concentrations of EC and OC could be disturbed by the air temperature fluctuation and snowfall. Our study indicates that the lightabsorbing impurities contributed 1.6%-5.1% to the reduction in sea ice albedo with melt during the measurement period. The significant period oflight-absorbing impurities influencing on sea ice albedo begins with the rapid melting of overlying snow and ends before the melt ponds formed widely, which lasted for about 10 days in Barrow, 2015.Tingfeng Dou Cunde Xiao Zhiheng Du J. 11Engineering broad-spectrum disease-resistant rice by editing multiple susceptibility genes显示文摘Rice blast and bacterial blight are important diseases of rice(Oryza sativa)caused by the fungus Magnaporthe oryzae and the bacterium Xanthomonas oryzae pv.oryzae(Xoo),respectively.Breeding rice varieties for broadspectrum resistance is considered the most effective and sustainable approach to controlling both diseases.Although dominant resistance genes have been extensively used in rice breeding and production,generating diseaseresistant varieties by altering susceptibility(S)genes that facilitate pathogen compatibility remains unexplored.Here,using CRISPR/Cas9 technology,we generated loss-of-function mutants of the S genes Pi21 and Bsr-d1 and showed that they had increased resistance to M.oryzae.We also generated a knockout mutant of the S gene Xa5 that showed increased resistance to Xoo.Remarkably,a triple mutant of all three S genes had significantly enhanced resistance to both M.oryzae and Xoo.Moreover,the triple mutant was comparable to the wild type in regard to key agronomic traits,including plant height,effective panicle number per plant,grain number per panicle,seed setting rate,and thousand-grain weight.These results demonstrate that the simultaneous editing of multiple S genes is a powerful strategy for generating new rice varieties with broadspectrum resistance.Hui Tao Xuetao Shi Feng He Dan Wang Ning Xiao Hong Fang Ruyi Wang Fan Zhang Min Wang Aihong Li Xionglun Liu Guo-Liang Wang Yuese Ning 2021Journal of Integrative Plant Biology2021,63,9:4

13Rapid Urbanization Induced Extensive Forest Loss to Urban Land in the Guangdong-Hong Kong-Macao Greater Bay Area, China显示文摘China has experienced rapid urbanizations with dramatic land cover changes since 1978. Forest loss is one of land cover changes, and it induces various eco-environmental degradation issues. As one of China’s hotspot regions, the Guangdong-Hong KongMacao Greater Bay Area(GBA) has undergone a dramatic urban expansion. To better understand forest dynamics and protect forest ecosystem, revealing the processes, patterns and underlying drivers of forest loss is essential. This study focused on the spatiotemporal evolution and potential driving factors of forest loss in the GBA at regional and city level. The Landsat time-series images from 1987 to2017 were used to derive forest, and landscape metrics and geographic information system(GIS) were applied to implement further spatial analysis. The results showed that: 1) 14.86% of the total urban growth area of the GBA was obtained from the forest loss in1987–2017;meanwhile, the forest loss area of the GBA reached 4040.6 km2, of which 25.60%(1034.42 km2) was converted to urban land;2) the percentages of forest loss to urban land in Dongguan(19.14%), Guangzhou(18.35%) and Shenzhen(15.81%) were higher than those in other cities;3) the forest became increasingly fragmented from 1987–2007, and then the fragmentation decreased from2007 to 2017);4) the landscape responses to forest changes varied with the scale;and 5) some forest loss to urban regions moved from low-elevation and gentle-slope terrains to higher-elevation and steep-slope terrains over time, especially in Shenzhen and Hong Kong.Urbanization and industrialization greatly drove forest loss and fragmentation, and, notably, hillside urban land expansion may have contributed to hillside forest loss. 14Autophagy targets Hd1 for vacuolar degradation to regulate rice flowering显示文摘Flowering time(heading date)is a critical agronomic trait that determines the yield and regional adaptability of crops.Heading date 1(Hd1)is a central regulator of photoperiodic flowering in rice(Oryza sativa).However,how the homeostasis of Hd1 protein is achieved is poorly understood.Here,we report that the nuclear autophagy pathway mediates Hd1 degradation in the dark to regulate flowering.Loss of autophagy function results in an accumulation of Hd1 and delays flowering under both short-day and long-day conditions.In the dark,nucleus-localized Hd1 is recognized as a substrate for autophagy and is subjected to vacuolar degradation via the autophagy protein OsATG8.The Hd1-0sATG8 interaction is required for autophagic degradation of Hd1 in the dark.Our study reveals a new mechanism by which Hd1 protein homeostasis is regulated by autophagy to control rice flowering.Our study also indicates that the regulation of flowering by autophagic degradation of Hd1 orthologs may have arisen over the course of mesangiosperm evolution,which would have increased their flexibility and adaptability to the environment by modulating flowering time.Zhi Hu Zhipan Yang Yi Zhang Aihong Zhang Qingtao Lu Ying Fang Congming Lu 2022Molecular Plant2022,15,7:3
15LncRNA HOTAIR promotes endometrial fibrosis by activating TGF-β1/Smad pathway显示文摘Homeobox transcript antisense RNA(HOTAIR)is a long non-coding RNA associated with a number of fibrosis-related diseases.The aim of this study was to investigate the specific role of HOTAIR in the development of endometrial fibrosis and to identify the molecular mechanisms underlying this process.Reverse transcription-quantitative polymerase chain reaction(RT-qPCR)was used to determine the expression levels of HOTAIR in samples of intrauterine adhesion(lUA)tissue and in endometrial stromal cells(ESCs)that had been treated with transforming growth factor beta 1(TGF-β1).Additionally,we transfected ESCs with either overexpression plasmid(pcDNA-HOTAIR)or silencing construct(si-HOTAIR)and then treated these cells with TGF-β1.We then performed RTqPCR and western blot analysis,along with cell proliferation and apoptosis assays,to investigate the effects of HOTAIR on the transdifferentiation of ESCs into myofibroblasts.The results showed that the expression levels of HOTAIR were significantly elevated in IUA tissue and in ESCs that had been treated with TGF-β1.The overexpression of HOTAIR had a pro-fibrotic effect on ESCs,while the silencing of HOTAIR exerted an anti-fibrotic effect.Most importantiy,the protein expression levels of p-Smad2 and p-Smad3 were significantly upregulated in TGF-β1.treated ESCs transfected with pcDNA-HOTAIR and were downregulated after transfection with si-HOTAIR constructs.These data indicate that HOTAIR promotes endometrial fibrosis by activating the TGF-β1/Smad signaling pathway,suggesting that the inhibition of HOTAIR may represent a promising therapeutic option for suppressing endometrial fibrosis.Jianhong Wu Lingge Jin Yudi Zhang Aihong Duan Juhong Liu Ziwen Jiang Liang Huang Jing Chen Zhaohui Liu Dan Lu Yinmei Dai 2020Acta Biochimica et Biophysica Sinica2020,52,12:3
16Study on drying and combustion process in grate-CFB incinerator显示文摘The drying and combustion process in the combined grate and circulating fluidized bed(grate-CFB) municipal solid waste(MSW) incinerator was investigated experimentally and mathematically.The drying grate bed was simulated by a muffle furnace,which could be controlled at a constant tempera-ture level.The kind of wastes,thickness of waste layer fed and temperature were chosen as the ad-justable parameters to study their effect on the drying process.The experimental results indicated that the hydrophilic wastes were more difficult to be dried than the hydrophobic wastes.The higher the temperature is the easier the waste is to be dried.The thinner waste layer is favorable to drying the waste.The pyrolysis experiment in the furnace showed that the higher temperature level could reduce the conversion rate of carbon to carbon monoxide.The semi-empirical mathematical model that in-cluded the bed material distribution submodel,volatile matter release submodel,carbon particle combustion submodel and so on was proposed.A 260 t/d grate-CFB incinerator was modeled and the model predicted bulk density agreed with the measured value from industrial field test.The predicted flue gas(e.g.CO2,CO) concentration deviated slightly from the industrial test data.The parameters and variables used in the model were determined by the experiments or practical field test.This model can be used to design the grate-CFB incinerator and guide its operation.QingHai Li YanGuo Zhang MeiQian Chen AiHong Meng ChangHe Chen 2009Science China(Technological Sciences)2009,52,5:3
17Pentatricopeptide repeat protein PHOTOSYSTEM I BIOGENESIS FACTOR2 is required for splicing of ycf3显示文摘To gain a better understanding of the molec-ular mechanisms of photosystem!(PSI)biogenesis,wecharacterized the Arabidopsis thaliana photosystem l bio-genesis factor 2(pbf2)mutant,which lacks PSl complex.PBF2 encodes a P-class pentatricopeptide repeat(PPR)protein.In the pbf2 mutants,we observed a striking de-crease in the transcript level of only one gene,thechloroplast gene ycf3,which is essential for PSl assembly.Further analysis of ycf3 transcripts showed that PBF2 isspecifically required for the splicing of ycf3 intron 1.Computational prediction of binding sequences and elec-trophoretic mobility shift assays reveal that PBF2 specifi-cally binds to a sequence in ycf3 intron 1.Moreover,we found that PBF2 interacted with two general factors forgroup ll intron splicing CHLOROPLAST RNA SPLICING2-AsSOCIATED FACTOR1(CAF1)and CAF2,and facilitated theassociation of these two factors with ycf3 intron 1.Ourresults suggest that PBF2 is specifically required for thesplicing of ycf3 intron 1 through cooperating with CAF1and CAF2.Our results also suggest that additional proteinsare required to contribute to the specificity of CAF-dependent group ll intron splicing.Xuemei Wang Zhipan Yang Yi Zhang Wen Zhou Aihong Zhang Congming Lu 2020Journal of Integrative Plant Biology2020,62,11:2
18Genome-wide analysis of NDR1/HIN1-like genes in pepper(Capsicum annuum L.)and functional characterization of CaNHL4 under biotic and abiotic stresses显示文摘Plant NDR1/HIN1-like(NHL)genes play an important role in triggering plant defenses in response to biotic stresses.In this study,we performed a genome-wide identification of the NHL genes in pepper(Capsicum annuum L.)and characterized the functional roles of these CaNHL genes in response to abiotic stresses and infection by different pathogens.Phylogenetic analysis revealed that CaNHLs can be classified into five distinct subgroups,with each group containing generic and specific motifs.Regulatory element analysis showed that the majority of the promoter regions of the identified CaNHLs contain jasmonic acid(JA)-responsive and salicylic acid(SA)-responsive elements,and transcriptomic analysis revealed that CaNHL genes are expressed in all the examined tissues of pepper.The CaNHL1,CaNHL4,CaNHL6,CaNHL10,CaNHL11,and CaNHL12 genes were significantly upregulated under abiotic stress as well as in response to different pathogens,such as TMV,Phytophthora capsici and Pseudomonas syringae.In addition,we found that CaNHL4 localizes to the plasma membrane.CaNHL4-silenced pepper plants display significantly increased susceptibility to TMV,Phytophthora capsici and Pseudomonas syringae,exhibiting reduced expression of JA-related and SA-related genes and reduced ROS production.However,transient overexpression of CaNHL4 in pepper increases the expression of JArelated and SA-related genes,enhances the accumulation of ROS,and inhibits the infection of these three pathogens.Collectively,for the first time,we identified the NHL genes in pepper and demonstrated that CaNHL4 is involved in the production of ROS and that it also regulates the expression of JA-related and SA-related genes in response to different pathogens,suggesting that members of the CaNHL family play an essential role in the disease resistance of pepper.Changyun Liu Haoran Peng Xinyu Li Chaolong Liu Xing Lv Xuefeng Wei Aihong Zou Jian Zhang Guangjin Fan Guanhua Ma Lisong Ma Xianchao Sun 2020Horticulture Research2020,7,1:2
19Effect of three tree species on UV weathering of wood flour-HDPE composites显示文摘Eucalyptus urophylla, Acacia mangium, and Pinus caribaea are the primary species for the wood industry in Vietnam. Wood residues of these species were used to reinforce high-density polyethylene(HDPE) composites. The flexural or bending property, impact strength and surface color were evaluated after exposure to accelerated ultraviolet weathering up to 2000 h. The weathered surface was characterized by scanning electron microscopy(SEM) and Fourier transform infrared(FTIR) spectroscopy. The results indicate that A. mangium/HDPE composites had the lowest color change and least fading,and remained stable after 1500 h exposure. FTIR spectroscopy showed that the oxidation of the composites increased in parallel with duration of exposure by assessing the concentration of carbonyl groups on the surfaces. SEM showed that all three species reinforced composites exhibited similar severe cracks after 1000 h;however, at the end of the weathering test, E. urophylla and P. caribaea composites were more severely cracked than A. mangium composites. A. mangium also had the highest flexural strength, impact strength and crystallinity duringweathering. 20Accurate construction of cell membrane biomimetic graphene nanodecoys via purposeful surface engineering to improve screening efficiency of active components of traditional Chinese medicine显示文摘Biomimetic nanoengineering presents great potential in biomedical research by integrating cell membrane(CM) with functional nanoparticles. However, preparation of CM biomimetic nanomaterials for custom applications that can avoid the aggregation of nanocarriers while maintaining the biological activity of CM remains a challenge. Herein, a high-performance CM biomimetic graphene nanodecoy was fabricated via purposeful surface engineering, where polyethylene glycol(PEG) was used to modifying magnetic graphene oxide(MGO) to improve its stability in physiological solution, so as to improve the screening efficiency to active components of traditional Chinese medicine(TCM). With this strategy, the constructed PEGylated MGO(PMGO) could keep stable at least 10 days, thus improving the CM coating efficiency. Meanwhile, by taking advantage of the inherent ability of He La cell membrane(HM) to interact with specific ligands, HM-camouflaged PMGO showed satisfied adsorption capacity(116.2 mg/g) and selectivity. Finally, three potential active components, byakangelicol, imperatorin,and isoimperatorin, were screened from Angelica dahurica, whose potential antiproliferative activity were further validated by pharmacological studies.
