| 2 | Identification of Ca2+ signaling components in neural stem/progenitor cells during differentiation into neurons and glia in intact and dissociated zebrafish neurospheres显示文摘The development of the CNS in vertebrate embryos involves the generation of different sub-types of neurons and glia in a complex but highly-ordered spatio-temporal manner. Zebrafish are commonly used for exploring the development, plasticity and regeneration of the CNS, and the recent development of reliable protocols for isolating and culturing neural stem/progenitor cells(NSCs/NPCs) from the brain of adult fish now enables the exploration of mechanisms underlying the induction/specification/differentiation of these cells. Here, we refined a protocol to generate proliferating and differentiating neurospheres from the entire brain of adult zebrafish. We demonstrated via RT-qPCR that some isoforms of ip3 r, ryr and stim are upregulated/downregulated significantly in differentiating neurospheres, and via immunolabelling that 1,4,5-inositol trisphosphate receptor(IP3 R) type-1 and ryanodine receptor(RyR) type-2 are differentially expressed in cells with neuron-or radial glial-like properties. Furthermore, ATP but not caffeine(IP3 R and RyR agonists, respectively), induced the generation of Ca^(2+) transients in cells exhibiting neuron-or glial-like morphology. These results indicate the differential expression of components of the Ca^(2+) -signaling toolkit in proliferating and differentiating cells. | 16 | Structure-based multi-ligand molecular modeling to predict the synergistic effects of limonin and obacunone from Simiao pill against nitric oxide synthase 3 associated with hyperuricemia显示文摘Hyperuricemia(HUA)mainly occurs because of purine metabolism disorders.We recently proposed that limonin from Simiao pill may have therapeutic effects on nitric oxide synthase 3(NOS3)that is related to HUA.Concurrently,our previous work employed a new method,structure-based multi-ligand molecular modeling,to identify potential agents from a herbal formula that may produce synergistic effects and may have the potential to develop combination drugs.Therefore,we employed multi-ligand modeling to seek compounds with potential synergistic effects with limonin against NOS3.We obtained 403 multi-ligand docking results between 403 compounds and the limonin-NOS3 complex(average affinity–8.297 kcal/mol).Then we selected the top 10 highest binding affinity compounds for virtual pharmacokinetic and toxicity screening and we found that only obacunone passed all filters.We further subjected obacunone,bound to limonin and NOS3,to molecular dynamics simulations.We found that the NOS3-limonin-obacunone complex was more stable than the NOS3-limonin complex,based on the root mean square deviation of backbone Cαatoms and root mean square fluctuation,which suggests that synergistic effects may exist between limonin and obacunone.Further cell and animal experimental research is required to verify our results. | Qing-Hong Chen Xiao-Mei Chen Xiao-Hu Chen Yan-Ting You Shu-Xuan Yang Chuang-Hai Wu Mei-Lin Chen Ming-Di Li Akari Komori Yan-Yan Liu Andrew Hung Xiao-Shan Zhao Hong Li | 2023 | Precision Medicine Research2023,5,3: | 0 |
| 17 | Controversies on Cornell Realism显示文摘This article examines the criticisms and debates about Cornell realism.While critics,like Shafer-Landau,Tropman,Oliveira and Perrine,reject the claim by Cornell realism that moral knowledge can be empirically investigated the same as natural science is,I argue that some of their arguments are not sufficient to refute Cornell realism.What is crucial in assessing Cornell realism is distinguishing normative ethics from empirical science.While ethics is normative in nature,that of empirical science is descriptive and predictive.I also show that the debate between Tropman and Long is at cross purposes in their discussion about the nature of moral knowledge.By clarifying different meanings of moral knowledge,I argue that while arguments by Cornell realism can be applied to moral psychology,the study of normative ethics through empirical investigation still faces the problem of an is-ought gap.Indeed,many of Cornell realist arguments are begging many questions.I have also examined recent debates on normativity objection by Parfit and Copp.I argue that Copp’s naturalism is very similar to Huemer’s intuitionism.Copp’s argument of non-analytical naturalism seems to support rather than refute moral intuitionism. | Andrew Tsz Wan Hung | 2023 | Fudan Journal of the Humanities and Social Sciences2023,16,2: | 0 |
| 18 | COVID-19 outbreaks in residential care homes in Hong Kong and effectiveness of vaccine against severe outcomes显示文摘Background:COVID-19 outbreaks in residential care homes for the elderly(RCHEs)and for persons with disability(RCHDs)have caused significant morbidity and mortality during 5th epidemic in Hong Kong.This article reviewed COVID-19 outbreaks situation and estimated the effectiveness of receiving at least two-dose of COVID-19 vaccine in preventing severe outcomes.Methods:To estimate attack rates and vaccination coverage,documentation on COVID-19 infection and their vaccination records of residential care homes(RCH)residents reported between December 31,2021 and May 31,2022 were reviewed,and infected cases were follow-up for 4 weeks for severe outcomes or death.Correlation between vaccination coverage against attack rate by types of homes was examined.Infected RCH residents with available information were included in the analysis of vaccine effectiveness against severe outcomes and death.Results:COVID-19 vaccination coverage was low in RCHDs(median 0.46,IQR:0.24-0.76)and very low in RCHEs(median 0.08,IQR:0.00-0.19).Higher attack rates were recorded among RCHE residents(median 0.84,IQR:0.64-0.93)and higher case fatality rate(CFR:28.1%)than in RCHDs(median 0.58,IQR:0.31-0.84;CFR:3.9%).The attack rate decreased when vaccination coverage increased for both RCHEs(ρ=−0.131,p<0.001)and RCHDs(ρ=−0.333,p<0.001).Comparing with infected residents who were unvaccinated/vaccinated with one-dose,receiving at least two-dose was estimated to be effective in reducing severe outcomes in 31%and 36%of infected RCHE and RCHD residents respectively;with greater reduction in mortality among RCHD than RCHE residents(54%and 38%,respectively).Vaccine effectiveness of two-dose of BNT162b2 against severe outcomes and death are higher than that of CoronaVac.Conclusions:Increasing COVID-19 vaccination could have significant impact on reducing the risk of COVID-19 outbreaks in RCHs.At least two-dose of COVID-19 vaccine is still effective in reducing severe outcomes and death among infected residents in RCHs during Omicron epidemic. | Mei Hung Joanna Leung Kin Hang Kung Ian Siu-kiu Yau Yan Yee Fung Kirran Nazesh Mohammad Cheong Chi Andrew Lau Ka Wing Albert Au Shuk Kwan Chuang | 2023 | Infectious Medicine2023,2,1: | 0 |
