1A seven-gene-deleted African swine fever virus is safe and effective as a live attenuated vaccine in pigs显示文摘African swine fever(ASF)is a devastating infectious disease in swine that is severely threatening the global pig industry.An efficacious vaccine is urgently required.Here,we used the Chinese ASFV HLJ/18 as a backbone and generated a series of genedeleted viruses.The virulence,immunogenicity,safety,and protective efficacy evaluation in specific-pathogen-free pigs,commercial pigs,and pregnant sows indicated that one virus,namely HLJ/18-7GD,which has seven genes deleted,is fully attenuated in pigs,cannot convert to the virulent strain,and provides complete protection of pigs against lethal ASFV challenge.Our study shows that HLJ/-18-7GD is a safe and effective vaccine against ASFV,and as such is expected to play an important role in controlling the spread of ASFV.Weiye Chen Dongming Zhao Xijun He Renqiang Liu Zilong Wang Xianfeng Zhang Fang Li Dan Shan Hefeng Chen Jiwen Zhang Lulu Wang Zhiyuan Wen Xijun Wang Yuntao Guan Jinxiong Liu Zhigao Bu 2020Science China(Life Sciences)2020,63,5:72
2Syntheses, structures and magnetic properties of three Co(Ⅱ)coordination architectures based on a flexible multidentate carboxylate ligand and different N-donor ligands显示文摘Three new cobalt complexes,{[Co5(tci)2(bimb)3(3-O)2(H2O)2]·3DMF·4H2O}n(1),{[Co3(tci)2(bib)]·2DMF·2H2O}n(2)and{[Co(Htci)(bpea)0.5]·H2O}n(3)(H3tci=tris(2-carboxyethyl)isocyanurate,bimb=4,4′-bis(imidazol-1-yl)biphenyl,bib=1,4-bis(imidazol-1-yl)benzene,bpea=1,2-bis(4-pyridyl)ethane,DMF=N,N′-dimethylformamide),have been successfully synthesized through the assembly of Co(II)ions,H3tci and different N-donor ligands,respectively.All complexes were structurally characterized by single crystal X-ray diffraction,elemental analyses,IR spectra,thermogravimetric(TG)analyses and X-ray powder diffraction(XRPD).Complex 1 exhibits a 3D three-fold parallel interpenetrated 3D→3D structure with(65·8)CdSO4topology.Complex 2 is built from[Co3(2-Ocarboxyl)2(CO2)4]clusters and linear bib ligands,displaying a two-fold parallel interpenetrated(3,8)-connected(43)2(46·618·84)topology,while complex 3 is a 3D pillar-layered structure involving an infinite–Co–(2-Ocarboxyl)(CO2)–Co–chain.The diverse structures of the three complexes indicate that the skeletons of different N-donor ligands play an important role in the assembly of such different frameworks.In addition,magnetic investigation indicates that besides spin-orbit coupling of Co(II)ions,there exist antiferromagnetic exchange interactions in Co5and Co3clusters of 1 and 2,respectively.TIAN Dan LIU SuiJun ZHANG DaShuai CHANG Ze HU TongLiang BU XianHe 2013Science China Chemistry2013,56,12:5

4Development and Application of a PCR Approach for Detection of Bovis, Sheep, Pig, and Chicken Derived Materials in Feedstuff显示文摘A PCR method for detection of bovis, sheep, pig, and chicken derived materials in feedstuff was established, and the existing method was improved according to the research on general primer and species-specific primers. First, general primer designed according to 16S rRNA gene sequence of bovis, sheep, pig, chicken, fish, and horse mtDNA was used for primary detection of animal derived materials in feedstuff. Species-specific primers designed according to conserved sequence of mtDNA of bovis, sheep, pig, and chicken were used for amplification of a 271, 274, 149, and 266 bp fragment, respectively. Further confirmation of the detection result was then carried out. PCR method for detection of animal derived materials in unknown feedstuff was developed by using general primer, relevant PCR system, and PCR condition. Also a PCR method for detection of each species (bovines, sheep, pig, and chicken) was designed by using our species- specific primers. High sensitivity and specificity of our method were confirmed with a minimum detection level of 0.1%. Method for detection of animal derived materials in this research is not only cheap and easy for operation but also precise and reliable results can be obtained. 11Immunogenicity of a recombinant VSV-Vectored SARS-CoV vaccine induced robust immunity in rhesus monkeys after single-dose immunization显示文摘Severe acute respiratory syndrome(SARS)is a highly contagious zoonotic disease caused by SARS coronavirus(SARS-Co V).Since its outbreak in Guangdong Province of China in 2002,SARS has caused 8096 infections and774 deaths by December 31st,2003.Although there have been no more SARS cases reported in human populations since 2004,the recent emergence of a novel coronavirus disease(COVID-19)indicates the potential of the recurrence of SARS and other coronavirus disease among humans.Thus,developing a rapid response SARS vaccine to provide protection for human populations is still needed.Spike(S)protein of SARS-Co V can induce neutralizing antibodies,which is a pivotal immunogenic antigen for vaccine development.Here we constructed a recombinant chimeric vesicular stomatitis virus(VSV)VSVΔG-SARS,in which the glycoprotein(G)gene is replaced with the SARS-Co V S gene.VSVΔG-SARS maintains the bullet-like shape of the native VSV,with the heterogeneous S protein incorporated into its surface instead of G protein.The results of safety trials revealed that VSVΔG-SARS is safe and effective in mice at a dose of 1×10^(6)TCID_(50).More importantly,only a single-dose immunization of 2×10^(7)TCID_(50)can provide high-level neutralizing antibodies and robust T cell responses to non-human primate animal models.Thus,our data indicate that VSVΔG-SARS can be used as a rapid response vaccine candidate.Our study on the recombinant VSV-vectored SARS-Co V vaccines can accumulate experience and provide a foundation for the new coronavirus disease in the future.Dan Shan Xiaoyan Tang Renqiang Liu Dan Pan Xijun Wang Jinying Ge Zhiyuan Wen Zhigao Bu 2022Virologica Sinica2022,37,2:0
