| 1 | Genetic variants of interferon regulatory factor 5 associated with chronic hepatitis B infection显示文摘AIM To investigate possible effects of IRF5 polymorphisms in the 3' UTR region of the IFR5 locus on susceptibilityto hepatitis B virus(HBV) infection and progression of liver diseases among clinically classified Vietnamese patients.METHODS Four IFR5 SNPs(rs13242262 A/T, rs77416878 C/T, rs10488630 A/G, and rs2280714 T/C) were genotyped in clinically classified HBV patients [chronic hepatitis B(CHB). n = 99; liver cirrhosis(LC), n = 131; hepatocellular carcinoma(HCC), n = 149] and in 242 healthy controls by direct sequencing and Taq Man realtime PCR assays. RESULTS Comparing patients and controls, no significant association was observed for the four IFR5 variants. However, the alleles rs13242262 T and rs10488630 G contributed to an increased risk of liver cirrhosis(LC vs CHB: OR = 1.5, 95%CI: 1.1-2.3, adjusted P = 0.04; LC vs CHB: OR = 1.7, 95%CI: 1.1-2.6, adjusted P = 0.019). Haplotype IRF5*TCGT constructed from 4 SNPs was observed frequently in LC compared to CHB patients(OR = 2.1, 95%CI: 1.2-3.3, adjusted P = 0.008). Haplotype IRF5*TCAT occurred rather among CHB patients than in the other HBV patient groups(LC vs CHB: OR = 0.4, 95%CI: 0.2-0.8, adjusted P = 0.03; HCC vs CHB: OR = 0.3, 95%CI: 0.15-0.7, adjusted P = 0.003). The IRF5*TCAT haplotype was also associated with increased levels of ALT, AST and bilirubin. | 6 | Novel CagA ELISA exhibits enhanced sensitivity of Helicobacter pylori CagA antibody显示文摘AIM To develop a novel Helicobacter pylori(H.pylori)CagA antibody enzyme-linked immunosorbent assay(ELISA)suitable for detecting serum anti-CagA antibodies with high sensitivity.METHODS Recombinant East Asian-type CagA protein was purified and immobilized for ELISA.Serum samples from 217Vietnamese individuals(110 H.pylori-infected and107 uninfected individuals)were applied.Conventional ELISA from Western-type CagA and our East Asian-type CagA ELISA were evaluated by comparing 38 subjects with the Western-type genotype and 72 subjects with the East Asian-type cagA genotype.Histological scores of the gastric mucosa were determined using the updated Sydney System to examine the relationship with anti-CagA antibody titers.RESULTS Recombinant 70-100 k Da fragments were immobilized on the ELISA plate.In ROC analysis,the area under the curve of our East Asian-type CagA ELISA was comparable to that of conventional CagA ELISA.The sensitivity of the two ELISAs differed depending on the cagA genotype.The sensitivity of East Asian-type Cag A ELISA was higher for subjects infected with East Asiantype cagA H.pylori(P<0.001),and the sensitivity of the conventional CagA ELISA tended to be higher for subjects infected with Western cagA H.pylori(P=0.056).The titer of anti-CagA antibody tended to correlate with monocyte infiltration scores(r=0.25,P=0.058)and was inversely correlated with H.pylori density(r=-0.26,P=0.043).CONCLUSION The novel ELISA is useful to detect anti-CagA antibodies in East Asian countries,and the titer may be a marker for predicting chronic gastritis. | Yuichi Matsuo Yasutoshi Kido Junko Akada Seiji Shiota Tran Thanh Binh Tran Thi Huyen Trang Ho DQ Dung Pham Huu Tung Tran Dinh Tri Ngo P Minh Thuan Le Quang Tam Bui Chi Nam Vu Van Khien Yoshio Yamaoka | 2017 | World Journal of Gastroenterology2017,23,1: | 1 |

| 10 | Combined transarterial chemoembolization and stereotactic body radiation therapy as a bridge therapy to liver transplant for hepatocellular carcinoma显示文摘Liver transplant(LT)is the curative treatment for patients with hepatocellular carcinoma(HCC).Bridge therapies are local treatments given to patients on the LT waitlist,to prevent tumor progression and to reduce the dropout rate.Case presentation:We reported a 40-year-old man diagnosed with Barcenola-Clinic Liver Cancer BCLC intermediate stage HCC and Child-Pugh A5 hepatitis B virus cirrhosis who underwent combined bridge therapies to LT.Firstly,the patient received transarterial chemoembolization(TACE)for two times and showed a partial response.Then he underwent stereotactic body radiation therapy(SBRT)with a total dose of 45 Gy in 3 fractions.Three months later,the tumor size and serum protein induced by Vitamin K absence or antagonists-II,alpha fetoprotein levels decreased gradually.In June 2019 a suitable donor was found and his LT was successfully performed.Conclusion:We propose that a combination of TACE and SBRT was feasible as bridge therapy for HCC patients on the LT waitlist. | Bui Quang Bieu Nguyen Dinh Chau Nguyen Xuan Kien Le Van Thanh Vu Van Quang Thai Doan Ky Nguyen Tien Thinh Mai Hong Bang | 2020 | Hepatoma Research2020,6,8: | 0 |
