| 8 | Characterization of maize leaf pyruvate orthophosphate dikinase using high throughput sequencing显示文摘In C_4 photosynthesis, pyruvate orthophosphate dikinase(PPDK) catalyzes the regeneration of phosphoenolpyruvate in the carbon shuttle pathway. Although the biochemical function of PPDK in maize is well characterized,a genetic analysis of PPDK has not been reported. In this study, we use the maize transposable elements Mutator and Ds to generate multiple mutant alleles of PPDK. Loss-offunction mutants are seedling lethal, even when plants were grown under 2% CO_2, and they show very low capacity for CO_2 assimilation, indicating C_4 photosynthesis is essential in maize. Using RNA-seq and GC-MS technologies, we examined the transcriptional and metabolic responses to a deficiency in PPDK activity. These results indicate loss of sPPDK results in downregulation of gene expression of enzymes of the C_4 cycle, the Calvin cycle, and components of photochemistry. Furthermore, the loss of PPDK did not change Kranz anatomy, indicating that this metabolic defect in the C_4 cycle did not impinge on the morphological differentiation of C_4 characters. However, sugar metabolism and nitrogen utilization were altered in the mutants. | 10 | Histology, physiology, and transcriptomic and metabolomic profiling reveal the developmental dynamics of annual shoots in tree peonies (Paeonia suffruticosa Andr.)显示文摘The development of tree peony annual shoots is characterized by“withering”,which is related to whether there are bud points in the leaf axillaries of annual shoots.However,the mechanism of“withering”in tree peony is still unclear.In this study,Paeonia ostii‘Fengdan’and P.suffruticosa‘Luoyanghong’were used to investigate dynamic changes of annual shoots through anatomy,physiology,transcriptome,and metabolome.The results demonstrated that the developmental dynamics of annual shoots of the two cultivars were comparable.The withering degree of P.suffruticosa‘Luoyanghong’was higher than that of P.ostii‘Fengdan’,and their upper internodes of annual flowering shoots had a lower degree of lignin deposition,cellulose,C/N ratio,showing no obvious sclerenchyma,than the bottom ones and the whole internodes of vegetative shoot,which resulted in the“withering”of upper internodes.A total of 36 phytohormone metabolites were detected,of which 33 and 31 were detected in P.ostii‘Fengdan’and P.suffruticosa‘Luoyanghong’,respectively.In addition,302 and 240 differentially expressed genes related to lignin biosynthesis,carbon and nitrogen metabolism,plant hormone signal transduction,and zeatin biosynthesis were screened from the two cultivars.Furtherly,36 structural genes and 40 transcription factors associated with the development of annual shoots were highly co-expressed,and eight hub genes involved in this developmental process were identified.Consequently,this study explained the developmental dynamic on the varied annual shoots through multi-omics,providing a theoretical foundation for germplasm innovation and the mechanized harvesting of tree peony annual shoots. | Ningning Tong Qingyan Shu Baichen Wang Liping Peng Zheng’an Liu | 2023 | Horticulture Research2023,10,9: | 0 |
| 11 | Lag compensation errors in active power filters based on DSP controllers显示文摘The growing problems of harmonic pollution on coal mine power lines caused by high-power DC drive systems has increased the use of active power filters.We analyzed compensation errors caused by the time lag in the detecting circuits of an active power filter based on DSP control.We derived a mathematical model for the compensation error starting from the error estimation when a single distortion frequency is present.This model was then extended to the case where multiple frequencies are present in the distortion.A formula for a general theory of compensation error with fixed load and fixed lag time is presented.The theoretical analysis and experimental results show that the delay time of an active power filter mainly arises from the sampling time.Lower sampling frequencies introduce larger compensation errors in the active power filter reference current. | WANG, Xuedan JIANG, Jianguo GAO, Baichen | 2010 | Mining Science and Technology2010,20,2: | 0 |
