112味止血中药对脂多糖诱导小鼠巨噬细胞产生一氧化氮的抑制作用显示文摘目的:研究12味止血中药对脂多糖(LPS)诱导小鼠巨噬细胞产生一氧化氮(NO)的抑制作用,并探讨其止血机制。方法:以1μg.mL-1LPS刺激小鼠巨噬细胞RAW264.7,22h后以Griess法测定NO的终产物亚硝酸盐的含量,观察LPS对NO生成的影响。结果:12味止血药均可抑制LPS诱导小鼠巨噬细胞RAW264.7中NO的生成。三七、地榆、仙鹤草、槐花、艾叶、蒲黄、侧柏叶及白茅根的抑制作用显著(P<0.05)。结论:本实验为12味止血中药对脂多糖诱导小鼠巨噬细胞产生NO的抑制作用提供了一定的理论依据。廖晖 Linda K Banbury David N Leach 2007中国药房2007,18,9:27
2小柴胡汤对体外培养人肝癌细胞株HepG2增殖的影响显示文摘目的探讨小柴胡汤45%乙醇提取物对体外培养人肝癌细胞株HepG2增殖的影响及其可能的机理。方法采用ATP-Lite法测定不同浓度小柴胡汤对体外培养的HepG2增殖的作用,同时利用氧自由基清除能(ORAC)法测定样品的抗氧化活性;以LPS刺激小鼠巨噬细胞RAW264.7产生一氧化氮(NO),Griess法测定NO的终产物亚硝酸盐的含量,观察样品对NO生成的影响。结果小柴胡汤在62.5~500mg/L浓度范围内可以显著抑制HepG2增殖,抑制作用呈剂量相关性,计算半数抑制浓度(IC50)为(90.7±23.7)mg/L。小柴胡汤提取物具有一定的抗氧化活性,其抗氧化能力为阳性对照维生素C的1.3倍。小柴胡汤在3.125~50mg/L浓度范围内均可以显著抑制NO的生成(P<0.05,P<0.01)。结论小柴胡汤具有体外抑制人肝癌细胞株HepG2增殖的作用,其抑制作用可能与其抗氧化活性及抑制NO生成有关。廖晖 David J de Vries Linda K Banbury David N Leach 2010中国中医药信息杂志2010,17,11:7
316味中药炮制前后抗氧化活性的比较研究显示文摘目的:比较16味中药炮制前后的抗氧化活性。方法:采用氧自由基清除能力(ORAC)法测定样品的抗氧化活性。荧光素(FL)为荧光探针。2,2-偶氮-2-氨基氢氯化丙烷在37℃时以稳定的速率产生过氧化氢(H2O2),H2O2氧化FL从而造成其荧光衰减。抗氧化剂的存在可以清除H2O2,延迟FL的荧光衰减,动态记录样品35min内的荧光变化。以水溶性维生素E为标准品绘制标准方程,根据Trolox标准方程计算样品的ORAC值。表儿茶素为阳性对照。结果:16味中药炮制前、后均具有一定的ORAC值,酒黄芩的ORAC值最高,为4241μmol TE·g-1;炒薏苡仁的ORAC值最低,为17μmol TE·g-1。10味中药炮制品的ORAC值低于生品,其中7味有显著性差异(P<0.05);6味炮制品的ORAC值高于生品,其中3味有显著性差异(P<0.05)。结论:16味中药炮制前、后抗氧化活性的变化可能与其所含的化学成分含量改变有关。廖晖 王孝敏 Linda K Banbury David N Leach 2010中国药房2010,21,47:5
4Effect of Honghua (Flos Carthami) on nitric oxide production in RAW 264.7 cells and α-glucosidase activity显示文摘OBJECTIVE: To study the effects of extracts from Honghua(Flos Carthami) on lipopolysaccharide induced nitric oxide(NO) production in RAW 264.7cells and the influence of the extracts on yeastα-glucosidase activity. The total flavonoid content of the extracts was also determined.METHODS: Cytotoxicity of the extracts to RAW264.7 cells was evaluated by the ATPlite? method.Inhibitory effects of the extracts on NO production were evaluated by Griess assay. Curcumin was used as a positive control. Screening of extracts for potential α-glucosidase inhibitors was done by a fluorometric assay.The assay was based on the hydrolysis of 4-methylumbelliferyl-a-D-glucopyranoside to form the fluorescent product, 4-methylumbelliferone. Acarbose was used as a positive control. The total flavonoid content was tested using kaempferol as the standard.RESULTS: There were significant inhibitory effects on NO production when the extracts were 25-100μg/mL(P<0.05) and curcumin was 2-4 μg/mL(P<0.001). The extracts showed an inhibitory effect onα-glucosidase activity at the concentrations of15.6-125 μg/mL with a half maximal(50%) inhibitory concentration(IC50) of(32.8±5.7) μg/mL, compared with the IC 50 of acarbose at(1.8±0.4) μg/mL.There was a significant difference between the two IC50values(P<0.001). The total content of flavonoids per gram of dried herb was1.14 mg.CONCLUSION:Honghua(Flos Carthami)showed in hibitory effects on NO production in activated RAW264.7 macrophage cells and an inhibitory effect on yeast α-glucosidase. 6Effects and Potential Mechanisms of Danzhi Xiaoyao Pill(丹栀逍遥丸) on Proliferation of MCF-7 Human Breast Cancer Cells in vitro显示文摘Objective:To investigate the effects of 50% ethyl alcohol(EtOH)extracts from Danzhi Xiaoyao Pill(丹栀逍遥丸,DXP)on the proliferation of MCF-7 human breast cancer cells and potential mechanisms.Methods:ATP-Lite assay was performed to test the proliferation of the MCF-7 breast cancer cell line;and antioxidant activity was measured by the oxygen radical absorbance capacity (ORAC).The effects of DXP on nitric oxide(NO)production were tested by lipopolysaccharide(LPS)- stimulated RAW 264.7 murine macrophages using the Griess reaction.Results:The 50% EtOH DXP extracts displayed a cytotoxic response on MCF-7 cells at 0.10,0.25 and 0.50 mg/mL dose- dependently with the proliferation inhibited by more than 85%.The ORAC value of the DXP was 820μmoL Trolox equivalent/g,about 40% of the vitamin C value.DXP extracts had significant inhibitory effect on NO production at the concentration from 0.0625 mg/mL to 0.5 mg/mL(P<0.05,P<0.01).Conclusion:The extracts of DXP could significantly inhibit the proliferation of MCF-7 cells,with the effect possibly related to its antioxidant activity and the inhibition of NO production.廖晖 Linda K Banbury David N Leach 2008Chinese Journal of Integrative Medicine2008,14,2:1
